| 1 | A nuclear import inhibitory peptide ameliorates the severity of cholecystokinin-induced acute pancreatitis显示文摘AIM: To assess the effect of our novel cell-permeable nuclear factor-kappaB (NF-κB) inhibitor peptide PN50 in an experimental model of acute pancreatitis. PN50 was produced by conjugating the cell-penetrating penetratin peptide with the nuclear localization signal of the NF-κB p50 subunit.METHODS: Pancreatitis was induced in male Wistar rats by administering 2×100 μg/kg body weight of cholecystokininoctapeptide (CCK) intraperitoneally (IP) at an interval of 1 h. PN50-treated animals received 1 mg/kg of PN50 IP 30 min before or after the CCK injections. The animals were sacrificed 4 h after the first injection of CCK.RESULTS: All the examined laboratory (the pancreatic weight/body weight ratio, serum amylase activity,pancreatic levels of TNF-α and IL-6, degree of lipid peroxidation, reduced glutathione levels, NF-κB binding activity, pancreatic and lung myeloperoxidase activity) and morphological parameters of the disease were improved before and after treatment with the PN50 peptide.According to the histological findings, PN50 protected the animals against acute pancreatitis by favoring the induction of apoptotic, as opposed to necrotic acinar cell death associated with severe acute pancreatitis.CONCLUSION: Our study implies that reversible inhibitors of stress-responsive transcription factors like NF-κB might be clinically useful for the suppression of the severity of acute pancreatitis. | Tamás Letoha Csaba Somlai Tamáas Takács Annamária Szabolcs Katalin Jármay Zoltán Rakonczay Jr Péter Hegyi Ilona Varga József Kaszaki István Krizbai Imre Boros Ern(?) | 2 | Proteoglycans in liver cancer显示文摘Proteoglycans are a group of molecules that contain at least one glycosaminoglycan chain,such as a heparan,dermatan,chondroitin,or keratan sulfate,covalently attached to the protein core.These molecules arecategorized based on their structure,localization,and function,and can be found in the extracellular matrix,on the cell surface,and in the cytoplasm.Cell-surface heparan sulfate proteoglycans,such as syndecans,are the primary type present in healthy liver tissue.However,deterioration of the liver results in overproduction of other proteoglycan types.The purpose of this article is to provide a current summary of the most relevant data implicating proteoglycans in the development and progression of human and experimental liver cancer.A review of our work and other studies in the literature indicate that deterioration of liver function is accompanied by an increase in the amount of chondroitin sulfate proteoglycans.The alteration of proteoglycan composition interferes with the physiologic function of the liver on several levels.This article details and discusses the roles of syndecan-1,glypicans,agrin,perlecan,collagen XVIII/endostatin,endocan,serglycin,decorin,biglycan,asporin,fibromodulin,lumican,and versican in liver function.Specifically,glypicans,agrin,and versican play significant roles in the development of liver cancer.Conversely,the presence of decorin could potentially provide protective effects. | Kornélia Baghy Péter Tátrai Eszter Regős Ilona Kovalszky | 2016 | World Journal of Gastroenterology2016，22，1： | 8 |
| 3 | Therapeutic proteasome inhibition in experimental acute pancreatitis显示文摘AIM: To establish the therapeutic potential of proteasome inhibition, we examined the therapeutic effects of MG132 (Z-Leu-Leu-Leu-aldehyde) in an experimental model of acute pancreatitis.METHODS: Pancreatitis was induced in rats by two hourly intraperitoneal (ip) injections of cholecystokinin octapeptide (CCK; 2 x 100 μg/kg) and the proteasome inhibitor MG132 (10 mg/kg ip) was administered 30 min after the second CCK injection. Animals were sacrificed 4 h after the first injection of CCK.RESULTS: Administering the proteasome inhibitor MG132 (at a dose of 10 mg/kg, ip) 90 min after the onset of pancreatic inflammation induced the expression of cell-protective 72 kDa heat shock protein (HSP72) and decreased DNA-binding of nuclear factor-kB (NF-kB). | 4 | Epidemiologic characteristics of Helicobacter pylori infection in southeast Hungary显示文摘BACKGROUND Epidemiologic studies have revealed a decrease in the prevalence of Helicobacter pylori(H.pylori)infection in Western Europe.AIM To obtain data regarding the prevalence of H.pylori in Csongrád and Békés Counties in Hungary,evaluate the differences in its prevalence between urban and rural areas,and establish factors associated with positive seroprevalence.METHODS One-thousand and one healthy blood donors[male/female:501/500,mean age:40(19–65)years]were enrolled in this study.Subjects were