| 1 | Three-dimensional-printed individualized porous implants:A new“implant-bone”interface fusion concept for large bone defect treatment显示文摘Bone defect repairs are based on bone graft fusion or replacement.Current large bone defect treatments are inadequate and lack of reliable technology.Therefore,we aimed to investigate a simple technique using three-dimensional(3D)-printed individualized porous implants without any bone grafts,osteoinductive agents,or surface biofunctionalization to treat large bone defects,and systematically study its long-term therapeutic effects and osseointegration characteristics.Twenty-six patients with large bone defects caused by tumor,infection,or trauma received treatment with individualized porous implants;among them,three typical cases underwent a detailed study.Additionally,a large segmental femur defect sheep model was used to study the osseointegration characteristics.Immediate and long-term biomechanical stability was achieved,and the animal study revealed that the bone grew into the pores with gradual remodeling,resulting in a long-term mechanically stable implant-bone complex.Advantages of 3D-printed microporous implants for the repair of bone defects included 1)that the stabilization devices were immediately designed and constructed to achieve early postoperative mobility,and 2)that osseointegration between the host bone and implants was achieved without bone grafting.Our osseointegration method,in which the“implant-bone”interface fusion concept was used instead of“bone-bone”fusion,subverts the traditional idea of osseointegration. | Teng Zhang Qingguang Wei Hua Zhou Zehao Jing Xiaoguang Liu Yufeng Zheng Hong Cai Feng Wei Liang Jiang Miao Yu Yan Cheng Daoyang Fan Wenhao Zhou Xinhong Lin Huijie Leng Jian Li Xinyu Li Caimei Wang Yun Tian Zhongjun Liu | 2021 | Bioactive Materials2021,6,11: | 9 |
| 2 | Activity after Site-Directed Mutagenesis of CD59 on Complement-Mediated Cytolysis显示文摘CD59 may inhibit the cytolytic activity of complement by binding to C8/C9 and protect host cell membranes against homologous membrane attack complex (MAC). However,CD59 is widely overexpressed on tumor cells,which has been implicated in tumorigenesis. The active site of CD59 relative to MAC is still confused. As reported the MAC binding site is located in the vicinity of a hydrophobic groove on the membrane distal face of the protein centered around residue W40. Here two site-directed mutagenesis were performed by overlapping extension PCR to delete residue W40 site (Mutant 1,M1) or to change C39W40K41 to W39W40W41 (Mutant 2,M2). Then we constructed mutant CD59 eukaryotic expression system and investigated their biological function on CHO cells compared with wild-type CD59. Stable populations of CHO cells expressing recombinant proteins were screened by immunotechnique. After 30 passages culturing,proteins could be tested. Dye release assays suggest that M1CD59 loses the activity against complement,while M2CD59 increases the anti-complement activity slightly. | 3 | RNA-binding protein LIN28B inhibits apoptosis through regulation of the AKT2/FOXO3A/BIM axis in ovarian cancer cells显示文摘LIN28B is an evolutionarily conserved RNA-binding protein that regulates mRNA translation and miRNA let-7 maturation in embryonic stem cells and developing tissues.Increasing evidence demonstrates that LIN28B is activated in cancer and serves as a critical oncogene.However,the underlying molecular mechanisms of LIN28B function in tumorigenesis are still largely unknown.Here we report that LIN28B was expressed in over half of the patients with epithelial ovarian cancer who were examined(n=584).Functional experiments demonstrated that LIN28B inhibited ovarian cancer cell apoptosis.Furthermore,we showed that the proapoptotic factor BIM played an essential role in the antiapoptotic function of LIN28B.RNA-IP microarray analysis suggested that LIN28B binds to mRNAs that are associated with the DNA damage pathway,such as AKT2,in ovarian cancer cells.By binding to AKT2 mRNA and enhancing its protein expression,LIN28B regulated FOXO3A protein phosphorylation and decreased the transcriptional level of BIM,which antagonized the antiapoptosis activity of LIN28B.Taken together,these results mechanistically linked LIN28B and the AKT2/FOXO3A/BIM axis to the apoptosis pathway.The findings may have important implications in the diagnosis and therapeutics of ovarian cancer. | Xiaojuan Lin Jianfeng Shen Dan Peng Xinhong He Congjian Xu Xiaojun Chen Janos LTanyi Kathleen Montone Yi Fan Qihong Huang Lin Zhang Xiaomin Zhong | 2018 | Signal Transduction and Targeted Therapy2018,3,1: | 8 |
