Study on the promotion of lymphocytes in patients with COVID-19 by broad-spectrum chemokine receptor inhibitor vMIP-Ⅱ and its Mechanism of signal transmission in vitro显示文摘Dear Editor,COVID-19 cases showed a significant decrease in the number of peripheral CD4^(+) and CD8^(+)T cells accompanied by overactivation of cells.1 HHV8-derived viral macrophage inflammatory protein vMIP-Ⅱ is the only broad-spectrum chemokine receptor inhibitor that binds to three subtribes of chemokine receptors, which can significantly increase the specific cell immune response in infected organisms and have an effect of reducing plasma viral load on the viremia period.24 Here we report the effect of vMIP-Ⅱ in COVID-19.Shiyu Li Shuting Liu Zhenyou Jiang Lixia Feng Yong Gao Youyu Chen Anding Xu Wenhua Huang Nuofu Zhang Hanxiao Sun 2021Signal Transduction and Targeted Therapy2021,6,4:5
Evaluation of human epidermal growth factor receptor 2 status of breast cancer using preoperative multidetector computed tomography with deep learning and handcrafted radiomics features显示文摘Objective:To evaluate the human epidermal growth factor receptor 2(HER2)status in patients with breast cancer using multidetector computed tomography(MDCT)-based handcrafted and deep radiomics features. Methods:This retrospective study enrolled 339 female patients(primary cohort,n=177;validation cohort,n=162)with pathologically confirmed invasive breast cancer.Handcrafted and deep radiomics features were extracted from the MDCT images during the arterial phase. After the feature selection procedures, handcrafted and deep radiomics signatures and the combined model were built using multivariate logistic regression analysis.Performance was assessed by measures of discrimination, calibration, and clinical usefulness in the primary cohort and validated in the validation cohort. Results:The handcrafted radiomics signature had a discriminative ability with a C-index of 0.739[95%confidence interval(95%CI):0.661-0.818]in the primary cohort and 0.695(95%CI:0.609-0.781)in the validation cohort. The deep radiomics signature also had a discriminative ability with a C-index of 0.760(95%CI:0.690-0.831)in the primary cohort and 0.777(95%CI:0.696-0.857)in the validation cohort. The combined model, which incorporated both the handcrafted and deep radiomics signatures, showed good discriminative ability with a C-index of 0.829(95%CI:0.767-0.890)in the primary cohort and 0.809(95%CI:0.740-0.879)in the validation cohort. Conclusions:Handcrafted and deep radiomics features from MDCT images were associated with HER2 status in patients with breast cancer. Thus, these features could provide complementary aid for the radiological evaluation of HER2 status in breast cancer.Xiaojun Yang Lei Wu Ke Zhao Weitao Ye Weixiao Liu Yingyi Wang Jiao Li Hanxiao Li Xiaomei Huang Wen Zhang Yanqi Huang Xin Chen Su Yao Zaiyi Liu Changhong Liang 2020Chinese Journal of Cancer Research2020,32,2:4
Survival difference between EGFR Del19 and L858R mutant advanced non-small cell lung cancer patients receiving gefitinib:a propensity score matching analysis显示文摘Objective: Although superior clinical benefits of epidermal growth factor receptor(EGFR) tyrosine kinase inhibitors(TKIs) in the treatment of advanced non-small-cell lung cancer(NSCLC) had been reported, the survival difference between exon 19 deletion(Del19) and exon 21 Leu858 Arg substitution(L858 R) remains controversial. The purpose of this study is to investigate the differences in progression-free survival(PFS) and overall survival(OS) between different EGFR mutant subtypes among advanced NSCLC patients receiving gefitinib. Methods: There were 204 advanced NSCLC patients with EGFR mutations treated with gefitinib were enrolled in this retrospective cohort study.Patients were divided into the EGFR Del19 group and the L858 R mutated group according to their mutant subtype.Propensity score matching(PSM) was conducted by using a nearest-neighbor algorithm(1:1) to adjust for demographical and clinical covariates.Survival curves were constructed with the Kaplan-Meier method and compared by using the log-rank test. Results: The PFS in Del19 group was similar to that in the L858 R group [before PSM 8.6 vs.7.2 months,P=0.072; after PSM 7.3 vs.7.2 months,P=0.155].No differences were detected in OS between the L858 R and the Del19 group(before PSM 17.8 vs.13.1 months,P=0.253; after PSM 16.9 vs.13.1 months,P=0.339).The Del19 group was significantly younger compared with the L858 R mutation group in age(P=0.015).Conclusions: No significant difference was found in the PFS or OS between the Del19 and L858 R mutant NSCLC patients receiving gefitinib. The age gap might contribute to the survival differences between Del19 and L858 R groups.PSM is of important value to the elimination of potential bias.Minglei Zhuo Qiwen Zheng Jun Zhao Meina Wu Tongtong An Yuyan Wang Jianjie Li Shuhang Wang Jia Zhong Xue Yang Hanxiao Chen Bo Jia Zhi Dong Emei Gao JingjingWang Ziping Wang 2017Chinese Journal of Cancer Research2017,29,6:4
