2Network pharmacology-based analysis of Chinese herbal Naodesheng formula for application to Alzheimer's disease显示文摘Naodesheng(NDS) formula, which consists of Rhizoma Chuanxiong, Lobed Kudzuvine, Carthamus tinctorius, Radix Notoginseng, and Crataegus pinnatifida, is widely applied for the treatment of cardio/cerebrovascular ischemic diseases, ischemic stroke, and sequelae of cerebral hemorrhage, etc. At present, the studies on NDS formula for Alzheimer's disease(AD) only focus on single component of this prescription, and there is no report about the synergistic mechanism of the constituents in NDS formula for the potential treatment of dementia. Therefore, the present study aimed to predict the potential targets and uncover the mechanisms of NDS formula for the treatment of AD. Firstly, we collected the constituents in NDS formula and key targets toward AD. Then, drug-likeness, oral bioavailability, and blood-brain barrier permeability were evaluated to find drug-like and lead-like constituents for treatment of central nervous system diseases. By combining the advantages of machine learning, molecular docking, and pharmacophore mapping, we attempted to predict the targets of constituents and find potential multi-target compounds from NDS formula. Finally, we built constituent-target network, constituent-target-target network and target-biological pathway network to study the network pharmacology of the constituents in NDS formula. 4Preparation and activity of conjugate of monoclonal antibody HAb18 against hepatoma F( ab′ )_2 fragment and staphylococcal enterotoxin A显示文摘AIM To prepare the conjugate of staphylococcal enterotoxin A (SEA) protein which is a bacterial SAg and the F(ab')2 fragment of mAb HAbl8 against human hepatocellular carcinoma (HCC), and identify its activity in order to use SAg in the targeting therapy of HCC.METHODS MAb HAbl8 was extracted from the abdominal dropsy of Balb/ c mice, and was purified through chromatography column SP-40HR with Fast protein liquid chromatography (FPLC) system. The F(ab')2 fragment of mAb HAb18 was prepared by papainic digestion method. The conjugate of mAb HAb18 F(ab')2fragment and SEA was prepared with chemical conjugating reagent N-succinimidyl-3-( 2-pyridyldithio) propionate (SPDP) and purified through chromatography column Superose 12with FPLC system. The molecular mass and purity of each collected peak were identified with SDS-PAGE assay. The protein content was assayed by Lowry's method. The antibody activity of HAb18 F (ab')2 against HCC in the conjugate was identified by indirect immunocytochemical ABC method, and the activity of SEA in the conjugate to activate peripheral blood mononuclear cells (PBMC) was identified with MTT assay.RESULTS The lgG mAb HAb18 was extracted,and purified successfully. Immunocytochemical staining demonstrated that it reacted with most of HHCC cells of human HCC cell line. There were two peaks in the process of purification of the prepared HAb18 F(ab)2-SEA conjugate. SDS-PAGE assay demonstrated that the molecular mass of the first peak was about 130 ku, and the second peak was the mixture of about 45 ku and a little 100 ku proteins. The immunocytochemical staining was similar in HAb18 F (ab ')2-SEAconjugate and HAb18 F (ab ')2, i.e., thecytoplasm and/or cell membranes of most HHCC cells were positively stained. The MTT assay showed that the optical absorbance (A) value at 490 nm of HAb18 F (ab')2-SEA conjugate was 0.182 ± 0.012, that of negative control was 0.033± 0.009, and there was significant difference between them ( P < 0.05).CONCLUSION SPDP is a good proteinconjugating reagent and can be used in preparing protein conjugate. 8Earthquake prediction from China's mobile gravity data显示文摘The relation between plate tectonics and earthquake evolution is analyzed systematically on the basis of 1998e2010 absolute and relative gravity data from the Crustal Movement Observation Network of China. Most earthquakes originated in the plate boundary or within the fault zone. Tectonic deformation was most intense and exhibited discontinuity within the tectonically active fault zone because of the differential movement; the stress accumulation produced an abrupt gravity change, which was further enhanced by the earthquake. The gravity data from China's Mainland since 2000 obviously reflected five major earthquakes(Ms > 7), all of which were better reflected than before 2000. Regional gravity anomalies and a gravity gradient change were observed in the area around the epicenter about 2 or 3 years before the earthquake occurred, suggesting that gravity change may be a seismic precursor. 9QTL Mapping of Isoflavone,Oil and Protein Contents in Soybean (Glycine max L.Merr.)