| 1 | Glutamate and aspartate alleviate testicular/epididymal oxidative stress by supporting antioxidant enzymes and immune defense systems in boars显示文摘Several potential oxidative agents have damaging effects on mammalian reproductive systems.This study was conducted to investigate the effects of glutamate(Glu)and aspartate(Asp)supplementation on antioxidant enzymes and immune defense systems in the outer scrotum of boars injected with H2O2.A total of 24 healthy boars were randomly divided into 4 treatment groups:control(basal diet,saline-treated),H2O2(basal diet,H2O2-challenged outer scrotum(1 m L kg^–1 BW)),Glu(basal diet+2%Glu,H2O2-challenged),and Asp(basal diet+2%Asp,H2O2-challenged).Our results showed that both Glu and Asp supplementation improved testicular morphology and decreased the genital index in the H2O2-treated boars.Glu and Asp administration increased the antioxidant enzyme activities and affected the testicular inflammatory cytokine secretion but had no effect on sex hormone levels.Furthermore,the m RNA expression of CAT,Cu Zn SOD,and GPx4 was altered in the testes and epididymis of boars treated with Asp and Glu.Glu and Asp supplementation also modulated the expression of TGF-β1,IL-10,TNF-α,IL-6 and IL-1βin the testis and epididymis.These results indicate that dietary Glu and Asp supplementation might enhance antioxidant capacity and regulate the secretion and expression of inflammatory cytokines to protect the testes and epididymis of boars against oxidative stress. | Wenjie Tang Jian Wu Shunshun Jin Liuqin He Qinlu Lin Feijun Luo Xingguo He Yanzhong Feng Binsheng He Pingping Bing Tiejun Li Yulong Yin | 2020 | Science China(Life Sciences)2020,63,1: | 9 |
| 2 | Assessment of the immune responses to Treponema pallidum Gpd DNA vaccine adjuvanted with IL-2 and chitosan nanoparticles before and after Treponema pallidum challenge in rabbits显示文摘Syphilis is a multistage,sexually transmitted disease caused by the spirochete,Treponema pallidum(Tp).A significantly high incidence of syphilis has been reported in several countries,including China,and there is an urgent need for the development of efficacious vaccines against syphilis.DNA vaccines are a major breakthrough in the field of vaccination with several advantages over traditional vaccines.Animal model studies of Tp DNA vaccines have not been reported elsewhere but our previous reports describe the development of a single-gene Tp DNA vaccine and preclinical immunization study.In this study,chitosan(CS) nanoparticles were used as a vector and an interleukin-2 expression plasmid(pIL-2) as an adjuvant to enhance a TpGpd DNA vaccine candidate(pTpGpd) in a rabbit Tp skin challenge model.At week 8 after the first immunization,three rabbits from each group were used to determine cytokine measurements and spleen lymphocyte proliferation assay.pTpGpd in combination with pIL-2 wrapped with CS led to the greatest enhancement of anti-TpGpd antibodies and T-cell proliferation.During infection,levels of anti-TpGpd antibodies and T-cell proliferation were measured.Both the serum special IgG and IL-2,interferon-γ were significantly increased by the co-injection of the IL-2 plasmid compared with the injection of TpGpd DNA alone(P<0.05).Furthermore,IL-2 plasmid coinjection efficiently enhanced the antigen-specific lymphocyte proliferation response.Additionally,the ratios of positive skin lesions and ulcer lesions in groups immunized with pTpGpd were significantly lower than those of the pIL-2,CS or pIL-2 mixed with CS control groups(P<0.001).CS vectored and pIL-2 adjuvanted pTpGpd immunized animals exhibited the lowest rates of positive skin tests(8.33%) and ulcer lesions(4.17%) and the fastest recovery(42 d).These experiments indicate that co-injection of a pIL-2 plasmid with pTpGpd DNA vaccine wrapped with CS can significantly strengthen the long-term stability of immune response during infection,efficiently improve the protective effect against T.pallidum spirochetes infection and attenuate syphilitic lesion development. | ZHAO FeiJun ZHANG XiaoHong LIU ShuangQuan ZENG TieBing YU Jian GU WeiMing ZHANG YueJun CHEN Xi WU YiMou | 2013 | Science China(Life Sciences)2013,56,2: | 8 |
