1Micelle or polymersome formation by PCL-PEG-PCL copolymers as drug delivery systems显示文摘Polye-caprolactone)-b-poly(ethylene glycol)-b-poly(e-caprolactone)(PCL-b-PEG-b-PCL,PCEC) triblock copolymers have been widely investigated in last several decades.Here,by altering the weight ratio of monomers in ring-opening polymerization,a series of PCEC triblock copolymers with varying hydrophobicity were synthesized,which were characterized by FTIR,1 H NMR,GPC and DSC.When PCEC copolymers with different weight ratios of PCL/PEG were dispersed in different aqueous solutions,they could self-assemble and form two distinctive nanoparticular structures:micelles or polymersomes.We then chose paclitaxel(PTX) as the model drug and encapsulate PTX into PCEC polymeric micelles and polymersomes.The physicochemical characterizations of the nanoparticles such as morphology,the size and distribution,zeta potential,drug loading content,and encapsulation efficiency were also performed.Our results showed that polymeric micelles or polymersomes from PCEC both displayed narrow size distributions and could achieve high drug loading efficiencies.In vitro cellular uptake results suggested that Nile Red loaded polymeric micelles or polymersomes displayed more internalization after 24 h incubation than those after 4 h incubation.These findings suggest that polymeric micelles and polymersomes based on PCL-b-PEG-b-PCL copolymers have great potential to effectively delivery hydrophobic drugs.Chunyan Hu Zhuo Chen Shengjie Wu Yanfeng Han Hai Wang Hongfan Sun Deling Kong Xigang Leng Chun Wang Linhua Zhang Dunwan Zhu 2017Chinese Chemical Letters2017,28,9:4
2A competitive RT-PCR-based approach to quantifying promoter activity of stress gene显示文摘Chloramphenicol acetyltransferase (CAT) as a stable reporter has been broadly adopted to detect promoter activity of many stress genes in eukaryotic cells. A simplified, convenient and relatively accurate assay to reveal CAT activities driven by heat shock gene promoter upon cell stress is reported here. A CAT mutant control plasmid pMCAT is constructed out of pBLCAT3, with a DNA segment within the CAT gene deleted. The control plasmid pMCAT along with the wild type CAT reporter gene driven by the regulatory sequence of hsp90β gene are co-transfected into Jurkat cells. Following total cellular RNA extracted and reverse transcribed, PCR is carried out to amplify two individual fragments with the same pair of synthesized primers, which bind to the identical sequences in both pMCAT and the CAT reporter plasmids. The two PCR fragments amplified from the reporter and control RNAs are identified by their size differences in agarose gel electrophoresis. The ratio of intensity of the two PCR bands in the same lane is taken as 'relative units of promoter efficiency' of a gene. The transcripts instead of CAT activity thus detected, may not only provide a closer look at the promoter activity of the stress gene but also normalize the efficiency of transfection simultaneously. With experimental conditions optimized, visible and reliable results can be obtained without using any sophisticated instruments. Since the results obtained with the RT-PCR approach are reproducible, much less troublesome and time-consuming, and comparable to our previous data, we thus take it as a conventional procedure for promoter activity assay of human hsp gene upon heat shock. 3Highly-rough surface carbon nanofibers film as an effective interlayer for lithium–sulfur batteries显示文摘Lithium–sulfur(Li–S)battery with a new configuration is demonstrated by inserting a flexible nitrogen-doping carbon nanofiber(N-CNFs)interlayer between the sulfur cathode and the separator.The N-CNFs film with high surface roughness and surface area is fabricated by electrospinning and a subsequent calcination process.The N-CNFs film interlayer not only effectively traps the shuttling migration of polysulfides but also gives the whole battery reliable electronic conductivity,which can effectively enhance the electrochemical performance of Li–S batteries.Finally,Li–S batteries with long cycling stability of 785 mAh/g after 200 cycles and good rate capability of 573 mAh/g at 5 C are achieved.Hongfan Zhu Mo Sha Huaping Zhao Yuting Nie Xuhui Sun Yong Lei 2020Journal of Semiconductors2020,41,9:3
