| 2 | COP9 signalosome subunit PfCsnE regulates secondary metabolism and conidial formation in Pestalotiopsis fici显示文摘The COP9 signalosome(CSN) is a highly conserved multiprotein complex in all eukaryotes and involved in regulation of organism development. In filamentous fungi, several lines of evidence indicate that fungal development and secondary metabolism(SM) are mediated by the fifth subunit of CSN, called CsnE. Here we uncover a connection with CsnE and conidial formation as well as SM regulation in the plant endophytic fungus Pestalotiopsis fici. A homology search of the P. fici genome with CsnE, involved in sexual development and SM in Aspergillus nidulans, identified PfCsnE. Deletion of PfcsnE resulted in a mutant that stopped conidial production, but the conidia are recovered in a PfcsnE complemented strain. This indicates that PfCsnE is required for the formation of conidia. Secondary metabolite analysis demonstrated that the ΔPfcsnE strain produced more chloroisosulochrin, less ficiolide A production in comparison to wild type(WT). Transcriptome analysis of WT andΔPfcsnE strains indicated that PfcsnE impacts the expression levels of 8.37% of 14,797 annotated genes. Specifically, nine biosynthetic gene clusters(BGCs) were up-regulated and three BGCs were down-regulated by PfCsnE. | 9 | Lower synaptic density associated with gaming disorder:an^(18)F-SynVesT-1 PET imaging study显示文摘Background Internet gaming disorder(IGD)is an ideal model to study the mechanisms underlying synaptic deficits in addiction as it eliminates the confounding effects of substance use.Synaptic loss and deficits are hypothesised to underlie the enduring maladaptive behaviours and impaired cognitive function that contribute to IGD.Aims This study aimed to determine whether subjects with IGD have lower synaptic density than control subjects and the relationship between synaptic density and IGD severity.Methods Eighteen unmedicated subjects diagnosed with current IGD according to the Diagnostic and Statistical Manual of Mental Disorders,Fifth Edition criteria and 16 demographically matched healthy controls(HCs)participated in the study and underwent 18F-labelled difluoro-analogue of UCB-J(18F-SynVesT-1)positron emission tomography scans to assess the density of synaptic vesicle glycoprotein 2A(SV2A).The Internet Gaming Disorder Scale-Short Form(IGDS9-SF),Hamilton Rating Scale for Depression(HAMD),Hamilton Anxiety Rating Scale(HAMA),Barratt Impulsiveness Scale Version 11(BIS-11),Stroop Colour-Word Test(SCWT),stop-signal paradigms and N-back tasks were administered to all subjects.Results Patients with IGD had significantly higher scores on the IGDS9-SF,HAMD,HAMA and BIS-11 than HCs.HCs performed better on the two-back and SCWT tests as well as in terms of stop-signal reaction times(SSRTs)in the stop-signal paradigms than patients with IGD.Lower uptake was found in the bilateral putamen,right pregenual anterior cingulate cortex and Rolandic operculum of patients with IGD compared with HCs.Furthermore,in the IGD group,IGDS9-SF scores and daily gaming hours were negatively correlated with the standardised uptake value ratios of 18F-SynVesT-1 in the bilateral putamen.Longer SSRTs were significantly associated with lower SV2A density in the right pregenual anterior cingulate cortex and right Rolandic operculum.Conclusions The in vivo results in this study suggest that lower synaptic density contributes to the severity and impairments in inhibitory control of IGD.These findings may provide further incentive to evaluate interventions that restore synaptic transmission and plasticity to treat IGD. | Jiale Hou Qian Xiao Ming Zhou Ling Xiao Ming Yuan Na Zhong Jiang Long Tao Luo Shuo Hu Huixi Dong | 2023 | General Psychiatry2023,36,5: | 0 |
| 10 | Characteristics and geochemical implications of light hydrocarbons from ultra-deep Ordovician oils in the North Shuntuoguole area, Tarim Basin显示文摘The ultra-deep Ordovician reservoirs in North Shuntuoguole Oilfield(or Shunbei Oilfield)of Sinopec have achieved annual production of one million ton,and the oil&gas in different faults show different physical properties and fluid phases.In this study,the 28 oil samples from the ultra-deep Ordovician were analyzed using whole oil chromatography.The heptane and isoheptane values of the oil samples were in the range of 29.79%‒46.86%and 1.01%-3.06%,respectively,indicating the oils are high mature.The maturity that calculated based on light hydrocarbon values was higher than which calculated by using aromatic hydrocarbon parameters,suggesting the light hydrocarbon maturity mostly reflects the maturity of the late charged hydrocarbon.The 2M-/3M-C5 and 2M-/3M-C_(6) ratios varied in the ranges of 1.41‒1.81 and 0.79-1.09,respectively,and the iC_(5)/nC_(5) and 3M-C_(5)/nC_(6) ratios were 0.31‒0.90 and 0.16-0.37,respectively,indicating that ultra-deep Ordovician reservoirs have not experienced biodegradation.The Mango parameter K_(1) of the oil samples ranges 0.96‒1.01 except for the oil from Well SB4,which suggests that most of the reservoirs have not suffered thermochemical sulfur reduction(TSR).Meanwhile,the oils have not experienced evaporative fractionation since the toluene/nC_(7) and nC_(7)/MCC_(6) ratios range from 0.10-0.38 and 1.50‒1.80,respectively.The close correlation between P_(3) and P_(2)+N_(2) and between P_(2) and N_(2)/P_(3) indicates that the oils from different faults have the same origin.According to the characteristics of LHs rich in n-alkane,as well as other biomarkers,such as aryl isoprenoids,and aromatic hydrocarbon parameters,the oil originated from the source rock of Lower Cambrian Yu’ertusi Formation.Meanwhile,the source rocks in different fault zones slightly differed in organic facies. | Li DONG Anlai MA Huixi LIN Lu YUN Weilong PENG Xiuxiang ZHU | 2022 | Frontiers of Earth Science2022,16,3: | 0 |