| 4 | Chemical genomics reveals inhibition of breast cancer lung metastasis by Ponatinib via c-Jun显示文摘Metastasis is the leading cause of human cancer deaths.Unfortunately,no approved drugs are available for antimetastatic treatment.In our study,high-throughput sequencing-based high-throughput screening(HTS^2)and a breast cancer lung metastasis(BCLM)-associated gene signature were combined to discover anti-metastatic drugs.After screening of thousands of compounds,we identified Ponatinib as a BCLM inhibitor.Ponatinib significantly inhibited the migration and mammosphere formation of breast cancer cells in vitro and blocked BCLM in multiple mouse models.Mechanistically,Ponatinib represses the expression of BCLM-associated genes mainly through the ERK/c-Jun signaling pathway by inhibiting the transcription of JUN and accelerating the degradation of c-Jun protein.Notably,JUN expression levels were positively correlated with BCLM-associated gene expression and lung metastases in breast cancer patients.Collectively,we established a novel approach for the discovery of anti-metastatic drugs,identified Ponatinib as a new drug to inhibit BCLM and revealed c-Jun as a crucial factor and potential drug target for BCLM.Our study may facilitate the therapeutic treatment of BCLM as well as other metastases. | Wei Shao Shasha Li Lu Li Kequan Lin Xinhong Liu Haiyan Wang Huili Wang Dong Wang | 2019 | Protein & Cell2019,10,3: | 7 |
| 5 | To inhibit coffee ring effect in inkjet printing of light-emitting polymer films by decreasing capillary force显示文摘Polymer thin film with uniform thickness and flat surface profile is the key point for polymer light emitting diodes(PLEDs) by inkjet printing. However, the coffee ring effect is usually observed due to the mismatch between the evaporation of the solvent and the decrease of solution volume, which promotes the formation of radial flow from the interior of the drop to the edge. In this paper, coffee ring effects of inkjet printed poly(spirobifluorene) films were proposed to be restrained by decreasing capillary force by adding co-solvent with high boiling point and high viscosity to the main solvent. The low evaporation rate of the co-solvent can reduce the driving force of the radial flow; meanwhile the high viscosity of the co-solvent can increase the resistance of the radial flow. Thus, polymer films with improve uniformity can be obtained due to the suppression of the radial flow. | 6 | Practical strategy to construct anti-osteosarcoma bone substitutes by loading cisplatin into 3D-printed titanium alloy implants using a thermosensitive hydrogel显示文摘Surgical resection and perioperative adjuvant chemotherapy-based therapies have improved the prognosis of patients with osteosarcoma;however,intraoperative bone defects,local tumour recurrence,and chemotherapy-induced adverse effects still affect the quality of life of patients.Emerging 3D-printed titanium alloy(Ti6Al4V)implants have advantages over traditional implants in bone repair,including lower elastic modulus,lower stiffness,better bone conduction,more bone in-growth,stronger mechanical interlocking,and lager drug-loading capacity by their inherent porous structure.Here,cisplatin,a clinical first-line anti-osteosarcoma drug,was loaded into Ti6Al4V implants,within a PLGA-PEG-PLGA thermo-sensitive hydrogel,to construct bone substitutes with both anti-osteosarcoma and bone-repair functions.The optimal concentrations of cisplatin(0.8 and 1.6 mg/mL)were first determined in vitro.Thereafter,the