| 1 | Current developments in pharmacological therapeutics for chronic constipation显示文摘Chronic constipation is a common gastrointestinal disease severely affecting the patient's quality of life. The traditional treatment of constipation is the use of laxatives. Recently, several new drugs including lubiprostone, linaclotide and prucalopride have been approved for treatment of chronic constipation. However, a significant unmet medical need still remains, particularly among those patients achieving poor results by current therapies. The 5-HT4 receptor modulators velusetrag and naronapride,the guanylate cyclase C agonist plecanatide and the ileal bile acid transporter inhibitor elobixibat are recognized as the most promising drugs under investigation. | 2 | Predicting plant traits and functional types response to grazing in an alpine shrub meadow on the Qinghai-Tibet Plateau显示文摘The identification of easily measured plant functional types (PFTs) that consistently predict grazing response would be a major advance.The responses to grazing of individual traits and PFTs were analyzed along a grazing gradient in an alpine shrub meadow on the Qinghai-Tibet Plateau,China.Three response types were identified;grazing increaser (GI),grazing decreaser (GD),and neutral (NE) for both traits and PFTs.Seven traits were measured:plant height,economic group,cotyledon type,plant inclination,growth form,life cycle,and vegetative structure.The first five were significantly affected by grazing.Ordinal regressions for grazing response of the seven traits showed that the best single predictors of response were growth form (including the attributes 'Scattered','Bunched' or 'Closely Bunched'),and plant inclination ('Rosette','Prostrate',or 'Erect'),followed by economic group ('Shrub','Grass','Sedge','Legume','Forb',or 'Harmful') and plant height ('Tall','Medium',or 'Small').Within the four optimal traits,the summed dominance ratio (SDR) of small plants,forbs,rosette and bunched plants,invariably increased,while that of tall plants,shrubs,grasses,and erect plants decreased,when grazing pressure was enhanced.Canonical correspondence analysis (CCA) identified eleven explanatory PFTs based on 195 defined PFTs,by combining the different attributes of the four optimal traits.Among explanatory PFTs,the most valuable in predicting the community response to grazing were Tall×Shrub×Erect×Scattered and Small×Forb×Rosette,as these have the closest connections with grazing disturbance and include fewer species.Species richness,diversity,and community evenness,did not differ among grazing treatments because turnover occurred in component species and their relative abundances along the grazing gradient.We have demonstrated that a minimum set of PFTs resulting from optimal individual traits can provide consistent prediction of community responses to grazing in this region.This approach provides a more accurate indicator of change within a changing environment than do univariate measures of species diversity.We hope to provide a link between management practices and vegetation structure,forming a basis for future,large scale,plant trait comparisons. | ZHU ZhiHong WANG XiaoAn LI YingNian WANG Gang GUO Hua | 2012 | Science China Earth Sciences2012,55,5: | 14 |
| 3 | Scale dependence of plant species richness and vegetation-environment relationship along a gradient of dune stabilization in Horqin Sandy Land, Northern China显示文摘Ecological patterns and processes in dune ecosystems have been a research focus in recent years, however the information on how dune stabilization influences the spatial scale dependence of plant diversity is still lacking. In this study, we measured the plant species richness, soil properties and altitude across four spatial scales(1, 10, 100 and 1,000 m2) at three different dune stabilization stages(mobile dune, semi-fixed dune and fixed dune) in Horqin Sandy Land, Northern China. We also examined the relationships between plant species richness, community composition and environmental factors along the gradient of dune stabilization. Our results showed that plant species richness increased with the increase of spatial scales in each dune stabilization stage, as well as with the increase of dune stabilization degrees. Canonical correspondence analysis(CCA) showed that plant distribu- tions in the processes of dune stabilization were determined by the combined environmental gradient in relation to soil organic carbon(SOC), total nitrogen(TN), carbon/nitrogen(C/N), pH, electrical conductivity(EC), soil water content(SWC), fine sand(FS), very fine sand(VFS), silt and clay(SC), and altitude. Plant species richness was significantly and positively correlated to SOC and TN in mobile dune, and significantly and positively correlated to SOC, TN, C/N, VFS and SC in semi-fixed dune. However, no significant correlation between plant species richness and environmental factors was observed in fixed dune. In addition, plant species richness in different dune stabili- zation