显示文摘Soybean(Glycine max L.Merr.) is the world's foremost source of edible plant oil and proteins,meantime,the biologically active secondary metabolites such as saponins and isoflavones are benefit to human health.The objective of this study was to identify quantitative trait loci(QTL) and epistatic interactions associated with isoflavone,protein,and oil contents in soybean seeds.An F13 recombinant inbred line(RIL) comprising 474 lines was derived from a cross between Jindou 23 and Huibuzhi cultivars.SSR technique was employed for mapping of the QTLs.The QTLs for isoflavone,protein,and oil contents were analyzed and 23 QTLs were detected based on the constructed linkage map.Six QTLs for isoflavone content were localized in linkage groups J,N,D2,and G,eleven QTLs for oil content were localized in the linkage groups A1,A2,B2,C2,and D2,and six QTLs for protein content were localized in linkage groups B2,C2,G,and H1.The correlative analysis demonstrated that the isoflavone content had significant correlation with protein content,while significantly negative correlations was existed between oil and protein content,and significantly positive correlations was existed between protein and oil content.All these findings have laid an important basis for the marker assisted breeding in soybean.The phenotypic correlations of quantitative traits may be resulted from the correlation of the QTL controlling those traits.LIANG Hui-zhen YU Yong-liang WANG Shu-feng LIAN Yun WANG Ting-feng WEI Yan-li GONG Peng-tao LIU Xue-yi FANG Xuan-jun ZHANG Meng-chen 2010Agricultural Sciences in China2010,9,8:13
10Effect of Electro-acupuncture on Expression of IRS-1/PI3K/GLUT4 Pathway in Ovarian Granulosa Cells of Infertile Patients with Polycystic Ovary Syndrome-Insulin Resistance of Phlegm-Dampness Syndrome显示文摘Objective:To evaluate the effect of electro-acupuncture(EA)in infertile patients with phlegmdampness polycystic ovary syndrome-insulin resistance(PCOS-IR).Methods:Seventy-six PCOS-IR patients who underwnet in vitro fertilization and embryo transfer(IVF-ET)were equally assigned to two groups according to a random digital table:the EA group and the control group,with 38 cases in each group.Before undergoing IVF,the two groups were treated with EA or pseudo-acupuncture,respectively,for 3 menstrual cycles.The intervention was 25 min twice a week until the day of oocyte collection.The selected acupoints were Zhongwan(RN 12),Tianshu(ST 25),Daheng(SP 15),Daimai(GB 26),Qihai(CV 6),Guanyuan(CV 4),and bilateral points including Xuehai(SP 10),Fenglong(ST 40),Zusanli(ST 36),and Yinlingquan(SP 9).Evaluation of phlegm-dampness syndrome score and IR score were carried out before and after treatment.Additionally,the number of oocytes retrieved,transplantable embryo rate,high-quality embryo rate,clinical pregnancy rate and live birth rate were compared between the two groups.Real-time polymerase chain reaction analysis was used to monitor the m RNA expression of the insulin receptor substrate(IRS-1),phosphatidylinositiol 3-kinase(PI3 K)and glucose transport factor 4(GLUT4)in ovarian granulosa cells.Results:EA treatment reduced the phlegm-dampness syndrome score as well as the IR scores compared with the control group(P<0.05).No significant differences in the number of oocytes retrieved and clinical pregnancy rate between the two groups(P>0.05).Moreover,the transplantable embryo rate[49.0%(284/580)vs.41.9%(273/652)],high-quality embryo rate[36.6%(104/284)vs.27.8%(76/273)],and live birth rate[50%(19/38)vs.26.3%(10/38)]in the EA group were significantly higher than in the control group(P<0.05).Gene expression analyses revealed significantly elevated IRS-1,PI3 K and GLUT4 m RNA in ovarian granulosa cells of the EA group compared with the control group(P<0.05).Conclusions:EA may ameliorate the effects of phlegm-dampness syndrome and ovarian IR in PCOS-IR patients.Mechanistically,this effect might be through an upregulation of the IRS-1/PI3 K/GLUT4 signaling pathway,which may result in improved oocyte quality and embryonic development potential.(Registration No.Chi CTR1800015453)XIANG Shan XIA Ming-feng SONG Jing-yan LIU Dan-qi LIAN Fang 2021Chinese Journal of Integrative Medicine2021,27,5:11
11Effects of Electroacupuncture on Expression of PI3K/Akt/Foxo3a in Granulosa Cells from Women with Shen(Kidney) Deficiency Syndrome Undergoing in vitro Fertilization-Embryo Transfer显示文摘Objective: To observe the effects of electroacupuncture(EA) on reproductive outcomes in women with Shen(Kidndy) deficiency syndrome after in vitro fertilization-embryo transfer(IVF-ET),and explore the underlying molecular mechanism.Methods: Sixty-six infertile patients with Shen de?ciency syndrome undergoing IVF-ET were divided into EA or control groups according to a random table,33 cases in each group.Before undergoing IVF,patients in the EA and control groups received EA therapy and placebo needle puncture,respectively,for 3 menstrual cycles.Shen de?ciency syndrome scores were assessed.Other outcome measures included the number of retrieved oocytes and fertilization,high-quality embryo and clinical pregnancy rates.Follicular ?uid was collected on the day of oocyte retrieval,and granulosa cell expression of phosphatidylinositide3-kinases(PI3 K),serine-threonine kinase(Akt) and forkhead box O3(Foxo3 a) m RNA were measured by reverse transcribed and quantitative real-time polymerase chain reaction.Results: Syndrome scores for pre-versus post-treatments decreased significantly(16.53±1.75 to 8.67±1.61) in the EA group(P<0.05),but showed no signi?cant change in the control group(17.18±1.58 to 14.74±1.58).A signi?cant difference in score change was found between the EA and control groups(P<0.05).High-quality embryo and clinical