anti-tumour effect and biosafety of the cisplatin/hydrogel-loaded implants,as well as their bone-repair potential were evaluated in vivo in tumour-bearing mouse,and bone defect rabbit models,respectively.The loading of cisplatin reduced tumour volume by more than two-thirds(from 641.1 to 201.4 mm3)with negligible organ damage,achieving better anti-tumour effects while avoiding the adverse effects of systemic cisplatin delivery.Although bone repair was hindered by cisplatin loading at 4 weeks,no difference was observed at 8 weeks in the context of implants with versus without cisplatin,indicating acceptable long-term stability of all implants(with 8.48%-10.04%bone in-growth and 16.94%-20.53%osseointegration).Overall,cisplatin/hydrogel-loaded 3D-printed Ti6Al4V implants are safe and effective for treating osteosarcoma-caused bone defects,and should be considered for clinical use. | Zehao Jing Renhua Ni Jiedong Wang Xinhong Lin Daoyang Fan Qingguang Wei Teng Zhang Yufeng Zheng Hong Cai Zhongjun Liu | 2021 | Bioactive Materials2021,6,12: | 3 |

| 10 | Analysis of the chemoprophylactic effect on close contacts of patients with active tuberculosis and positive tuberculin skin tests显示文摘Objective:The current study analyzed the chemoprophylactic effect of isoniazide on close contacts of patients with active tuberculosis and positive tuberculin skin tests(TSTs).Methods:A total of 1206 close contacts of patients with active tuberculosis and strongly-pos-itive TSTs were enrolled.The patients had chest X-ray examinations.Patients without tuberculosis and other diseases were divided into the following groups:90 patients in the prophylaxis group,who were given 300 mg of isoniazid qd(3-5 mg/kg for children)over a 10-month treatment course;and 89 patients in the control group without drug therapy.Both groups were followed for 10 years.Results:(1)There were 568 patients with negative results and 638 with positive results,includ-ing 445 with ordinarily-and moderately-positive results,and 193 with strongly-positive results(a positive rate of 52.9%[638/1206]and a strongly-positive rate of 16.0%[193/1206]).Fourteen tuber-culosis patients were identified(tuberculosis detection rate of 1.1%).(2)During the 3-year period of follow-up,there were 4 patients in the prophylaxis group and 12 patients in the control group who acquired tuberculosis(a morbidity rate of 4.7%[4/84]and 13.4%[12/89],respectively),and the dif-ference was statistically significant(χ^(2)=3.916,P=0.048).Six patients in the prophylaxis group,all of whom were children,discontinued medication use during the course of treatment due to adverse drug reactions,for an adverse drug reaction occurrence rate of 6.6%(6/90),a medication completion rate of 93.3%(84/90),and a 3-year protection ratio of 64.6%.(3)During the 4-6 year period,there were two patients in the prophylaxis group and three patients in the control group who acquired tubercu-losis(a morbidity rate of 2.5%[2/78]and 4.1%[3/73],respectively),two in the prophylaxis group and four in the control group who were lost to follow-up(a loss to follow-up rate of 2.5%[2/80]and 5.1%[4/77],respectively),and the difference was not statistically significant(χ^(2)=0.006,P=0.940;χ^(2)=0.215,P=0.643).(4)During the 7-10 year study,there was one patient in the prophylaxis group and two patients in the control group who acquired tuberculosis(a morbidity rate of 1.3%[1/72]and 3.1%[2/64],respectively),and four in the prophylaxis group and six in the control group who were lost to follow-up(a loss to follow-up rate of 5.2%[4/76]and 8.5%[6/70],respectively),and the dif-ference was not statistically different(χ^(2)=0.011,P=0.918;χ^(2)=0.176,P=0.675).(5)Within 10 years,there were 7 patients in the prophylaxis group and 17 patients in the control group who acquired tu-berculosis(a morbidity rate of 8.3%[7/84]and 21.5%[17/79],respectively;χ^(2)=4.770,P=0.029),and 6 in the prophylaxis group and 10 in the control group were lost to follow-up(the loss to follow-up rate was 7.1%[6/84]and 11.2%[10/89],respectively;χ^(2)=0.863,P=0.353).Conclusion:Close contacts of patients with active tuberculosis are at high-risk for acquir-ing tuberculosis.It is safe and effective for patients with strongly-positive TST results to undergo isoniazid chemoprophylaxis for 10 months. | Cunzhi Lin Fangfang Wang Jinfeng Li Jianxin Du Hairong Wang Xinhong Zhu | 2014 | Family Medicine and Community Health2014,2,3: | 0 |
