| 1 | Rapid generation of genetic diversity by multiplex CRISPR/Cas9 genome editing in rice显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated endonuclease 9(CRISPR/Cas9) system has emerged as a promising technology for specific genome editing in many species. Here we constructed one vector targeting eight agronomic genes in rice using the CRISPR/Cas9 multiplex genome editing system. By subsequent genetic transformation and DNA sequencing, we found that the eight target genes have high mutation efficiencies in the T_0 generation. Both heterozygous and homozygous mutations of all editing genes were obtained in T_0 plants. In addition, homozygous sextuple, septuple, and octuple mutants were identified. As the abundant genotypes in T_0 transgenic plants, various phenotypes related to the editing genes were observed. | 2 | Characteristics of authigenic pyrite and its sulfur isotopes influenced by methane seep at Core A, Site 79 of the middle Okinawa Trough显示文摘The anaerobic oxidation of methane(AOM) has strongly developed at Core A, Site 79 of the middle Okinawa Trough, East China Sea, and a large amount of authigenic pyrite is preserved in the surface sediment. In this study, we analyze the characteristics of the authigenic pyrite and its sulfur isotopic values. The authigenic pyrite is stripy and tubular, and there were foraminifera compartments filled with pyrite. The pyrite is extracted using chromium reduction, and the values of ? 34 S are found to lie between ?41.20‰ and 8.92‰ V-CDT. The bulk pyrite tends to be more enriched in 34 S with increasing depth. Particularly, the ? 34 S value of the pyrite lies between ?32.73‰ and ?41.20‰ V-CDT above 278 cmbsf, but it quickly increases below this depth(?21.49‰–8.92‰ V-CDT). At the same time, the total sulfur content of the pyrite shows an abrupt increase above 100 cmbsf but is otherwise stable between 1.04% and 0.55% below 100 cmbsf. The stable and negative values of ? 34 S and the decreasing values of total sulfur above 278 cmbsf indicate reduced AOM activities in 17.18–5.3 ka. In addition, the increasing ? 34 S and pyrite content indicate strong AOM development and methane seep below 278 cmbsf in 18.8–17.18 ka. In particular, the highest positive value of ? 34 S occurring in 18.78 ka indicates the most intense AOM activity. | 11 | Oscillation quenching and physical explanation on freeplay-based aeroelastic airfoil in transonic viscous flow显示文摘Limit Cycle Oscillation(LCO)quenching of a supercritical airfoil(NLR 7301)considering freeplay is investigated in transonic viscous flow.Computational Fluid Dynamics(CFD)based on Navier-Stokes equations is implemented to calculate transonic aerodynamic forces.A loosely coupled scheme with steady CFD and an efficient graphic method are developed to obtain the aerodynamic preload.LCO quenching phenomenon is observed from the nonlinear dynamic aeroelastic response obtained by using time marching approach.As the airspeed increases,LCO appears then quenches,forming the first LCO branch.Following the quenching region,LCO occurs again and sustains until the divergence of the response,forming the second LCO branch.The quenching of LCOs was addressed physically based on the aerodynamic preload and the linear flutter characteristic.An“island”of stable region is observed in the flutter boundary,i.e.the flutter speed versus the mean Angle of Attack(AoA).The LCO quenches when the aerodynamic preload crosses this stable region with the increasing of airspeed.The LCO quenching of this model in transonic flow is essentially induced by destabilizing effect from aerodynamic preload,since the flutter speed is sensitive to AoA due to aerodynamic nonlinearity. | Yayun SHI Shun HE Gaowei CUI Gang CHEN Yan LIU | 2023 | Chinese Journal of Aeronautics2023,36,10: | 0 |
