| 9 | Production of neutralizing monoclonal antibody against human vascular endothelial growth factor receptor Ⅱ显示文摘AIM: To prepare neutralizing monoclonal antibody (mAb) against extracellular immunoglobulin (Ig)-like domain Ⅲ of vascular endothelial growth factor receptor KDR and study its biological activity. METHODS: Soluble KDR Ig domain III (KDR-Ⅲ) fusion protein was expressed in E Coli and purified from the bacterial periplasmic extracts via an affinity chromatography. Monoclonal antibodies against KDR-Ⅲ were prepared by hybridoma technique. ELISA and FACS analysis were used to identify its specificity. Immunoprecipitation and [^3H]-thymidine incorporation assay were also used to detect the activity of anti-KDR mAb blocking the phosphorylation of KDR tyrosine kinase receptor and the influence on vascular endothelial growth factor-induced mitogenesis of human endothelial cells. RESULTS: A monoclonal antibody, Ycom1D3 (IgG1), was generated from a mouse immunized with the recombinant KDR-Ⅲ protein. YcomlD3 bound specifically to both the soluble KDR-Ⅲ and the cell-surface expressed KDR. Ycom1D3 effectively blocked VEGF/KDR interaction and inhibited VEGF-stimulated KDR activation in human endothelial cells. Furthermore, the antibody efficiently neutralized VEGF-induced mitogenesis of human endothelial cells. | 18 | Decabromodiphenyl ether induces the chromosome association disorders of spermatocytes and deformation failures of spermatids in mice显示文摘The environmental presence of decabromodiphenyl ether(BDE-209),which is toxic to the male reproductive system,is widespread.The current study investigated its mechanism of toxicity in mice.The results showed,that BDE-209 induced DNA damage,decreased the expression of the promoter of meiosis spermatogenesis-and oogenesis-specific basic helix-loop-helix 1(Sohlh1),meiosis related-factors Lethal(3)malignant brain tumor like 2(L3MBTL2),PIWI-like protein 2(MILI),Cyclin-dependent kinase 2(CDK2),Cyclin A,synaptonemal complex protein 1(SYCP1)and synaptonemal complex protein 3(SYCP3),and caused spermatogenic cell apoptosis,resulting in a decrease in sperm quantity and quality.Furthermore,BDE-209 downregulated the levels of anaphase-promoting complex/cyclosome(APC/C),increased the expression of PIWI-like protein 1(MIWI)in the cytoplasm of elongating spermatids,and decreased the nuclear levels of RING finger protein 8(RNF8),ubiquitinated(ub)-H2A/ub-H2B,and Protamine 1(PRM1)/Protamine 2(PRM2),while increasing H2A/H2B nuclear levels in spermatids.The reproductive toxicity was persistent for 50 days following the withdrawal of BDE-209 exposure.The results suggested that BDE-209 inhibits the initiation of meiosis by decreasing the expression of Sohlh1.Furthermore,the reduced expression of L3MBTL2 inhibited the formation of chromosomal synaptonemal complexes by depressing the expression of meiosis regulators affecting the meiotic progression and also inhibited histone ubiquitination preventing the replacement of histones by protamines,by preventing RNF8 from entering nuclei,which affected the evolution of spermatids into mature sperm. | Jinglong Xue Xiangyang Li Yafei Chi Leqiang Gao Yue Zhang Yan Wang Moxuan Zhao Jialiu Wei Zhixiong Shi Xianqing Zhou | 2024 | Journal of Environmental Sciences2024,,4: | 0 |
| 19 | The Puccinia striiformis effector Hasp98 facilitates pathogenicity by blocking the kinase activity of wheat TaMAPK4显示文摘The obligate biotrophic fungus Puccinia striiformis f.sp.tritici(Pst)employs virulence effectors to disturb host immunity and causes devastating stripe rust disease.However,our understanding of how Pst effectors regulate host defense responses remains limited.In this study,we determined that the Pst effector Hasp98,which is highly expressed in Pst haustoria,inhibits plant immune responses triggered by flg22or nonpathogenic bacteria.Overexpression of Hasp98 in wheat(Triticum aestivum)suppressed avirulent Pst-triggered immunity,leading to decreased H2O2accumulation and promoting P.striiformis infection,whereas stable silencing of Hasp98 impaired P.striiformis pathogenicity.Hasp98 interacts with the wheat mitogenactivated protein kinase TaMAPK4,a positive regulator of plant resistance to stripe rust.The conserved TEY motif of TaMAPK4 is important for its kinase activity,which is required for the resistance function.We demonstrate that Hasp98inhibits the kinase activity of TaMAPK4 and that the stable silencing of TaMAPK4 compromises wheat resistance against P.striiformis.These results suggest that Hasp98 acts as a virulence effector to interfere with the MAPK signaling pathway in wheat,thereby promoting P.striiformis infection. | Jinping Wei Xiaodong Wang Zeyu Hu Xiaojie Wang Jialiu Wang Jianfeng Wang Xueling Huang Zhensheng Kang Chunlei Tang | 2023 | Journal of Integrative Plant Biology2023,65,1: | 0 |
