1Acute complications of spinal cord injuries显示文摘The aim of this paper is to give an overview of acute complications of spinal cord injury(SCI). Along with motor and sensory deficits, instabilities of the cardiovascular, thermoregulatory and broncho-pulmonary system are common after a SCI. Disturbances of the urinary and gastrointestinal systems are typical as well as sexual dysfunction. Frequent complications of cervical and high thoracic SCI are neurogenic shock, bradyarrhythmias, hypotension, ectopic beats, abnormal temperaturecontrol and disturbance of sweating, vasodilatation and autonomic dysreflexia. Autonomic dysreflexia is an abrupt, uncontrolled sympathetic response, elicited by stimuli below the level of injury. The symptoms may be mild like skin rash or slight headache, but can cause severe hypertension, cerebral haemorrhage and death. All personnel caring for the patient should be able to recognize the symptoms and be able to intervene promptly. Disturbance of respiratory function are frequent in tetraplegia and a primary cause of both short and long-term morbidity and mortality is pulmonary complications. Due to physical inactivity and altered haemostasis, patients with SCI have a higher risk of venous thromboembolism and pressure ulcers. Spasticity and pain are frequent complications which need to be addressed. The psychological stress associated with SCI may lead to anxiety and depression. 2Ephrin-B2 is differentially expressed in the intestinal epithelium in Crohn's disease and contributes to accelerated epithelial wound healing in vitro显示文摘AIM: Eph receptor tyrosine kinases and their membrane bound receptor-like ligands, the ephrins, represent a bi-directional cell-cell contact signaling system that directs epithelial movements in development. The meaning of this system in the adult human gut is unknown. We investigated the Eph/ephrin mRNA expression in the intestinal epithelium of healthy controls and patients with inflammatory bowel disease (IBD).METHODS: mRNA expression profiles of all Eph/ephrin family members in normal small intestine and colon were established by real-time RT-PCR. In addition, differential expression in IBD was investigated by cDNA array technology, and validated by both real-time RT-PCR and immunohistochemistry. Potential effects of enhanced EphB/ephrin-B signaling were analyzed in an in vitro IEC-6 cell scratch wound model.RESULTS: Human adult intestinal mucosa exhibits a complex pattern of Eph receptors and ephrins. Beside the known prominent co-expression of EphA2 and ephrinA1,we found abundantly co-expressed EphB2 and ephrin-B1/2.Interestingly, cDNA array data, validated by real-time PCR and immunohistochemistry, showed upregulation of ephrin-B2 in both perilesional and lesional intestinal epithelial cells of IBD patients, suggesting a role in epithelial homeostasis. Stimulation of ephrin-B signaling in ephrinB1/2 expressing rat IEC-6-cells with recombinant EphB1Fc resulted in a significant dose-dependent acceleration of wound closure. 3Associations between the physical environment and park-based physical activity:A systematic review显示文摘Background:With an increase in the evidence for the associations between park-based physical activity(PA)and physical environments(especially park and neighborhood environments),researchers face an important challenge in interpreting and summarizing the evidence to develop environ?mental change interventions.An updated review is needed to better inform policymaking and environmental interventions.The current study aimed to systematically review the research on the associations of park-based PA with park and neighborhood environmental characteristics.Methods:We targeted English peer-reviewed articles from 5 electronic databases using keywords related to park-based PA,park environments,and neighborhood environments.Of the 4071 identified papers,25 studies published between 2008 and 2016 met all the eligibility criteria and were included in this review.Results:The characteristics of physical environment that received consistent support included paths/trails,lighting,and incivilities(e.g.,broken glasses and litter).Mixed findings were revealed for 6 park environmental factors(unspecified active facilities,playgrounds and skating areas,fit?ness stations,picnic areas,greenness,and park size)and 2 neighborhood environmental factors(park density and park proximity).Conclusion:It can be concluded that paths/trails,lighting,and incivilities are 3 key physical environmental attributes of park-based PA.Given the inconsistent findings on park and neighborhood environmental factors,more robust designs such as prospective investigation are required.Ru Zhang Hagen Wulff Yanping Duan Petra Wagner 2019Journal of Sport and Health Science2019,8,5:7
