1High-speed image reconstruction for optically sectioned,super-resolution structured illumination microscopy显示文摘Super-resolution structured illumination microscopy(SR-SIM)is an outstanding method for visualizing the subcellular dynamics in living cells.To date,by using elaborately designed systems and algorithms,SR-SIM can achieve rapid,optically sectioned,SR observation with hundreds to thousands of time points.However,real-time observation is still out of reach for most SIM setups as conventional algorithms for image reconstruction involve a heavy computing burden.To address this limitation,an accelerated reconstruction algorithm was developed by implementing a simplified workflow for SR-SIM,termed joint space and frequency reconstruction.This algorithm results in an 80-fold improvement in reconstruction speed relative to the widely used Wiener-SIM.Critically,the increased processing speed does not come at the expense of spatial resolution or sectioning capability,as demonstrated by live imaging of microtubule dynamics and mitochondrial tubulation.Zhaojun Wang Tianyu Zhao Huiwen Hao Yanan Cai Kun Feng Xue Yun Yansheng Liang Shaowei Wang Yujie Sun Piero RBianco Kwangsung Oh Ming Lei 2022Advanced Photonics2022,4,2:9

3Creep-Fatigue Interaction in an Fe-25Ni-15Cr Based Alloy显示文摘The interaction between high temperature creep and low cycle fatigue in an Fe-25Ni-15Cr basedalloy GH132 has been studied under three different loading schedules: (a) continuous cycliccreep, (b) prior creep followed by fatigue, and (c) prior fatigue followed by creep. It is found thatthe interaction appears nearly linear for all the three schedules. The behaviour of interactionin GH132 alloy is different from that previously observed in a GH30 alloy (Nimonic 75 type),in which although the interaction at schedules (a) and (c) was linear, a pronounced negativeinteraction occurred at the schedule (b), i.e., prior creep followed by fatigue. This differenceis found to be associated with the difference in the evolution of microstructure. 4Rapid,artifact-reduced,image reconstruction for super-resolution structured illumination microscopy显示文摘Super-resolution structured ilumination microscopy(SR-SIM)is finding increasing application in biomedical research due to its superior ability to visualize subcellular dynamics in living cells.However,during image reconstruction artifacts can be introduced and when coupled with time-consuming postprocessing procedures,limits this technique from becoming a routine imaging tool for biologists.To address these issues,an accelerated,artifact-reduced reconstruction algorithm termed joint space frequency reconstruction-based artifact reduction algorithm(JSFR-AR-SIM)was developed by integrating a high-speed reconstruc tion framework with a high-fidelity optimization approach designed to suppress the sidelobe artifact.Consequently,JSFR-AR-SIM produces high-quality,super-resolution images with minimal artifacts,and the reconstruction speed is increased.We anticipate this algorithm to facilitate SR-SIM becoming a routine tool in biomedical laboratories.Zhaojun Wang Tianyu Zhao Yanan Cai Jingxiang Zhang Huiwen Hao Yansheng Liang Shaowei Wang Yujie Sun Tongsheng Chen Piero R.Bianco Kwangsung Oh Ming Lei 2023The Innovation2023,4,3:0

10Long-lasting photoluminescence quantum yield of cesium lead halide perovskite-type quantum dots显示文摘Cesium lead halide perovskite(CsPbX_(3),X=Cl,Br,I)quantum dots(QDs)and their partly Mn^(2+)-substituted QDs(CsPb1–xMnxX3)attract considerable attention owing to their unique photoluminescence(PL)efficiencies.The two types of QDs,having different PL decay dynamics,needed to be further investigated in a form of aggregates to understand their solid-state-induced exciton dynamics in conjunction with their behaviors upon degradation to achieve practical applications of those promising QDs.However,thus far,these QDs have not been sufficiently investigated to obtain deep insights related to the long-term stability of their PL properties as aggregated solid-states.Therefore,in this study,we comparatively examined CsPbX_(3)-and CsPb1–xMnxX_(3)-type QDs stocked for>50 d under dark ambient conditions by using excitation wavelength-dependent PL quantum yield and time-resolved PL spectroscopy.These investigations were performed with powder samples in addition to solutions to determine the influence of the inter-QD interaction of the aged QD aggregates on their radiative decays.It turns out that the Mn^(2+)-substituted QDs exhibited long-lasting PL quantum efficiencies,while the unsubstituted CsPbX_(3)-type QDs exhibited a drastic reduction of their PL efficiencies.And the obtained PL traces were clearly sensitive to the sample status.This is discussed with the possible interaction depending on the size and distance of the QD aggregates.Yonghyun Kim Huiwen Liu Yi Liu Boa Jin Hao Zhang Wenjing Tian Chan Im 2021Frontiers of Chemical Science and Engineering2021,15,1:0
11PTEN-induced putative kinase 1 regulates mitochondrial quality control and is essential for the maturation of human induced pluripotent stem cell-derived cardiomyocytes显示文摘Human induced pluripotent stem cell-derived cardiomyocytes(hiPSC-CMs)have attracted attention in the field of regenerative medicine due to their potential ability to repair damaged hearts.However,the immature phenotype of these cells limits their clinical application.Cardiomyocyte maturation is accompanied by changes in mitochondrial quality.PTEN-induced putative kinase 1(PINK1)has been linked to mitochondrial quality control.However,whether the changes in mitochondrial quality in hiPSC-CMs are associated with PINK1,and the impact of PINK1 on hiPSC-CMs development are not clear.In this study,we found that knockdown of PINK1 in hiPSC-CMs resulted in mitochondrial fragmentation and impaired mitochondrial functions such as mitophagy and mitochondrial biogenesis.PINK1 deletion also inhibited the maturation of hiPSC-CMs,reverting them to a naive structural and functional state.We found that restoring the mitochondrial structure did not completely rescue the mitochondrial dysfunction caused by PINK1 deletion,while activation of PINK1 kinase activity using kinetin promoted mitochondrial fusion,increased the mitochondrial membrane potential and ATP production,and maintained the development and maturation of hiPSC-CMs.In conclusion,PINK1 regulates the mitochondrial structure and function of hiPSC-CMs,and is essential for the maturation of hiPSC-CMs.Huiwen Liu Yanting Sun Hao Xu Bin Tan Qin Yi Jie Tian Jing Zhu 2023Genes & Diseases2023,10,5:0
