| 1 | Positioning accuracy assessment for the 4GEO/5IGSO/2MEO constellation of COMPASS显示文摘Determined to become a new member of the well-established GNSS family,COMPASS(or BeiDou-2) is developing its capabilities to provide high accuracy positioning services.Two positioning modes are investigated in this study to assess the positioning accuracy of COMPASS' 4GEO/5IGSO/2MEO constellation.Precise Point Positioning(PPP) for geodetic users and real-time positioning for common navigation users are utilized.To evaluate PPP accuracy,coordinate time series repeatability and discrepancies with GPS' precise positioning are computed.Experiments show that COMPASS PPP repeatability for the east,north and up components of a receiver within China's Mainland is better than 2 cm,2 cm and 5 cm,respectively.Apparent systematic offsets of several centimeters exist between COMPASS precise positioning and GPS precise positioning,indicating errors remaining in the treatments of COMPASS measurement and dynamic models and reference frame differences existing between two systems.For common positioning users,COMPASS provides both open and authorized services with rapid differential corrections and integrity information available to authorized users.Our assessment shows that in open service positioning accuracy of dual-frequency and single-frequency users is about 5 m and 6 m(RMS),respectively,which may be improved to about 3 m and 4 m(RMS) with the addition of differential corrections.Less accurate Signal In Space User Ranging Error(SIS URE) and Geometric Dilution of Precision(GDOP) contribute to the relatively inferior accuracy of COMPASS as compared to GPS.Since the deployment of the remaining 1 GEO and 2 MEO is not able to significantly improve GDOP,the performance gap could only be overcome either by the use of differential corrections or improvement of the SIS URE,or both. | ZHOU ShanShi CAO YueLing ZHOU JianHua HU XiaoGong TANG ChengPan LIU Li GUO Rui HE Feng CHEN JunPing WU Bin | 2012 | Science China(Physics,Mechanics & Astronomy)2012,55,12: | 31 |

| 3 | Development of small molecule extracellular signal-regulated kinases(ERKs) inhibitors for cancer therapy显示文摘The mitogen-activated protein kinase(MAPK)/extracellular signal-regulated kinase 1/2(ERK1/2) signaling pathway is widely activated by a variety of extracellular stimuli, and its dysregulation is associated with the proliferation, invasion, and migration of cancer cells. ERK1/2 is located at the distal end of this pathway and rarely undergoes mutations, making it an attractive target for anticancer drug development. Currently, an increasing number of ERK1/2 inhibitors have been designed and synthesized for antitumor therapy, among which representative compounds have entered clinical trials. When ERK1/2 signal transduction is eliminated, ERK5 may provide a bypass route to rescue proliferation, and weaken the potency of ERK1/2 inhibitors. Therefore, drug research targeting ERK5 or based on the compensatory mechanism of ERK5 for ERK1/2 opens up a new way for oncotherapy. | 5 | DC-CIK联合化疗治疗非小细胞肺癌的临床疗效评价(英文)显示文摘Objective: The aim of the study was to evaluate the safety and therapeutic effects of autologous dendritic cells co-cultured with cytokine-induced killer cells (DC-CIK) combined with chemotherapy in advanced non-small cell lung cancer (NSCLC) patients. Methods: Fifty patients with advanced NSCLC (stages III to IV), who had received therapies in our Center (Department of Biotherapy, Affiliated to Cancer Hospital of Shanxi Medical University, Taiyuan, China) from August 2008 to January 2010, were treated by DC-CIK + chemotherapy as the combined treatment group; fifty advanced NSCLC patients treated with chemotherapy at the same time served as controls. The immunologic function, short-term therapeutic effects, the 1-year survival rate, the life quality, the chemotherapy side effects were compared between the two groups, the safety and therapeutic effects of DC-CIK cells therapy were observed too. Results: There was no obvious change of subsets of T cells in peripheral blood before and after therapy in DC-CIK + chemotherapy group, and IFN-γ was improved after therapy in this group (P < 0.05); in chemotherapy alone group, the ratios of CD3+CD4+, CD3+CD8+, CD3-CD56+ cells and the secretion of IL-2, TNF-α decreased significantly after therapy (P < 0.05); the ratios of CD3+CD8+, CD3+CD56+ were improved after cell culture (P < 0.05). The disease control rate (DCR) of DC-CIK + chemotherapy group was higher than that in the chemotherapy alone group (78.0% vs 56.0%, P < 0.05); the 1-year survival rates of DC-CIK + chemotherapy group and chemotherapy alone group were 50% and 44% respectively, had no significant difference. Compared with chemotherapy alone group, the occurrence of chemotherapy side effects (including bone marrow suppression, nausea and vomiting, peripheral nerve toxicity) was less in the DC-CIK + chemotherapy group (P < 0.05). The physical and appetite were better in DC-CIK + chemotherapy group after therapy. | 6 | Determining antimicrobial resistance profiles and identifying novel mutations of Neisseria gonorrhoeae genomes obtained by multiplexed MinION sequencing显示文摘Gonorrhea is one of the most common sexually transmitted diseases worldwide. To cure infection and prevent transmission,timely and appropriate