1lncRNA NEAT1 promotes the Taxol resistance of breast cancer via sponging the miR-23a-3p-FOXA1 axis显示文摘Breast cancer is the most prevalent malignancy among women worldwide.Paclitaxel(Taxol)is a widely applied chemotherapeutic agent against breast cancer.Although Taxol therapy has achieved improvements recently,development of chemoresistance of breast cancer patients is a major obstacle,leading to therapeutic failure.Long non-coding RNAs(lncRNAs)play pivotal roles in tumorigenesis and progresses of breast cancer.However,the biological roles and molecular targets of lncRNA NEAT1 in Taxol-resistant breast cancer remain unclear.Here,we report that NEAT1 is significantly upregulated in breast tumors and cell lines.In addition,silencing NEAT1 effectively sensitizes breast cancer cells to Taxol.Bioinformatical analysis and luciferase assay demonstrated that miR-23a-3p could be sponged and downregulated by NEAT1.We demonstrated that miR-23a-3p was downregulated and functioned as a tumor suppressor in breast cancer.Furthermore,in the established Taxol-resistant MDA-MB-231 breast cancer cell line,we detected significantly increased NEAT1 expression and downregulated miR-23a-3p expression.Importantly,FOXA1 was identified and validated as a direct target of miR-23a-3p in breast cancer cells.Rescue experiments demonstrated that the restoration of miR-23a-3p in NEAT1-overexpressing Taxol-resistant breast cancer cells successfully overcame the NEAT1-promoted Taxol resistance.Taken together,our results revealed the clinical roles and molecular mechanisms for the NEAT1-mediated chemoresistance,providing new insights into the development of non-coding RNA-based therapeutic strategies for enhancing the anti-cancer effects of traditional chemotherapeutic drugs.Lin Zhu Fengchun Wang Wenhui Fan Zhi Jin Chao Teng Jianxin Zhang 2021Acta Biochimica et Biophysica Sinica2021,53,9:7

3Fluid Inclusion and Carbon-Oxygen Isotope Studies of the Hujiayu Cu Deposit,Zhongtiao Mountains,China:Implications for Synmetamorphic Copper Remobilization显示文摘The Hujiayu Cu deposit,representative of the 'HuBi-type' Cu deposits in the Zhongtiao Mountains district in the southern edge of the North China Craton,is primarily hosted in graphitebearing schists and carbonate rocks.The ore minerals comprise mainly chalcopyrite,with minor sphalerite,siegenite[(Co,Ni)_3S_4],and clausthalite[Pb(S,Se)].The gangue minerals are mainly quartz and dolomite,with minor albite.Four fluid inclusion types were recognized in the chalcopyrite-pyrite-dolomite-quartz veins,including CO_2-rich inclusions(type Ⅰ),low-salinity,liquid-dominated,biphase aqueous inclusions(type Ⅱ),solid-bearing aqueous inclusions(type Ⅲ),and solid-bearing aqueous-carbonic inclusions(type Ⅳ).Type I inclusion can be further divided into two sub-types,i.e.,monophase CO_2 inclusions(type Ⅰa) and biphase CO_2-rich inclusions(with a visible aqueous phase),and type Ⅲ inclusion is divided into a subtype with a halite daughter mineral(type Ⅲa) and a subtype with multiple solids(type Ⅲb).Various fluid inclusion assemblages(FIAs) were identified through petrographic observations,and were classified into four groups.The group-1 FIA,consisting of monophase CO_2 inclusions(type Ⅰa),homogenized into the liquid phase in a large range of temperatures from-1 to 28℃,suggesting post-entrapment modification.The group-2 FIA consists of type Ⅰb,Ⅲb and Ⅳ inclusions,and is interpreted to reflect fluid immiscibility.The group-3 FIA comprises type Ⅱ and Ⅲa inclusions,and the group-4FIA consists of type Ⅱ inclusions with consistent phase ratios.The group-1 and group-2 FIAs are interpreted to be entrapped during mineralization,whereas group-3 and group-4 FIAs probably represent the post-mineralization fluids.The solid CO_2 melting temperatures range from-60.6 to56.6℃ and from-66.0 to-63.4℃ for type Ⅰa and type Ⅳ inclusions,respectively.The homogenization temperatures for type Ⅱ inclusions range from 132 to 170℃ for group-3 FIAs and115 to 219℃ for group-4 FIAs.The halite melting temperatures range from 530 to 562℃ for typeⅢ b and Ⅳ inclusions,whereas those for type Ⅲa inclusions range from 198 to 398℃.Laser Raman and SEM-EDS results show that the gas species in fluid inclusions are mainly CO_2 with minor CH_4,and the solids are dominated by calcite and halite.The calcite in the hosting marble and dolomite in the hydrothermal veins