| 3 | Electrospinning of hydroxypropyl methylcellulose trimellitate solutions显示文摘Nonwoven fiber mats of hydroxypropyl methylcellulose trimellitate(HPMCT) with potential applications in controlled delivery of drugs and scaffolds for tissue cultures have been successfully fabricated by electrospinning of HPMCT solutions.The formation and diameters of HPMCT fibers fabricated by electrospinning were strongly influenced by the solvents employed,electrostatic field strength,and solution concentrations.The electrospun products generated from all HPMCT solutions with various weight-average molecular weights in methanol/dichloromethane(1:1) solvent consisted of clustered beads,while methanol/dichloromethane(1:5) solvent helped to generate smooth,uniform,submicron fibers.From the relationship between specific viscosity and molecular weight in different solvents,it could be concluded that methanol/dichloromethane(1:5) solvent enhanced the molecular entanglements,improved the viscosity of the solutions,thus helped to generate smooth fibers.When all the other variables were kept constant,the concentration range suitable for electrospinning was between 10% and 16%(wt),and the average fiber diameter increased as the concentration increased.When all the other variables were kept constant,the average fiber diameter decreased as the strength of the electrostatic field increased.Electrospinning the solutions of two compounds with similar structure,hydroxypropyl methylcellulose phthalate(HPMCP) and HPMCT,we found that the fibers electrospun from HPMCP were thicker than that from HPMCT,which is attributed to the fact that the solution's conductivity of HPMCT(containing two free carboxyl groups) is higher than that of HPMCP(containing only one free carboxyl group). | MA Gang,SHAO ZiQiang,WANG WenJun,WANG FeiJun,TAN LingLing & LIAO Bing School of Material,Beijing Institute of Technology,Beijing 100081,China | 2010 | Science China Chemistry2010,53,1: | 3 |

| 7 | Natural alleles of a uridine 5'-diphospho-glucosyltransferase gene responsible for differential endosperm development between upland rice and paddy rice显示文摘Traditional upland rice generally exhibits insufficient grains resulting from abnormal endosperm development compared to paddy rice. However, the underlying molecular mechanism of this trait is poorly understood. Here,we cloned the uridine 5’-diphospho(UDP)-glucosyltransferase gene EDR1(Endosperm Development in Rice) responsible for differential endosperm development between upland rice and paddy rice by performing quantitative trait loci analysis and map-based cloning. EDR1 was highly expressed in developing seeds duringgrain filling. Natural variations in EDR1 significantly reduced the UDP-glucosyltransferase activity of EDR1 YZNcompared to EDR1 YD1,resulting in abnormal endosperm development in the near-isogenic line, accompanied by insufficient grains and changes in grain quality.By analyzing the distribution of the two alleles EDR1 YD1 and EDR1 YZNamong diverse paddy rice and upland rice varieties, we discovered that EDR1 was conserved in upland rice, but segregated in paddy rice. Further analyses of grain chalkiness in the alleles of EDR1 YD1 and EDR1 YZNvarieties indicated that rice varieties harboring EDR1 YZNand EDR1 YD1 preferentially showed high chalkiness, and low chalkiness,respectively. In this review, we systemically summarized the efficacy of BR on cardiovascular diseases(CVDs), hyperlipidemia, hypertension, diabetes, obesity, osteoporosis, neurodegenerative diseases, cancer, immune disorders, inflammation, anxiety, liver and stomach damage, etc. Underlying mechanisms have also been analyzed, which include regulating lipid synthesis and metabolism, improving insulin levels and increasing glucose metabolism, anti-oxidation, promoting the immune system, transcription and non-transcriptional regulation, blocking cell cycle, thereby improving disease symptoms and reducing its recurrence rate. Generally, BR has been almost disappeared in modern society, but it is promising to exploit the potential to become a functional food. | 20 | Identification and fine mapping of rtms1-D,a gene responsible for reverse thermosensitive genic male sterility from Diannong S-1X显示文摘Thermosensitive genic male sterility(TGMS)has been widely used in two-line hybrid rice breeding.Due to hybrid seed production being highly affected by changeable environments,its application scope is limited to some extent.Thus,it is of great importance to identify potential TGMS genes in specific rice varieties.Here,Diannong S-1 xuan(DNS-1 X),a reverse TGMS(RTGMS)japonica male sterile line,was identified from Diannong S-1.Genetic analysis showed that male sterility was tightly controlled by a single recessive gene,which was supported by the phenotype of the F;and F;populations derived from the cross between DNS-1 X and Yunjing 26(YJ26).Combining simple sequence repeat(SSR)markers and bulked segregation analysis(BSA),we identified a 215 kb region on chromosome 10 as a candidate reverse TGMS region,which was designated as rtmsl-D.It was narrower than the previously reported RTGMS genes rtmsl and tms6(t).The fertility conversion detected in the natural environment showed that DNS-1 X was sterile below 28-30℃;otherwise,it was fertile.Histological analysis further indicated that the pollen abortion was occurred in the young microspore stage.This study will provide new resources for two-line hybrid rice and pave the way for molecular breeding of RTGMS lines. | Xiao Zhang Guimei Chang Zihao Wu Jinpeng Wan Jun Yang Feijun Wang Fang Wang Diqiu Yu Peng Xu | 2022 | Plant Diversity2022,44,2: | 0 |