stages was also determined by the combined gradient of soil properties and altitude. These results suggest that plant species richness has obvious scale dependence along the gradient of dune stabilization. | 4 | Hypoxia-induced factor-1 alpha upregulates vascular endothelial growth factor C to promote lymphangiogenesis and angiogenesis in breast cancer patients显示文摘Hypoxia-induced factor-1 alpha(HIF-1α)affects many effector molecules and regulates tumor lymphangiogenesis and angiogenesis during hypoxia.The aim of this study was to investigate the role of HIF-1αin the regulation of vascular endothelial growth factor C(VEGF-C)expression and its effect on lymphangiogenesis and angiogenesis in breast cancer.Lymphatic vessel density(LVD),microvessel density(MVD)and the expressions of HIF-1αand VEGF-C proteins were evaluated by immunohistochemistry in 75 breast cancer samples.There was a significant correlation between HIF-1αand VEGF-C(P=0.014,r=0.273,Spearman's coefficient of correlation).HIF-1αand VEGF-C overexpression was significantly correlated with higher LVD(P=0.003 and P=0.017,respectively),regional lymph nodal involvement(P=0.002 and P=0.004,respectively)and advanced tumor,node,metastasis(TNM)classification(P=0.001 and P=0.01,respectively).Higher MVD was observed in the group expressing higher levels of HIF-1αand VEGF-C(P=0.033 and P=0.037,respectively).Univariate analysis showed shorter survival time in patients expressing higher levels of HIF-1αand VEGF-C.HIF-1αwas also found to be an independent prognostic factor of overall survival in multivariate analysis.The results suggest that HIF-1αmay affect VEGF-C expression,thus acting as a crucial regulator of lymphangiogenesis and angiogenesis in breast cancer.This study highlights promising potential of HIF-1αas a therapeutic target against tumor lymph node metastasis. | Xiaojian Ni Yingchun Zhao Jingjing Ma Tiansong Xia Xiaoan Liu Qiang Ding Xiaoming Zha Shui Wang | 2013 | The Journal of Biomedical Research2013,27,6: | 10 |
| 5 | Effect of spatial scale and topography on spatial heterogeneity of soil seed banks under grazing disturbance in a sandy grassland of Horqin Sand Land, Northern China显示文摘Soil seed banks play an important role in the distribution and composition of plant communities in semiarid grassland ecosystems.However,information on how spatial scale influences the spatial heterogeneity of soil seed banks in a grassland under grazing disturbance is still lacking.Based on field sampling and greenhouse germination,we measured the species composition and seed density of soil seed banks at different spatial scales (30 m×30 m,30 m×60 m and 30 m×90 m)along a topographical gradient in a sandy grassland in Horqin Sand Land, Northern China.By applying geostatistical methods,we examined how spatial scale and topography affected the spatial distribution of soil seed banks in the study area.Our results showed that the total number of species in soil seed banks,as well as the number of dominant annuals,increased with the increase of spatial scales.Seed density in soil seed banks decreased with the increase of spatial scales due to an increase in the slopes and relative heights of the sampling points.Geostatistical analysis showed that the relative structural variance(C/(C0+C))of seed density and species richness were over 65%for all spatial scales,indicating that these variables had an obvious spatial autocorrelation and the spatial structured variance accounted for the largest proportion of the total sample variance.Spatial autocorrelation of seed density in soil seed banks increased with the increase of measured scales,while that of species richness showed a reverse trend.These results suggest that the total number of species in soil seed banks is spatial scale dependent and lower topography may accommodate more seeds.Spatial distribution of seed density in soil seed banks is also scale dependent due to topographic variation.Grassland management,therefore,needs to consider local grazing disturbance regime,spatial scale and topography. | XiaoAn ZUO ShaoKun WANG XueYong ZHAO WenJin LI Johannes KNOPS Amy KOCHSlEK | 2012 | Journal of Arid Land2012,4,2: | 9 |

| 7 | Analytical geometry method of planetary gear trains显示文摘This paper presents an analytical geometry method for kinematics and efficiency of planetary gear trains (PGTs). The novel method which is capable of evolution and contrast analysis of mechanism kinematics, can be applied to any typical one-and two-degree-of-freedom plane PGTs containing any number of simple, compound or complex-compound planetary gear sets. The efficiency analysis of this method features a systematized and programmed process and its independence of the speed ratio. The primary contribution of this work lies in the integration of quantitative calculation, qualitative evolution and comparative analysis of kinematics of PGTs into one diagram, and in the integration of kinematics and efficiency analysis into a single method system. First, the analytical