| 2 | Natural Variation in CCD4 Promoter Underpins Species-Specific Evolution of Red Coloration in Citrus Peel显示文摘Carotenoids and apocarotenoids act as phytohormones and volatile precursors that influence plant development and confer aesthetic and nutritional value critical to consumer preference.Citrus fruits display considerable natural variation in carotenoid and apocarotenoid pigments.In this study,using an integrated genetic approach we revealed that a 5;c/s-regulatory change at CCD4b encoding CAROTENOID CLEAVAGE DIOXYGENASE 4b is a major genetic determinant of natural variation in C3 0 apocarotenoids responsible for red coloration of citrus peel.Functional analyses demonstrated that in addition the known role in synthesizing 3-citraurin,CCD4b is also responsible for the production of another important C3 0 apocarotenoid pigment,p-citraurinene.Furthermore,analyses of the CCD4b promoter and transcripts from various citrus germplasm accessions established a tight correlation between the presence of a putative 5'c/s-regulatory enhancer within an MITE transposon and the enhanced allelic expression of CCD4b in C3 0 apocarotenoid-rich red-peeled accessions.Phylogenetic analysis provided further evidence that functional diversification of CCD4b and naturally occurring variation of the CCD4b promoter resulted in the stepwise evolution of red peels in mandarins and their hybrids.Taken together,our findings provide new insights into the genetic and evolutionary basis of apocarotenoid diversity in plants,and would facilitate breeding efforts that aim to improve the nutritional and aesthetic value of citrus and perhaps other fruit crops. | Xiongjie Zheng Kaijie Zhu Quan Sun Weiyi Zhang Xia Wang Hongbo Cao Meilian Tan Zongzhou Xie Yunliu Zeng Junli Ye Lijun Chai Qiang Xu Zhiyong Pan Shunyuan Xiao Paul D.Fraser Xiuxin Deng | 2019 | Molecular Plant2019,12,9: | 18 |
| 3 | CPSF30-L-mediated recognition of mRNA m^(6)A modification controls alternative polyadenylation of nitrate signaling-related gene transcripts in Arabidopsis显示文摘N6-methyladenosine(m^(6)A),a ubiquitous internal modification of eukaryotic mRNAs,plays a vital role in almost every aspect of mRNA metabolism.However,there is little evidence documenting the role of m^(6)A in regulating alternative polyadenylation(APA)in plants.APA is controlled by a large protein-RNA complex with many components,including CLEAVAGE AND POLYADENYLATION SPECIFICITY FACTOR30(CPSF30).In Arabidopsis,CPSF30 has two isoforms and the longer isoform(CPSF30-L)contains a YT512-B Homology(YTH)domain,which is unique to plants.In this study,we showed that CPSF30-L YTH domain binds to m^(6)A in v itro.In the cpsf30-2 mutant,the transcripts of many genes including several important nitrate signaling-related genes had shifts in polyadenylation sites that were correlated with m^(6)A peaks,indicating that these gene transcripts carrying m^(6)A tend to be regulated by APA.Wild-type CPSF30-L could rescue the defects in APA and nitrate metabolism in cpsf30-2,but m^(6)A-binding-defective mutants of CPSF30-L could not.Taken together,our results demonstrated that m^(6)A modification regulates APA in Arabidops is and revealed that the m^(6)A reader CPSF30-L affects nitrate signaling by controlling APA,shedding new light on the roles of the m^(6)A modification during RNA 3-end processing in nitrate metabolism. | Yiteng Hou Jing Sun Baixing Wu Yangyang Gao Hongbo Nie Zhentian Nie Shuxuan Quan Yong Wang Xiaofeng Cao Sisi Li | 2021 | Molecular Plant2021,14,4: | 12 |
| 4 | Virtual network function scheduling via multilayer encoding genetic algorithm with distributed bandwidth allocation显示文摘Network function virtualization represents a revolutionary approach to network service deployment. This software-oriented approach for virtual network functions(VNFs) deployment enables more flexible and dynamic network services to meet diversified demands. To minimize the execution time of all VNFs in service function chains, VNF scheduling must be addressed. In this paper, we improve upon the flexible job-shop model by introducing the process of bandwidth allocation. First, we propose a multilayer encoding genetic algorithm to solve the VNF scheduling model. In addition, we design a distributed method for bandwidth allocation based on the Nash bargaining solution. Finally, by combining the genetic algorithm with distributed bandwidth allocation, we present a heuristic algorithm that solves the VNF scheduling problem in one stage. Using a multilayer encoding genetic algorithm, we simplify the constraints of the VNF scheduling problem and reduce its time complexity. At the same time, our Nash game solution refines the granularity of bandwidth allocation to further reduce the transmission delay between VNFs. The effectiveness of our proposed heuristic algorithm is verified through numerical evaluation. | 5 | Flavanols from the Camellia sinensis var.assamica and their hypoglycemic and hypolipidemic activities显示文摘α-Glucosidase and lipase inhibitors play important roles in the treatment of hyperglycaemia and dyslipidemia. To identify novel naturally occurring inhibitors, a bioactivity-guided phytochemical research was performed on the pu-erh tea. One new flavanol, named(–)-epicatechin-3-O-(Z)-coumarate(1), and 16 known analogs(2–17) were isolated from the aqueous extract of the pu-erh tea. Their structures were determined by spectroscopic and chemical methods. Furthermore, the water extract of pu-erh tea and its fractions exhibited inhibitory activities against α-glucosidases and lipases in vitro;compound 15 showed moderate inhibitory effect against sucrase with an IC_(50) value of 32.5 μmol/L and significant inhibitory effect against maltase with an IC_(50) value of 1.3 μmol/L. [Method] Different treatments on weed control were conducted in tobacco fields in Anshun City, Guizhou Province, from 2017 to 2019. [Result] Various treatments had no negative effect on tobacco plant growth in the field, and had different degrees of control effects on five dominant weeds, including Digitaria sanguinalis, Setaria viridis, Fagopyrum dibotrys, Commelina communis and Chenopodium album. White mulching film and 50% butralin·clomazone EC 160 m L/667 m^(2) + white mulching film had the worst performance, which had extremely significant or significant differences with other treatments. There was no sig-nificant difference among most treatments, and the overall effects were comprehensive(multi-factor) treatment > double factor treatment > single factor treatment.
