| 1 | N^6 -methyl-adenosine (m^6 A) in RNA: An Old Modification with A Novel Epigenetic Function显示文摘N6 -methyl-adenosine (m6A) is one of the most common and abundant modifications on RNA molecules present in eukaryotes. However, the biological significance of m6A methylation remains largely unknown. Several independent lines of evidence suggest that the dynamic regulation of m6A may have a profound impact on gene expression regulation. The m6A modification is catalyzed by an unidentified methyltransferase complex containing at least one subunit methyltransferase like 3 (METTL3). m6A modification on messenger RNAs (mRNAs) mainly occurs in the exonic regions and 3'-untranslated region (3'-UTR) as revealed by high-throughput m6A-seq. One significant advance in m6A research is the recent discovery of the first two m6A RNA demethylases fat mass and obesity-associated (FTO) gene and ALKBH5, which catalyze m6A demethylation in an a-ketoglutarate (a-KG)-and Fe2+-dependent manner. Recent studies in model organisms demonstrate that METTL3, FTO and ALKBH5 play important roles in many biological processes, ranging from development and metabolism to fertility. Moreover, perturbation of activities of these enzymes leads to the disturbed expression of thousands of genes at the cellular level, implicating a regulatory role of m6A in RNA metabolism. | 4 | Compound Zhebei granules combined with chemotherapy for the treatment of refractory acute leukemia: a randomized clinical trial显示文摘OBJECTIVE: To observe the effect of compound Zhebei granules(CZBG) with chemotherapy in the treatment of refractory acute leukemia.METHODS: In this multicenter, double-blind, placebo-controlled clinical trial, we used a central(online) randomization system to assign 235 patients to two treatment groups. A total of 118 patients received chemotherapy combined with CZBG(4 g,twice daily) and 117 patients received chemotherapy plus placebo. The clinical efficacy was evaluated at the end of one chemotherapeutic cycle.RESULTS: In the full analysis set, in which deaths due to disease progression were regarded as inefficacy, the rates of complete remission(CR) and partial remission(CR + PR) were 32.35% and 50.00%,respectively, for the chemotherapy combined with CZBG group, and 23.08% and 35.58%, respectively,for the chemotherapy plus placebo group. There was a statistically significant difference between the two groups according to a ysis set(PPχ2test(P < 0.05). | 5 | Resequencing of 145 Landmark Cultivars Reveals Asymmetric Sub-genome Selection and Strong Founder Genotype Effects on Wheat Breeding in China显示文摘Controlled pedigrees and the multi-decade timescale of national crop plant breeding programs offer a unique experimental context for examining how selection affects plant genomes.More than 3000 wheat cultivars have been registered,released,and documented since 1949 in China.In this study,a set of 145 elite cultivars selected from historical points of wheat breeding in China were re-sequenced.A total of 43.75 Tb of sequence data were generated with an average read depth of 17.94x for each cultivar,and more than 60.92 million SNPs and 2.54 million InDels were captured,based on the Chinese Spring RefSeq genome v1.0.Seventy years of breeder-driven selection led to dramatic changes in grain yield and related phenotypes,with distinct genomic regions and phenotypes tar-geted by different breeders across the decades.There are very clear instances illustrating how introduced Italian and otherforeign germplasm was integrated into Chinese wheat programs and reshaped the genomic landscape of local modern cultivars.Importantly,the resequencing data also highlighted significant asymmetric breeding selec-tion among the three sub-genomes:this was evident in both the collinear blocks for homeologous chromosomes and among sets of three homeologous genes.Accumulation of more newly assembled genes in newer cultivars implied the potential value of these genes in breeding.Conserved and extended sharing of linkage disequilibrium(LD)blocks was highlighted among pedigree-related cultivars,in which fewer haplotype differences were detected.Fixation or replacement of haplotypes from founder genotypes after generations of breeding was related to their breeding value.Based on the haplotype frequency changes in LD blocks of pedigree-related cultivars,we propose a strategy for evaluating the breeding value of any given line on the basis of the accumulation(pyramiding)of bene-ficial haplotypes.Collectively,our study demonstrates the influence of 'founder genotypes' on the output of breeding efforts over many decades and also suggests that founder genotype perspectives are in fact more dy-namic when applied in the context of modern genomics-informed breeding. | Chenyang Hao Chengzhi Jiao Jian Hou Tian Li Hongxia Liu Yuquan Wang Jun Zheng Hong Liu Zhihong Bi Fengfeng Xu Jing Zhao Lin Ma Yamei Wang Uzma Majeed Xu Liu Rudi Appels Marco Maccaferri Roberto Tuberosa Hongfeng Lu Xueyong Zhang | 2020 | Molecular Plant2020,13,12: | 9 |
