| 2 | LORELEI-LIKE GPI-ANCHORED PROTEINS 2/3 Regulate Pollen Tube Growth as Chaperones and Coreceptors for ANXUR/BUPS Receptor Kinases in Arabidopsis显示文摘Pollen tube growth is crucial for successful fertilization.In Arabidopsis thaliana,the ANXUR(ANX)/BUPS receptor kinase complex controls and maintains pollen tube growth in response to autocrine rapid alkalin-ization factor 4/19(RALF4/19)signaling;however,the molecular and cellular mechanisms underlying the ANX/BUPS-mediated regulation of pollen tube growth remain unclear.In this study,we found that pollen-specific LORELEI-like GPI-anchored proteins 2 and 3(LLG2/3)promote pollen tube growth in vitro and in vivo.LLG2/3 interacte with ANX/BUPS in a RALF4-concentration-dependent manner,suggesting that ANX/BUPS and LLG2/3 might form a receptor-coreceptor complex for perceiving RALF peptide signals.Disruption of the ANX/BUPS-LLG2/3 interaction led to the cytoplasmic retention of ANX1/2,in either llg2/3 knockdown mutants or in anx1/2 mutants lacking the J region,which mediates the ANX/BUPS-LLG2/3 interaction.Moreover,we found that RALF4 induced the production of reactive oxygen species(ROS),which stimulate pollen tube growth and reduce pollen burst rate.ROS levels are reduced in the pollen tubes of LLG2/3 RNAi lines,and application of exogenous H2O2 could partially rescue the defective pollen tube growth of LLG2/3 RNAi lines.Taken together,our study identifies LLG2/3 as novel regulatory components of pollen tube growth,and shows that they chaperone ANX/BUPS for secretion to the apical plasma membrane of pollen tube and act as coreceptors of ANX/BUPS in the acti-vation of ROS production for promoting pollen tube growth. | Hanqian Feng Chen Liu Rong Fu Minmin Zhang Hui Li Lianping Shen Qiqi Wei Xiang Sun Lin Xu Bin Ni Chao Li | 2019 | Molecular Plant2019,12,12: | 10 |
| 3 | Development of SA-533 Type B CL. 1+SA-240 Type 304L roll-bonded clad steel plate for safety injection tank of CAP1400 nuclear power plant显示文摘Aiming to meet the demand of the country's nuclear demonstration project on the CAP1400 nuclear power plant,Baosteel uses the roll-bonding technology and develops the SA-533 Type B CL. 1+SA-240 Type 304L high-strength and high-toughness clad steel plate with a shear strength of over 310 MPa for the nuclear power plant's safety injection tank. The properties of the quenched and tempered and the simulated post-weld heat treatment states are systematically studied herein through a comprehensive inspection and evaluation of the composition,microstructure,and properties of the clad steel plate. The results show that the bonding interface has high shear strength and that the base metal has high strength and good toughness at low temperatures. Hence,the performance fully meets the technical requirements of the CAP1400 nuclear power plant's safety injection tank in the country's nuclear demonstration project. The results show the cladding interface of the S30403+Q345R roll-bonded clad steel plate has high shear strength,the base metal has good properties,and the mechanical properties of the steel plate head and tail are uniform. | 5 | Production of a monoclonal antibody specific for high molecular weight glutenin subunits (HMW-GS) in wheat and its antigenic determinant显示文摘Wheat high molecular weight glutenin subunits (HMW-GS) 1Bx14 and 1By15 isolated by preparative SDS-PAGE are used as antigen to immunize BALB/c mice. Subcutaneous inoculation of the antigen is performed. The intra-peritoneal injection is completed 3 days before fusion with myeloma cell (SP2/0) via PEG-1500. The fusion cells are selected by indirect enzyme-linked immuno-sorbent assay (ELISA). Positive hybrid cells are further verified three times by limit dilution of the culture cells. A hybridoma cell line is successfully obtained. The monoclonal antibody belongs to IgG1 subclass. In immunoblotting, the antibody binds to all HMW-GS of T.aestivum cultivars, but does not bind to other storage proteins in seeds of wheat. This result is consisting with the high homology in amino acid sequences among the HMW glutenin subunits in wheat. The antibody also binds to HMW-GS storage proteins in Aegilops squarrosa and T. durum (durum wheat). Furthermore, it also binds to HMW storage proteins in Secale cereale (rye),Hordeum vulgare (barley). | 12 | Activation of the Melanocortin-4 receptor signaling byα-MSH stimulates nervedependent mouse digit regeneration显示文摘Background:Expression of Mc4r in peripheral organs indicates it has broader roles in organ homeostasis and regeneration.However,the expression and function of Mc4r in the mouse limb and digit has not been fully investigated.Our previous work showed that Mc4r−/−mice fail to regenerate the digit,but whether activation of MC4R signaling could rescue digit regeneration,or stimulate proximal digit regeneration is not clear.Results:We analyzed the expression dynamics of Mc4r in the embryonic and postnatal mouse limb and digit using the Mc4r-gfp mice.We found that Mc4r-GFP is mainly expressed in the limb nerves,and in the limb muscles that are undergoing secondary myogenesis.Expression of Mc4r-GFP in the adult mouse digit is restricted to the nail matrix.We also examined the effect ofα-MSH on mouse digit regeneration.We found that administration ofα-MSH in the Mc4r+/−mice rescue the delayed regeneration of distal digit tip.α-MSH could rescue distal digit regeneration in denervated hindlimbs.In addition,α-MSH could stimulate regeneration of the proximally amputated digit,which is non-regenerative.Conclusions:Mc4r expression in the mouse limb and digit is closely related to nerve tissues,andα-MSH/MC4R signaling has a neurotrophic role in mouse digit tip regeneration. | Hanqian Xu Hailin Zhang Yanqing Fang Huiran Yang Ying Chen Chao Zhang Gufa Lin | 2021 | Cell Regeneration2021,10,1: | 1 |

The results indicate that this steel is characterized by a bainite microstructure,in which several types of carbides precipitate along the ferrite laths. As the temperature increases from room temperature to 375 ℃,the strength of the steel increases slightly and the fracture toughness clearly decreases. However,when the temperature continues to increase up to 500 ℃,the strength decreases and the fracture toughness increases. At all the temperatures investigated,the strength and toughness of the developed 12Cr2Mo1R steel were capable of meeting the design requirements of a high-temperature gas-cooled reactor. | 19 | The effect of different cooling and tempering conditions on the strength and toughness of SA533B steel显示文摘The microstructure and mechanical properties of SA533B low-alloy steel were investigated under different cooling and tempering conditions. Steel plates cooled at 40℃ /s and 25℃ /min,exibited microstructures of auto-tempered martensite and bainite,respectively. During tempering,the strength of steels quenched at the two different cooling rates decreased continuously with an increase in the tempering parameter,which is defined by T(20 +log t),where T is the temperature and t is the holding time.However,the rate of change of the strength scales with the degree to which the virgin microstructure deviates from equilibrium. The strength of the martensitic steel decreased faster and finally obtained a value close to that of the bainitic steel. The ductility and toughness gradually improved during tempering,passed through a maximum value,and finally diminished. The level of maximum toughness was also found to depend on the cooling rates of SA533B steel.