geometry method is defined, its basic properties are given, and the systematization procedure to perform kinematic analysis is demonstrated. As an application, analytical geometry diagrams of common PGTs are exhibited in the form of a list, whose kinematic characteristics and general evolution tendency are discussed. Then, with the mapping of PGTs onto the angular speed plane, the efficiency formula of analytical geometry, which has an extremely concise form, and a simple method for power flow estimation are put forward. Moreover, a general procedure is provided to analyze the efficiency and power flow. | 8 | Synergistic antitumor activity of artesunate and HDAC inhibitors through elevating heme synthesis via synergistic upregulation of ALAS1 expression显示文摘Artemisinin and its derivatives(ARTs) were reported to display heme-dependent antitumor activity. On the other hand, histone deacetylase inhibitors(HDACi) were known to be able to promote heme synthesis in erythroid cells. Nevertheless, the effect of HDACi on heme homeostasis in nonerythrocytes remains unknown. We envisioned that the combination of HDACi and artesunate(ARS)might have synergistic antitumor activity through modulating heme synthesis. In vitro studies revealed that combination of ARS and HDACi exerted synergistic tumor inhibition by inducing cell death. Moreover, this combination exhibited more effective antitumor activity than either ARS or HDACi monotherapy in xenograft models without apparent toxicity. Importantly, mechanistic studies revealed that HDACi coordinated with ARS to increase 5-aminolevulinate synthase(ALAS1) expression, and subsequent heme production, leading to enhanced cytotoxicity of ARS. Notably, knocking down ALAS1 significantly blunted the synergistic effect of ARS and HDACi on tumor inhibition, indicating a critical role of ALAS1 upregulation in mediating ARS cytotoxicity. Collectively, our study revealed the mechanism of synergistic antitumor action of ARS and HDACi. | 10 | Land cover changes and the effects of cultivation on soil properties in Shelihu wetland,Horqin Sandy Land,Northern China显示文摘Land cover change plays an essential role in the alternation of soils properties.By field investigation and applying satellite images,land cover information in the Shelihu wetland was carried out in an area of 2,819 hm 2 in 1985,1995,2000,2005,2010 and 2011,respectively,in Horqin Sandy Land.A total of 57 soil sampling sites across Shelihu were chosen in wet meadow(CL0),cropland(CL) and sandy land(SL) according to the spatial characteristics of water body change.Soil texture,organic carbon(SOC),total nitrogen(TN) and total phosphorus(TP) contents,electrical conductivity(EC) and pH were measured at the soil depths of 0-10,10-20 and 20-40 cm to examine the influence of agricultural conversion and continuous cultivation on soil properties.The results showed that the study area was covered by water body in 1985,which gradually declined afterwards and then reclaimed rapidly at a mean annual rate of 132.1 hm 2 /a from wet meadow to cropland since 1995.In 2011,water body was drained and the area was occupied by 10.8% of CL0,76.9% of CL and 12.3% of SL.Large amounts of SOC,TN and TP were accumulated in the above depths in CL0.Soil in CL0 also had higher EC and silt and clay fractions,lower pH than in SL and CL.Soil in SL was seriously degraded with lower contents of SOC,TN and TP than in CL and CL0.SOC,TN content and EC in CL decreased with the increase of cultivation age,while pH showed a reverse trend with significance at plough horizon.The agricultural conversion in Shelihu was driven by the comprehensive factors of precipitation reduction,economic development and intense competitions for irrigation water.Continuous cultivation in this process is not sustainable because of SOC degradation and nutrient content reduction.The key point is that conventional tillage and removal of residuals induced further land degradation.Wetland reclamation for immediate economic interests led to greater costs in the long-term environmental restoration in Horqin Sandy Land. | Jie LIANI XueYong ZHAO XiaoAn ZUO ShaoKun WANG XinYuan WANG YongQing LUO | 2013 | Journal of Arid Land2013,5,1: | 4 |
| 11 | The genome of Magnolia biondii Pamp. provides insights into the evolution of Magnoliales and biosynthesis of terpenoids显示文摘Magnolia biondii Pamp.