1miRNA-711-SP1-collagen-I pathway is involved in the anti-fibrotic effect of pioglitazone in myocardial infarction显示文摘Although microRNAs(miRNAs) have been intensively studied in cardiac fibrosis,their roles in drug-mediated anti-fibrotic therapy are still unknown.Previously,Pioglitazone attenuated cardiac fibrosis and increased miR-711 experimentally.We aimed to explore the role and mechanism of miR-711 in pioglitazone-treated myocardial infarction in rats.Our results showed that pioglitazone significantly reduced collagen-I levels and increased miR-711 expression in myocardial infarction heart.Pioglitazone increased the expression of miR-711 in cardiac fibroblasts,and overexpression of miR-711 suppressed collagen-I levels in angiotensin II(Ang II)-treated or untreated cells.Transfection with antagomir-711 correspondingly abolished the pioglitazone-induced reduction in collagen-I levels.Bioinformatics analysis identified SP1,which directly promotes collagen-I synthesis,as the putative target of miR-711.This was confirmed by luciferase assay and western blot analysis.Additionally,increased SP1 expression was attenuated by pioglitazone in myocardial infarction heart.Furthermore,transfection of antagomir-711 attenuated pioglitazone-reduced SP1 expression in cardiac fibroblasts with or without Ang II stimulation.We conclude that pioglitazone up-regulated miR-711 to reduce collagen-I levels in rats with myocardial infarction.The miR-711-SP1-collagen-I pathway may be involved in the anti-fibrotic effects of pioglitazone.Our findings may provide new strategies for miRNA-based anti-fibrotic drug research.ZHAO Na YU HaiYi YU HaiTao SUN Min ZHANG YouYi XU Ming GAO Wei 2013Science China(Life Sciences)2013,56,5:9
2Genome editing in Drosophila melanogaster: from basic genome engineering to the multipurpose CRISPR-Cas9 system显示文摘Nowadays, genome editing tools are indispensable for studying gene function in order to increase our knowledge of biochemical processes and disease mechanisms. The extensive availability of mutagenesis and transgenesis tools make Drosophila melanogaster an excellent model organism for geneticists. Early mutagenesis tools relied on chemical or physical methods,ethyl methane sulfonate(EMS) and X-rays respectively, to randomly alter DNA at a nucleotide or chromosomal level. Since the discovery of transposable elements and the availability of the complete fly genome, specific genome editing tools, such as P-elements, zinc-finger nucleases(ZFNs) and transcription activator-like effector nucleases(TALENs), have undergone rapid development. Currently, one of the leading and most effective contemporary tools is the CRISPR-cas9 system made popular because of its low cost, effectiveness, specificity and simplicity of use. 3GEOMETRY AND DIMENSION OF SELF-SIMILAR SET显示文摘The authors show that the self-similar set for a finite family of contractive similitudes (sim-ilarities, i.e., |fi(x) - fi(y)| = ai|x - y|, x,y ∈ RN, where 0 < ai < 1) is uniformly perfectexcept the case that it is a singleton. As a corollary, it is proved that this self-similar set haspositive Hausdorff dimension provided that it is not a singleton. And a lower bound of theupper box dimension of the uniformly perfect sets is given. 5Spectroscopic analysis of high electric field enhanced ionization in laser filaments in air for corona guiding显示文摘We report on a systematic experimental study on the fluorescence spectra produced from a femtosecond laser filament in air under a high electric field. The electric field alone was strong enough to create corona discharge(CD). Fluorescence spectra from neutral and ionic air molecules were measured and compared with pure high-voltage CD and pure laser filamentation(FIL). Among them, high electric field assisted laser FIL produced nitrogen fluorescence more efficiently than either pure CD or pure FIL processes. The nonlinear enhancement of fluorescence from the interaction of the laser filament and corona discharging electric field resulted in a more efficient ionization along the laser filament zone, which was confirmed by the spectroscopic measurement of both ionization-induced fluorescence and plasma-scattered 800 nm laser pulses. 