2Investigating different bio-responses of the upper ocean to Typhoon Haitang using Argo and satellite data显示文摘The responses of the upper ocean to Typhoon Haitang in July 2005 are investigated using Argo float and multiplatform satellite data.The results show decreasing sea surface temperature(SST),a deepening of the mixed layer depth(MLD),and enhanced Chlorophyll-a(Chl-a)concentration.Two extreme cool regions are identified.While the magnitude of SST cooling in the two regions is similar,the biological response(Chla enhancement)differs.To facilitate comparisons,the region to the northeast of Taiwan is defined as region A and the region east of Taiwan as region B.Ekman pumping and the intrusion of the Kuroshio play an important role in the enhancement of Chl-a in region A.Cold eddies provide the material source for the formation of the cold center in region B,where mixing is dominant.Because of the relatively high translation speed(5 m/s)in region B,Ekman pumping has little influence on the cooling and Chl-a enhancement processes.Moreover,the MLD is shallower than the nutricline,which means that mixing does not result in a marked increase in nutrients in the euphotic layer(where the nutrient concentration is uniformly depleted).Sea temperatures,in contrast,gradually decrease with depth below the bottom of the mixed layer.In contrast to region A,region B showed no significant enhancement of Chl-a but strong SST cooling.Haixia Shan Yuping Guan Jianping Huang 2014Chinese Science Bulletin2014,59,8:6
32017 Chinese expert consensus on the clinical application ofserum marker for thyroid cancer显示文摘In recent years, the clinical incidence of thyroid cancer has been increasing year by year, and its risk assessment and clinical management methods have also been accordingly modified and constantly improved. There are great differences between the clinical diagnostic and therapeutic modes and disease management of thyroid cancer employed by various medical institutions in China, particularly with regard to the clinical application of serum marker of thyroid cancer. 13Variant rs8400 enhances ALKBH5 expression through disrupting miR-186 binding and promotes neuroblastoma progression显示文摘Objective:AlkB homolog 5(ALKBH5)has been proven to be closely related to tumors.However,the role and molecular mechanism of ALKBH5 in neuroblastomas have rarely been reported.Methods:The potential functional single-nucleotide polymorphisms(SNPs)in ALKBH5 were identified by National Center for Biotechnology Information(NCBI)dbSNP screening and SNPinfo software.TaqMan probes were used for genotyping.A multiple logistic regression model was used to evaluate the effects of different SNP loci on the risk of neuroblastoma.The expression of ALKBH5 in neuroblastoma was evaluated by Western blotting and immunohistochemistry(IHC).Cell counting kit-8(CCK-8),plate colony formation and 5-ethynyl-2'-deoxyuridine(EdU)incorporation assays were used to evaluate cell proliferation.Wound healing and Transwell assays were used to compare cell migration and invasion.Thermodynamic modelling was performed to predict the ability of miRNAs to bind to ALKBH5 with the rs8400 G/A polymorphism.RNA sequencing,N6-methyladenosine(mA)sequencing,mA methylated RNA immunoprecipitation(MeRIP)and a luciferase assay were used to identify the targeting effect of ALKBH5 on SPP1.Results:ALKBH5 was highly expressed in neuroblastoma.Knocking down ALKBH5 inhibited the proliferation,migration and invasion of cancer cells.miR-186-3p negatively regulates the expression of ALKBH5,and this ability is affected by the rs8400 polymorphism.When the G nucleotide was mutated to A,the ability of miR-186-3p to bind to the 3'-UTR of ALKBH5 decreased,resulting in upregulation of ALKBH5.SPPI is the downstream target gene of the ALKBH5 oncogene.Knocking down SPP1 partially restored the inhibitory effect of ALKBH5 downregulation on neuroblastoma.Downregulation of ALKBH5 can improve the therapeutic efficacy of carboplatin and etoposide in neuroblastoma.Conclusions:We first found that the rs8400 G>A polymorphism in the m6A demethylase-encoding gene ALKBH5 increases neuroblastoma susceptibility and determines the related mechanisms.The aberrant regulation of ALKBH5 by miR-186-3p caused by this genetic variation in ALKBH5 promotes the occurrence and development of neuroblastoma through the ALKBH5-SPP1 axis.Qian Guan Huiran Lin Wenfeng Hua Lei Lin Jiabin Liu Linqing Deng Jiao Zhang Jiwen Cheng Zhonghua Yang Yong Li Jun Bian Haixia Zhou Suhong Li Li Li Lei Miao Huimin Xia Jing He Zhenjian Zhuo 2023Chinese Journal of Cancer Research2023,35,2:0
