1Biocatalytic desymmetrization of 3-substituted glutaronitriles by nitrilases. A convenient chemoenzymatic access to optically active (S)-Pregabalin and (R)-Baclofen显示文摘Desymmetrization of prochiral 3-substituted glutaronitriles offers a new approach to access(S)-Pregabalin and(R)-Baclofen. A number of nitrilases from diverse sources were screened with 3-isobutylglutaronitriles(1a) or 3-(4′-chlorophenyl)glutaronitriles(1b) as the substrate. Some nitrilases were found to catalyze the desymmetric hydrolysis of 1a and 1b to form optically active 3-(cyanomethyl)-5-methylhexanoic acid(2a) and 3-(4′-chlorophenyl)-4-cyanobutanoic acid(2b) with high enantiomeric excesse(ee), respectively. This cannot be achieved using traditional chemical hydrolysis. Among them, AtNIT3 generated(R)-2b, whereas BjNIT6402 and HsNIT produced the opposite(S)-enantiomer with high conversions and ee values. Not only the nitrilases showed different activities and stereoselectivities toward these 3-substituted glutaronitriles, the 3-substituent of the substrates also exerted great effect on the enzyme activity and stereoselectivity.(S)-2a and(S)-2b were prepared with high yields and ee values using BjNIT6402 and HsNIT as the biocatalysts, respectively. A straightforward Curtius rearrangement of(S)-2a and(S)-2b, followed by the acidic hydrolysis, afforded(S)-Pregabalin and(R)-Baclofen. 5Efficient enzymatic synthesis of(S)-1-(30-bromo-20-methoxyphenyl)ethanol,the key building block of lusutrombopag显示文摘(S)-1-(30-Bromo-20-methoxyphenyl)ethanol((S)-1b)is the key precursor for the synthesis of Lusutrombopag.The bioreduction of 1-(30-bromo-20-methoxyphenyl)ethanone(1a)offers an attractive method to access this important compound.Through screening the available carbonyl reductases,we obtained a carbonyl reductase from Novosphingobium aromaticivorans(CBR),which could completely convert 100 g/L of 1a to(S)-1b.Furthermore,a carbonyl reductase from Novosphingobium sp.Leaf2(NoCR)was identified to completely convert 200 g/L of 1a to(S)-1b with excellent enantioselectivity(>99%ee)and 77%isolated yield using FDH/formate system for NADH regeneration.The K_(m) and k_(cat) of recombinant NoCR towards 1a were 0.66 mmol/L and 7.5 s-1,and the catalytic efficiency k_(cat)/K_(m) was 11.3 mmol/s.L.Meanwhile,NoCR showed high catalytic activity and stereoselectivity towards acetophenone derivatives with halogen or methoxy substitution on the benzene ring,indicating that NoCR is a valuable biocatalyst with potential practical applications.Yunfeng Cui Yangyang Ji Xi Chen Jianjiong Li Jinhui Feng Qing Zhao Peiyuan Yao Qiaqing Wu Dunming Zhu 2023Green Synthesis and Catalysis2023,4,3:0
