| 1 | An experimental investigation on the mechanism of fluid flow through single rough fracture of rock显示文摘The structure of fractures in nature rock appears irregular and induces complicated seepage flow behavior.The mechanism and quantitative description of fluid flow through rock fractures is a difficult subject that has been greatly concerned in the fields of geotechnical,mining,geological,and petroleum engineering.In order to probe the mechanism of fluid flow and the effects of rough structures,we conducted a few laboratory tests of fluid flow through single rough fractures,in which the Weierstrass-Mandelbrot fractal function and PMMA material were employed to produce the fracture models with various fractal roughnesses.A high-speed video camera was employed to record the fluid flow through the entire single rough fracture with a constant hydraulic pressure.The properties of fluid flow varying with the fracture roughness and the influences of the rough structure were analyzed.The components of flow resistance of a single rough fracture were discussed.A fractal model was proposed to relate the fluid resistance to the fracture roughness.A fractal equivalent permeability coefficient of a single rough fracture was formulated.This study aims to provide an experimental basis and reference for better understanding and quantitatively relating the fluid flow properties to the structures of rock fractures. | JU Yang ZHANG QinGang YANG YongMing XIE HePing GAO Feng WANG HuiJie | 2013 | Science China(Technological Sciences)2013,56,8: | 28 |
| 2 | Adoptive cell transfer therapy for hepatocellular carcinoma显示文摘Hepatocellular carcinoma (HCC) is one of the most common cancers worldwide. This malignancy is associated with poor prognosis and high mortality. Novel approaches for prolonging the overall survival of patients with advanced HCC are urgently needed. The antitumor activities of adoptive cell transfer therapy (ACT), such as strategies based on tumor-infiltrating lymphocytes and cytokine-induced killer cells, are more effective than those of traditional strategies. Currently, chimeric antigen receptor T-cell (CAR-T) immunotherapy has achieved numerous breakthroughs in the treatment of hematological malignancies, including relapsed or refractory lymphoblastic leukemia and refractory large B-cell lymphoma? Nevertheless, this approach only provides a modest benefit in the treatment of solid tumors. The clinical results of CAR-T immunotherapy for HCC that could be obtained at present are limited. Some published studies have demonstrated that CAR-T could inhibit tumor growth and cause severe side effects. | 3 | Chimeric antigen receptor T cell targeting EGFRvIII for metastatic lung cancer therapy显示文摘Lung cancer is the most common incident cancer and the leading cause of cancer death. In recent years, the development of tumor immunotherapy especially chimeric antigen receptor T (CAR-T) cell has shown a promising future. Epidermal growth factor receptor variant III (EGFRvlll) is a tumor-specific mutation expressed in various types of tumors and has been detected in non-small cell lung cancer with a mutation rate of 10%. Thus, EGFRvIII is a potential antigen for targeted lung cancer therapy. In this study, CAR vectors were constructed and transfected into virus-packaging cells. Then, activated T cells were infected with retrovirus harvested from stable virus-producing single clone cell lines. CAR expression on the surfaces of the T cells was detected by flow cytometry and Western blot. The function of CAR-T targeting EGFRvIII was then evaluated. The EGFRvIII-CAR vector was successfully constructed and confirmed by DNA sequencing. A stable virus-producing cell line was produced from a single clone by limited dilution. The culture conditions for the cell line, including cell density, temperature, and culture medium were optimized. After infection with retrovirus, CAR was expressed on more than 90% of the T cells. The proliferation of CAR-T cells were induced by cytokine and specific antigen in vitro. More importantly, EGFRvIII-CART specifically and efficiently recognized and killed A549-EGFRvIII cells with an effector/target ratio of 10:1 by expressing and releasing cytokines, including perforin, granzyme B, IFN-γ, and TNF-α. The in vivo study indicated that the metastasis of A549-EGFRvIII cells in mice were inhibited by EGFRvIII-CART cells, and the survival of the mice was significantly prolonged with no serious side effects. EGFRvIII-CART showed significantly efficient antitumor activity against lung cancer cells expressing EGFRvlll in vivo and in vitro. | 10 | Effect of low-level laser therapy on tooth-related pain and somatosensory function evoked by orthodontic treatment显示文摘Low-level laser therapy(LLLT) may have an effect on the pain associated with orthodontic treatment. The aim of this study was to evaluate the effect of LLLT on pain and somatosensory sensitization induced by orthodontic treatment. Forty individuals(12–33 years old; mean ± standard deviations: 20.8 ± 5.9 years) scheduled to receive orthodontic treatment were randomly divided into a laser group(LG) or a placebo group(PG)(1:1). The LG received LLLT(810-nm gallium-aluminium-arsenic diode laser in continuous mode with the power set at 400 mW, 2 J·cm–2) at 0 h, 2 h, 24 h, 4 d, and 7 d after treatment, and the PG received inactive treatment at the same time points. In both groups, the non-treated side served as a control. A numerical rating scale(NRS) of pain, pressure pain thresholds(PPTs), cold detection thresholds(CDTs), warmth detection thresholds(WDTs), cold pain thresholds(CPTs), and heat pain thresholds(HPTs) were tested on both sides at the gingiva and canine tooth and on the hand. The data were analysed by a repeated measures analysis of variance(ANOVA). The NRS pain scores were significantly lower in the LG group(P = 0.01). The CDTs,CPTs, WDTs, HPTs, and PPTs at the gingiva and the PPTs at the canine tooth were significantly less sensitive on the treatment side of the LG compared with that of the PG(P < 0.033). The parameters tested also showed significantly less sensitivity on the nontreatment side of the LG compared to that of the PG(P < 0.043). There were no differences between the groups for any quantitative sensory testing(QST) measures of the hand. The application of LLLT appears to reduce the pain and sensitivity of the tooth and gingiva associated with orthodontic treatment and may