(Magnoliaceae,magnoliids)is a phylogenetically,economically,and medicinally important ornamental tree species widely grown and cultivated in the north-temperate regions of China.Determining the genome sequence of M.biondii would help resolve the phylogenetic uncertainty of magnoliids and improve the understanding of individual trait evolution within the Magnolia genus.We assembled a chromosome-level reference genome of M.biondii using~67,~175,and~154Gb of raw DNA sequences generated via Pacific Biosciences single-molecule real-time sequencing,10X Genomics Chromium,and Hi-C scaffolding strategies,respectively.The final genome assembly was~2.22Gb,with a contig N50 value of 269.11 kb and a BUSCO complete gene percentage of 91.90%.Approximately 89.17%of the genome was organized into 19 chromosomes,resulting in a scaffold N50 of 92.86Mb.The genome contained 47,547 protein-coding genes,accounting for 23.47%of the genome length,whereas 66.48%of the genome length consisted of repetitive elements.We confirmed a WGD event that occurred very close to the time of the split between the Magnoliales and Laurales.Functional enrichment of the Magnolia-specific and expanded gene families highlighted genes involved in the biosynthesis of secondary metabolites,plant–pathogen interactions,and responses to stimuli,which may improve the ecological fitness and biological adaptability of the lineage.Phylogenomic analyses revealed a sister relationship of magnoliids and Chloranthaceae,which are sister to a clade comprising monocots and eudicots.The genome sequence of M.biondii could lead to trait improvement,germplasm conservation,and evolutionary studies on the rapid radiation of early angiosperms. | Shanshan Dong Min Liu Yang Liu Fei Chen Ting Yang Lu Chen Xingtan Zhang Xing Guo Dongming Fang Linzhou Li Tian Deng Zhangxiu Yao Xiaoan Lang Yiqing Gong Ernest Wu Yaling Wang Yamei Shen Xun Gong Huan Liu Shouzhou Zhang | 2021 | Horticulture Research2021,8,1: | 4 |

| 13 | Response of Polygonum viviparum Species and Community Level to Long-term Livestock Grazing in Alpine Shrub Meadow in Qinghai-Tibet Plateau显示文摘Grazing by domestic herbivores is generally recognized as a major ecological factor and an important evolutionary force in grasslands. Grazing has both extensive and profound effects on individual plants and communities. We investigated the response patterns of Polygonum viviparum species and the species diversity of an alpine shrub meadow in response to long-term livestock grazing by a field manipulative experiment controlling livestock numbers on the Qinghai-Tibet Plateau in China. Here, we hypothesize that within a range of grazing pressure, grazing can alter relative allocation to different plant parts without changing total biomass for some plant species if there is life history trade-offs between plant traits. The same type of communities exposed to different grazing pressures may only alter relative species’ abundances or species composition and not vary species diversity because plant species differ in resistant capability to herbivory. The results show that plant height and biomass of different organs differed among grazing treatments but total biomass remained constant. Biomass allocation and absolute investments to both reproduction and growth decreased and to belowground storage increased with increased grazing pressure, indicating the increasing in storage function was attained at a cost of reducing reproduction of bulbils and represented an optimal allocation and an adaptive response of the species to long- term aboveground damage. Moreover, our results showed multiform response types for either species groups or single species along the gradient of grazing intensity. Heavy grazing caused a 13.2% increase in species richness. There was difference in species composition of about 18%–20% among grazing treatment. Shannon-Wiener (H ) diversity index and species evenness (E) index did not differ among grazing treatments. An oxidation assisted crack initiation and propagation phenomenon were found to explain the shorted life under TMF cycles. In order to characterize the stress and strain deformations under TMF loadings, a modified Chaboche's constitutive model was applied. | 15 | A small scale expression screen identifies tissue specific markers in the Dugesia japonica strain Pek-1显示文摘Freshwater planaria has tremendous capacity to reform the missing part of the body and therefore is considered as one of the most important model organism for regeneration study. At present, Schmidtea mediterranea and Dugesia japonica are the two major species utilized for laboratory manipulations. Dugesia japonica flatworms are widely distributed in the Far East including Cherry Valley region in the northwest area of Beijing, China. We reported here the establishment of an asexual Dugesia japonica strain Pek1, as a suitable system for regeneration study. Using morphological, karyotypical as well as phylogenetic analyses, we confirmed that these flatworms indeed belonged to Dugesia japonica. We went on to show that the commonly used in situ probes and immunohistochemistry reagents and protocols were applicable to the Pek1 strain. Using this strain, we carried out small scale analysis on EST, RNAi and gene expression. We identified 193 unique EST sequences and 65 of them had not been reported in planarian. By RNAi analysis, we showed that 48 genes, when downregulated individually, had no effect on regeneration. Furthermore, we identified 3 groups of tissue specific expressing genes that were useful for cell lineage analysis. | 17 | Preharvest application of 1-methylcyclopropene and Ethephon altered cuticular