2A Review of the Design and Architecture of Starch-Based Dietary Foods显示文摘Carbohydrates,which are mostly present in sugar,starch,and fiber,are one of the main ingredients of food and the primary source of energy in the human diet.Among these three main sources,starch stands out as one of the most abundant reserves of carbohydrates.Investigating starch would not only enhance our understanding of the functionality of starch in the human body but also aid in the design of novel starch-based dietary foods.The present review first provides a state-of-the-art understanding of the various classifications of dietary starches,including rapidly digesting starch(RDS),slowly digesting starch(SDS),and resistant starch(RS).Moreover,both the in vivo and in vitro determination methods of the digestibility of starch-based dietary foods are discussed.Based on the current understanding,present research strategies to design novel starch-based dietary foods through either the direct addition of modified starch or the alteration of processing conditions are highlighted.Furthermore,certain perspectives related to the future research directions of starch-based foods are also included.Yuxiang Bai Xiaoxiao Li Hangyan Ji Yu Wang Danni Zheng Yanli Wang Zhengyu Jin 2021Engineering2021,7,5:2

12Supramolecular complex glycoconjugate vaccine generates self-enhancement effects for carbohydrate antigen delivery显示文摘Targeting delivery of tumor-associated carbohydrate antigen(TACA)-based vaccine to antigen-presenting cells(APCs)mediated by endogenous antibodies can improve the immunogenicity of TACA.However,an essential requirement of this approach is to generate high titers of endogenous antibodies in vivo through pre-immunization,which complicates the immunization procedure and may cause side effects.Herein,we report a new generation of APC-targeting TACA-based supramolecular complex vaccine,assembled by sialyl Thomsen-nouveau-bovine serum albumin-adamantine(sTn-BSA-Ada)and heptavalent rhamnose(Rha)-modifiedβ-cyclodextrin(β-CD)via host-guest interaction.The complex vaccine retained anti-Rha antibodies recruiting capability and facilitated the APCs uptake of the vaccine via the interaction of the Fc-domain with the Fc receptors on APCs.We demonstrate that direct immunization of complex vaccine elicited anti-Rha and anti-sTn specific immune response synchronously,generating a novel self-enhancement effect that can improve the antigen delivery to APCs in high efficacy.The structure-activity relationship(SAR)study proved that complex vaccine 4 with polyethylene glycol 6(PEG 6)linker in host molecule provoked a robust and specific sTn immune response comparable to the pre-immunization approach.The antisera induced by complex vaccine,either through direct immunization or pre-immunization,exhibited equal potency of cytotoxicity against the sTn expression cancer cells.This study provides a general platform for TACA-based vaccines with self-enhancement effects without the need for pre-immunization.Han Lin Yanchun Li Kun Zhou Dan Li Hangyan Yu Jie Zhao Haofei Hong Zhifang Zhou Zhimeng Wu 2024Chinese Chemical Letters2024,35,2:0
13novel amylolytic enzyme from Palaeococcus ferrophilus with malto-oligosaccharide forming ability belonging to subfamily GH13_20显示文摘By heterologous expression of a gene from Palaeococcus ferrophilus,a novel recombinant enzyme designated AMPf was obtained and proved to be an amylolytic enzyme with unique catalytic characteristics.The optimal temperature and pH of AMPf were 50℃ and 7.0 respectively.Although sequence analysis and acarbose hydrolysis ability indicated that AMPf belongs to the subfamily GH13_20,interestingly,this enzyme hardly acts on cyclodextrins and pullulan distinguishing it from most enzymes in this subfamily.AMPf hydrolyzes starches to glucose,maltose,maltotriose,and maltotetrose as main products.AMPf mainly liberates glucose from starch with the concentration of 1%(w/v),while it shows malto-oligosaccharide forming ability with higher starch concentration of 4%(w/v).Also,the 4,6-ethylidene-4-nitrophenyl-maltoheptaose hydrolysis ability further indicates the unique combination endo-acting and glucose releasing exo-acting activtity of AMPf.AMPf could utilize maltose and maltotriose to produce malto-oligosaccharides by transglycosylation activity.It was proven AMPf has application protential in malto-oligosaccharides production.Hangyan Ji Xiaoxiao Li Tong Jiang Qi Fang Yuxiang Bai Jie Long Long Chen Zhengyu Jin 2022Food Bioscience2022,45,1:0
