1Wound Healing Assay Using Microfluidic Channels显示文摘An approach was described to produce a wound area in vitro using laminar flow technique to selectively remove cells in microfluidic channels.Cell migration which plays an important role in the process of wound healing can be observed dynamically by this method.In order to prove our system,we have studied its properties by injecting NIH-3T3 cells into main channel and added several reagents to observe the mediatory influence.The results reveal that fibroblast growth factor-2(FGF-2),insulin-like growth factor 1(IGF-I),platelet-derived growth factor BB(PDGF-BB) and ascorbic acid(Vc) groups can promote cell migration compared with the control group.But the average migration distance was diminished in dexamethasone group.It can be concluded that this system can be used to analyze the process of wound healing.XI Yan-li WANG Shu-tao SU Bin HA Qing NIU Feng-lan NIE Fu-qiang 2012Chemical Research in Chinese Universities2012,28,3:2

3Hepatocyte growth factor gene transfer effects on the femoral and intramuscular nerve in a canine model of lower limb ischemia显示文摘BACKGROUND: Recent advancements in gene therapy have provided new methodology for treating ischemia in lower extremities. Gene transfer of angiogenic factors to ischemic tissues may promote local proliferation of new vessels and form collateral circulation. OBJECTIVE: To observe histopathological changes in the femoral and intramuscular nerve three months after intramuscular injection of hepatocyte growth factor (HGF) into the peripheral skeletal muscle in a canine model of lower limb ischemia. DESIGN: Randomized occlusion modelled and verification animal study. SETTING: Experimental Center, Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA. MATERIALS: This study was performed at Animal Experimental Center, Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA from September to November 2006. A total of eight male mongrel dogs, weighing 12–15 kg and 1.5–3 years of age, were selected for this study. This experimental study was in accordance with local ethics standards. Recombinant plasmid carrying HGF (pUDKH) and occlusion model plasmid (pUDK) were provided by the Third Laboratory of Radiation Medical Institute, Academy of Military Medical Sciences of PLA. METHODS: Grouping and model establishment: under anesthesia, complete vascular occlusion models were established on the left lower extremities. The experimental dogs were randomly divided into a model group and a pUDKH treatment group, with four dogs in each group. Dogs in the pUDKH group were injected with 0.15 mg/kg pUDKH. Ten minutes later, intramuscular injections were performed at three spots into the peripheral skeletal muscle of the left hind limb, as well as lateral injections at two spots. The injection volume at each spot was 0.2 mL. Dogs in the model group were injected with pUDK, and dosage and injection method were identical to the treatment group. MAIN OUTCOME MEASURES: Histopathological changes in the femoral nerve, as well as internal and external intramuscular nerve tissues in the hind limb of dogs three months after plasmid injection under optic microscope. RESULTS: (1) Histopathological changes in the femoral nerve: tiny nerves from the femoral nerve to the intramuscular nerve exhibited marked degeneration in the model group. The degenerating features included neurites, myelin sheaths, and Schwann cell nuclei. Neuropathy in the pUDKH treatment group was not detected. (2) Histopathological changes of the intramuscular nerve: large and irregular vacuoles were present on several longitudinal sections of intramuscular nerve fibers in the model group, as well as annular-shaped blank regions on transverse sections of peripheral neurites. In the pUDKH treatment group, large, blank regions were present in several segments of partial nerve fibers of the longitudinal intramuscular nerve region, but only a few nerve fibers exhibited annular-shaped blank regions on the transverse section of peripheral neurites. 