tested for H.pylori IgG antibody positivity via enzyme-linked immunosorbent assay.Subgroup analysis by age,gender,smoking habits,alcohol consumption,and urban vs nonurban residence was also performed.RESULTS The overall seropositivity of H.pylori was 32%.It was higher in males(34.93%vs 29.2%,P=0.0521)and in rural areas(36.2%vs 27.94%,P=0.0051).Agricultural/industrial workers were more likely to be positive for infection than office workers(38.35%vs 30.11%,P=0.0095)and rural subjects in Békés County than those in Csongrád County(43.36%vs 33.33%,P=0.0015).CONCLUSION Although the prevalence of H.pylori infection decreased in recent decades in Southeast Hungary,it remains high in middle-aged rural populations.Generally accepted risk factors for H.pylori positivity appeared to be valid for the studied population. | Lenke Bálint Andrea Tiszai Gábor Kozák Ilona Dóczi Veronika Szekeres Orsolya Inczefi Georgina Ollé Krisztina Helle Richárd Róka András Rosztóczy | 2019 | World Journal of Gastroenterology2019，25，42： | 5 |
| 5 | Biological treatment of chicken feather waste for improved biogas production显示文摘A two-stage system was developed which combines the biological degradation of keratin-rich waste with the production of biogas.Chicken feather waste was treated biologically with a recombinant Bacillus megaterium strain showing keratinase activity prior to biogas production.Chopped,autoclaved chicken feathers (4%,W/V) were completely degraded,resulting in a yellowish fermentation broth with a level of 0.51 mg/mL soluble proteins after 8 days of cultivation of the recombinant strain.During the subsequent anaerobic batch digestion experiments,methane production of 0.35 Nm 3 /kg dry feathers (i.e.,0.4 Nm 3 /kg volatile solids of feathers),corresponding to 80% of the theoretical value on proteins,was achieved from the feather hydrolyzates,independently of the prehydrolysis time period of 1,2 or 8 days.Cultivation with a native keratinase producing strain,Bacillus licheniformis resulted in only 0.25 mg/mL soluble proteins in the feather hydrolyzate,which then was digested achieving a maximum accumulated methane production of 0.31 Nm 3 /kg dry feathers.Feather hydrolyzates treated with the wild type B.megaterium produced 0.21 Nm 3 CH 4 /kg dry feathers as maximum yield. | Gergely Forgcs Saeid Alinezhad Amir Mirabdollah Elisabeth Feuk-Lagerstedt Ilona Srvri Horvth | 2011 | Journal of Environmental Sciences2011，23，10： | 4 |
| 6 | Elevated miR-33a and miR-224 in steatotic chronic hepatitis Cliver biopsies显示文摘AIM:To assess the expression of selected microRNAs(miRNA) in hepatitis C,steatotic hepatitis C,noninfected steatotic and normal liver tissues.METHODS:The relative expression levels of miR-21,miR-33 a,miR-96,miR-122,miR-125 b,miR-221 and miR-224 were determined in 76 RNA samples isolated from 18 non-steatotic and 28 steatotic chronic hepatitis C(CHC and CHC-Steatosis,respectively) cases,18 non-infected,steatotic liver biopsies of metabolic origin(Steatosis) and 12 normal formalin-fixed paraffin-embedded liver tissues using TaqMan MicroRNA Assays.All CHC biopsy samples were obtained prior to initiating therapy.Patients' serum biochemical values,which included glucose,triglyceride,cholesterol,alanine aminotransferase(ALT),aspartate aminotransferase(AST),gamma-glutamyl-transferase(GGT),alkaline phosphatase(AP),were obtained and correlated with relative miRNA expression.RESULTS:When compared with control non-infected liver samples,miR-122 and miR-221 levels were reduced in CHC-Steatosis(P < 0.03) and in CHC,CHCSteatosis and Steatosis(P < 0.01).Alternatively,the expression of miR-33 a and miR-224 were elevated in CHC-Steatosis and Steatosis in comparison to control tissue(P < 0.01).The levels of miR-33 a and miR-224 in CHC-Steatosis(P < 0.02) and miR-224 in Steatosis(P < 0.001) were increased in comparison to CHC samples.By contrast,the expression of miR-21 did not differ statistically between diseased and normal liver samples.Levels of miR-33 a correlated negatively with serum AST and AP levels in Steatosis as well as with necroinflammatory grade in CHC,whereas miR-21 correlated positively with AST in Steatosis and displayed negative correlation with triglyceride level in CHC-Steatosis.In contrast,miRNA levels were not correlated with ALT,GGT,cholesterol levels or fibrosis stage.CONCLUSION:Differences in miRNA expression were observed between CHC and steatotic CHC,CHC and steatotic liver,but not between steatotic CHC and steatotic liver of metabolic origin. | Gabor Lendvai Katalin Jármay Gizella Karácsony Tünde Halász Ilona Kovalszky Kornélia Baghy Tibor Wittmann Zsuzsa Schaff András Kiss | 2014 | World Journal of Gastroenterology2014，20，41： | 2 |
