1Controllable preparation of particles with microfluidics显示文摘This paper reviews recent development and achievements in controllable preparation of nanoparticles, micron spherical and non-spherical particles, using microfluidics. A variety of synthesis strategies are presented and compared, including single-phase and multiphase microflows. The main structures of microfluidic devices and the fundamental principles of microflows for particle preparation are summarized and identifled. The controllability of particle size, size distribution, crystal structure, morphology, physical and chemical properties, is examined in terms of the special features of microfluidic reactors. 2Laboratory simulation on acoustic well-logging with phased array transmitter显示文摘Two small scale acoustic phased arrays with 4 elements have been designed and assembled in the laboratory. Experiments have been carried out with them. It is found that both directivity and radiation lobe width of the phased array can be regulated by changing the time delay between the input signals on neighboring elements. Results measured are in good agreement with those calculated. By using the phased array as an acoustic transmitter and hydrophone as a receiver, small scale acoustic well-logging simulations have been carried out both on an aluminum model well and on a concrete one. Experimental results show that, by increasing the time delay of the input signals on neighboring elements, the steered radiation angle of the phased array becomes larger and larger, and generation conditions of the refracted compressional wave and the refracted shear wave are reached successively, and the refracted compressional wave, the refracted shear wave and the Stoneley wave axe strengthened, respectively. 3In situ preparation of hydrophobic CaCO_3 nanoparticles in a gas-liquid microdispersion process显示文摘This study presents a novel process of in situ surface modification of CaCCh nanoparticles using a multipleorifice dispersion microreactor.CO_2/Ca(OH)_2 precipitation reaction was employed to prepare CaCO_3nanoparticles with sodium stearate surfactant.Synthesized CaCCh products were characterized by thermogravimetric analysis(TGA),infra-red(IR),X-ray diffraction(XRD),transmission electron microscopy(TEM) and Brunauer-Emmet-Teller analysis(BET).The effect of various operation parameters on nanoparticles and the dosage of sodium stearate were determined.The results showed that the preparation process could be precisely controlled with efficient mass transfer process.The particles were highly hydrophobic with a contact angle of 117 and monodisperse with an average size of 30 nm.The adsorptions of sodium stearate and calcium ion on solid particles during the in situ surface modification process were investigated.Le Du Yujun Wang Guangsheng Luo 2013Particuology2013,11,4:8
4Recent developments in microfluidic device-based preparation, functionalization, and manipulation of nano- and micro-materials显示文摘Microfluidic systems provide a powerful platform for preparing,functionalizing,and manipulating nano-/micro-materials.In this review,we summarize the characteristics of state-of-the-art microfluidic techniques and their application to processing novel nano-/micro-materials with tunable size,shape,morphology,composition,and properties.A variety of synthesis strategies based on novel microfiuidic devices are presented and compared,including their integration with real-time monitoring,external fields,and automatic feedback control.Novel functional nano-/micro-materials with complex structures have been controllably prepared and post-processed to obtain high-quality products.Strategies for controlling and tuning physical and chemical properties are also examined in terms of the design and function of microfluidic platforms.We provide an outlook on future developments in the processing of nano-/micro-materials by microfluidic technologies.Guangsheng Luo Le Du Yujun Wang Kai Wang 2019Particuology2019,17,4:5
5Determination of the Circulation for a Large-Scale Wind Turbine Blade Using Computational Fluid Dynamics显示文摘The determination of the circulation for wind turbine blades is an important problem in engineering.In the present study,we develop a specific approach to evaluate the integral that represents mathematically the circulation.First the potentialities of the method are assessed using a two-dimensional NACA64_A17 airfoil as a testbed and evaluating the influence of different integration paths and angles of attack on the circulation value.Then the method is applied to blades with different relative heights in order to provide useful reference data to be used for the optimization and reverse design of wind turbine blades.As shown by the results,the integral value changes with the integral path,and an“optimal circle radius”exists.We calibrate the integral value by comparing its value with the lift formula.In this was we succeed in showing that there is a certain error when the radius is too small.However,the error can increase rapidly when the radius is too large.When the radius of the circle is 1–6 times the chord length,the error of all integral values is less than 5%.The optimal radius varies with the angle of attack.Hao Cheng Guangsheng Du Meng Zhang Kun Wang Wenbin Bai 2020Fluid Dynamics & Materials Processing2020,16,4:3