12Inner Co Synergizing Outer Ru Supported on Carbon Nanotubes for Efficient pH-Universal Hydrogen Evolution Catalysis显示文摘Exploring highly active but inexpensive electrocatalysts for the hydrogen evolution reaction(HER)is of critical importance for hydrogen production from electrochemical water splitting.Herein,we report a multicomponent catalyst with exceptional activity and durability for HER,in which cobalt nanoparticles were in-situ confined inside bamboo-like carbon nanotubes(CNTs)while ultralow ruthenium loading(~2.6μg per electrode area~cm^(−2))is uniformly deposited on their exterior walls(Co@CNTsǀRu).The atomic-scale structural investigations and theoretical calculations indicate that the confined inner Co and loaded outer Ru would induce charge redistribution and a synergistic electron coupling,not only optimizing the adsorption energy of H intermediates(ΔGH*)but also facilitating the electron/mass transfer.The as-developed Co@CNTsǀRu composite catalyst requires overpotentials of only 10,32,and 63 mV to afford a current density of 10 mA cm^(−2) in alkaline,acidic and neutral media,respectively,representing top-level catalytic activity among all reported HER catalysts.The current work may open a new insight into the rational design of carbon-supported metal catalysts for practical applications.Jian Chen Yuan Ha Ruirui Wang Yanxia Liu Hongbin Xu Bin Shang Renbing Wu Hongge Pan 2022Nano-Micro Letters2022,14,11:1
13Human Papillomavirus 16E6 Oncogene Mutation in Cervical Cancer显示文摘Objective:Cervical cancer(CC) is the second most common type of cancer in women worldwide,after breast cancer.High-risk human papillomaviruses(HR-HPVs) are considered to be the major causes of cervical cancer.HPV16 is the most common type of HR-HPVs and HPV16 E6 gene is one of the major oncogenes.Specific mutations are considered as dangerous factors causing CC.This study was designed to find mutations of HPV16 E6 and the relationship between the mutations and the happening of CC.Methods:The tissue DNA was extracted from 15 biopsies of CC.Part of HPV16 E6 gene(nucleotide 201-523) was amplified by polymerase chain reaction(PCR) from the CC tissue DNA.The PCR fragments were sequenced and analyzed.Results:The result of PCR showed that the positive rate of HPV16 E6 was 93.33%(14/15).After sequencing and analyzing,in the 13 out of 14 PCR fragments,4 maintained prototype(30.77%),8 had a same 350G mutation(61.54%),and 1 had a 249G mutation(7.69%).Conclusion:This study suggest that there is a high infection rate of HPV in cervical cancer and most of the HPV16 E6 gene has mutations.Those mutations may have an association with the development of cervical cancer.Feng Sun Xiao-qin Ha Tong-de Lv Chuan-ping Xing Bin Liu Xiao-zhe Cao 2009Chinese Journal of Cancer Research2009,21,2:1

16Low-temperature synthesis of Fe_(2)(MoO_(4))_(3) nanosheets: A cathode for sodium ion batteries with kinetics enhancement显示文摘Sodium ion batteries (SIBs) are alternatives to lithium ion batteries (LIBs), and offer some significant benefits such as cost reduction and a lower environmental impact;however, to compete with LIBs, further research is required to improve the performance of SIBs. In this study, an orthorhombic Na super ionic conductor structural Fe_(2)(MoO_(4))_(3) nanosheet with amorphous-crystalline core-shell alignment was synthesized using a facile low-temperature water-vapor-assisted solid-state reaction and applied as a cathode material for SIBs. The obtained material has a well-defined three-dimensional stacking structure, and exhibits a high specific capacity of 87.8 mAh·g^(−1) at a current density of 1 C = 91 mA·g^(−1) after 1,000 cycles, which is due to the considerable contribution of extra surface-related reaction such as the pseudo-capacitive process. This material shows significantly improved cycling and rated behavior as well as enhanced performance under high- and low-temperature conditions, as compared to the same materials prepared by the conventional high-temperature solid-state reaction. 17母亲颗粒物空气污染暴露与足月出生体重:效应与异质性的多国评估显示文摘[背景]越来越多的证据显示,母亲空气污染暴露与对胎儿生长的不良影响之间存在关联;但是,现有的文献并不一致。[目的]研究9个国家14个中心的数据,量化母亲颗粒物空气污染暴露与足月出生体重和低出生体重(LBW)之间的关联,并探讨此关联中地点特征和暴露评估方法对中心之间异质性的影响。[方法]空气污染与妊娠结局国际合作(ICAPPO)的各中心采用共同的分析方案,对PM10和PM2.5(颗粒物≤10和2.5μm)与足月LBW和连续出生体重进行效应评估。采用meta分析将各个中心(~300万例出生)的效应评估结合起来,并采用meta回归分析在报告的效应评估中,中心特征和暴露评估方法对中心之间异质性的影响。[结果]随机效应meta分析显示,调整母亲的社会经济地位后,在整个妊娠期间,足月LBW与PM10[比值比(OR)=1.03;95%CI:1.01~1.05]和PM2.5[OR=1.10;95%CI:1.03~1.18]每增加10μg/m3呈正相关。在充分调整随机效应的meta分析中,作为一个连续性的结果,PM10暴露每增加10μg/m3与足月出生体重呈负相关(-8.9g;95%CI:-13.2^-4.6 g)。meta回归分析显示,具有较高平均PM2.5水平和PM2.5∶PM10比率的中心,以及采用时间暴露评估(与时空相比)的中心,所报告的关联更强。[结论]在研究人群中,母亲颗粒物污染暴露与足月LBW相关联。有3个地点特征和暴露评估方法学方面的不同导致各个中心报告的关联的差异。