4Adhesive anastomosis for organ transplantation显示文摘The recent development of tough tissue adhesives has stimulated intense interests among material scientists and medical doctors.However,these adhesives have seldom been tested in clinically demanding surgeries.Here we demonstrate adhesive anastomosis in organ transplantation.Anastomosis is commonly conducted by dense sutures and takes a long time,during which all the vessels are occluded.Prolonged occlusion may damage organs and even cause death.We formulate a tough,biocompatible,bioabsorbable adhesive that can sustain tissue tension and pressurized flow.We expose the endothelial surface of vessels onto a gasket,press two endothelial surfaces to the adhesive using a pair of magnetic rings,and reopen the bloodstream immediately.The time for adhesive anastomosis is shortened compared to the time for sutured anastomosis.We have achieved adhesive anastomosis of a great vein in transplanting the liver of a pig.After the surgery,the adhesive is absorbed,the vein heals,and the pig lives for over one month.Kang Liu Hang Yang Gaobo Huang Aihua Shi Qiang Lu Shanpei Wang Wei Qiao Haohua Wang Mengyun Ke Hongfan Ding Tao Li Yanchao Zhang Jiawei Yu Bingyi Ren Rongfeng Wang Kailing Wang Hui Feng Zhigang Suo Jingda Tang Yi Lv 2022Bioactive Materials2022,7,7:3

6Study design,general characteristics of participants,and preliminary findings from the metabolome,microbiome,and dietary salt intervention study(MetaSalt)显示文摘Background:High sodium intake is an important risk factor for hypertension and cardiovascular disease.However,the association between gut microbiota composition and metabolomic profiles with dietary sodium intake and blood pressure(BP)is not well-understood.The metabolome,microbiome,and dietary salt intervention(MetaSalt)study aimed to investigate microbial and metabolomic profiles related to dietary sodium intake and BP regulation.Methods:This family-based intervention study was conducted in four communities across three provinces in rural northern China in 2019.Probands with untreated prehypertension or stage-1 hypertension were identified through community-based BP screening,and family members including siblings,offspring,spouses,and parents were subsequently included.All participants participated in a 3-day baseline examination with usual diet consumption,followed by a 10-day low-salt diet(3 g/d of salt or 51.3 mmol/d of sodium)and a 10-day high-salt diet(18 g/d of salt or 307.8 mmol/d of sodium).Differences in mean BP levels were compared according to the intervention phases using a paired Student’s t-test.Results:A total of 528 participants were included in this study,with a mean age of 48.1 years,36.7%of whom were male,76.8%had a middle school(69.7%)or higher(7.1%)diploma,23.4%had a history of smoking,and 24.4%were current drinkers.The mean arterial pressure at baseline was 97.2±10.5 mm Hg for all participants,and significantly decreased during the low-salt intervention(93.8±9.3,P<0.0001)and subsequently increased during the high-salt intervention(96.4±10.0,P<0.0001).Conclusions:Our dietary salt intervention study has successfully recruited participants and will facilitate to evaluate the effects of gut microbiota and metabolites on BP regulation in response to sodium burden,which will provide important evidence for investigating the underlying mechanisms in the development of hypertension and subsequent cardiovascular diseases.Trial registration:The study was registered in the Chinese Clinical Trial Registry database(ChiCTR1900025171).Zengliang Ruan Jianxin Li Fangchao Liu Jie Cao Shufeng Chen Jichun Chen Keyong Huang Yaqin Wang Hongfan Li Yan Wang Zhongyu Xue Laiyuan Wang Jianfeng Huang Dongfeng Gu Xiangfeng Lu 2021Chronic Diseases and Translational Medicine2021,7,4:1

9DNA Barcoding Assessment of Green Macroalgae in Coastal Zone Around Qingdao, China显示文摘An assessment with assistance of DNA barcoding was conducted on green macroalgae in coastal zone around Qingdao, China, during the period of April- December, 2011. Three markers were applied in molecular discrimination, including the plastid elongation factor tufA gene, the internal transcribed spacer(ITS) region of the ribosomal cistron and rubisco large subunit gene 3' regions(rbcL-3P). DNA barcoding discriminated 8 species, excluding species of genus Cladophora and Bryopsis due to failures in amplification. We ascertained and corrected 4 species identified by morphological methods for effectively assisting the classification. The gene tufA presented more advantages as an appropriate DNA marker with the strongest amplification success rate and species discrimination power than the other two genes. The poorest sequencing success largely handicapped the application of ITS. Samples identified by tufA and rbcL as Ulva flexuosa were clustered into the clade of U. prolifera by ITS in the neighbor-joining tree. Confusion with discrimination of the complex of U. linza, U. procera and U. prolifera(as the LPP complex) still existed for the three DNA markers. Based on our results, rbcL is recommended as a preferred marker for assisting tufA to discriminate green macroalgae. In distinguishing green-tide-forming Ulva species, the free-floating sample collected from the green tide in 2011 was proved to be identical with U. prolifera in Yellow Sea for ITS and rbcL genes. The scheme makes both sides in equal positions during the course of electronic transaction.
