2trans-2-Enoyl-CoA Reductase Tecr-Driven Lipid Metabolism in Endothelial Cells Protects against Transcytosis to Maintain Blood-Brain Barrier Homeostasis显示文摘The transport and metabolism of lipids in cerebrovascular endothelial cells(ECs)have been hypothesized to regulate blood-brain barrier(BBB)maturation and homeostasis.Long-chain polyunsaturated fatty acids(LCPUFAs)as the important lipids components of cell membranes are essential for the development and function of BBB,but the direct links of lipid metabolism and ECs barrier function remain to be established.Here,we comprehensively characterize the transcriptomic phenotype of developmental cerebrovascular ECs in single-cell resolution and firstly find that trans-2-enoyl-CoA reductase(Tecr),a verylong-chain fatty acid synthesis,is highly expressed during barriergenesis and decreased after BBB maturation.EC-specific knockout of Tecr compromises angiogenesis due to delayed vascular sprouting.Importantly,EC-specific deletion of Tecr loss restrictive quality of vascular permeability from neonatal stages to adulthood,with high levels of transcytosis,but maintains the vascular tight junctions.Moreover,lipidomic analysis shows that the expression of Tecr in ECs is associated with the containing of omega-3 fatty acids,which directly suppresses caveolae vesicles formation.These results reveal a protective role for Tecr in BBB integrity and suggest that Tecr as a novel therapeutic target in the central nervous system(CNS)diseases associated with BBB dysfunction.Jinxuan Wang Jianxiong Xu Guangchao Zang Tao Zhang Qi Wu Hongping Zhang Yidan Chen Yi Wang Weixi Qin Shuang Zhao Erdai Qin Juhui Qiu Xiaojuan Zhang Lin Wen Yeqi Wang Guixue Wang 2022Research2022,,3:1

5Biogas residue biochar integrated with phosphate from its ash for the effective recovery of nutrients from piggery biogas slurry显示文摘Magnesium ammonium phosphate(MAP)precipitation generally requires an external phosphorus source to increase the recovery of NH_(4)^(+)-N from biogas slurry.However,P-rich piggery biogas residue has been ignored as a phosphorus source.In this study,biogas residue was carbonized into biogas residue biochar(BRC),followed by acid leaching to synthesize functionalized BRC and release PO_(4)^(3−)-P from its ash as the phosphorus source.The effects of different acids on the leaching efficiency and morphological changes of P in BRC were investigated,and NH_(4)^(+)-N and PO_(4)^(3−)-P in the biogas slurry were recovered with functionalized BRC and MAP precipitation.The results showed that oxalic acid-hydrochloric acid mixed acid could leach more than 96%of P in BRC,while weakening the inhibitory effect of Ca^(2+)on MAP precipitation.The BRC was mainly composed of inorganic P,and most nonapatite IP and apatite P(Ca_(3)(PO_(4))2)were leached during acid leaching,with the latter more easily leached.Under optimal recovery conditions,the method had a significant recovery effect on NH_(4)^(+)-N(96.4%)and PO_(4)^(3−)-P(99.3%)in biogas slurry.The recovery of NH_(4)^(+)-N and PO_(4)^(3−)-P by functionalized BRC was mainly through chemical precipitation(forming NH_(4)MgPO_(4)·H_(2)O precipitate)while bonding with-OH,C=O and C-H functional groups.The final recovery product was also a BRC-based slow-release N-P fertilizer rich in struvite.This study solved the disposal problems of P-rich biogas residue and N-rich biogas slurry while providing an innovative technology for the resource utilization of faecal sewage at pig farms.Zifeng Luo Hongping Wen Haotian Zhang Yunliang Li Xueyi Mai Yulong Zhang Jinjin Wang Yongtao Li Zhen Zhang 2022Biochar2022,4,1:1
6Schaftoside inhibits 3CL^(pro)and PL^(pro)of SARS-CoV-2 virus and regulates immune response and inflammation of host cells for the treatment of COVID-19显示文摘It is an urgent demand worldwide to control the coronavirus disease 2019(COVID-19)pandemic caused by the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)virus.The 3-chymotrypsin-like protease(3CL^(pro))and papain-like protease(PL^(pro))are key targets to discover SARS-CoV-2 inhibitors.After screening 12 Chinese herbal medicines and 125 compounds from licorice,we found that a popular natural product schaftoside inhibited 3CL^(pro)and PL^(pro)with IC_(50)values of 1.73±0.22 and 3.91±0.19μmol/L,respectively,and inhibited SARS CoV-2 virus in Vero E6 cells with EC_(50)of 11.83±3. 23μmol/L. Hydrogen-deuterium exchange mass spectrometry analysis, quantum mechanics/molecular mechanics calculations, together with site-directed mutagenesis indicated the antiviral activities of schaftoside were related with non-covalent interactions with H41, G143 and R188 of3CLpro, and K157, E167 and A246 of PLpro. Moreover, proteomics analysis and cytokine assay revealed that schaftoside also regulated immune response and inflammation of the host cells. The antiinflammatory activities of schaftoside were confirmed on lipopolysaccharide-induced acute lung injury mice. 7Effects of Different Feeding Patterns on the Growth of Micropterus salmoides显示文摘Three experimental groups with different feeding frequencies,different feeding rates and compensatory feeding were set up to study their effects on the growth of Micropterus salmoides.The results showed that different feeding rates of 2%,3%and 4%had no significant effects on the growth of M.salmoides.Therefore,before the individual weight of 50 g,the fish could be fed with a feeding rate of 4%,which could be gradually decreased to 2%in the later period.Under the condition of relatively fixed feeding rate,different feeding frequencies had an impact on the growth of M.salmoides.Under the conditions of suitable water temperature,it is reasonable to feed twice daily.The experimental fish fed for 5 d and then deprived of feed for 1 d showed fully compensatory growth,and there were no significant differences in weight gain rate,daily weight gain and specific growth rate compared with the continuous feeding group(P>0.05).Therefore,according to the feeding conditions of fish in the breeding process,stopping feeding for 1 d every 5 d could save feed and labor and improve breeding efficiency.Yuzhang HAN Zhourui WEN Aiwu YI Jun HUANG Zhiqiang ZHU Wenyu TONG Hongping ZHENG 2023Agricultural Biotechnology2023,12,2:0