7A novel molecular classification method for osteosarcoma based on tumor cell differentiation trajectories显示文摘Subclassification of tumors based on molecular features may facilitate therapeutic choice and increase the response rate of cancer patients.However,the highly complex cell origin involved in osteosarcoma(OS)limits the utility of traditional bulk RNA sequencing for OS subclassification.Single-cell RNA sequencing(sc RNA-seq)holds great promise for identifying cell heterogeneity.However,this technique has rarely been used in the study of tumor subclassification.By analyzing sc RNA-seq data for six conventional OS and nine cancellous bone(CB)samples,we identified 29 clusters in OS and CB samples and discovered three differentiation trajectories from the cancer stem cell(CSC)-like subset,which allowed us to classify OS samples into three groups.The classification model was further examined using the TARGET dataset.Each subgroup of OS had different prognoses and possible drug sensitivities,and OS cells in the three differentiation branches showed distinct interactions with other clusters in the OS microenvironment.In addition,we verified the classification model through IHC staining in 138 OS samples,revealing a worse prognosis for Group B patients.Furthermore,we describe the novel transcriptional program of CSCs and highlight the activation of EZH2 in CSCs of OS.These findings provide a novel subclassification method based on sc RNA-seq and shed new light on the molecular features of CSCs in OS and may serve as valuable references for precision treatment for and therapeutic development in OS.Hao Zhang Ting Wang Haiyi Gong Runyi Jiang Wang Zhou Haitao Sun Runzhi Huang Yao Wang Zhipeng Wu Wei Xu Zhenxi Li Quan Huang Xiaopan Cai Zaijun Lin Jinbo Hu Qi Jia Chen Ye Haifeng Wei Jianru Xiao 2023Bone Research2023,11,1:0
8Neon venting of activated NEG-coated beam pipes in IHEP显示文摘Background In High Energy Photon Source(HEPS)the beam vacuum pipes employ TiZrVHf non-evaporable getter(NEG)coatings in order to meet the operation requirements.In many occasions it is inevitable to bring the sectors to atmosphere in order to open detectors,insert or change faulty parts.Consequently,NEG coatings will be saturated and followed with activation to restore the vacuum.Such an intervention would require heating up the whole sector to hundreds of degree and last from 2 to 4 weeks.Purpose Therefore,a Neon venting system is developed in HEPS in order to perform fast interventions without losing much of the performance of the activated NEG coating.Methods Preliminary experiments has been carried out to test the venting with Neon of different purity by comparing pumping speed lost and vacuum recovery before and after Neon venting.Results Results show that Neon with ultrahigh purity(purified by NEG cartridge purifier)could preserve full pumping performance of the NEG coated pipes.Neon with 6 N purity could preserve half pumping performance of the NEG coated pipes,but vacuum could be fully restored in only 2 days.Neon with 5 N purity could saturate the NEG coated pipes,leading to full pumping performance lost.Therefore,Neon with 6 N purity or above is promising in applications of efficient operations in HEPS.Yuchen Yang Haichang Duan Tao Huang Yongsheng Ma Fei Sun Haiyi Dong Ping He 2022Radiation Detection Technology and Methods2022,6,4:0
9Seroprevalence of human enterovirus A71 in Guangzhou,China,2019-2021显示文摘Enterovirus A71 (EV-A71) is a significant hand-foot-mouth disease (HFMD) etiology. The inactivated EV-A71 vaccines were approved in China in 2016. However, the seroprevalence of EV-A71 after the vaccine application and its potential association with the EV-A71 epidemic in the population are rarely studied. In this study, we analyzed the incidence of EV-A71 infection and seroepidemiology in Guangzhou City, China. From 2019 to 2021, 167,920 clinically confirmed HFMD cases were reported in Guangzhou. In 6,868 enterovirus-positive samples, Coxsackievirus A6 and Coxsackievirus A16 were dominant genotypes, and only 3 EV-A71-positive samples were detected, highlighting the deficient epidemic activity of EV-A71. Microneutralization assay was performed on 1,000 representative serum samples. Notably, the seroprevalence and geometric mean titer (GMT) decreased significantly in 2020, and that in the < 3-year age group were increased and even higher than that in 3–5-year age group in 2019 and 2021, which was contrary to our previous surveillance result and other studies in Guangzhou. Furthermore, a moderate decline of GMT level was observed following the vaccination, but the seropositive serums were still detected for 49 months after second immunization, suggesting the long-term persistence of the immunity. Our seroepidemiology study revealed relatively higher neutralizing antibody activity in the susceptible population after the EV-A71 vaccine was adopted in 2016 in Guangzhou.