Mixture Toxicities of Three Pesticides Having Different Time-Toxicity Profiles显示文摘In the environment, chemical pollutants always exist as mixtures. To evaluate mixture toxicity,we should ex-amine the concentration ratios and the concentration levels of mixture components as well as the interactive time between the components and testing organism.Selecting three pesticides, metribuzin(MET),dodine(DOD)and ri-domil(RID),as mixture components,Vibrio qinghaiensis sp.-Q67 as testing organism, and 96-well microplate as exposure experiment carrier, the luminescence inhibition toxicities of the pesticides and their binary mixtures at six exposure time points(0.25,2,4,8,12,and 16 h)were determined. The results showed that the three pesticides have different time-toxicity profiles. The toxicity of MET increases over time, and those of DOD and RID show a slight decrease or increase in the first 4 h,respectively, and then remain unchanged to 16 h.The toxicities of the binary mixtures consisting of the three pesticides depend on not only the mixture composition but also the exposure time. The time-toxicity profiles of all rays in MET-DOD mixture system are U-shaped curves, those in MET-RID system are basically monotonically increased and those in DOD-RID system are basically monotonically decreased.How-ever,our findings indicate that for the test substances, mixture toxicity can be predicted by the extended concentra-tion addition model.Mo Yu Shushen Liu Mengchao Wang Fu Chen Hanxiao Tang 2014Chinese Journal of Chemistry2014,32,6:2
Clinical outcomes of atezolizumab in combination with etoposide/platinum for treatment of extensive-stage small-cell lung cancer:A real-world,multicenter,retrospective,controlled study in China显示文摘Objective:Atezolizumab along with chemotherapy has prolonged the survival of patients with extensive-stage small-cell lung cancer(ES-SCLC)worldwide,although real-world(RW)data are lacking in China. This study was designed to evaluate the efficacy and clinical outcomes of atezolizumab plus etoposide/platinum(EP).Methods:Data obtained in this retrospective study were captured from six oncology units of five medical facilities from January 2019 to April 2022.For first-line treatments, atezolizumab combined with EP vs.EP alone,we primarily evaluated progression-free survival(PFS);other efficacy indicators, including overall survival(OS),objective response rate(ORR),and patterns of SCLC progression and adverse events(AEs)were assessed. Results:The primary analysis included data from 225 patients,of whom 133 received EP along with atezolizumab(atezolizumab group)and 92 received EP alone(EP group). The PFS duration of the atezolizumab group[7.10 months;95%confidence interval(95%CI),6.53-9.00]exceeded that of the EP group(6.50 months;95%CI,4.83-7.53).Overall,the hazard ratio(HR)was 0.69(95%CI,0.49-0.97)(P=0.029);particularly,the HR was 0.54(95%CI,0.36-0.80)among patients undergoing≥4 chemotherapy cycles and 0.33(95%CI,0.20-0.56)among individuals with atezolizumab maintenance. The ORR and disease-control rate(DCR)were similar between the two groups.Because of incomplete OS data, the median OS was not determined for either group.Bone marrow suppression was the most common AE detected(58.6%)in the atezolizumab group.Immune-related AEs occurred in 19 patients in the atezolizumab group(14.3%),with only one case of grade 3 encephalitis. Conclusions:This RW study in China demonstrated improved clinical outcomes of atezolizumab along with EP for ES-SCLC,particularly in the chemosensitive population. These results align with the results of the IMpower133 study, although the impact of this treatment modality on OS warrants additional follow-up studies.Hanxiao Chen Xiangjuan Ma Jie Liu Yu Yang Yong Fang Liping Wang Jian Fang Jun Zhao Minglei Zhuo 2022Chinese Journal of Cancer Research2022,34,4:1
Supercritical fluid-assisted controllable fabrication of open and highly interconnected porous scaffolds for bone tissue engineering显示文摘Recently tremendous progress has been evidenced by the advancements in developing innovative three-dimensional(3 D)scaffolds using various techniques for addressing the autogenous grafting of bone. In this work, we demonstrated the fabrication of porous polycaprolactone(PCL) scaffolds for osteogenic differentiation based on supercritical fluid-assisted hybrid processes of phase inversion and foaming. This eco-friendly process resulted in the highly porous biomimetic scaffolds with open and interconnected architectures. Initially, a 2^3 factorial experiment was designed for investigating the