4Wound-Induced Shoot-to-Root Relocation of JA-Ile Precursors Coordinates Arabidopsis Growth显示文摘Multicellular organisms rely on the movement of signaling molecules across cells,tissues,and organs to communicate among distal sites.In plants,localized leaf damage activates jasmonic acid(JA)-dependent transcriptional reprogramming in both harmed and unharmed tissues.Although it has been indicated that JA species can translocate from damaged into distal sites,the identity of the mobile compound(s),the tissues through which they translocate,and the effect of their relocation remain unknown.Here,we found that following shoot wounding,the relocation of endogenous jasmonates through the phloem is essential to initiate JA signaling and stunt growth in unharmed roots of Arabidopsis thaliana.By employing grafting experiments and hormone profiling,we uncovered that the hormone precursor cis-12-oxo-phytodienoic acid(OPDA)and its derivatives,but not the bioactive JA-Ile conjugate,translocate from wounded shoots into undamaged roots.Upon root relocation,the mobile precursors cooperatively regulated JA responses through their conversion into JA-Ile and JA signaling activation.Collectively,our findings demonstrate the existence of long-distance translocation of endogenous OPDA and its derivatives,which serve as mobile molecules to coordinate shoot-to-root responses,and highlight the importance of a controlled redistribution of hormone precursors among organs during plant stress acclimation.Adina Schulze Marlene Zimmer Stefan Mielke Hagen Stellmach Charles WMelnyk Bettina Hause Debora Gasperini 2019Molecular Plant2019,12,10:7
5Tight junction disruption: Helicobacter pylori and dysregulation of the gastric mucosal barrier显示文摘Long-term chronic infection with Helicobacter pylori(H. pylori) is a risk factor for gastric cancer development. In the multi-step process that leads to gastric cancer,tight junction dysfunction is thought to occur and serve as a risk factor by permitting the permeation of luminal contents across an otherwise tight mucosa. Mechanisms that regulate tight junction function and structure in the normal stomach,or dysfunction in the infected stomach,however,are largely unknown. Although conventional tight junction components are expressed in gastric epithelial cells,claudins regulate paracellular permeability and are likely the target of inflammation or H. pylori itself. There are 27 different claudin molecules,each with unique properties that render the mucosa an intact barrier that is permselective in a way that is consistent with cell physiology. Understanding the architecture of tight junctions in the normal stomach and then changes that occur during infection is important but challenging,because most of the reports that catalog claudin expression in gastric cancer pathogenesis are contradictory. Furthermore,the role of H. pylori virulence factors,such as cytotoxin-associated gene A and vacoulating cytotoxin,in regulating tight junction dysfunction during infection is inconsistent in different gastric cell lines and in vivo,likely because non-gastric epithelial cell cultures were initially used to unravel the details of their effects on the stomach. Hampering further study,as well,is the relative lack of cultured cell models that have tight junction claudins that are consistent with native tissues. 6Effects of insulin and pathway inhibitors on the PI3K-Akt- mTOR phosphorylation profile in acute myeloid leukemia cells显示文摘The phosphatidylinositol 3-kinase(PI3K)-Akt-mechanistic target of rapamycin(mTOR)pathway is constitutively activated in human acute myeloid leukemia(AML)cells and is regarded as a possible therapeutic target.Insulin is an agonist of this pathway and a growth factor for AML cells.We characterized the effect of insulin on the phosphorylation of 10 mediators in the main track of the PI3K-Akt-mTOR pathway in AML cells from 76 consecutive patients.The overall results showed that insulin significantly increased the phosphorylation of all investigated mediators.However,insulin effects on the pathway activation profile varied among patients,and increased phosphorylation in all mediators was observed only in a minority of patients;in other patients,insulin had divergent effects.Global gene expression profiling and proteomic/phosphoproteomic comparisons suggested that AML cells from these two patient subsets differed with regard to AML cell differentiation,transcriptional regulation,RNA metabolism,and cellular metabolism.Strong insulin-induced phosphorylation was associated with weakened antiproliferative effects of metabolic inhibitors.PI3K,Akt,and mTOR inhibitors also caused divergent effects on the overall pathway phosphorylation profile in the presence of insulin,although PI3K and Akt inhibition caused a general reduction in Akt pT308 and 4EBP1 pT36/pT45 phosphorylation.For Akt inhibition,the phosphorylation of upstream mediators was generally increased or unaltered.In contrast,mTOR inhibition reduced mTOR pS2448 and S6 pS244 phosphorylation but increased Akt pT308 phosphorylation.In conclusion,the effects of both insulin and PI3K-Akt-mTOR inhibitors differ between AML patient subsets,and differences in insulin responsiveness are associated with differential susceptibility to metabolic targeting.Ina Nepstad Kimberley Joanne Hatfield Ida Sofie Grønningsæter Elise Aasebø Maria Hernandez-Valladares Karen Marie Hagen Kristin Paulsen Rye Frode SBerven Frode Selheim Håkon Reikvam Øystein Bruserud 2019Signal Transduction and Targeted Therapy2019,4,1:4

