| 1 | Scalable volumetric imaging for ultrahigh-speed brain mapping at synaptic resolution显示文摘The speed of high-resolution optical imaging has been a rate-limiting factor for meso-scale mapping of brain structures and functional circuits,which is of fundamental importance for neuroscience research.Here,we describe a new microscopy method of Volumetric Imaging with Synchronized on-the-fly-scan and Readout(VISo R)for high-throughput,high-quality brain mapping.Combining synchronized scanning beam illumination and oblique imaging over cleared tissue sections in smooth motion,the VISo R system effectively eliminates motion blur to obtain undistorted images.By continuously imaging moving samples without stopping,the system achieves high-speed 3D image acquisition of an entire mouse brain within1.5 hours,at a resolution capable of visualizing synaptic spines.A pipeline is developed for sample preparation,imaging,3D image reconstruction and quantification.Our approach is compatible with immunofluorescence methods,enabling flexible cell-type specific brain mapping and is readily scalable for large biological samples such as primate brains.Using this system,we examined behaviorally relevant whole-brain neuronal activation in 16 c-Fos-sh EGFP mice under resting or forced swimming conditions.Our results indicate the involvement of multiple subcortical areas in stress response.Intriguingly,neuronal activation in these areas exhibits striking individual variability among different animals,suggesting the necessity of sufficient cohort size for such studies. | Hao Wang Qingyuan Zhu Lufeng Ding Yan Shen Chao-Yu Yang Fang Xu Chang Shu Yujie Guo Zhiwei Xiong Qinghong Shan Fan Jia Peng Su Qian-Ru Yang Bing Li Yuxiao Cheng Xiaobin He Xi Chen Feng Wu Jiang-Ning Zhou Fuqiang Xu Hua Han Pak-Ming Lau Guo-Qiang Bi | 2019 | National Science Review2019,6,5: | 7 |
| 2 | Detection and characterization of diverse alpha-and betacoronaviruses from bats in China显示文摘Bats have been implicated as important reservoir hosts of alpha- and betacoronaviruses. In this study, diverse coronaviruses(CoVs) were detected in 50 of 951(positive rate 5.3%) intestinal specimens of eight bat species collected in four provinces and the Tibet Autonomous Region of China by pan-coronavirus RT-PCR screening. Based on 400-nt RNA-dependent RNA polymerase(Rd RP) sequence analysis, eight belonged to genus Alphacoronavirus and 42 to Betacoronavirus.Among the 50 positive specimens, thirteen gave rise to CoV full-length Rd RP gene amplification for further sequence comparison, of which three divergent sequences(two from a unreported province) were subjected to full genome sequencing. Two complete genomes of betacoronaviruses(JTMC15 and JPDB144) and one nearly-complete genome of alphacoronavirus(JTAC2) were sequenced and their genomic organization predicted. The present study has identified additional numbers of genetically diverse bat-borne coronaviruses with a wide distribution in China. | 5 | Rapid and Sparse Labeling of Neurons Based on the Mutant Virus-Like Particle of Semliki Forest Virus显示文摘Sparse labeling of neurons contributes to uncovering their morphology, and rapid expression of a fluorescent protein reduces the experiment range. To achieve the goal of rapid and sparse labeling of neurons in vivo, we established a rapid method for depicting the fine structure of neurons at 24 h post-infection based on a mutant viruslike particle of Semliki Forest virus. Approximately 0.014 fluorescent focus-forming units of the mutant virus-like particle transferred enhanced green fluorescent protein into neurons in vivo, and its affinity for neurons in vivo was stronger than for neurons in vitro and BHK21(baby hamster kidney) cells. Collectively, the mutant virus-likeparticle provides a robust and convenient way to reveal the fine structure of neurons and is expected to be a helper virus for combining with other tools to determine their connectivity. | 6 | Experimental Studies on Fuel Spray Characteristics of Pressure-Swirl Atomizer and Air-Blast Atomizer显示文摘In this work,the effects of fuel temperatures and pressure drops on the flow field and spray characteristics of a pressure-swirl atomizer were discussed using the Particle Imaging Velocimetry(PIV),Planar Laser Induced Fluorescence(PLIF)and Laser Particle Size Analyzer(LPSA)methods.Then the air-blast atomizer was selected to study the interaction of initial atomization and flow field.The effect of fuel-air ratio on the air-blast atomizer were also