| 6 | Mode of débitage and technical cognition of hominids at the Guanyindong site显示文摘Exploring and interpreting the mode of cognition and behavioral patterns of prehistoric hunter-gatherers is always one of the main goals of current Paleolithic research. In the 1940s, French prehistorians proposed the technological method for studying lithic assemblages which opened a brand-new perspective on the prehistoric domain. In the 1960s, French ethnographer and anthropologist André Leroi-Gourhan proposed the concept 'chane opératoire' and established the theory of lithic technological study. After the 1980s, the concept 'chane opératoire' was substituted for two different but complementary concepts: techno-cognition and techno-economy, which contributed not only to revealing the technical knowledge and know-how applied during lithic production but also to interpreting the technical behavior of prehistoric hunter-gatherers from social and economic viewpoints. The lithic technological study of the Guanyindong site, presented in this paper, was carried out for the first time by applying lithic technological method developed by French prehistorians to the Chinese materials. Our analytical results demonstrated that the débitage system of Guanyindong is quite distinct from the concept Levallois largely used in Europe, Near-East and Africa. | 8 | Clinical characteristics and changes in living quality of patients with radiation encephalopathy induced by radiation therapy for treating nasopharyngeal carcinoma显示文摘BACKGROUND: Radiation encephalopathy (RE) caused by radiation therapy of nasopharyngeal carcinoma severely influences patients' quality of life (QOL). The factors, which influence such patients' QOL, have not been confirmed. OBJECTIVE: To observe the clinical and imageological characteristics of patients with radiation therapy of nasopharyngeal carcinoma-induced RE and the changes in QOL, and analyze QOL influencing factors. DESIGN: Retrospective case analysis. SETTING: Department of Neurology, the Second Affiliated Hospital of Sun Yat-sen University. PARTICIPANTS: Eighty-nine inpatients or outpatients with RE induced by radiation therapy of nasopharyngeal carcinoma admitted to Sun Yat-sen University Cancer Center and Department of Neurology, the Second Affiliated Hospital of Sun Yat-sen University from March 1994 to August 2004 were involved in this experiment. They all met the diagnosis criteria of RE from MERRITT'S neurology (10th edition). Thirty-three involved patients were randomly chosen as RE group. Another 34 concurrent inpatients or outpatients with nasopharyngeal carcinoma who received radiation therapy but without RE were chosen as control group. Informed consents of detected items were obtained from all the involved subjects. METHODS: ① Patients were evaluated when they were followed up. The World Health Organization Quality of Life Questionnaire abbreviated version (WHOQOL-BREF) was used for on-the-spot evaluation. High points of WHOQOL-BREF indicated better QOL. The Late Effects on Normal Tissues - Subjective, Objective, Management and Analytic (LENT-SOMA) scale for evaluating radiation injury was used to evaluate headache and neurologic disorder of patients with RE induced by radiation therapy of nasopharyngeal carcinoma. The evaluation was graded into 5 degrees. High degrees indicted severer clinical symptoms. ② Disease latency (i.e. time interval from symptoms and body signs appearing or radiation therapy ending to onset), initial symptoms, common symptoms, imageological characteristics, QOL and other related factors of patients were recorded. ③ Statistical management was carried out with SPSS 10.0 software. MAIN OUTCOME MEASURES: QOL and clinical characteristics of patients with RE induced by radiation therapy of nasopharyngeal carcinoma as well as QOL influencing factors. RESULTS: All the involved subjects participated in the final analysis. ① Disease latency of patients with RE ranged from 0 to 24 years, and 95% confidence interval was 0 to 15 years. There were significant differences in amount of invaded cases between 6 months after one radiation therapy and 6 months after two or more radiation therapies (χ2=36.76,P < 0.01). ② The common initial symptom of patients with RE was glossopharyngeal paralysis (33 cases, 37%). ③ The first 3 common symptoms of patients with RE were glossopharyngeal paralysis (52 cases, 58%), limb inertia and sensory disorder (25 cases, 28%) and headache (25 cases, 28%).