Payam Dadvand Jennifer Parker Michelle L. 18Groundwater status and associated issues in the Mekong-Lancang River Basin: International collaborations to achieve sustainable groundwater resources显示文摘Groundwater is an important and readily available source of fresh water in the Mekong-Lancang River Basin. With a rapid population growth and increasing human activities, an increasing number of countries in the Mekong-Lancang River Basin are experiencing depleted and degraded groundwater supplies. In transboundary river basins, such as the Mekong-Lancang River, prioritizing the use of the shared aquifer by one riparian government may affect the opportunities of other riparian governments and lead to potential water conflicts between neighboring countries. To promote the sharing of strategies and information for the sustainable and equitable use of water resources of the shared basin, international collaborative workshops on groundwater resources have been organized for all Mekong-Lancang River countries. These workshops provide an opportunity to communicate and discuss nationally sensitive issues on groundwater by the associated countries, with topics covering multiple aspects of groundwater, such as the groundwater status in the basin, quality issues, water use conflicts, hydrological information gaps, management policies and capacity building for successful water resource management. Consensus has been reached by all countries on the importance of catchment-based groundwater management and the need for close communication among the countries. Strategies for managing transboundary aquifer issues must foster international collaboration based on the regional network, influence national networks and enhance the capacity to building maps and monitoring systems based on associated databases. The sustainability of water resources cannot be achieved without the integrated involvement and contributions by multiple countries and various stakeholders. 19Lead acetate in drinking water is toxic to hippocampal tissue Measuring relative protein changes using tissue array detection显示文摘BACKGROUND: Lead can cause structural changes in the hippocampus, followed by damage to learning and memory functions, but its specific mechanisms are not yet clear. OBJECTIVE: To observe long-term toxicity of high-dose lead in drinking water on hippocampal tissue in rats, and analyze the potential association of oxidative damage, cell apoptosis, and pathology. DESIGN, TIME AND SETTING: A randomized, controlled animal experiment was performed at the Center for Medical Experiment, Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA from May 2007 to October 2008. MATERIALS: Rabbit anti Bcl-2, Bax, and inducible nitric oxide synthase (iNOS) polyclonal antibodies were purchased from Santa Cruz Biotechnology, USA. An streptavidin-peroxidase immunohistochemistry kit and concentrated DAB kit were purchased from Beijing Zhongshan Biotechnology Company Limited, China. Crystal violet was purchased from Sigma, USA. METHODS: A total of 72 Wistar rats, aged 3 months, were randomly divided into control, low-, middle-, and high-dose lead groups, with 18 rats per group. Animal models were established through free drinking water contaminated by Pb2+ for 1, 3, and 6 months, respectively. MAIN OUTCOME MEASURES: The general toxicity of lead was dynamically observed; the levels of Pb2+ in the blood and brain tissue homogenate were detected using atomic absorption method; pathological changes were observed using hematoxylin-eosin staining and tigroid body staining; the protein expression levels of Bcl-2, Bax, and iNOS were dynamically observed using streptavidin-peroxidase immunohistochemistry of the hippocampus. RESULTS: Lead exposure reduced autonomic activities, produced a slumped appearance, slow responses, and lusterless fur, especially in the high-dose group. The amount of ingestion and hydroposia showed a decreasing trend, especially in middle- and high-dose groups. Lead levels in whole blood and brain homogenate were higher than controls (P < 0.01). Lead caused degeneration of hippocampal neurons and pyknosis, with fewer tigroid bodies, especially in high-dose lead group. Bcl-2 expression decreased with increasing lead dose (P < 0.01), whereas lead dose-dependently increased Bax levels (P < 0.01) and iNOS levels (P < 0.05). CONCLUSION: High levels of Pb2+ may disrupt hippocampal structure by passing through the blood brain barrier.