| 11 | Formal semantic description standard family for Chinese KOS(Ⅱ): Research and implementation of Classification Scheme Sharing Service System显示文摘In the context of globalization of network information, it is an urgent need to consider networked development and sharing for classification schemes. Since 2000, the international mainstream classification schemes have published their Web versions one after another, while there is still a lack of an integrated, reusable and Semantic-Web-oriented solution for classification schemes and classification-like knowledge organization systems. By using computer programs to automatically analyze the source files of the classification schemes in the MARC or HTML format, this paper adopts CNKOS to produce the semantic description of the classification schemes. On account of this, this paper employs international advanced open-source technologies, takeing the representative domestic CLC as an example, to construct the CLSS prototype system. It is proved that this solution can be put into practical use and can facilitate the quick deployment of other classification-like Chinese knowledge organization systems. The successful implementation of the system verifies the scientificity and practicality of CNKOS, and it plays a demonstrating and promoting role and provides a reusable solution for other domestic classification schemes and classification-like knowledge organization systems. | 14 | Effects of Yishenjiangzhuo granules on immunity and bone metabolism in patients with stage 3-4 chronic kidney disease显示文摘OBJECTIVE:To determine the effects of Yishenjiangzhuo granules(YJG)on bone metabolism and to explore the changes in levels of bone Gla protein(BGP),tartrate-resistant acid phosphatase(TRAP),as well as their relationships with levels of B cells,regulatory T cells(Treg)and interleukin(IL)-17 in patients with stage 3-4 chronic kidney disease(CKD)before and after treatment.METHODS:Fifty-three stage 3-4 CKD patients were divided randomly into two groups:YJG treatment and control.Peripheral blood was taken from two groups of CKD patients and 21 healthy subjects in the normal group.The parameters determined were the levels of CD4+,CD19+,CD19+CD69+,CD19+AV,Treg(CD4+CD25+CD127lo),BGP,TRAP,IL-17,calcium,phosphate,blood urea nitrogen,serum creatinine(SCr),hemoglobin(Hb)in peripheral blood,and urinary creatinine.Calcium-phosphate products and endogenous creatinine clearance rate(CCr)were calculated according to standard protocols.RESULTS:In YJG and control groups,SCr levels were lowered(P<0.01)after treatment,whereas CCr(P<0.05)as well as Hb and albumin levels(P<0.01)were increased.The changes in levels of CCr and SCr in the YJG group were more significant.After treatment,CD19+CD69+and Treg levels in the two groups varied(P<0.01)compared with those of the normal group;the level of CD19+increased but the levels of CD4+and CD19+AV decreased(P<0.01)in both groups.Compared with the control group,the changes of CD19+and CD19+AV in the YJG group were more apparent(P<0.05).Compared with the normal group,levels of IL-17 in both groups increased significantly(P<0.01),and the difference in the control group was more significant(P<0.05).After treatment,the TRAP level increased(P<0.05),but the difference in BGP level(P>0.05)was not significant.CONCLUSION:In stage 3-4 CKD patients,B cells and IL-17 participated in the induction of osteoclast activation.YJG could also elevate the level of B cells and decrease their apoptosis,but showed no significant effects on active B cells,IL-17 or osteoclast activity. | Jing Zheng Yingda Lin Lu Huang Caifeng Chen Xuemin Zheng Xinhong Wu Ciyun Liu | 2013 | Journal of Traditional Chinese Medicine2013,33,5: | 0 |