④Imageology of patients with RE was characterized by long T1 and T2 signal shadow on magnetic resonance. ⑤Total scores of health, of daily life and of social relationship, measured with WHOQOL-BREF, of patients in RE group were significantly lower than those in the control group, respectively [(2.06±0.86)points vs. (2.59±0.66)points; (2.45±0.75)points vs. (2.91±0.75)points; (51.67±15.24) points vs. (59.22±13.03)points, P < 0.05]. Patients undergoing two or more radiation therapies were inferior to those undergoing one radiation therapy in total scores of health, and of daily life, scores of physiology, and of psychology (P < 0.05). ⑥Glossopharyngeal paralysis was negatively correlated with scores of psychology, total scores of daily life and of health in WHOQOL-BREF (P < 0.05); Head was negatively correlated with scores of psychology in WHOQOL-BREF (P < 0.05); Neurologic deficit wasnegatively correlated with scores of psychology, and total scores of health in WHOQOL-BREF (P < 0.05). CONCLUSION: ①The latency of RE of patients undergoing two or more radiation therapies is remarkably shortened, and QOL is decreased, but there is no difference in QOL between two genders. ② Brain edema is common in imageology of RE after radiation therapy. | 11 | Verification of Mutagen Function of Zeocin in Nannochloropsis oceanica Through Transcriptome Analysis显示文摘Zeocin can cause double strand breaks of DNA and thus is frequently used as a selective antibiotic of eukaryotic Sh ble transformants. In non-transformation system, Zeocin may function as a mutagen if not totally lethal. To verify such function of Zeocin, we mutated Nannochloropsis oceanica by increasing the concentration of Zeocin in medium gradually, and isolated a N. oceanica strain(single cell culture) which survived Zeocin up to 10.0μg mL^(-1). The Zeocin-tolerant strain entered the exponential growth phase later and grew slower than the wild strain. Transcriptome profiling showed that the Zeocin-tolerant N. oceanica strain survived Zeocin mainly by adapting(heritable), rather than acclimating(plastic) to Zeocin. Hence mutating N. oceanica with Zeocin was approved effective. Meanwhile, the physiological characteristics of this Zeocin-tolerant strain were demonstrated. As we proposed, N. oceanica tolerated Zeocin by strengthening its protein degradation and antioxidation. The genes controlling cell division and cellular response to stimuli may also have played important roles in the reduction of growth and the tolerance to Zeocin. Our findings evidenced that Zeocin can serve as an appropriate mutagen of microalgae. | 12 | New materials of the steppe mammoth, Mammuthus trogontherii, with discussion on the origin and evolutionary patterns of mammoths显示文摘Recently found materials indicate that the steppe mammoth, Mammuthus trogontherii, survived in northern China into the late Pleistocene. East Asia is the key area of mammoth evolution after the initial radiation of early forms out of Africa and into Eurasia at the beginning of the late Pliocene (c. 3.5–3.0 Ma). M. rumanus, M. meridionalis, M. trogontherii, and M. primige- nius probably formed a continuous and transitional evolutionary lineage within the pan-Eurasian mammoth radiation in East Asia. Each speciation event of the Eurasian mammoths was followed by a rapid and large-scale dispersal event: out of East Asia. Allopatric speciation is the main speciation pattern of Mammuthus. However, the functions and mechanisms of miR-214-3p on GCs are unclear. This study focuses on miR-214-3p in terms of the effects on GCs proliferation and estradiol synthesis.Results: Our findings revealed that miR-214-3p promotes proliferation and inhibits estradiol synthesis in porcine GCs. MiR-214-3p can increase the percentage of S-phase cells, the number of EdU labeled positive cells, and cell viability. However, E2 