relative significance of various processing parameters and achieving better control over the porosity as well as the compressive mechanical properties of the scaffold. Then, single factor experiment was carried out to understand the effects of various processing parameters on the morphology of scaffolds. On the other hand, we encapsulated a growth factor, i.e., bone morphogenic protein-2(BMP-2), as a model protein in these porous scaffolds for evaluating their osteogenic differentiation. In vitro investigations of growth factor loaded PCL scaffolds using bone marrow stromal cells(BMSCs) have shown that these growth factor-encumbered scaffolds were capable of differentiating the cells over the control experiments. Furthermore, the osteogenic differentiation was confirmed by measuring the cell proliferation, and alkaline phosphatase(ALP) activity, which were significantly higher demonstrating the active bone growth. Together, these results have suggested that the fabrication of growth factor-loaded porous scaffolds prepared by the eco-friendly hybrid processing efficiently promoted the osteogenic differentiation and may have a significant potential in bone tissue engineering.Hanxiao Tang Ranjith Kumar Kankala Shibin Wang Aizheng Chen 2019Science China(Life Sciences)2019,62,12:1

Collagen-based materials in reproductive medicine and engineered reproductive tissues显示文摘Collagen, the main component of mammal skin, has been traditionally used in leather manufacturing for thousands of years due to its diverse physicochemical properties.Collagen is the most abundant protein in mammals and the main component of the extracellular matrix(ECM).The properties of collagen also make it an ideal building block for the engineering of materials for a range of biomedical applications.Reproductive medicine, especially human fertility preservation strategies and reproductive organ regeneration, has attracted significant attention in recent years as it is key in resolving the growing social concern over aging populations worldwide.Collagen-based biomaterials such as collagen hydrogels, decellularized ECM(dECM),and bioengineering techniques including collagen-based 3D bio-printing have facilitated the engineering of reproductive tissues. This review summarizes the recent progress in apply-ing collagen-based biomaterials in reproductive.Furthermore,we discuss the prospects of collagen-based materials for engineering artificial reproductive tissues, hormone replacement therapy, and reproductive organ reconstruction, aiming to inspire new thoughts and advancements in engineered reproductive tissues research.Hanxiao Chen Luqi Xue Guidong Gong Jiezhou Pan Xiaoling Wang Yaoyao Zhang Junling Guo Lang Qin 2022Journal of Leather Science and Engineering2022,4,1:0
Differential immune responses in pregnant patients recovered from COYID-19显示文摘Pregnant women are generally more susceptible to viral infection.Although the impact of SARS-CoV-2 in pregnancy remains to be determined, evidence indicates that the risk factors for severe COVID-19 are similar in pregnancy to the general population. Here we systemically analyzed the clinical characteristics of pregnant and non-pregnant female COVID-19 patients who were hospitalized during the same period and found that pregnant patients developed marked lymphopenia and higher inflammation evident by higher C-reactive protein and IL-6.To elucidate the pathways that might contribute to immunopathology or protective immunity against COVID-19 during pregnancy,we applied single-cell mRNA sequencing to profile peripheral blood mononuclear cells from four pregnant and six non-pregnant female patients after recovery along with four pregnant and three non-pregnant healthy donors. We found normal clonal expansion of T cells in the pregnant patients, heightened activation and chemotaxis in NK,NKT,and MAIT cells, and differential interferon responses in the monocyte compartment. Our data present a unique feature in both innate and adaptive immune responses in pregnant patients recovered from COVID-19.Ge Chen Yiming Zhang Yaoyao Zhang Jihui Ai Bin Yang Mengge Cui Qiuyue Liao Hanxiao Chen Hualin Bai Dashing Shang Jing Chen ChaoYang Sun Haiyi Liu Fengyuan Liu Bin Mao Guoqiang Sun Lu Chen Jing-wen Lin Kezhen Li 2021Signal Transduction and Targeted Therapy2021,6,8:0