antimicrobial therapy is necessary. Unfortunately, Neisseria gonorrhoeae, the etiological agent of gonorrhea, has acquired nearly all known mechanisms of antimicrobial resistance(AMR), thereby compromising the efficacy of antimicrobial therapy. Treatment failure resulting from AMR has become a global public health concern. Whole-genome sequencing is an effective method to determine the AMR characteristics of N. gonorrhoeae. Compared with next-generation sequencing, the MinION sequencer(Oxford Nanopore Technologies(ONT)) has the advantages of long read length and portability. Based on a pilot study using MinION to sequence the genome of N. gonorrhoeae, we optimized the workflow of sequencing and data analysis in the current study. Here we sequenced nine isolates within one flow cell using a multiplexed sequencing strategy. After hybrid assembly with Illumina reads, nine integral circular chromosomes were obtained. By using the online tool Pathogenwatch and a BLAST-based workflow, we acquired complete AMR profiles related to seven classes of antibiotics. We also evaluated the performance of ONT-only assemblies. Most AMR determinants identified by ONT-only assemblies were the same as those identified by hybrid assemblies. Moreover, one of the nine assemblies indicated a potentially novel antimicrobial-related mutation located in mtrR which results in a frame-shift, premature stop codon, and truncated peptide.In addition, this is the first study using the MinION sequencer to obtain complete genome sequences of N. gonorrhoeae strains which are epidemic in China. This study shows that complete genome sequences and antimicrobial characteristics of N.gonorrhoeae can be obtained using the MinION sequencer in a simple and cost-effective manner, with hardly any knowledge of bioinformatics required. | 8 | The Browsing Pattern and Review Model of Online Consumers Based on Large Data Analysis显示文摘In the context of online shopping, commodity information and consumer reviews are main factors that will affect purchasing behavior. Started from the preference of commodity information browsing and the inherent property of online reviews, this paper focuses on the browsing data for statistical analysis and the interval distribution of consumer reviews based on the real data of360 buy which is the domestic large-scale B2 C commerce website in China. Researches find that commodity information browsing time distribution on Internet is fragment and can be depicted by the fat tail effect. It also demonstrated that user's browsing patterns are related to the type of information and the displaying of the information,which means that the pieces of the picture and the length of the titles affecting the clicks rate. Reviews on the interval distribution can be depicted by the power-law function and there is a monotonically increasing relationship between power-exponent and the customers' concerns with the corresponding commodity, the higher the exponent,the higher the degree of consumer attention. | 13 | Transcriptome profile of Dunaliella salina in Yuncheng Salt Lake reveals salt-stress-related genes under diff erent salinity stresses显示文摘Salt stress is an abiotic stress to plants in especially saline lakes.Dunaliella,a halophilic microalga distributed throughout salt lakes and seas,can respond to different salinity stresses by regulating the expression of some genes.However,these genes and their function and biological processes involved remain unclear.Profi ling these salt-stress-related genes in a high-salt-tolerant Dunaliella species will help clarify the salt tolerance machinery of Dunaliella.Three D.salina_YC salt-stress groups were tested under low(0.51 mol/L),moderate(1.03 mol/L),and high(3.42 mol/L)NaCl concentrations and one control group under very low(0.05 mol/L)NaCl concentration and 3 transcriptome results that were deep sequenced and de novo assembled were obtained per group.Twelve high-quality RNA-seq libraries with 46585 upregulated and 47805 downregulated unigenes were found.Relative to the control,188 common differentially expressed genes(DEGs)were screened and divided into four clusters in expression pattern.Fifteen of them annotated in the significant enriched Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)were validated via qPCR.Their qPCR-based relative expression patterns were similar to their RNA-seq-based patterns.Two significant DEGs,the geranylgeranyl diphosphate synthase coding gene(1876-bp cDNA)and diacylglycerol O-acyltransferase coding gene(2968-bp cDNA),were cloned and analyzed in silico.The total lipid content,superoxide dismutase specific activity,and betacarotene content of D.salina_YC increased gradually with increasing salinity.In addition,the expression of 11 validated genes involved in fatty acid biosynthesis/degradation,active oxygen or carotenoid metabolisms showed significant changes.In addition,algal photochemical efficiency was diminished with increasing salinity,as well as the expression of 4 photosynthesis-related genes.These results could help clarify the molecular mechanisms underlying D.salina responses to the Yuncheng Salt Lake environment and lay a foundation for further utilization of this algal resource. | Fan GAO Fangru NAN Jia FENG Junping LÜ Qi LIU Xudong LIU Shulian XIE | 2021 | Journal of Oceanology and Limnology2021,39,6: | 1 |
