1Presence of Porphyromonas gingivalis in gingival squamous cell carcinoma显示文摘Periodontal disease has been recently linked to a variety of systemic conditions such as diabetes, cardio-vascular disease, preterm delivery, and oral cancer. The most common bacteria associated with periodontal disease, Porphyromonas gingivalis (P. gingivalis) has not yet been studied in the malignant gingival tissues. The objective of this study was to investigate the presence of P. gingivalis in specimens from squamous cell carcinoma patients. We have performed immunohistochemical staining to investigate the presence of P. gingivalis and Streptococcus gordonii (S. gordonii), a non invasive oral bacteria, in paraffin embedded samples of gingival squamous cell carcinoma (n=10) and normal gingiva (n=5). Staining for P. gingivalis revealed the presence of the bacteria in normal gingival tissues and gingival carcinoma, with higher levels (more than 33%, P<0.05) detected in the carcinoma samples. The staining intensity was also significantly enhanced in the malignant tissue by 2 folds (P<0.023) compared to specimens stained for the non-invasive S. gordonii. P. gingivalis is abundantly present in malignant oral epithelium suggesting a potential association of the bacteria with gingival squamous cell carcinoma.Joseph Katz Mairelys D. Onate Kaleb M. 2Optimisation of Adventitious Shoot Regeneration and Agrobacterium-mediated Transformation in Canna × generalis(Canna Lily)显示文摘Canna being ornamental plants has a significant role in agriculture, medical, economy and food industry. Canna has a limited vase life due to the rapid loss of moisture from its perianth. For improving its market value, cuticularisation of the perianth can be achieved by the expression of a heterologous cutin producing gene, using the tissue culture and transformation protocol developed in this study. Efficient, rapid and direct adventitious shoot regeneration was successfully established in Canna × generalis using recalcitrant rhizome explants. The explants were cultured on MS medium supplemented with 6-benzylaminopurine(6-BA), thidiazuron(TDZ), and kinetin. Among the four genotypes taken for tissue culture, the ‘Trinacria variegata' was the best responding cultivar. And 2 mg · L^(-1)6-BA or 1.5 mg · L^(-1) TDZ along with 0.1 mg · L^(-1) IAA was optimum for their regeneration. The highest regeneration was achieved in ‘Trinacria variegata'(36%) on 6-BA, 33% on TDZ while kinetin failed to evoke any regenerative responses. Regeneration was enhanced by supplementation of 100 mg · L^(-1) ascorbic acid(AsA), while, 100 mg · L^(-1) of l-cysteine or 100 mg · L^(-1) dithiothreitol(DTT), inhibited regeneration. Shoots were observed to develop 3–5 fibrous roots on MS medium supplemented with 0.5 mg · L^(-1) indole-3-butyric acid(IBA). The plantlets were transplanted into pots and acclimatised in glasshouse with 100%survival. For transformation of Canna, rhizome explants were co-cultivated for 60 min in Agrobacterium suspension. The explants were washed with 500 mg · L^(-1) cefotaxime solution, subjected to 100 mg · L^(-1) kanamycin selection followed by excision of the shoots and culturing them on IBA-supplemented media for root development. 202,4-D Hyper Accumulation Induced Cellular Responses of Azolla pinnata R. Br. to Sustain Herbicidal Stress显示文摘In the present experiment with ongoing concentration(0µM,100µM,250µM,500µM and 1000µM)of 2,4-D,the responses of Azolla pinnata R.Br.was evaluated based on cellular functions.Initially,plants were significantly tolerated up to 1000µM of 2,4-D with its survival.This was accompanied by a steady decline of indole acetic acid(IAA)concentration in tissues with 78.8%over the control.Membrane bound H^(+)-ATPase activity was over expressed within a range of 1.14 to 1.25 folds with activator(KCl)and decreased within a range of 57.3 to 74.6%in response to inhibitor(Vanadate)application.With regards to IAA metabolism,plants recorded a linear increase with wall bound oxidase activity up to maximum concentration of 2,4-D.The variations were more moderated when wall bound IAA-oxidase recorded a linear increase proportionate to the 2,4-D concentrations.This was more extended with the presence of different isoforms of IAA-oxidase which was much more pronounced with distinct polymorphisms of expressed proteins,however,not independent to the 2,4-D concentrations.Polyamines like spermine,spermidine and putrescine(spm,spd and put)were not consistent in concentration with the dosages of 2,4-D.Besides these,plants were induced to apoplastic NAD(P)H oxidase activity maximally by 1.6 folds under 500µM 2,4-D over control.Still,putrescine responded more or less consistently and recorded maximally 11.9 folds at 500µM 2,4-D as compared to the control.NAD(P)H oxidase activity recorded maximally 1.6 folds against control and remain consistent throughout the concentrations of 2,4-D.GPX along with APX were more linear in responses through the concentration of 2,4-D except CAT as compared to control.On enzymatic antioxidative activity,peroxidases(GPX and APX)were overexpresed in a similar manner except for catalase with a non-significant rise.In stabilization of cellular redox,glutathione reductase attended maximum value by 2.45 folds at 1000µM evidenced with significant variations in protein polymorphism.The sensitivity of 2,4-D also appeared in Azolla with a maximum loss of nucleic acids as documented by the comet assay.Moreover,the Azolla might have some DNA damage protective activity as evident using frond extract with plasmid nick assay.Therefore,Azolla plants with its cellular responses is evident to sustain against the 2,4-D herbicidal stress and may be granted in bio remediation process for the contaminated soil.Arnab Kumar De Arijit Ghosh Debabrata Dolui Indraneel Saha Malay Kumar Adak 2020Phyton-International Journal of Experimental Botany2020,89,4:0