concentration was reduced after miR-214-3p agomir treatment. We also found that miR-214-3p up-regulates the expression of cell cycle genes including cell cycle protein B(Cyclin B), cell cycle protein D(Cyclin D), cell cycle protein E(Cyclin E), and cyclin-dependent kinase 4(CDK4) at the transcription and translation levels, but down-regulates the mRNA and protein levels of cytochrome P450 family 11 subfamily A member 1(CYP11A1), cytochrome P450 family 19 subfamily A member 1(CYP19A1), and steroidogenic acute regulatory protein(StAR)(i.e., the key enzymes in estradiol synthesis). The results showed that patients who have suffered from large-artery atherosclerosis, relative to small-artery occlusion patients, were characterized by elevated serum uric acid but reduced high-density lipoprotein cholesterol and triglyceride levels. Logistic regression showed that elevated uric acid and lower triglyceride levels were the main risk factors for patients with large-artery atherosclerosis. | 19 | Determining antimicrobial resistance profiles and identifying novel mutations of Neisseria gonorrhoeae genomes obtained by multiplexed MinION sequencing显示文摘Gonorrhea is one of the most common sexually transmitted diseases worldwide. To cure infection and prevent transmission,timely and appropriate antimicrobial therapy is necessary. Unfortunately, Neisseria gonorrhoeae, the etiological agent of gonorrhea, has acquired nearly all known mechanisms of antimicrobial resistance(AMR), thereby compromising the efficacy of antimicrobial therapy. Treatment failure resulting from AMR has become a global public health concern. Whole-genome sequencing is an effective method to determine the AMR characteristics of N. gonorrhoeae. Compared with next-generation sequencing, the MinION sequencer(Oxford Nanopore Technologies(ONT)) has the advantages of long read length and portability. Based on a pilot study using MinION to sequence the genome of N. gonorrhoeae, we optimized the workflow of sequencing and data analysis in the current study. Here we sequenced nine isolates within one flow cell using a multiplexed sequencing strategy. After hybrid assembly with Illumina reads, nine integral circular chromosomes were obtained. By using the online tool Pathogenwatch and a BLAST-based workflow, we acquired complete AMR profiles related to seven classes of antibiotics. We also evaluated the performance of ONT-only assemblies. Most AMR determinants identified by ONT-only assemblies were the same as those identified by hybrid assemblies. Moreover, one of the nine assemblies indicated a potentially novel antimicrobial-related mutation located in mtrR which results in a frame-shift, premature stop codon, and truncated peptide.In addition, this is the first study using the MinION sequencer to obtain complete genome sequences of N. gonorrhoeae strains which are epidemic in China. This study shows that complete genome sequences and antimicrobial characteristics of N.gonorrhoeae can be obtained using the MinION sequencer in a simple and cost-effective manner, with hardly any knowledge of bioinformatics required. | 20 | Spectrofluorimetric analysis of captopril based on its obstruction effect of the nanomaterial surface energy transfer between acridine orange and gold nanoparticles显示文摘A simple and sensitive method for detection of captopril was established based on its obstructive effect on nanomaterial surface energy transfer(NSET). It was found that the acridine orange(AO) could be adsorbed onto the surface of citrated-gold nanoparticles(Au NPs) through electrostatic interaction. Incidentally, the fluorescence of AO was quenched owing to the dipole-dipole interaction of NSET between AO fluorophore and the Au NPs. However, captopril could obstruct the occurrence of NSET between AO and Au NPs effectively with the formation of Au-S covalent bonds between it and the Au NPs. Consequently, AO molecules were moved away from the surface of Au NPs leading to a decline of the energy transfer efficiency. Moreover, the fluorescence of AO could be gradually restored with the addition of captopril. Under the optimal conditions, the recovered fluorescence intensity correlated linearly with the concentration of captopril in the range of 400 nmol/L–2.0 μmol/L with a detection limit of 71 nmol/L. Besides, the proposed method was successfully applied for the detection of captopril in troches with the recovery of 93%–102% and the RSD lower than 2.24%.
