| 1 | Transformative hyaluronic acid-based active targeting supramolecular nanoplatform improves long circulation and enhances cellular uptake in cancer therapy显示文摘Hyaluronic acid(HA) is a natural ligand of tumor-targeted drug delivery systems(DDS) due to the relevant CD44 receptor overexpressed on tumor cell membranes. However, other HA receptors(HARE and LYVE-1) are also overexpressing in the reticuloendothelial system(RES). Therefore,polyethylene glycol(PEG) modification of HA-based DDS is necessary to reduce RES capture.Unfortunately, pegylation remarkably inhibits tumor cellular uptake and endosomal escapement,significantly compromising the in vivo antitumor efficacy. Herein, we developed a Dox-loaded HA-based transformable supramolecular nanoplatform(Dox/HCVBP) to overcome this dilemma. Dox/HCVBP contains a tumor extracellular acidity-sensitive detachable PEG shell achieved by a benzoic imine linkage.The in vitro and in vivo investigations further demonstrated that Dox/HCVBP could be in a 'stealth' state at blood stream for a long circulation time due to the buried HA ligands and the minimized nonspecific interaction by PEG shell. However, it could transform into a 'recognition' state under the tumor acidic microenvironment for efficient tumor cellular uptake due to the direct exposure of active targeting ligand HA following PEG shell detachment. | 5 | Experimental study on the shear performance of quasi-NPR steel bolted rock joints显示文摘Quasi-NPR(negative Poisson’s ratio)steel is a new type of super bolt material with high strength,high ductility,and a micro-negative Poisson’s effect.This material overcomes the contrasting characteristics of the high strength and high ductility of steel and it has significant energy-absorbing characteristics,which is of high value in deep rock and soil support engineering.However,research on the shear resistance of quasi-NPR steel has not been carried out.To study the shear performance of quasi-NPR steel bolted rock joints,indoor shear tests of bolted rock joints under different normal stress conditions were carried out.Q235 steel and#45 steel,two representative ordinary bolt steels,were set up as a control group for comparative tests to compare and analyze the shear strength,deformation and instability mode,shear energy absorption characteristics,and bolting contribution of different types of bolts.The results show that the jointed rock masses without bolt reinforcement undergo brittle failure under shear load,while the bolted jointed rock masses show obvious ductile failure characteristics.The shear deformation ca-pacity of quasi-NPR steel is more than 3.5 times that of Q235 steel and#45 steel.No fracture occurs in the quasi-NPR steel during large shear deformation and it can provide stable shear resistance.However,the other two types of control bolts become fractured under the same conditions.Quasi-NPR steel has significant energy-absorbing characteristics under shear load and has obvious advantages in terms of absorbing the energy released by shear deformation of jointed rock masses as compared with ordinary steel.In particular,the shear force plays a major role in resisting the shear deformation of Q235 steel and#45 steel,therefore,fracture failure occurs under small bolt deformation.However,the axial force of quasi-NPR steel can be fully exerted when resisting joint shear deformation;the steel itself does not break when large shear deformation occurs,and the supporting effect of the jointed rock mass is effectively guaranteed. | Manchao He Shulin Ren Haotian Xu Senlin Luo Zhigang Tao Chun Zhu | 2023 | Journal of Rock Mechanics and Geotechnical Engineering2023,15,2: | 1 |

| 8 | Petrogenesis and Tectonic Implications of Jurassic Granites in the Xingcheng Area,Northeastern North China Craton显示文摘New integrated geochemical studies are reported for Jurassic granites of the Xingcheng area in the northeastern North China Craton.U-Pb zircon data indicate that the Huashan and Taili monzogranites were emplaced during the Early(189±2 Ma)and Late(155±1 Ma)Jurassic,respectively.They are typical of high-K calc-alkaline series rocks and I-type granites,according to our whole-rock geochemical researches.Both Early and Late Jurassic monzogranites show adakitic rock characteristics because of their high Sr contents(221-347 ppm)and Sr/Y ratios(28.7-37.5),and low Y contents(7.83-14.7 ppm).The Early Jurassic monzogranite samples have an(^(87)Sr/^(86)Sr)i ratio of 0.7046,ε_(Nd)(t)values of -11.62 to -11.51,and ε_(Hf)(t)values of -13.6 to-6.4,whereas the Late Jurassic monzogranites have higher(87Sr/86Sr)i ratios of 0.7069-0.7071and lower ε_(Nd)(t)(-20.65 to-20.46)and ε_(Hf)(t)(-27.6 to-20.0)values.We suggest that the Early Jurassic adakitic rocks were derived from partial melting of thickened lower crust contaminated with mantle-derived materials,related to subduction of the Paleo-Pacific Plate.The Late Jurassic adakitic rocks were derived from partial melting of thickened lower crust in an extensional tectonic setting associated with an active continental margin. | MU Maosong YANG Debin XU Wenliang QUAN Yikang HAO Leran YANG Haotian WANG Anqi | 2021 | Acta Geologica Sinica(English Edition)2021,95,5: | 1 |
