| 2 | APOLIPOPROTEIN E GENE POLYMORPHISMS AND RISK FOR CORONARY ARTERY DISEASE IN CHINESE XINJIANGUYGUR AND HAN POPULATION显示文摘Objective To examine the relationship between apolipoprotein E (Apo E) gene polymorphism and risk of coronary artery disease (CAD), analyzing association of polymorphism with classical risk factors. Methods A total of 124 patients (including 84 Han population and 40 Uygur population) with angiographically verified CAD or myocardial infarction were prospectively evaluated. Data referring to hypertension, diabetes, and tobacco consump-tion were recorded. The levels of total cholesterol (TC), high density lipoprotein (HDL) cholesterol, Apo A1 and B, and triglycerides (TG) were determined. DNA was obtained from 124 patients and 70 controls. In order to determine Apo E genotypes, DNA was PCR amplified and digested with HhaI. The genetic polymorphism of Apo E is due to three common alleles, epsilon(ε) 2, ε3, ε4, at a single autosomal gene locus. These alleles determine the six phenotypes E2/2, E3/3, E4/4, E4/2, E4/3, and E3/2. Results In Uygur population, the frequency of the ε2, ε3, and ε4 was 0.155, 0.648, and 0.197 respectively. In Han po-pulation, the frequency of the ε2, ε3, and ε4 was 0.081, 0.772, and 0.146 respectively. In the patient group, the frequency of the ε2, ε3, and ε4was 0.060, 0.758, and 0.182 respectively. In the control group, the frequency of the ε2, ε3, and ε4 was 0.193, 0.671, and 0.136 respectively. ε2 frequency of Uygur’ patients and controls was 0.050 and 0.290 respectively. Serum low density lipoprotein (LDL) cholesterol, TC, and TG values tended to decrease from the Apo E-4 phenotypes to Apo E-2 phenotypes. When deletion polymorphism of ε2 was compared with the common risk factors for CAD, its risk ratio (RR) is 4.38. Conclusions These studies confirm and find that Apo E phenotype distribution in Uygur population differs significantly from that in Han population in Xinjiang. CAD patients have significantly lower ε2 allele and slightly higher ε3 or ε4 allele frequency than controls, especially in Uygur population. | 3 | TAOK1 negatively regulates IL-17-mediated signaling and inflammation显示文摘Interleukin 17(IL-17)is an important cytokine that can induce tissue inflammation and is involved in the pathogenesis of numerous autoimmune diseases.However,the regulation of its signaling transduction has not been well described.In this study,we report that thousand and one kinase 1(TAOK1)functions as a negative regulator of IL-17-mediated signal transduction and inflammation.TAOK1 knockdown promotes IL-17-induced cytokine and chemokine expression and the activation of mitogen-activated protein kinases and nuclear factor-κB.We further demonstrate that TAOK1 interacts with IL-17 receptor A(IL-17RA)independent of its kinase activity,and TAOK1 dose-dependently prevents the formation of the IL-17R-Act1(nuclear factor activator 1,also known as tumor necrosis factor receptor-associated factor 3 interacting protein 2)complex.Consistent with this,TAOK1 deficiency exacerbates colitis in the 2,4,6-trinitrobenzenesulfonic acid-induced experimental model of inflammatory bowel disease,likely by its promotion of the IL-17-mediated signaling pathway.TAOK1 expression is decreased in the colons of ulcerative colitis patients.In conclusion,these findings suggest that TAOK1 is involved in the development of IL-17-related autoimmune disorders. | Zhaoru Zhang Zhen Tang Xianwei Ma Kai Sun Liping Fan Jie Fang Jianping Pan Xiaojian Wang Huazhang An Jun Zhou | 2018 | Cellular & Molecular Immunology2018,15,8: | 10 |
| 4 | Effect of the graphitic degree of carbon supports on the catalytic performance of ammonia synthesis over Ba-Ru-K/HSGC catalyst显示文摘A series of high surface area graphitic carbon materials(HSGCs) were prepared by ball-milling method. Effect of the graphitic degree of HSGCs on the catalytic performance of Ba-Ru-K/HSGC-x(x is the ball-milling time in hour) catalysts was studied using ammonia synthesis as a probe reaction. The graphitic degree and pore structure of HSGC-x supports could be successfully tuned via the variation of ball-milling time. Ru nanoparticles of different Ba-Ru-K/HSGC-x catalysts are homogeneously distributed on the supports with the particle sizes ranging from 1.6 to 2.0 nm. | 7 | MEF2C promotes M1 macrophage polarization and Th1 responses显示文摘The polarization of macrophages to the M1 or M2 phenotype has a pivotal role in inflammation and host defense;however,the underlying molecular mechanism remains unclear.Here,we show that myocyte enhancer factor 2 C(MEF2C)is essential for regulating M1 macrophage polarization in response to infection and inflammation.Global gene expression analysis demonstrated that MEF2C deficiency in macrophages downregulated the expression of M1 phenotypic markers and upregulated the