7Single-shot AAV-vectored vaccine against SARS-CoV-2 with fast and long-lasting immunity显示文摘Due to the insufficient long-term protection and significant efficacy reduction to new variants of current COVID-19 vaccines,the epidemic prevention and control are still challenging.Here,we employ a capsid and antigen structure engineering(CASE)strategy to manufacture an adenoassociated viral serotype 6-based vaccine(S663V-RBD),which expresses trimeric receptor binding domain(RBD)of spike protein fused with a biological adjuvant RS09.Impressively,the engineered S663V-RBD could rapidly induce a satisfactory RBD-specific IgG titer within 2 weeks and maintain the titer for more than 4 months.Compared to the licensed BBIBP-CorV(Sinopharm,China),a single-dose S663V-RBD induced more endurable and robust immune responses in mice and elicited superior neutralizing antibodies against three typical SARS-CoV-2 pseudoviruses including wild type,C.37(Lambda)and B.1.617.2(Delta).More interestingly,the intramuscular injection of S663V-RBD could overcome pre-existing immunity against the capsid.Given its effectiveness,the CASE-based S663VRBD may provide a new solution for the current and next pandemic.Fuhua Wu Shuang Luo Yongshun Zhang Yangsen Ou Hairui Wang Zhaofei Guo Chunting He Shuting Bai Penghui He Min Jiang Xiaoyan Chen Guangsheng Du Xun Sun 2023Acta Pharmaceutica Sinica B2023,13,5:2

9Effects of ferrites on magnetic transport properties in La_(2/3)Sr_(1/3)MnO_3/ferrites composites显示文摘Effects of soft-magnetic MnZn ferrite (Mn0.5Zn0.5Fe2O4, MZF) and hard-magnetic Ba ferrite (BaO·6Fe2O3, BaM) on the structure and magnetic transport properties of [La2/3Sr1/3MnO3] (LSMO)/(x) [ferrites] (ferrites=MZF, BaM) composites have been investigated. It was found that the inclusion of MZF phase reduces magnetization and ferromagnetic-paramagnetic transition temperature (Tc) of the composites. With increasing the content of the dopants, the high-temperature magnetoresistance (MR) decreases, whereas low-temperature MR increases and reaches 42% at 150 K and x=0.1. However, for the LSMO/BaM composites, magnetization and ferromagnetic- paramagnetic transition temperature (Tc) decrease firstly as x<5%, and then increase as x>5%. The resistivity of the composites increases by five orders of magnitude at x=1% and is out of measured range at x=5%. 11Recent progress in application of nanovaccines for enhancing mucosal immune responses显示文摘Mucosal vaccines that stimulate both mucosal and systemic immune responses are desirable,as they could prevent the invading pathogens at their initial infection sites in a convenient and userfriendly way. Nanovaccines are receiving increasing attention for mucosal vaccination due to their merits in overcoming mucosal immune barriers and in enhancing immunogenicity of the encapsulated antigens.Herein, we summarized several nanovaccine strategies that have been reported for enhancing mucosal immune responses, including designing nanovaccines that have superior mucoadhesion and mucus penetration capacity, designing nanovaccines with better targeting efficiency to M cells or antigen-presenting cells, and co-delivering adjuvants by using nanovaccines. The reported applications of mucosal nanovaccines were also briefly discussed, including prevention of infectious diseases, and treatment of tumors and autoimmune diseases. 14The effects of bonding conditions on the propagation of acoustic waves along a cased borehole显示文摘Based on the wave equations in cylindrically layered structures and boundary conditions, the frequency equation for axisymmetric guided waves and the expression for sound fields in a cased borehole excited by a monopole or multipole source have been derived. The synthetic full waveforms excited by the monopole and dipole source are simulated using a real axis integration and FFT method. According to the axisymmetric guided wave modes, the synthetic full waveforms and the effects of the interface conditions on the sound field in a cased borehole have been analyzed and studied respectively. Numerical results indicate that it may be difficult to distinguish well bonded, poorly bonded or unbonded intermediate layer between the steel pipe and formation if only using a monopole source or dipole source. 16Combination of AAV-delivered tumor suppressor PTEN with anti-PD-1 loaded depot gel for enhanced antitumor immunity显示文摘Recent clinical studies have shown that mutation of phosphatase and tensin homolog deleted on chromosome 10(PTEN)gene in cancer cells may be associated with immunosuppressive tumor microenvironment(TME)and poor response to immune checkpoint blockade(ICB)therapy.Therefore,efficiently restoring PTEN gene expression in cancer cells is critical to improving the responding rate to ICB therapy.Here,we screened an adeno-associated virus(AAV)capsid for efficient PTEN gene delivery into B16F10 tumor cells.We demonstrated that intratumorally injected AAV6-PTEN successfully restored the tumor cell PTEN gene expression and effectively inhibited tumor progression by inducing tumor cell immunogenic cell death(ICD)and increasing immune cell infiltration.Moreover,we developed an anti-PD-1 loaded phospholipid-based phase separation gel(PPSG),which formed an in situ depot and sustainably release anti-PD-1 drugs within 42 days in vivo.In order to effectively inhibit the recurrence of melanoma,we further applied a triple therapy based on AAV6-PTEN,PPSG^(@anti-PD-1)and CpG,and showed that this triple therapy strategy enhanced the synergistic antitumor immune effect and also induced robust immune memory,which completely rejected tumor recurrence.We anticipate that this triple therapy could be used as a new tumor combination therapy with stronger immune activation capacity and tumor inhibition efficacy.Yongshun Zhang Lan Yang Yangsen Ou Rui Hu Guangsheng Du Shuang Luo Fuhua Wu Hairui Wang Zhiqiang Xie Yu Zhang Chunting He Cheng Ma Tao Gong Ling Zhang Zhirong Zhang Xun Sun 2024Acta Pharmaceutica Sinica B2024,14,1:0