have contralateral effects within the trigeminal system but no generalized QST effects. | 11 | Biodegradation of indole by a newly isolated Cupriavidus sp. SHE显示文摘Indole, a typical nitrogen heterocyclic aromatic pollutant, is extensively spread in industrial wastewater. Microbial degradation has been proven to be a feasible approach to remove indole, whereas the microbial resources are fairly limited. A bacterial strain designated as SHE was isolated and found to be an efficient indole degrader. It was identified as Cupriavidus sp. according to 16 SrRNA gene analysis. Strain SHE could utilize indole as the sole carbon source and almost completely degrade 100 mg/L of indole within 24 hr. It still harbored relatively high indole degradation capacity within p H 4–9 and temperature 25°C–35°C. Experiments also showed that some heavy metals such as Mn2+, Pb2+and Co2+did not pose severe inhibition on indole degradation. Based on high performance liquid chromatography–mass spectrum analysis, isatin was identified as a minor intermediate during the process of indole biodegradation. A major yellow product with m/z 265.0605(C15H8N2O3) was generated and accumulated, suggesting a novel indole conversion pathway existed. Genome analysis of strain SHE indicated that there existed a rich set of oxidoreductases, which might be the key reason for the efficient degradation of indole. | 12 | Molecular genetic and genomic analysis of wheat milling and end-use traits in China:Progress and perspectives显示文摘Wheat is the most widely cultivated staple food crop, and multiple types of food derivatives are processed and consumed globally. Wheat grain quality(WGQ) is central to food processing and nutritional value, and is a decisive factor for consumer acceptance and commercial value of wheat cultivars. Hence, improvement in WGQ traits is top priority for both conventional and molecular wheat breeding. In this review we will focus on two important WGQ traits, grain milling and end-use, and will summarize recent progress in China. Chinese scientists have invested substantial effort in molecular genetic and genomic analysis of these traits and their effects on end-use properties. The insights and resources generated have contributed to the understanding and improvement of these traits. As high-quality genomics information and powerful genome engineering tools are becoming available for wheat, more fundamental breakthroughs in dissecting the molecular and genomic basis of WGQ are expected. | 13 | Facile one-step synthesis of Cu_2O@Cu sub-microspheres composites as anode materials for lithium ion batteries显示文摘Cu_2O@Cu sub-microspheres composites with a narrow particle size distribution from 300 to 500 nm was successfully fabricated by one-step synthesis through the direct thermal decomposition of copper nitrate(Cu(NO_3)_2) in octadecylamine(ODA) solvent. As anode materials for lithium ion batteries, the Cu_2 O@Cu composites obviously possess high specific capacity, excellent cyclic stability and rate capability. The coulombic efficiency is about 84% in the 1 st cycle and increases significantly up to 97.8% during successive cycles at various current densities. Even under a high current density of 500 m A g^(-1), the discharge capacity of Cu_2 O@Cu composites remains up to 200 m Ah g^(-1). | 18 | Improved Multi-objective Ant Colony Optimization Algorithm and Its Application in Complex Reasoning显示文摘The problem of fault reasoning has aroused great concern in scientific and engineering fields.However,fault investigation and reasoning of complex system is not a simple reasoning decision-making problem.It has become a typical multi-constraint and multi-objective reticulate optimization decision-making problem under many influencing factors and constraints.So far,little research has been carried out in this field.This paper transforms the fault reasoning problem of complex system into a paths-searching problem starting from known symptoms to fault causes.Three optimization objectives are considered simultaneously: maximum probability of average fault,maximum average importance,and minimum average complexity of test.Under the constraints of both known symptoms and the causal relationship among different components,a multi-objective optimization mathematical model is set up,taking minimizing cost of fault reasoning as the target function.Since the problem is non-deterministic polynomial-hard(NP-hard),a modified multi-objective ant colony algorithm is proposed,in which a reachability matrix is set up to constrain the feasible search nodes of the ants and a new pseudo-random-proportional rule and a pheromone adjustment mechinism are constructed to balance conflicts between the optimization objectives.At last,a Pareto optimal set is acquired.Evaluation functions based on validity and tendency of reasoning paths are defined to optimize noninferior set,through which the final fault causes can be identified according to decision-making demands,thus realize fault reasoning of the multi-constraint and multi-objective complex system.Reasoning results demonstrate that the improved multi-objective ant colony optimization(IMACO) can realize reasoning and locating fault positions precisely by solving the multi-objective fault diagnosis model,which provides a new method to solve the problem of multi-constraint and multi-objective fault diagnosis and reasoning of complex system. | WANG Xinqing ZHAO Yang WANG Dong ZHU Huijie ZHANG Qing | 2013 | Chinese Journal of Mechanical Engineering2013,26,5: | 3 |
| 19 | Sliding friction of shale rock on dry quartz sand particles显示文摘The sliding friction of rock, involving all kinds of particles at the contact surface, is relevant to many problems, ranging from those in artificial engineering to earthquake dynamics. In this work, the frictional performance of the shale rock–dry quartz sand contact was investigated using a self-developed testing device. The study showed that the coefficient of friction of the contact increases with nominal stress and that the corresponding friction force increases approximately linearly with nominal stress, which is directly related to the contact stress between each single sand particle and rock shale. An overall dynamic coefficient, γ, reflecting the response of friction force to nominal stress, first decreases and then increases with area ratio, which is determined by not only the contact stress but also the interparticle friction force.