considered,where the fuel-air ratio was varied by adjusting mass flow rate of the air and fuel respectively.The results show that the spray angle of the pressure-swirl atomizer increases first and changes a little after the pressure drop higher than 0.5 MPa.However,more fuel concentrate on the central region,which is mainly caused by the increase of the proportion of small droplets with lower centrifugal force.The fuel temperature can improve the spray angle only in lower pressure drop,and it has a little effect under higher pressure drops.In addition,the fuel pressure drop has an obvious influence on the fuel distribution and flow field near the nozzle exit compared with the downstream.For the air-blast atomizer,the spray angle increases compared with the pressure-swirl atomizer for the introduction of swirl air.Furthermore,the spray angle decreases with the air mass rate increasing,and it increases with the fuel mass rate increasing.The distribution of velocity and droplet near the nozzle exit is influenced by the air mass rate,and the fuel mass rate mainly affects the distribution in the downstream.The fuel accumulates in the annular area below the nozzle,and the distribution of it changes little with the development along the axial direction. | WANG Kaixing FAN Xiongjie LIU Fuqiang LIU Cunxi LU Haitao XU Gang | 2021 | Journal of Thermal Science2021,30,2: | 1 |
| 7 | Spatiotemporal variations of carbon flux and nitrogen deposition flux linked with climate change at the centennial scale in China显示文摘The spatial and temporal variations of the vegetation carbon flux in China from 1901 to 2005 were studied using the vegetation net primary production(NPP)values from seven Earth-system models.In addition,the temporal and spatial changes in the nitrogen deposition flux in China were studied using the NHx and NOy fluxes from 1901 to 2005.The relationship between changes in the carbon flux,nitrogen flux and climate was analyzed.The results show that(1)over the past 100 years,NPP in China has shown an upward trend.The average trend coefficient is 0.88 and the NPP distribution trend is generally low in the north and high in the south,with a gradual increase from the northwest to the southeast.Temperature,precipitation and radiation are all conducive to plant growth in the direction of the gradient.The correlation coefficients between the ensemble model mean NPP and temperature,precipitation,longwave radiation and shortwave radiation are 0.88,0.73,0.91 and 0.67,respectively.(2)In the past 100 years,the NHx and NOy fluxes in China have shown an upward trend,with trend coefficients of 0.98 and 0.98,respectively,which pass the 99.9%confidence level of the t-test.NHx and NOy fluxes are also generally low in the north and high in the south,with a gradual increase from the northwest to the southeast in a step-like pattern.(3)The spatial distribution of the correlation coefficients between the NHx and NOy fluxes and air temperature is similar,with only slight differences in values.The spatial distribution of the correlation coefficients between the NHx and NOy fluxes and precipitation is similar in overall pattern,but the pattern is relatively complicated,with a positive-negative-positive-negative-positive pattern occurring across the monsoon region from north to south,and a negative-positive-negative-positive pattern occurring beyond the monsoon region from east to west.(4)The spatial distribution of the correlation coefficients between the NHx and NOy fluxes and NPP shows a generally consistent pattern,but the pattern is relatively uneven.The average distribution of the ensemble model mean is positive correlation in northeast China and southwest China,and alternating positive and negative correlation in other regions. | Dongdong GAO Li DAN Guangzhou FAN Hanqin TIAN Jing PENG Xiujing YANG Fuqiang YANG Yueyue LI | 2020 | Science China Earth Sciences2020,63,5: | 1 |

| 15 | Optimization of spectral distortion in a ytterbium-doped mode-locked fiber laser system显示文摘A method for optimizing the spectral distortion of an ultrafast pulse in a polarization-maintaining picosecond linear-cavity fiber laser with a one-stage fiber amplifier is proposed and demonstrated. The mechanism of control of the spectral distortion in the fiber system has been investigated. The experimental and theoretical results illustrate that the filtering effect of a fiber Bragg grating can effectively decrease the spectral oscillatory distortion accumulated by self-phase modulation.
