1'Climate effect' of the northeast cold vortex and its influences on Meiyu显示文摘The Northeast Cold Vortex (NECV) is an important weather system in the middle and high latitudes in East Asia. Its time scale is synoptic, yet the frequent activities of NECV have significant 'climate effect' which influences not only the monthly temperature in the lower troposphere in Northeast China but also the Meiyu rainfall in East Asia. On the basis of ERA-40 reanalysis data provided by ECMWF, the 'climate effect' of NECV and its relationship with Meiyu in East Asia are studied. It is shown that there is significant correlation between NECV during the Meiyu period and rainfall amount: strong NECV corresponds to more Meiyu rainfall and weak NECV corresponds to less rainfall. In strong NECV years, the dry and cold air from the north is led to the south by NECV, converges with the lower-level warm and wet southwesterly on the north verge of Meiyu region, thus forms an unstable stratification of 'upper dryness and lower wetness' . Triggered by ascending motion, the Meiyu rainfall amount is more than usual. It is on the contrary in weak NECV years. The anomalous SST in north Pacific in the previ-ous year may be a factor that results in the anomalous NECV at Meiyu period. The land-sea thermal contrast in summer facilitates NECV, while that in winter inhibits NECV. 2Molecular breeding of water lily: engineering cold stress tolerance into tropical water lily显示文摘Water lilies(order Nymphaeales)are rich in both economic and cultural values.They grow into aquatic herbs,and are divided into two ecological types:tropical and hardy.Although tropical water lilies have more ornamental and medicinal values compared to the hardy water lily,the study and utilization of tropical water lilies in both landscaping and pharmaceutical use is greatly hindered due to their limited planting area.Tropical water lilies cannot survive the winter in areas beyond 24.3°N latitude.Here,the transgenic pipeline through the pollen-tube pathway was generated for water lily for the first time.To improve cold stress tolerance of tropical water lilies,a gene encoding choline oxidase(CodA)driven by a cold stress-inducible promoter was transformed into a tropical water lily through the pollen-tube transformation.Six independent transgenic lines were tested for survival rate during two winter seasons from 2015 to 2017 in Hangzhou(30.3°N latitude).PCR and southern blot detection revealed that the CodA gene had been integrated into the genome.Reverse transcription PCR showed that CodA gene was induced after cold stress treatment,and further quantitative real-time PCR revealed different expressions among six 4 lines and line 3 had the highest expression.Multiple physiological experiments showed that after cold stress treatment,both the conductivity and malondialdehyde(MDA)levels from transgenic plants were significantly lower than those of non-transgenic plants,whereas the content of betaine and the activity of superoxide dismutase,catalase,and peroxidase were higher than those from non-transgenic plants.These results suggest that expression of exogenous CodA gene significantly improved the cold stress tolerance of tropical water lilies through a wide range of physiological alterations.Our results currently expanded a six-latitude cultivating area of the tropical water lilies.These results not only illuminate the bright future for water lily breeding but will also facilitate the functional genomic studies.Cuiwei Yu Guirong Qiao Wenmin Qiu Dongbei Yu Shirong Zhou Yan Shen Guanchun Yu Jing Jiang Xiaojiao Han Mingying Liu Liangsheng Zhang Fei Chen Yuchu Chen Renying Zhuo 2018Horticulture Research2018,5,1:4
3Nonuniform stretch processing for the range profile of a target with micro-motion显示文摘Complex motions such as micro-motions induce complex range-Doppler coupling effect in radar imaging. Hence, it is a challenge to reconstruct a clear range profile of a target with micro-motion. Stretch processing with fine motion compensation cannot perform well while it ties to eliminate the distortion of range profile caused by Doppler effect, because it is difficult to accurately estimate each scattering center velocity during coherent processing interval. Based on nonuniform stretch processing (NSP), a new technology is proposed to decouple range, time and Doppler and then achieve range profile of target with micro-motion. The new technology uses a new synthetic wide-band waveform and has an innovative mathematic interpretation. Statistical properties of the new approach are primarily examined. Typical models of target with micro-motion for high resolution radar are presented. 5PrP mutants with different numbers of octarepeat sequences are more susceptible to the oxidative stress显示文摘One of the physiological functions of cellular prion protein(PrP C )is believed to work as a cellular resistance to oxidative stress,in which the octarepeats region within PrP plays an important role.However,the detailed mechanism is less clear.In this study,the expressing plasmids of wild-type PrP (PrP-PG5)and various PrP mutants containing 0(PrP-PG0),9(PrP-PG9)and 12(PrP-PG12)octarepeats were generated and PrP proteins were expressed both in E.coli and in mammalian cells.Protein aggregation and formation of carbonyl groups were clearly seen in the recombinant PrPs expressed from E.coli after treatment of H2O2.MTT and trypan blue staining assays revealed that the cells expressing the mutated PrPs within octarepeats are less viable than the cells expressing wild-type PrP.Statistically significant high levels of intracellular free radicals and low levels of glutathione peroxidase were observed in the cells transfected with plasmids containing deleted or inserted octarepeats.Remarkably more productions of carbonyl groups were detected in the cells expressing PrPs with deleted and inserted octarepeats after exposing to H2O2.Furthermore,cells expressing wild-type PrP showed stronger resistant activity to the challenge of H2O2 at certain extent than the mutated PrPs and mock.