wax biosynthesis and fruit quality of apples at harvest and during cold storage显示文摘To investigate the roles of Harvista(a sprayable 1-methylcyclopropene,1-MCP,available component is 150 g·hm^(-2))and Ethephon(1 mL·L^(-1))applied at preharvest in apple(Malus domestica Borkh.'Golden Delicious')fruit cuticular wax biosynthesis,the expression of genes related to fruit cuticular wax biosynthesis and ethylene biosynthesis and signaling,ethylene production rate,respiration rate,wax constituents and structure,and fruit quality were determined at harvest and during cold storage.The results showed that 1-MCP inhibited the expression levels of ethylene biosynthesis and signaling-related genes,decreased fruit ethylene production,and inhibited the expression of Md LACS1,Md CER6,Md CER4 and Md WSD1,which resulted in decreases in alcohols,acids,and esters content in fruit cuticular wax.1-MCP also reduced fruit dropping rate from 17.17%to 12%;maintained fruit firmness,soluble solids,titratable acidity during cold storage;showed about as one fifth in the total length and one third in the widest width of wax crack as that in control at harvest.In contrast,Ethephon produced the opposite effects.In conclusion,1-MCP inhibited fruit ethylene biosynthesis and signaling,and influenced fruit cuticular wax biosynthesis.Thus,the fruit cuticular wax constituents and structure was altered,and the fruit quality were maintained at harvest and during cold storage.The results provide a new technology for improving apple fruit harvest and postharvest quality by preharvest application of sprayable 1-MCP. | Fujun Li Xinyue Zhang Jihan Wang Yaping Jiang Xinhua Zhang Xiaoan Li | 2022 | Horticultural Plant Journal2022,8,2: | 2 |
| 18 | Effects of tillage practices on nutrient loss and soybean growth in red-soil slope farmland显示文摘Field experiments were conducted to examine the effect of tillage practices on sediment and nutrient loss and soybean growth under natural rainfall conditions.Three tillage practices were applied:downslope ridge(check),downslope ridge+contour living hedgerow,and cross ridge.Cross ridge tillage reduced surface runoff by 69%and sediment yield by 86%,compared to the check treatment.The downslope ridge with a contour living hedgerow reduced surface runoff by 24%and sediment yield by 53%.Additionally,compared to the check plot,nutrient losses carried by runoff were reduced by over 68%and that carried in the sediment was reduced more than 85%in the cross ridge plot.Nutrient losses in runoff were reduced by 20%to 30%in the downslope ridge and contour living hedgerow plot and those carried in the sediment were reduced by 44%to 57%.Cross ridge tillage soybean yields exceeded those of the downslope ridge and downslope ridge+contour living hedgerow treatments by 16%-18%.Cross ridge tillage could contribute to the prevention sediment and nutrient loss and could improve crop yield,and thus it is recommended to be applied to mild slopes in the red soil region. | Yang Jie Zheng Haijin Chen Xiaoan Shen Le | 2013 | International Soil and Water Conservation Research2013,1,3: | 2 |

| 20 | Substrate stiffness-dependent regulatory volume decrease and calcium signaling in chondrocytes显示文摘The pericellular matrix stiffness is strongly associated with its biochemical and structural changes during the aging and osteoarthritis progress of articular cartilage.However,how substrate stiffness modulates the chondrocyte regulatory volume decrease(RVD)and calcium signaling in chondrocytes remains unknown.This study aims to investigate the effects of substrate stiffness on the chondrocyte RVD and calcium signaling by recapitulating the physiologically relevant substrate stiffness.Our results showed that substrate stiffness induces completely different dynamical deformations between the cell swelling and recovering progresses.Chondrocytes swell faster on the soft substrate but recovers slower than the stiff substrate during the RVD response induced by the hypo-osmotic challenge.We found that stiff substrate enhances the cytosolic Ca^(2+)oscillation of chondrocytes in the iso-osmotic medium.Furthermore,chondrocytes exhibit a distinctive cytosolic Ca^(2+)oscillation during the RVD response.Soft substrate significantly improves the Ca^(2+)oscillation in the cell swelling process whereas stiff substrate enhances the cytosolic Ca^(2+)oscillation in the cell recovering process.Our work also suggests that the TRPV4 channel is involved in the chondrocyte sensing substrate stiffness by mediating Ca^(2+)signaling in a stiffness-dependent manner.This helps to understand a previously unidentified relationship between substrate stiffness and RVD response under the hypo-osmotic challenge.A better understanding of substrate stiffness regulating chondrocyte volume and calcium signaling will aid the development of novel cell-instructive biomaterial to restore cellular functions. | Min Zhang Xiaoan Wu Genlai Du Weiyi Chen Quanyou Zhang | 2022 | Acta Biochimica et Biophysica Sinica2022,54,1: | 2 |