| 9 | Exploring the relationship of hyaluronic acid molecular weight and active targeting efficiency for designing hyaluronic acid-modified nanoparticles显示文摘Although it is reported that the targeting ability of hyaluronic acid(HA)-based nanoparticles(NPs) is molecular weight(MW) dependent,the influence of HA MW on targeting efficiency of HA-functionalized NPs and the underlying mechanism remain elusive. In this study,we constituted three HA-functionalized Dox-loaded NPs(Dox/HCVs) different HA MWs(7,63,and 102 k Da) and attempted to illustrate the effects of HA MW on the targeting efficiency.The three Dox/HCVs had similar physiochemical and pharmaceutical characteristics,but showed different affinity to CD44 receptor. Furthermore,Dox/HCV-63 exerted the best targeting effect and the highest cytotoxicity compared with Dox/HCV-7 and Dox/HCV-102. It was interesting to found that both the HA-CD44 binding affinity and induced CD44 clustering by HA-based NPs were HA MW-dependent,the two of which determine the apparent targeting efficacy of Dox/HCV NPs in the conflicting directions. | 10 | Moving target defense of routing randomization with deep reinforcement learning against eavesdropping attack显示文摘Eavesdropping attacks have become one of the most common attacks on networks because of their easy implementation. Eavesdropping attacks not only lead to transmission data leakage but also develop into other more harmful attacks. Routing randomization is a relevant research direction for moving target defense, which has been proven to be an effective method to resist eavesdropping attacks. To counter eavesdropping attacks, in this study, we analyzed the existing routing randomization methods and found that their security and usability need to be further improved. According to the characteristics of eavesdropping attacks, which are “latent and transferable”, a routing randomization defense method based on deep reinforcement learning is proposed. The proposed method realizes routing randomization on packet-level granularity using programmable switches. To improve the security and quality of service of legitimate services in networks, we use the deep deterministic policy gradient to generate random routing schemes with support from powerful network state awareness. In-band network telemetry provides real-time, accurate, and comprehensive network state awareness for the proposed method. | 11 | Unbiased transcriptomic analyses reveal distinet effects of immune deficiency in CNS function with and without injury显示文摘The mammalian central nervous system (CNS) is considered an immune privileged system as it is separated from the periphery by the blood brain barrier (BBB). Yet, immune functions have been postulated to heavily influence the functional state of the CNS, especially after injury or during neurodegeneration. There is controversy regarding whether adaptive immune responses are beneficial or detrimental to CNS injury repair. In this study, we utilized immunocompromised SCID mice and subjected them to spinal cord injury (SCI). We analyzed motor function, electrophysiology, histochemistry, and performed unbiased RNA-sequencing. SCID mice displayed improved CNS functional recovery compared to WT mice after SCI. Weighted gene-coexpression network analysis (WGCNA) of spinal cord transcriptomes revealed that SCID mice had reduced expression of immune function-related genes and heightened expression of neural transmission-related genes after SCI, which was confirmed by immunohistochemical analysis and was consistent with better functional recovery. Transcriptomic analyses also indicated heightened expression of neurotransmission-related genes before injury in SCID mice, suggesting that a steady state of immune-deficiency potentially led to CNS hyper-connectivity. Consequently, SCID mice without injury demonstrated worse performance in Morris water maze test. Taken together, not only reduced inflammation after injury but also dampened steady-state immune function without injury heightened the neurotransmission program, resulting in better or worse behavioral outcomes respectively. | 16 | Qualitative and quantitative evaluation of Flos Puerariae by using chemical fingerprint in combination with chemometrics method显示文摘In order to better control the quality of Flos Puerariae(FP),qualitative and quantitative analyses were initially performed by using chemical fingerprint and chemometrics methods in this study.First,the fingerprint of FP was developed by HPLC and the chemical markers were screened out by similarity analysis(SA),hierarchical clustering analysis(HCA),principal components analysis(PCA),and orthogonal partial least squares discriminant analysis(OPLS-DA).Next,the chemical constituents in FP were profiled and identified by HPLC coupled to Fourier transform ion cyclotron resonance mass spectrometry(HPLCFT-ICR MS).Then,the characteristic constituents in FP were quantitatively analyzed by HPLC.As a result,31 common peaks were assigned in the fingerprint and 6 of them were considered as qualitative markers.A total of 35 chemical constituents were detected by HPLC-FT-ICR MS and 16 of them were unambiguously identified by comparing retention time,UV absorption wavelength,accurate mass,and MS/MS data with those of reference standards.Subsequently,the contents of glycitin,genistin,tectoridin,glycitein,genistein,and tectorigenin in 13 batches of FP were detected,ranging from 0.4438 to 11.06 mg/g,0.955 to 1.726 mg/g,9.81 to 57.22 mg/g,3.349 to 41.60 mg/g,0.3576 to 0.989 mg/g,and 2.126 to 9.99 mg/g,respectively.In conclusion,fingerprint analysis in combination with chemometrics methods could discover chemical markers for improving the quality control standard of FP.It is expected that the strategy applied in this study will be valuable for further quality control of other traditional Chinese medicines. | Jing Han Ke Xu Quanxiang Yan Wenwen Sui Haotian Zhang Sijie Wang Zan Zhang Ziyun Wei Fei Han | 2022 | Journal of Pharmaceutical Analysis2022,12,3: | 0 |