14Analysis of Predictability of Local Rainstorm in Qinghai during August 19-20,2019显示文摘Based on conventional meteorological observation data,ECMWF model data,etc.,the causes of rainstorm in the northeast of Qinghai from the afternoon to the night of August 19,2019 were analyzed,and the causes and predictability of major forecast errors were discussed.The results show that there was high temperature,high humidity and high energy in the middle and low layers in the early period of rainstorm.As the subtropical high retreated eastwards,the high-altitude trough of the westerly belt and the vertical shear line of the plateau moved eastwards to make ground cold air move eastwards,and local short-term heavy precipitation appeared under the influence of this weather system.At the same time,the high-level dry and cold air moved into convective unstable areas.The wind speed at 700 hPa strengthened,and the low-level jet stream appeared to aggravate the instability of atmospheric stratification,which was more conducive to the generation of convection.The forecast results of precipitation based on various numerical models were significantly smaller,and the predicted western cold air was obviously weaker,which was also one of the reasons why the weak transportation of the western water vapor resulted in a small value of predicted precipitation.In the adjustment of model forecast,the proportion and amount of convective precipitation should be adjusted to be large,which has a certain indicating significance for the forecast of convective precipitation.In short-term nowcasting,the deviation of major weather systems predicted based on the model should be corrected in real time,and attention should be paid to the location of ground specific humidity and the convergence line.The falling area of heavy precipitation can be judged according to the high-value area of specific humidity,and the maximum of specific humidity appeared 1-2 h earlier than short-term heavy rainfall.Qingping LI Liang XU Qin GUAN Yao HU Chenghong MEI Jinhai LI Chengzhen SHI Haixia QI 2020Meteorological and Environmental Research2020,11,5:0

16CYP2S1 is a synthetic lethal target in BRAFV600E-driven thyroid cancers显示文摘BRAF^(V600E)is the most common genetic alteration and has become a major therapeutic target in thyroid cancers;however,intrinsic feedback mechanism limited clinical use of BRAF^(V600E)specific inhibitors.Synthetic lethal is a kind of interaction between two genes,where only simultaneously perturbing both of the genes can lead to lethality.Here,we identified CYP2S1 as a synthetic lethal partner of BRAF^(V600E)in thyroid cancers.First,we found that CYP2S1 was highly expressed in papillary thyroid cancers(PTCs)compared to normal thyroid tissues,particularly in conventional PTCs(CPTCs)and tall-cell PTCs(TCPTCs),and its expression was positively associated with BRAFV600E mutation.CYP2S1 knockdown selectively inhibited cell proliferation,migration,invasion and tumorigenic potential in nude mice,and promoted cell apoptosis in BRAFV600E mutated thyroid cancer cells,but not in BRAF wildtype ones.Mechanistically,BRAF^(V600E)-mediated MAPK/ERK cascade upregulated CYP2S1 expression by an AHR-dependent pathway,while CYP2S1 in turn enhanced transcriptional activity of AHR through its metabolites.This AHR/CYP2S1 feedback loop strongly amplified oncogenic role of BRAF^(V600E)in thyroid cancer cells,thereby causing synthetic lethal interaction between CYP2S1 and BRA^(FV600E).Finally,we demonstrated CYP2S1 as a potential therapeutic target in both BRAF^(V600E)-drived xenograft and transgenic mouse models by targetedly delivering CYP2S1-specific siRNA.Altogether,our data demonstrate CYP2S1 as a synthetic lethal partner of BRAFV600E in thyroid cancers,and indicate that targeting CYP2S1 will provide a new therapeutic strategy for BRAFV600E mutated thyroid cancers.Yiqi Li Xi Su Chao Feng Siyu Liu Haixia Guan Yue Sun Nongyue He Meiju Ji Peng Hou 2020Signal Transduction and Targeted Therapy2020,5,1:0
17Comparative study of the effects of Tartary buckwheat seed and sprout consumption on the physiological indices and gut microbiota of C57BL/6J mice显示文摘Tartary buckwheat(Fagopyrum tataricum)is a well-known pseudocereal for its health and economic value.However,abundant antinutritional factors(ANFs)reduces its health benefits.As reported,germination can improve the nutritional profile of grains.In this study,we systematically evaluate the safety of Tartary buckwheat seeds(TB)and Tartary buckwheat sprouts(TBS)used as high active ingredients.After evaluating nutrition levels,bioactive compounds and ANFs in TBS during germinating,5^(th)-day TBS were selected as the raw material.C57BL/6J mice were gavaged daily with distilled water,TB,or TBS for 6 weeks.The physiological indices related to ANFs were determined.Results showed that the TB intake tends to generate negative effects on the gut microbiota,and organs.Additionally,upon TB intake,the Fe^(3+)content in serum,trypsin activity in pancreas and jejunum decreased,while the cytokine,IgE,and histamine levels in serum,water content in faeces,cytokine levels in liver and jejunum increased.Conversely,TBS did not induce any obvious negative effects on the above relevant indices and showed better lipid-lowering effect.Altogether,TBS are safer and more effective as a raw material to produce the functional food for long-term consumption with the intention of preventing and treating hyperlipidaemia.Guohui Nan Haixia Zhao Qiong Wu Lisong Liu Zichao Guan Chenglei Li Huala Wu Dabing Xiang Qi Wu 2024Food Science and Human Wellness2024,13,2:0
