| 2 | Enzymatic activity characterization of SARS coronavirus 3C-like protease by fluorescence resonance energy transfer technique显示文摘Aim: To characterize enzymatic activity of severe acute respiratory syndrome (SARS) coronavirus (CoV) 3C-like protease (3CL^pro) and its four site-directedmutants. Methods: Based on the fluorescence resonance energy transfer (FRET)principle using 5-[(2'-aminoethyl)-amino] naphthelenesulfonic acid (EDANS) and4-[[4-(dimethylamino) phenyl] azo] benzoic acid (Dabcyl) as the energy transferpair, one fluorogenic substrate was designed for the evaluation of SARS-CoV3CL^pro proteolytic activity. Results: The kinetic parameters of the fluorogenicsubstrate have been determined as Kin=404μmol·L^-1, kcat=1.08min^-1, and kcat/Km=2.7mmol^-1·L·min^-1. SARS-CoV 3CL^pro showed substantial pH and temperature-triggered activity switches, and site-directed mutagenesis analysis of SARS-CoV3CL^pro revealed that substitutions of His^41, Cys^145, and His^163 resulted in complete loss of enzymatic activity, while replacement of Met^162 with Ala caused strongly increased activity. Conclusion: This present work has provided valuable information for understanding the catalytic mechanism of SARS-CoV 3CL^pro. | 3 | Efficient XML Query and Update Processing Using A Novel Prime-Based Middle Fraction Labeling Scheme显示文摘XML data can be represented by a tree or graph and the query processing for XML data requires the structural information among nodes. Designing an efficient labeling scheme for the nodes of Order-Sensitive XML trees is one of the important methods to obtain the excellent management of XML data. Previous labeling schemes such as region and prefix often sacrifice updating performance and suffer increasing labeling space when inserting new nodes. To overcome these limitations, in this paper we propose a new labeling idea of separating structure from order. According to the proposed idea, a novel Prime-based Middle Fraction Labeling Scheme(PMFLS) is designed accordingly, in which a series of algorithms are proposed to obtain the structural relationships among nodes and to support updates. PMFLS combines the advantages of both prefix and region schemes in which the structural information and sequential information are separately expressed. PMFLS also supports Order-Sensitive updates without relabeling or recalculation, and its labeling space is stable. | 6 | Fast-response fluorescent probe with favorable water solubility for highly sensitive imaging of endogenous tyrosinase in living cells and zebrafish model显示文摘Tyrosinase(TYR) is an important polyphenolic oxidase enzyme and usually regards as a biomarker of melano ma cancer.Highly effective tracking TYR activity in vivo will help to study the mechanism of TYR in living organisms and forecasts related diseases.In this study,we present a novel TYR-activatable fluo rescent probe(CHMC-DOPA) for tracking TYR activity in vitro and in vivo.CHMC-DOPA is constructed by incorporating dopamine(DOPA) moiety into a fluorescent chloro-hydroxyl-merocyanine(CHMC)scaffold.Upon exposure to TYR,the dopamine unit in CHMC-DOPA is oxidized to a dopaquinone derivative,and an intramolecular photo-induced electron transfer(PET) process between CHMC fluorophore and o-dopaquinone will take place,the fluorescence of CHMC-DOPA is quenched rapidly.Therefore,the evaluation of TYR activity is established in terms of the relationship between fluorescence quenching efficiency and TYR activity.In our experiments,CHMC-DOPA shows various advantages,such as fast response(8 min),low concentration of TYR activation(0.5 U/mL),good water-solubility,as well as the lowest detection limit(0.003 U/mL) compared with previously reported works.Furthermore,CHMCDO PA also exhibits excellent cell membrane permeability and low cytotoxicity,which is successfully used to monitor endogenous TYR activity in living cancer cells and zebrafish models.CHMC-DOPA performs well,and we anticipate that this newly designed novel platform will provide an alternative for high effective monitoring TYR activity in biosystems. | Zheng Li Xiaofeng Xia Yu You Cuifen Lu Guichun Yang Chao Ma Junqi Nie Qi Sun Shuilin Wu Jun Ren Feiyi Wang | 2021 | Chinese Chemical Letters2021,32,5: | 1 |

| 11 | A comparative study of seismic tomography models of Southwest China显示文摘The margin of the Tibetan Plateau of Southwest China is one of the most seismically active regions of China and is the location of the China Seismic Experimental Site(CSES).Many studies have developed seismic velocity models of Southwest China,but few have compared and evaluated these models which is important for further model improvement.Thus,we compared six published seismic shear-wave velocity models of Southwest China on absolute velocity and velocity perturbation patterns.The models are derived from different types of data(e.g.,surface waves from ambient noise and earthquakes,body-wave travel times,receiver functions) and inversion methods.We interpolated the models into a uniform horizontal grid(0.5° × 0.5°) and vertically sampled them at 5,10,20,30,40,and 60 km depths.We found significant differences between the six models.Then,we selected three of them that showed greater consistency for further comparison.Our further comparisons revealed systematic biases between models in absolute velocity that may be related to different data types.The perturbation pattern of the model is especially divergent in the shallow part,but more consistent in the deep part.We conducted synthetic and inversion tests to explore possible causes and our results imply that systematic differences between the data,differences in methods,and other factors may directly affect the model.Therefore,the Southwest China velocity model still has considerable room for improvement,and the impact of inconsistency between different data types on the model needs further research.Finally,we proposed a new reference shear-wave velocity model of Southwest China(SwCM-S1.0) based on the three selected models with high consistency.We believe that this model is a better representation of more robust features of the models that are based on different data sets. | Xuezhen Zhang Xiaodong Song Feiyi Wang | 2023 | Earthquake Science2023,36,1: | 0 |