expression of M2 phenotypic markers.MEF2C significantly promoted the expression of interleukin-12 p35 subunit(Il12a)and interleukin-12 p40 subunit(Il12b).Myeloid-specific Mef2c-knockout mice showed reduced IL-12 production and impaired Th1 responses,which led to susceptibility to Listeria monocytogenes infection and protected against DSS-induced IBD in vivo.Mechanistically,we showed that MEF2C directly activated the transcription of Il12a and Il12b.These findings reveal a new function of MEF2C in macrophage polarization and Th1 responses and identify MEF2C as a potential target for therapeutic intervention in inflammatory and autoimmune diseases. | Xibao Zhao Qianqian Di Han Liu Jiazheng Quan Jing Ling Zizhao Zhao Yue Xiao Han Wu Zherui Wu Wengang Song Huazhang An Weilin Chen | 2022 | Cellular & Molecular Immunology2022,19,4: | 4 |

| 9 | La_2Ce_2O_7 supported ruthenium as a robust catalyst for ammonia synthesis显示文摘La_2Ce_2O_7 nanoparticles were prepared by citric acid complexation method followed by calcination at varied temperatures. Then, supported with 4 wt% Ru, they were evaluated as the catalysts for ammonia synthesis under conditions similar with industry. With La_2Ce_2O_7 being calcinated at 700 or 800℃, the experimental results indicate that the Ru/La_2Ce_2O_7 catalyst exhibits much higher ammonia concentration or ammonia synthesis rate than that of Ru/CeO_2 and Ru/La_2O_3. In addition, Ru/La_2Ce_2O_7 possesses high stability under over-heating test. In the absence of any promotor, ammonia concentration of Ru/La_2Ce_2O_7 catalyst approaches 14% at 450℃, GHSV of 10000 h^(-1) and pressure of 10 MPa. The rate-determining step of ammonia synthesis, dissociation of N_2 is significantly facilitated by the strong metalesupport interaction(SMSI) between Ru and La_2Ce_2O_7. Due to the interaction, La_2Ce_2O_7 tends to donate electrons to Ru,resulting in the high electron density over the surface of Ru active sites which is favorable for the dissociation of N_2. | 10 | A Generalized Acceleration and Deceleration Approach for Continuous Small Line Blocks with Look-ahead显示文摘Generally complex 3D contours are divided into a lot of continuous small line blocks by CAD/CAM software. The defective CNTs-D supported Ba–Ru/CNTs-D catalysts exhibit superior catalytic performance in ammonia synthesis with a TOF be increased up to 0.30 s-1, which is 2.5 times of oxidized CNTs-O supported Ba–Ru/CNTs-O catalysts and 5 times of the Ba–Ru/CNTs. The characterizations by CO chemisorption, transmission electron microscope, Raman, and X-ray photoelectron spectroscopy revealed that the uniformly well dispersed Ru NPs can be stabilized on the defective sites of CNTs-D. | 12 | EFFECT OF OXIDIZED-LDL ON NF-κB NUCLEAR TRANSLOCATION IN AORTIC SMOOTH MUSCLE CELLS ORIGINATED FROM RATS OF DIFFERENT AGES显示文摘Objective To investigate the molecular mechanism of atherosclerosis that related to age. Methods Immunohistochemistry staining and Western blot were adopted to determine the nuclear translocation of nuclear factor-kappa B (NF-κB) and expression of platelet-derived growth factor B (PDGF-B) in smooth muscle cells (SMCs) co-cultured with low density lipoprotein (LDL), oxidized LDL (ox-LDL), and ox-LDL+high density lipoprotein (HDL) originated from rats of 2 and 10 months old respectively. Fat stain was used to identify the lipid intake in SMCs. Results The optimal stimulation time of ox-LDL to SMCs was 12 hours. NF-κB intensity increased in most nuclei of SMCs that originated from rats of either 2 or 10 months old co-cultured with ox-LDL. The intensity of NF-κB and the amount of intracellular lipid taken in SMCs were more obvious in cells from 10-month-old rats than from the younger ones. Change of PDGF-B expression in SMCs was not remarkable in each group of rats. Conclusions The 10-month-old rats are more susceptive to ox-LDL than 2-month-old rats in activating nuclear transloca- tion of NF-κB. Maybe this is one of the important reasons contributing to the difference between the older and younger rats on the initiation and development of atherosclerosis lesion. | 14 | Synthesis and characterization of semiconductor zinc sulfide nanotubes by soft-template method显示文摘ZnS nanotubes have been successfully synthe-sized from solutions containing a surfactant, Triton X-100(t-octyl-(OCH2CH2)χOH, χ=9, 10). X-ray diffraction (XRD),transmission electron microscope (TEM) and selected areaelectron diffraction (SAED) are employed to characterize thestructure and morphology of as-synthesized product. XRDand SAED pattern indicate that as-obtained products consist of pure polycrystalline cubic-phase ZnS structures. TEM images reveal that most of the products are tubular struc-tures, with diameters ranging between 37--52 nm andlengths up to 3 μm. The wall thickness of as-obtained ZnSnanotube is around 9 nm.