Coded aperture compressive imaging array applied for surveillance systems显示文摘This paper proposes an application of compressive imaging systems to the problem of wide-area video surveillance systems.A parallel coded aperture compressive imaging system and a corresponding motion target detection algorithm in video using compressive image data are developed.Coded masks with random Gaussian,Toeplitz and random binary are utilized to simulate the compressive image respectively. For compressive images,a mixture of the Gaussian distribution is applied to the compressed image field to model the background.A simple threshold test in compressive sampling image is used to declare motion objects.Foreground image retrieval from underdetermined measurement using the total variance optimization algorithm is explored. The signal-to-noise ratio (SNR) is employed to evaluate the image quality recovered from the compressive sampling signals, and receiver operation characteristic (ROC)curves are used to quantify the performance of the motion detection algorithm.Experimental results demonstrate that the low dimensional compressed imaging representation is sufficient to determine spatial motion targets.Compared with the random Gaussian and Toeplitz mask, motion detection algorithms using the random binary phase mask can yield better detection results. However using the random Gaussian and Toeplitz phase mask can achieve high resolution reconstructed images.Jing Chen Yongtian Wang Hanxiao Wu 2013Journal of Systems Engineering and Electronics2013,24,6:0
Magnetic Fields Affect Alcoholic Liver Disease by Liver Cell Oxidative Stress and Proliferation Regulation显示文摘It is well known that alcohol consumption leads to substantially increased free radical levels and health risks, which lacks effective treatment besides alcohol abstinence. Here,we compared different static magnetic field(SMF)settings and found that a downward quasi-uniform SMF of~0.1 to 0.2 T could effectively alleviate alcohol-induced liver damage and lipid accumulation and improve hepatic function.SMFs of two different directions can reduce the inflammation, reactive oxygen species levels, and oxidative stress in the liver, while the downward SMF had more obvious effects. Moreover,we found that the upward direction SMF of~0.1 to 0.2 T could inhibit DNA synthesis and regeneration in hepatocytes, which caused detrimental effects on the lifespan of'heavy drinking'mice. In contrast, the downward SMF prolongs survival of'heavy drinking'mice. On one hand, our study shows that~0.1 to 0.2 T moderate quasi-uniform SMFs with a downward direction have great promises to be developed into a physical method to reduce alcohol-induced liver damage;on the other hand, although the internationally recognized upper limit for SMF public exposure is 0.4 T,people should also pay extra attention to SMF strength, direction, and inhomogeneity that could generate harmful effects on specific severe pathological conditions.Chao Song Hanxiao Chen Biao Yu Lei Zhang Junjun Wang Chuanlin Feng Xingxing Yang Xiaofei Tian Yixiang Fan Xinmiao Ji Hua Wang Can Xie Xin Zhang 2023Research2023,,4:0
First Records of Late Triassic Conodont Fauna andδ13Ccarb from the Dengdengqiao Section,Dangchang County,Gansu Province,Northwestern China显示文摘Based on a study of 49 conodont and 57 geochemical samples from the Upper Triassic,carbonate-dominated Dengdengqiao Formation,Qinling Basin,China, the Carnian conodonts and carbon isotope records are first reported. Two genera and four species have been identified amongst 87 conodont elements:Mosherella praebudaensis,Mo.longnanensis sp.nov.,Mo.sp.,and'Misikella'longidentata.The presence of Mo.praebudaensis indicates that the lower part(bed 2)of the formation is attributable to the Julian(lower Carnian)substage.A radiolarian fauna identified in a previous study belongs to the upper Carnian, but the sampling horizon is unclear.Theδ13Ccarb curve shows a~1.8‰negative excursion beginning from upper part of bed 3,but its stratigraphic location is uncertain. The Dengdengqiao Formation is clearly at least partly of Carnian age but could include younger strata. The abundant calcareous algae at the section is probably due to some transport rather than preserved in site. The unusual ecosystem with rare marine organisms may reflect long-term stressful and unfavorable conditions at Dengdengqiao.Hanxiao Li Miaoyan Wang Muhui Zhang Paul B.Wignall Manuel Rigo Yanlong Chen Xianlang Wu Zhumin Ouyang Baojin Wu Zhaoyang Yi Zaitian Zhang Xulong Lai 2021Journal of Earth Science2021,32,3:0
Group 2 innate lymphoid cells can engulf and destroy bacteria显示文摘Group 2 innate lymphoid cells(ILC2s)are now known to play prominent roles in many inflammatory responses involving the production of Th2-type cytokines, which contribute to parasitic infections, tissue repair, and metabolic homeostasis[1].More recently,it has emerged that ILC2s may act as antigen-presenting cells, although corresponding evidence of phagocytic activity of these cells is currently scarce[2,3].Herein,we investigated whether ILC2s can phagocytose bacteria, through in vitro experiments.Yiran Yang Yan Li Yingjie Xu Hanxiao Zhang Yuhan Diao Shihao Chen Ye Cui Chris J.Corrigan Wei Wang Sun Ying 2021Cellular & Molecular Immunology2021,18,11:0