| 12 | Current Situation and Prospect of the Application of Real-World Evidence in Health Care显示文摘Objective To summarize the application of real-world evidence(RWE)in the medical and healthcare field of various countries,including relevant policies,application scenarios and application methods.Methods Relevant policies and application scenarios were obtained by consulting the official websites and public documents of various countries’healthcare institutions.Systematic literature retrieval was adopted to search PubMed,EMBASE,Cochrane Library,CNKI,CBM and Wanfang databases,and all papers related to real-world study and application were included.Then,these papers were classified and analyzed by country and application method.Results and Conclusion The RWE was mainly applied to supporting the preliminary approval of a new drug,expanding drug indications,accelerating approval or supporting conditional marketing authorizations and drug safety evaluation,etc.The United Kingdom,the United States,Germany,the Netherlands,Italy,Sweden,and France admitted RWE,but they treated the data obtained from RWE with caution.After systematic literature retrieval,a total of 701 articles were obtained,including relevant studies from 36 countries,among which the United States published 264 in total.The most common study was about using real-world data(RWD)to calculate treatment-related costs,which had a total of 259 studies.Secondly,158 articles were used for epidemiological analysis.Then,138 articles were about establishing risk models to analyze disease risk factors.A total of 70 articles were real-world efficacy evaluation of the drug treatment schemes,54 articles were about pharmacoeconomic evaluation with RWD as parameters.A total of 29 articles used RWD to build predictive models,and 15 articles used RWD to evaluate the health-related quality of life in patients.The application of RWE has been used widely in the medical and healthcare field of various countries.The application scenarios are gradually diversified,the application methods of RWD become mature,and the evidence quality of RWE is also improved greatly. | Xiao Feiyi Zhang Fang Li Xue Dong Li | 2023 | Asian Journal of Social Pharmacy2023,18,4: | 0 |

| 14 | Genetic diversity,population structure and core collection analysis of Hunan tea plant germplasm through genotyping-by-sequencing显示文摘Tea is considered to be a well-known and widely consumed beverage and Hunan province is rich in tea plant germplasm.In order to better conserve and utilize Hunan tea plant resources,110 tea accessions from seven geographical origins were used to assess genetic diversity of Hunan tea plant germplasm through genotyping by sequencing(GBS)technology.As a result,a total of 311,044 high-quality single nucleotide polymorphism(SNP)markers were obtained.Population structure,phylogenetic relationships and principal component analysis(PCA)divided the entire accessions into three groups.The genetic diversity and population differentiation analysis showed that the mean observed heterozygosity(Ho)ranged from 0.16 to 0.24,while the mean polymorphic information content(PIC)ranged from 0.14 to 0.17,and mean minor allele frequency(MAF)ranged from 0.11 to 0.14.Analysis of molecular variance(AMOVA)indicated that 81.38%of the total variance was derived from within populations,which suggested a rich genetic diversity in Hunan tea germplasms.Furthermore,a core tea germplasm set was developed,which was comprised of 22 tea plant accessions and maintained the whole genetic diversity of the entire collection.This work should be valuable for conservation and utilization of tea germplasm in Hunan。 | Feiyi Huang Jihua Duan Yu Lei Zhen Liu Yankai Kang Yi Luo Yingyu Chen Youyong Li Shuoqian Liu Saijun Li Zhonghua Liu | 2022 | Beverage Plant Research2022,2,1: | 0 |
| 15 | A deep-learning-based approach for seismic surface-wave dispersion inversion(SfNet)with application to the Chinese mainland显示文摘Surface-wave tomography is an important and widely used method for imaging the crust and upper mantle velocity structure of the Earth.In this study,we proposed a deep learning(DL)method based on convolutional neural network(CNN),named SfNet,to derive the vS model from the Rayleigh wave phase and group velocity dispersion curves.Training a network model usually requires large amount of training datasets,which is labor-intensive and expensive to acquire.Here we relied on synthetics generated automatically from various spline-based vS models instead of directly using the existing vS models of an area to build the training dataset,which enhances the generalization of the DL method.In addition,we used a random sampling strategy of the dispersion periods in the training dataset,which alleviates the problem that the real data used must be sampled strictly according to the periods of training dataset.Tests using synthetic data demonstrate that the proposed method is much faster,and the results for the vS model are more accurate and robust than those of conventional methods.We applied our method to a dataset for the Chinese mainland and obtained a new reference velocity model of the Chinese continent(ChinaVs-DL1.0),which has smaller dispersion misfits than those from the traditional method.The high accuracy and efficiency of our DL approach makes it an important method for vS model inversions from large amounts of surface-wave dispersion data. | Feiyi Wang Xiaodong Song Mengkui Li | 2023 | Earthquake Science2023,36,2: | 0 |