have δ^(13)C_(V-pdb) values of-0.2 to 1.2‰ and-1.2 to-6.3‰,and δ^(18)O_(v-smow) values of 14.0 to 20.8 ‰ and 13.2 to 14.3‰,respectively.The fluid inclusion and carbon-oxygen isotope data suggest that the ore-forming fluids were probably derived from metamorphic fluids,which had reacted with organic matter in sedimentary rocks or graphite and undergone phase separation at 1.4-1.8 kbar and 230-240℃,after peak metamorphism.It is proposed that the Hujiayu Cu deposit consists of two mineralization stages.The early stage mineralization,characterized by disseminated and veinlet copper sulfides,probably took place in an environment similar to sediment-hosted stratiform copper mineralization.Ore minerals formed in this precursor mineralization stage were remobilized and enriched in the late metamorphic hydrothermal stage,leading to the formation of thick quartz-dolomite-sulfides veins.QIU Zhengjie FAN Hongrui LIU Xuan WEN Bojie HU Fangfang YANG Kuifeng GUO Shuanglong ZHAO Fengchun 2015Acta Geologica Sinica(English Edition)2015,89,3:1
4Dose escalation guided by^(18)F-FDG PET/CT for esophageal cancer显示文摘Objective:To assess the feasibility of dose escalation guided by ^(18)F-deoxyglucose positron emission tomography/computer tomography(^(18)F-FDG PET/CT)for esophageal cancer(EC).Methods and materials:Ten random patients treated with definitive chemoradiotherapy and pre-therapeutic ^(18)FFDG PET/CT were included in this study.Retrospectively,a threshold of 50%of SUVmax was used to define the high FDG uptake region of the GTV(GTVPET).Three intensity-modulated radiation therapy(IMRT)plans were generated,delivering three dose levels to three different planning target volumes(PTVs).50.4 Gy delivered to PTV50.4 was defined on computed tomography(CT)as Plan50.4,63 Gy was delivered to PTV63 was defined as GTV plus a uniform margin of 0.5 cm as Plan63,and 70 Gy delivered to PTV70 was defined as GTVPET plus a 0.5 cm margin as Plan70.A dosimetric comparison was performed based on normal tissue complication probability(NTCP)for the lung and heart.Results:Clinically acceptable dose escalation failed for 2 of 10 patients in Plan63 due to heart dose constraints.One patient failed heart dose constraint for Plan70.Two patients failed spinal cord constraint for Plan63.Three patients failed lung dose constraints for both Plan63 and Plan70,two of which were even not suitable for Plan50.4 for the same reason.NTCP modeling for lung showed increased risk for Plan63 and Plan70 compared to Plan50.4.The difference between Plan63 and Plan70 was insignificant.NTCP modeling for heart showed an increased risk from 6.38%of Plan50.4 to 8.88%of Plan70(P=0.009)or 9.79%of Plan63(P=0.007).The risk of heart mortality was significantly higher for Plan63 than Plan70(P=0.047).Conclusions:Selective boosting of sub-volumes based on ^(18)F-FDG PET/CT is feasible way with a modest increase in the risk of cardiac and lung toxicities.Bingjie Fan Chengqiang Li Fengchun Mu Wenru Qin Linlin Wang Xindong Sun Chunni Wang Bing Zou Shijiang Wang Wanlong Li Man Hu 2021Radiation Medicine and Protection2021,2,2:0
5Microstructural design in LaCe misch-metal substituted 2:14:1-type sintered magnets by dual-alloy method显示文摘LaCe-based sintered magnets with different microstructural features and distinct rare earth elemental distribution were designed by dual-alloy method.The sample prepared by fine LaCe-containing powder and coarse LaCe-free powder possesses higher remanence(~13.41 kGs),whereas another sample prepared by fine LaCe-free powder and coarse LaCe-containing powder possesses higher coercivity(~5.67 kOe).Additionally,these samples are with the same nominal compositions and their elemental distribution features are obviously different in matrix grains respectively.Their remanence difference is mainly affected by the saturation magnetization difference caused by the distribution variation of the rare earth elements at the matrix phase.The coercivity difference is affected by the component of the grain boundary phase between the adjacent grains and the distribution variation of the rare earth elements at the matrix phase.These findings may provide a new prospect for the utilization of LaCe mischmetal in 2:14:1-type permanent magnets.Kan Chen Shuai Guo Hongliang Zhao Xiaodong Fan Fengchun Fan Guangfei Ding Renjie Chen Xiwen Zheng Aru Yan 2021Journal of Rare Earths2021,39,3:0
