1Targeting a novel inducible GPX4 alternative isoform to alleviate ferroptosis and treat metabolic-associated fatty liver disease显示文摘Metabolic-associated fatty liver disease(MAFLD),which is previously known as non-alcoholic fatty liver disease(NAFLD),represents a major health concern worldwide with limited therapy.Here,we provide evidence that ferroptosis,a novel form of regulated cell death characterized by iron-driven lipid peroxidation,was comprehensively activated in liver tissues from MAFLD patients.The canonical-GPX4(cGPX4),which is the most important negative controller of ferroptosis,is downregulated at protein but not mRNA level.Interestingly,a non-canonical GPX4 transcript-variant is induced(inducible-GPX4,iGPX4)in MAFLD condition.The high fat-fructose/sucrose diet(HFFD)and methionine/choline-deficient diet(MCD)-induced MAFLD pathologies,including hepatocellular ballooning,steatohepatitis andfibrosis,were attenuated and aggravated,respectively,in cGPX4-and iGPX4-knockin mice.cGPX4 and iGPX4 isoforms also displayed opposing effects on oxidative stress and ferroptosis in hepatocytes.Knockdown of iGPX4 by siRNA alleviated lipid stress,ferroptosis and cell injury.Mechanistically,the triggered iGPX4 interacts with cGPX4 to facilitate the transformation of cGPX4 from enzymatic-active monomer to enzymatic-inactive oligomers upon lipid stress,and thus promotes ferroptosis.Co-immunoprecipitation and nano LC–MS/MS analyses confirmed the interaction between iGPX4 and cGPX4.Our results reveal a detrimental role of non-canonical GPX4 isoform in ferroptosis,and indicate selectively targeting iGPX4 may be a promising therapeutic strategy for MAFLD.Jie Tong Dongjie Li Hongbo Meng Diyang Sun Xiuting Lan Min Ni Jiawei Ma Feiyan Zeng Sijia Sun Jiangtao Fu Guoqiang Li Qingxin Ji Guoyan Zhang Qirui Shen Yuanyuan Wang Jiahui Zhu Yi Zhao Xujie Wang Yi Liu Shenxi Ouyang Chunquan Sheng Fuming Shen Pei Wang 2022Acta Pharmaceutica Sinica B2022,12,9:3
2Comparative Study on the Efficacies of 11 Chemical Agents against Taro Soft Rot Caused by Pectobacterium aroidearum显示文摘[Objective]The paper was to screen the effective agents for effectively controlling the occurrence and prevalence of taro soft rot.[Method]The fungicidal effects of 11 fungicides on taro soft rot were determined by inhibition zone method.[Result]0.3%Tetramycin,25%bromothalonil,3%Zhongshengmycin and 30%zinc thiazole had better antifungal effect on Pectobacterium aroidearum at recommended concentration.The indoor toxicity of four fungicides showed that the EC50 of 0.3%tetramycin AS was the smallest of 0.6μg/mL,indicating that tetramycin had the largest toxicity and the best inhibitory effect against P.aroidearum;followed by 25%bromothalonil EC and 3%Zhongshengmycin WP,with the EC_(50) of 2.57 and 97.72μg/mL,respectively;the inhibitory effect of 30%zinc thiazole SC against P.aroidearum was the poorest.[Conclusion]The study provides a reference for screening out new and efficient chemical agents against taro soft rot.Dongkun Wang Xiuting Ji Huaixu Zhan Chengpeng Zheng Hao Zong Xiaolei Tan Xiuchun Du Weimin Wang Jingjing Wu Fenglong Wang Xiaoqiang Wang 2021Plant Diseases and Pests2021,12,2:2
3Tough polyimide composites synergistically reinforced by carbon nanofiber-grafted carbon fiber and rGO for improved heat dissipation and electromagnetic interference shielding显示文摘The rapid miniaturization and a consequent increase in electromagnetic wave(EM)and heat emission in integrated electronic devices are now urgently desiring for multifunctional polymer composites that simultaneously possess high mechanical properties,excellent electromagnetic interference(EMI)shield-ing ability and heat dissipation capability.Herein,a series of novel polyimide-based composite films were constructed by rationally assembling the carbon nanofiber-grafted carbon fiber(NCF)and reduced graphene oxide(rGO)into a highly electrically and thermally conductive pathway within polyimide ma-trix via a sequential bidirectional freezing casting and hot-pressing strategy.The combination of high tensile strength and toughness was obtained by incorporating rGO into the NCF-reinforced PI composite,giving rise to a hierarchical reinforcing structure of NCF and crack deflection induced by rGO nanosheets.The high electrical conductivity(7.0×10^(3) S m^(-1))endows the PI/NCF30/rGO3.5 composite film with a good EMI shielding effectiveness of 45 dB.Moreover,the well-aligned thermally conductive NCF and rGO and strong interfacial bonding between the polymer matrix and reinforcing fibers give rise to improved thermal conductivity(λ),particularly along the in-plane direction.Typically,the PI/NCF30/rGO3.5 exhibits an in-plane thermal conductivity of 5.18 W m^(-1) K^(-1),∼4.7 times increment compared to the pure PI(0.91 W m^(-1) K^(-1)).Besides,the resultant PI/NCF30/rGO3.5 composite film presents a superior Joule heat-ing performance with some features of fast thermal response,ease of regulation and sufficient reliability.Accordingly,the developed multifunctional polyimide-based composite films demonstrate high potential as advanced EMI shielding materials with excellent heat dissipation.Zhiqiang Wu Jie Dong Xiuting Li Xin Zhao Wenjun Tan Chengchang Ji Qinghua Zhang 2023Journal of Materials Science & Technology2023,,18:0
4Inhibitory Effects of 12 Chemical Agents against Alternaria alternata显示文摘[Objective]The paper was to screen highly efficient fungicides for the prevention and control of tobacco brown spot(Alternaria alternata).[Method]The inhibitory effects of 12 chemical agents against A.alternata were determined indoors by mycelium growth rate method.[Result]Toxicity test showed that 30%kresoxim-methyl·boscalid SC had the best inhibitory effect,with an EC_(50)of 1.05μg/mL,followed by 9%pyraclostrobin CS,with an EC_(50)of 2.09μg/mL.As a broad spectrum protective fungicide,77%copper calcium sulfate WP showed a low sensitivity against A.alternata in the petri dish test since it had a mechanism of action different from that of other chemical agents.[Conclusion]The study lays the foundation for subsequent screening of fungicides for the prevention and control of A.alternata in the field.Shen Hong Zhang Jiatao Cui Zhiyan Sun Guangjun Li Bin Zhang Yongfeng Xu Chuantao Ai Yongfeng Song Guanglong Ji Xiuting Wang Xiaoqiang 2020Plant Diseases and Pests2020,11,4:0