10Epiphytic orchids and their ecological niche under anthropogenic influence in central Himalayas,Nepal显示文摘The survival chance of epiphytic orchids today not only depends on the natural site conditions required by the orchids but also on anthropogenic changes in site conditions.This study answers two questions:(1) What is the ecological niche of the different epiphytic orchid species?(2) What are the ecological factors that threaten epiphytic orchid's population under anthropogenic disturbances? Our study area was the Kathmandu valley,Nepal,with its subtropical forest.We established 156 systematically selected sampling points in the Kathmandu area covering different types of ecosystems under human impacts such as densely populated area,agricultural land,mixed agricultural and settled area,old tree patches,and a natural forest in a national park.The ecological niche of the orchid species was analyzed with a principal component analysis(PCA).The correlations between the different site factors were statistically significant.Spearman's rank correlation matrices showed that the variables land-use intensities with altitude,and height with diameter in breast height(dbh) of host had the highest significant positive correlation coefficient(0.67 and 0.64 respectively).On the other hand,host bark p H and altitude as well as land use had a significantly strong negative correlation coefficient(-0.80 and-0.61,respectively).Different epiphytic orchid species interact differently with the given set ofenvironmental factors:for occurrence of Vanda cristata there is no single environmental factor of special influence,while for Rhynchostylis retusa high bark p H and high light availability are important.First two axis of the PCA explained more than 50% of the total variance.Most orchid species occupy a specific,narrow niche in this ecological space.The main causes of anthropogenic influence of orchid population in the Kathmandu Valley are loss of adequate host trees(species and size) and increasing air pollution,resulting in increasing host bark p H.ADHIKARI Yagya Prasad FISCHER Anton FISCHER Hagen Siegfried 2016Journal of Mountain Science2016,13,5:3
11Techno-economic comparison of three technologies for precombustion CO2 capture from a lignite-fired IGCC显示文摘This paper compares the techno-economic performances of three technologies for CO2 capture from a lignite-based IGCC power plant located in the Czech Republic:(1)Physical absorption with a Rectisol-based process;(2)Polymeric CO2-selective membrane-based capture;(3)Low-temperature capture.The evaluations show that the IGCC plant with CO2 capture leads to costs of electricity between 91 and 120€·MWh−1,depending on the capture technology employed,compared to 65€·MWh−1 for the power plant without capture.This results in CO2 avoidance costs ranging from 42 to 84€·tCO2,avoided−1,mainly linked to the losses in net power output.From both energy and cost points of view,the low-temperature and Rectisol based CO2 capture processes are the most efficient capture technologies.Furthermore,partial CO2 capture appears as a good mean to ensure early implementation due to the limited increase in CO2 avoidance cost when considering partial capture.To go beyond the two specific CO2-selective membranes considered,a cost/membrane property map for CO2-selective membranes was developed.This map emphasise the need to develop high performance membrane to compete with solvent technology.Finally,the cost of the whole CCS chain was estimated at 54€·tCO2,avoided−1 once pipeline transport and storage are taken into consideration.Simon Roussanaly Monika Vitvarova Rahul Anantharaman David Berstad Brede Hagen Jana Jakobsen Vaclav Novotny Geir Skaugen 2020Frontiers of Chemical Science and Engineering2020,14,3:3

18Ephrin-B reverse signaling induces expression of wound healing associated genes in IEC-6 intestinal epithelial cells显示文摘AIM: Eph receptors and ephrin ligands play a pivotal role in development and tissue maintenance. Since previous data have indicated an involvement of ephrin-B2 in epithelial healing, we investigated the gene expression and downstream signaling pathways induced by ephrin-B mediated cell-cell signaling in intestinal epithelial cells.METHODS: Upon stimulation of ephrin-B pathways in IFC-6 cells with recombinant rat EphB1-Fc, gene expression was analyzed by Affymetrix(R) rat genome 230 high density arrays at different time points. Differentially expressed genes were confirmed by real-time RT-PCR. In addition, MAP kinase pathways and focal adhesion kinase (FAK) activation downstream of ephrin-B were investigated by immunoblotting and fluorescence microscopy.RESULTS: Stimulation of the ephrin-B reverse signaling pathway in IEC-6 cells induces predominant expression of genes known to be involved into wound healing/cell migration, antiapoptotic pathways, host defense and inflammation.
