| 2 | Advances in research into gamete and embryo-fetal origins of adult diseases显示文摘The fetal and infant origins of adult disease hypothesis proposed that the roots of adult chronic disease lie in the effects of adverse environments in fetal life and early infancy. In addition to the fetal period, fertilization and early embryonic stages, the critical time windows of epigenetic reprogramming, rapid cell differentiation and organogenesis, are the most sensitive stages to environmental disturbances. Compared with embryo and fetal development, gametogenesis and maturation take decades and are more vulnerable to potential damage for a longer exposure period. Therefore, we should shift the focus of adult disease occurrence and pathogenesis further back to gametogenesis and embryonic development events, which may result in intergenerational, even transgenerational, epigenetic re-programming with transmission of adverse traits and characteristics to offspring. | 4 | A combination of follicle stimulating hormone, estradiol and age is associated with the pregnancy outcome for women undergoing assisted reproduction: a retrospective cohort analysis显示文摘To investigate the effect of basal hormone levels including follicle stimulating hormone(FSH) and estradiol(E2) and age on the pregnancy outcomes in women undergoing in vitro fertilization and embryo transfer(IVF-ET), we designed a retrospective cohort analysis and collected information of patients undergoing IVF between January 2014 and April 2017. We found that young women had much better pregnancy outcomes than older women regardless of the hormone levels. The pregnancy outcomes declined in young women with high basal E2, even though they had normal FSH. Older women with higher FSH had a worse outcome of pregnancy, especially with increased E2 level. The results suggest that a combination of FSH, E2 and age could effectively predict the pregnancy outcome for women undergoing IVF-ET, and we should encourage infertile women with diminished ovarian reserve, especially young infertile women, to try the assisted reproduction as early as possible. | 5 | miR-28-5p targets MTSS1 to regulate cell proliferation and apoptosis in esophageal cancer显示文摘Esophageal cancer (EC) is one of the most common aggressive malignant diseases worldwide. miR-28-5p plays important regulatory roles in many cancers including human EC. However, the molecular mechanism and potential role of miR-28-5p in EC remain uncertain. In this study, qRT-PCR and western blot analysis revealed that miR-28-5p expression was up-regulated and metastasis suppressor-1 (MTSS1) was down-regulated in EC tissues relative to matched para-cancer tissues. Cell counting kit-8 (CCK-8) assay demonstrated that miR-28-5p mimics increased cell viability, and miR-28-5p inhibitor decreased it. Flow cytometry (FCM) assay indicated that miR-28-5p mimics promoted cell cycle entry, while miR-28-5p inhibitor reduced it and induced cell apoptosis. Moreover, miR-28-5p mimics up-regulated the expressions of cyclin A, cyclin dependent kinase 2 (CDK2), cyclin D1, and cyclin E but down-regulated the expressions of cleaved caspase-3 and cleaved caspase-9, which was abolished by miR-28-5p inhibitor. Furthermore, luciferase reporter assay verified that miR-28-5p directly targeted MTSS1 3′UTR and down-regulated its expression. MTSS1 overexpression in TE-1 cells inhibited cell proliferation and promoted apoptosis induced by miR-28-5p mimics, whereas silencing of MTSS1 reversed cell progression induced by miR-28-5p inhibitor. We also demonstrated that miR-28-5p could promote esophageal tumor formation in vivo. Hematoxylin–eosin staining, immunohistochemistry, and TUNEL assays confirmed that miR-28-5p antagomir inhibited cell growth and accelerated apoptosis. Our results suggest that miR-28-5p may induce cell proliferation and suppress apoptosis to promote EC tumor formation via decreasing MTSS1 expression. | 6 | Mechanism underlying the retarded nuclear translocation of androgen receptor splice variants显示文摘As shown in our previous study, two alternatively spliced androgen receptor(AR) variants, which are exclusively expressed in the granulosa cells of patients with polycystic ovary syndrome, exhibit retarded nuclear translocation compared with wild-type AR. However, researchers have not yet determined whether these abnormalities correlate with heat shock protein 90(HSP90)and importin α(the former is a generally accepted co-chaperone of AR, and the latter is a component of classical nuclear import complexes). Here, these two variants were mainly retained in cytoplasm with HSP90 and importin α in the presence of dihydrotestosterone(DHT), and their levels in nucleus were significantly reduced, according to the immunofluorescence staining. The binding affinity of two AR variants for importin α was consistently decreased, while it was increased in WT-AR following DHT stimulation, leading to reduced nuclear import, particularly for the insertion-AR(Ins-AR). However, the binding affinities of two AR variants for HSP90 were increased in the absence of DHT compared with WT-AR, which functioned to maintain spatial structural stability, particularly for the deletion-AR(Del-AR). | 18 | Study on the relationship between chronic tonsillitis and early stage of kidney failure显示文摘Objective: to investigate whether chronic tonsillitis is associated with early kidney failure;to analysis the reliability of urine microalbumin as a diagnostic indicator of chronic tonsillitis and early stage of kidney failure;either tonsillectomy is the effective way to prevent complications of chronic tonsillitis and early kidney failure. Methods: 84 patients with chronic tonsillitis who admitted to the department of Otorhinolaryngology, Qinghai University Affiliated Hospital from March 2012 to February 2013 were selected as subjects, 33 males and 51 females who aged between 7-55 years. It was divided into three group according to the course of disease, group A: duration is 1 to 3 years;group B: duration of 3 to 5 years;group C;duration> 5 years;in the immunological laboratory, 12 cases of normal control group were collected, male 7 cases , female 5 cases, aged 7 to 55 years old. The amount of microalbuminuria before and after operation was determined by turbidimetric method. The collected data were analyzed statistically. Results: There was no significant increase in urine microalbumin compared group A and control group, P> 0.05, the difference was not statistically significant. Group B and group C were significantly higher than those in the control group (P <0.05), the difference was statistically significant. The levels of microalbuminuria in group A pre-operation was significantly higher than that of in group C (P <0.05). The difference was statistically significant. The levels of microalbuminuria in group B were significantly higher than those in group C (P <0.05) during preoperation. The difference was statistically significant. The levels of microalbuminuria were significantly decreased in group A post-operation compared to the relatively group pre-operation, P <0.05, the difference was statistically significant. In group B, pre-operative and postoperative comparison, urine microalbumin was significantly decreased, P <0.05, the difference was statistically significant. The levels of microalbuminuria were significantly decreased in post-operative in group C than that of its pre-operation, P <0.05, the difference was statistically significant. Conclusion: Patients with chronic tonsillitis can be complicated by early stage of kidney failure. There is a certain clinical value of urine microalbumin used as a diagnostic indicator of chronic tonsillitis and early kidney failure, and its sensitivity gradually increased with the progress of disease.
