1Gastro-intestinal toxicity of chemotherapeutics in colorectal cancer:The role of inflammation显示文摘Chemotherapy-induced diarrhea(CID)is a common and often severe side effect experienced by colorectal cancer(CRC)patients during their treatment.As chemotherapy regimens evolve to include more efficacious agents,CID is increasingly becoming a major cause of dose limiting toxicity and merits further investigation.Inflammation is a key factor behind gastrointestinal(GI)toxicity of chemotherapy.Different chemotherapeutic agents activate a diverse range of pro-inflammatory pathways culminating in distinct histopathological changes in the small intestine and colonic mucosa.Here we review the current understanding of the mechanisms behind GI toxicity and the mucositis associated with systemic treatment of CRC.Insights into the inflammatory response activated during this process gained from various models of GI toxicity are discussed.The inflammatory processes contributing to the GI toxicity of chemotherapeutic agents are increasingly being recognised as having an important role in the development of anti-tumor immunity,thus conferring added benefit against tumor recurrence and improving patient survival.We review the basic mechanisms involved in the promotion of immunogenic cell death and its relevance in the treatment of colorectal cancer.Finally,the impact of CID on patient outcomes and therapeutic strategies to prevent or minimise the effect of GI toxicity and mucositis are discussed.Chun Seng Lee Elizabeth J Ryan Glen A Doherty 2014World Journal of Gastroenterology2014,20,14:10
2Genomic change in hepatitis B virus associated with development of hepatocellular carcinoma显示文摘AIM: To determine the genomic changes in hepatitis B virus(HBV) and evaluate their role in the development of hepatocellular carcinoma(HCC) in patients chronically infected with genotype C HBV.METHODS: Two hundred and forty chronic hepatitis B(CHB) patients were subjected and followed for a median of 105 mo. HCC was diagnosed in accordance with AASLD guidelines. The whole X, S, basal core promoter(BCP), and precore regions of HBV were sequenced using the direct sequencing method.RESULTS: All of the subjects were infected with genotype C HBV. Out of 240 CHB patients, 25(10%) had C1653 T and 33(14%) had T1753 V mutation in X region; 157(65%) had A1762T/G1764 A mutations in BCP region, 50(21%) had G1896 A mutation in precore region and 67(28%) had pre-S deletions. HCC occurred in 6 patients(3%). The prevalence of T1753 V mutation was significantly higher in patients who developed HCC than in those without HCC. 4东南亚地区五加科植物进化关系的初步研究(英文)显示文摘东南亚五加科包含14个属约500种,本文应用ITS片段对该区五加科植物的进化关系作了初步研究。研究显示该地区五加科植物具有复杂的起源,很多属属于亚洲掌状复叶类群或Hedereae族的一支中。该区特有类群Harmsiopanax形态上非常特殊,但其系统位置尚未不明朗。在Brassaiopsis属中,有几种形态差异较大的种,但它们属同一单系,加之各种间ITS序列差异较小,故应是新近起源于马来亚半岛和苏门达腊岛的种类。Wardenia simplex聚类在Brassaiopsis一支中,故不支持将Wardenia作为独立的属。东南亚地区对于Schefflera属的发育非常重要,已有的证据显示该区的Schefflera属植物属于该属的Heptapleurum类群。马来亚与泰国南部的Dendropanax lancifolius并没有与Dendropanax属的核心类群聚在一起,其系统地位需进一步研究。Macropanax maingayi是非常特殊的一个种,曾被独立分出,成立了单种属Hederopsis。本文的分析清楚表明它属于Macropanax属。Aralia merrillii因为其不同寻常的攀缘特性而被独立出来,建立了单种属Acanthophora,但ITS序列分析支持将它置于Aralia属中。新增的取样继续支持Arthrophyllum的单系性。Os-moxylon的原初分布范围在东南亚,它是五加科系统进化树上孤立的类群。文军 朱昱苹 Chunghee LEE Elizabeth WIDJAJA LENG Guan Saw 2008云南植物研究2008,30,4:5

7Association of overexpression of TIF1γ with colorectal carcinogenesis and advanced colorectal adenocarcinoma显示文摘AIM:To determine the expression and clinical significance of transcriptional intermediary factor 1 gamma (TIF1γ),Smad4 and transforming growth factor-beta (TGFβR) across a spectrum representing colorectal cancer (CRC) development.METHODS:Tissue microarrays were prepared from archival paraffin embedded tissue,including 51 colorectal carcinomas,25 tubular adenomas (TA) and 26 HPs,each with matched normal colonic epithelium.Immunohistochemistry was performed using antibodies against TIF1γ,Smad4 and TGFβ RⅡ.The levels of expression were scored semi-quantitatively (score 0-3 or loss and retention for Smad4).RESULTS:Overexpression of TIF1γ was detected in 5/26 (19%) HP;however,it was seen in a significantly higher proportion of neoplasms,15/25 (60%) TAs and 24/51 (47%) CRCs (P<0.05).Normal colonic mucosa,HP,and TAs showed strong Smad4 expression,while its expression was absent in 22/51 (43%) CRCs.Over-expression of TGFβ RⅡ was more commonly seen in neoplasms,13/25 (52%) TAs and 29/51 (57%) CRCs compared to 9/26 (35%) HP (P<0.05).Furthermore,there was a correlation between TIF1γ overexpression and Smad4 loss in CRC (Kendall tau rank correlation value=0.35,P<0.05).The levels of TIF1γ overexpression were significantly higher in stage Ⅲ than in stage Ⅰ and Ⅱ CRC (P<0.05).CONCLUSION:The findings suggest that over-expression of TIF1γ occurs in early stages of colorectal carcinogenesis,is inversely related with Smad4 loss,and may be a prognostic indicator for poor outcome.Shilpa Jain Shashideep Singhal Franto Francis Cristina Hajdu Jin-Hua Wang Arief Suriawinata Yin-Quan wang Miao Zhang Elizabeth H Weinshel Fritz Francois Zhi-Heng Pei Peng Lee Ru-Liang Xu 2011World Journal of Gastroenterology2011,17,35:3