18Nanoparticles with rough surface improve the therapeutic effect of photothermal immunotherapy against melanoma显示文摘Photothermal therapy has been intensively investigated for treating cancer in recent years.However,the long-term therapeutic outcome remains unsatisfying due to the frequently occurred metastasis and recurrence.To address this challenge,immunotherapy has been combined with photothermal therapy to activate anti-tumor immunity and relieve the immunosuppressive microenvironment within tumor sites.Here,we engineered silica-based core-shell nanoparticles(JQ-1@PSNs-R),in which silica cores were coated with the photothermal agent polydopamine,and a bromodomain-containing protein 4(BRD4) inhibitor JQ-1 was loaded in the polydopamine layer to combine photo thermal and immune therapy for tumor elimination.Importantly,to improve the therapeutic effect,we increased the surface roughness of the nanoparticles by hydrofluoric acid(HF) etching during the fabrication process,and found that the internalization of JQ-1@PSNs-R was significantly improved,leading to a strengthened photothermal killing effect as well as the increased intracellular delivery of JQ-1.In the animal studies,the multifunctional nanoparticles with rough surfaces effectively eradicated melanoma via photothermal therapy,successfully activated tumor-specific immune responses against residual tumor cells,and further prevented tumor metastasis and recurrence.Our results indicated that JQ-1@PSNs-R could serve as an innovative and effective strategy for combined cancer therapy.Jiao Xue Yining Zhu Shuting Bai Chunting He Guangsheng Du Yuandong Zhang Yao Zhong Wenfei Chen Hairui Wang Xun Sun 2022Acta Pharmaceutica Sinica B2022,12,6:0
19Determination of interfacial tension and viscosity under dripping flow in a step T-junction microdevice显示文摘Microfluidic approaches for the determination of interfacial tension and viscosity of liquid-liquid systems still face some challenges.One of them is liquid-liquid systems with low interfacial and high viscosity,because dripping flow in normal microdevices can’t be easily realized for the systems.In this work,we designed a capillary embedded step T-junction microdevice to develop a modified microfluidic approach to determine the interfacial tension of several systems,specially,for the systems with low interfacial tension and high viscosity.This method combines a classical T-junction geometry with a step to strengthen the shear force further to form monodispersed water/oil(w/o)or aqueous two-phase(ATP)droplet under dripping flow.For systems with low interfacial tension and high viscosity,the operating range for dripping flow is relative narrow whereas a wider dripping flow operating range can be realized in this step Tjunction microdevice when the capillary number of the continuous phase is in the range of 0.01 to 0.7.Additionally,the viscosity of the continuous phase was also measured in the same microdevice.Several different systems with an interfacial tension from 1.0 to 8.0 m N·m^(-1) and a viscosity from 0.9 to 10 m Pa·s were measured accurately.The experimental results are in good agreement with the data obtained from a commercial interfacial tensiometer and a spinning digital viscometer.This work could extend the application of microfluidic flows.Li Ma Yongjin Cui Lin Sheng Chencan Du Jian Deng Guangsheng Luo 2022Chinese Journal of Chemical Engineering2022,35,2:0
20An antigen self-assembled and dendritic cell-targeted nanovaccine for enhanced immunity against cancer显示文摘The rise of nanotechnology has opened new horizons for cancer immunotherapy.However,most nano vaccines fabricated with nanomaterials suffer from carrier-related concerns,including low drug loading capacity,unpredictable metabolism,and potential systemic toxicity,which bring obstacles for their clinical translation.Herein,we developed an antigen self-assembled nanovaccine,which was resulted from a simple acryloyl modification of the antigen to induce self-assembly.Furthermore,a dendritic cell targeting head mannose monomer and a mevalonate pathway inhibitor zoledronic acid(Zol)were integrated or absorbed onto the nanoparticles(denoted as MEAO-Z)to intensify the immune response.The synthesized nano vaccine with a diameter of around 70 nm showed successful lymph node transportation,high dendritic cell internalization,promoted costimulatory molecule expression,and preferable antigen cross-presentation.In virtue of the above superiorities,MEAO-Z induced remarkably higher titers of serum antibody,stronger cytotoxic T lymphocyte immune responses and IFN-γsecretion than free antigen and adjuvants.In vivo,MEAO-Z significantly suppressed EG7-OVA tumor groth and prolonged the survival time of tumor-bearing mice.These results indicated the translation promise of our self-assembled nano vaccine for immune potentiation and cancer immunotherapy.Yunting Zhang Min Jiangy Guangsheng Du Xiaofang Zhong Chunting He Ming Qin Yingying Hou Rong Liu Xun Sun 2023Acta Pharmaceutica Sinica B2023,13,8:0