pregnancy rates were both increased in the EA group compared with the control group [69.15%(195/282) vs.60.27%(176/292) and 66.67%(22/33) vs.42.42%(14/33),respectively,P<0.05].The fertilization rate was equivalent in EA and control groups.No difference was found in the number of retrieved oocytes between the two groups.Granulosa cell expression levels of PI3 K and Akt m RNA were signi?cantly increased in the EA group compared with the control group,while the expression of Foxo3 a was reduced(all P<0.05).Conclusions: For infertile patients with Shen de?ciency syndrome undergoing IVF,EA for tonifying Shen as an adjunct treatment may alleviate clinical symptoms and improve the high-quality embryo rate.The EA-induced mechanism may involve regulation of PI3 K/Akt/Foxo3 a expression in granulosa cells to improve the developmental microenvironment of oocytes and inhibit granulosa cell apoptosis,possibly contributing to the improved clinical pregnancy rate(Registration No.Chi CTR 1800016217).WU Hai-cui ZHANG Jian-wei SUN Zhen-gao XIANG Shan QIAO Yan LIAN Fang 2019Chinese Journal of Integrative Medicine2019,25,4:10

15Gravity observations along the eastern margin of the Tibetan Plateau and an application to the Lushan MS 7.0 earthquake显示文摘This paper introduces relative and absolute gravity change observations in the eastern portion of the Tibetan Plateau.We analyze and discuss a change that occurred in 2010in the gravity along the eastern margin of the plateau and the relationship between this change and the 2013 Lushan MS7.0earthquake.Our results show that:(1)before the Lushan MS7.0earthquake,gravity anomalies along the eastern margin of the Tibetan Plateau changed drastically.The Lushan earthquake occurred at the bend of the high gradient zone of gravity variation along the southern edge of the Longmenshan fault zone.(2)The 2013 Lushan earthquake occurred less than 100 km away from the epicenter of the 2008 Wenchuan earthquake.Lushan and Wenchuan are located at the center of a fourquadrant section with different gravity anomalies,which may suggest that restoration after the Wenchuan earthquake may have played a role in causing the Lushan earthquake.(3)A medium-term prediction based on changes in gravity anomalies was made before the Lushan MS7.0 earthquake,in particular,a prediction of epicenter location.Wei-feng Liang Yun-feng Zhao Yun-ma Xu Yi-qing Zhu Shu-song Guo Fang Liu Lian Liu 2013Earthquake Science2013,26,3:8

17Effect of Mg/Al atom ratio of support on catalytic performance of Co-Mo/MgO-Al_2O_3 catalyst for water gas shift reaction显示文摘Co-Mo-based catalysts supported on mixed oxide supports MgO-Al2O3 with different Mg/Al atom ratios for water gas shift reaction were studied by means of TPR, Raman, XPS and ESR. It was found that the octahedral Mo species in oxidized Co-Mo/MgO(x)-Al2O3 catalyst and the contents of Mo5+, Mo4+, S2- and S22- species in the functioning catalysts increased with increasing the Mg/Al atom ratio of the support under the studied experimental conditions. This is favorable for the formation of the active Co-Mo-S phase of the catalysts. 18Dielectric monitoring method for the drug release mechanism of drug-loading chitosan microspheres显示文摘The chitosan microspheres loaded with model drug, salicylate molecular, are prepared, and the dielectric spectroscopy of this kind of suspension is measured, meanwhile a significant relaxation is observed for the microspheres suspension. By the modeling analytic of the relaxation, the parameters, which can reflect the properties of microspheres and continuous medium, are obtained. Furthermore, the releasing process of microspheres in aqueous phase is monitored real-timely, namely, by analyzing the time-dependent variation of the parameters reflecting the relaxation characteristics and electrical parameters in each phase the releasing process is detected. The research shows that the controlled release process of chitosan microspheres loaded with salicylate in aqueous phase is divided into three phases according to different releasing mechanisms. 19Effects of Quicklime and Iron Tailings as Modifier on Composition and Properties of Steel Slag显示文摘Steel slag had lower activity and much lower hydration rate than cement.Quicklime and iron tailings were designed as modification agent to adjust the composition and properties of high temperature steel slag.The results show that quicklime as modifier can greatly increase the content of cementitious minerals in modified steel slag and also promote the decomposition of RO phases and transformation of MgO in RO phase to f-MgO.After high temperature modification with compound modifier of quicklime and iron tailings,steel slag shows the main mineral phases of C3 S,C2F and MgFe2O4.The activity index of modified steel slag at 28 days reaches 95.5% when the steel slag is modified by 15% of the compound modifier with the ratio of quicklime to iron tailings equal to 2∶1at 1 350℃.Moreover,the sample with the modified steel slag exhibits the dense structure of hydration paste and the main hydration products of C-S-H gels and Ca(OH)2 crystals.Zuo-shun ZHANG Fang LIAN Lai-jun MA Yu-sheng JIANG 2015Journal of Iron and Steel Research(International)2015,22,1:5
