| 1 | A novel hepatitis B virus genotyping system by using restriction fragment length polymorphism patterns of S gene amplicons显示文摘AIM: Traditional hepatitis B virus (HBV) genotyping methods using restriction fragment length polymorphism (RFLP) can reliably identify genotypes A to F. As HBV genotypes G and H have been recently identified, this study was to establish an accurate and simple genotyping method for all eight HBV genotypes (A to H).METHODS: Two hundred and forty HBV small S sequences obtained from GeneBank were analysed for restriction enzyme sites that would be genotype-specific. Restriction patterns following digestion with restriction enzymes BsrⅠ, StyⅠ, DpnⅠ, HpaⅡ, and EaeⅠ, were determined to identify all eight HBV genotypes. Mixed genotype infections were confirmed by cloning and further RFLP analysis.RESULTS: The new genotyping method could identify HBV genotypes A to H. Genotypes B and C could be determined by a single step digestion with BsrI and StyI in parallel. This was particularly useful in the Far East where genotypes B and C are predominant. Serum samples from 187 Chinese HBV carders were analysed with this genotyping system, and the genotype distribution was 1.1% (2), 51.9% (97), 40.6% (76) and 4.8% (9) for genotypes A, B, C, and D, respectively. Mixed genotypes were found in only 3 patients (1.6%). Sequence data analysis confirmed the validity of this new method.CONCLUSION: This HBV genotyping system can identify all eight HBV genotypes. | 2 | Effects of puerarin on number and activity of endothelial progenitor cells from peripheral blood显示文摘AIM: To investigate whether puerarin can augment endothelial progenitor cells (EPCs) numbers, promote EPC proliferative, migratory, adhesive, and in vitro vasculogenesis capacity. METHODS: EPCs were characterized as adherent cells by double staining of DiLDL-uptake and lectin binding under a laser scanning confocal microscope. Expression of KDR, VEGFR-2, and AC133 was detected by flow cytometry. EPCs proliferation, migration and in vitro vasculogenesis were determined with MTT assay, modified Boyden chamber assay, and in vitro vasculogenesis kit, respectively. EPCs adhesive assay was performed by replating those on fibronectin-coated dishes, then adherent cells were counted. RESULTS: Incubation of isolated human MNCs with puerarin 0.1-3 mmol/L increased the number of EPCs, EPC proliferative, migratory, adhesive, and in vitro vasculogenesis capacity in a concentration-and time-dependent manner, which reached maximum at 3 mmol/L, 24 h (approximately 1-fold increase, P<0.01). | 3 | Therapeutic efficacy of high-dose vitamin C on acute pancreatitis and its potential mechanisms显示文摘AIM: To observe the therapeutic efficacy of high-dose Vitamin C (Vit. C) on acute pancreatitis (AP), and to explore its potential mechanisms.METHODS: Eghty-four AP patients were divided into treatment group and control group, 40 healthy subjects were taken as a normal group. In the treatment group,Vit. C (10 g/day) was given intravenously for 5 days,whereas in the control group, Vit. C (1 g/day) was given intravenously for 5 days. Symptoms, physical signs, duration of hospitalization, complications and mortality rate were monitored. Meanwhile, serum amylase, urine amylase and leukocyte counts were also determined. The concentration of plasma vitamin C (P-VC), plasma lipid peroxide (P-LPO),plasma vitamin E (P-VE), plasma β-carotene (P-13-CAR),whole blood glutathione (WB-GSH) and the activity of erythrocyte surperoxide dimutase (E-SOD) and erythrocyte catalase (E-CAT) as well as T lymphocyte phenotype were measured by spectrophotometry in the normal group and before and after treatment with Vit. C in the treatment and the control group.RESULTS: Compared with the normal group, the average values of P-VC, P-VE, P-β-CAR, WB-GSH and the activity of E-SOD and E-CAT in AP patients were significantly decreased and the average value of P-LPO was significantly increased, especially in severe acute pancreatitis (SAP) patients (P<0.05. P-VC, P=0.045; P-VE, P=0.038; P=0.041;P-β-CAR, P=0.046; WB-GSH, P=0.039; E-SOD, P=0.019;E-CAT, P =0.020; P-LPO, P =0.038). Compared with the normal group, CD3 and CD4 positive cells in AP patients were significantly decreased. The ratio of CD4/CD8 and CD4 positive cells were decreased, especially in SAP patients (P<0.05. CD4/CD8, P =0.041; CD4, P =0.019). Fever and vomiting disappeared, and leukocyte counts and amylase in urine and blood become normal quicker in the treatment group than in the control group. Moreover, patients in treatment group also had a higher cure rate, a lower complication rate and a shorter in-ward days compared with those in he control group. Alter treatment, the average value of P-VC was significantly higher and the values of SIL-2R, TNF-α, IL-6 and IL-8 were significantly lower in the treatment group than in the control group (P<0.05 P-VC,P=0.045; SIL-2R, P=0.012; TNF-α, P=0.030; IL-6, P=0.015;and IL-8, P =0.043). In addition, the ratio of CD4/CD8 and CD4 positive cells in the patients of treatment group were significantly higher than that of the control group after treatment (P<0.05. CD4/CD8, P =0.039; CD4, P =0.024).CONCLUSION: High-dose vitamin C has therapeutic efficacy on acute pancreatitis. | 4 | Construction of phospholamban antisense RNA recombinant adenoassociated virus vector and its effects in rat cardiomyocytes显示文摘Aim: To construct a recombinant adeno-associated virus (rAAV) vector containing gene encoding phospholamban antisense RNA (asPLB), and analyse its effect on expression of PLB, expression and activity of sarco-endoplasmic reticulum Ca^2+-ATPase (SERCA), and the change of intracellular free Ca^2+ concentration ([Ca^2+]i) in rat cardiomyocytes. Methods: The target gene encoding PLB antisense RNA was inserted inversely into the adeno-associated virus plasmid pAAV-MCS digested by corresponding restricted endonuclease enzyme. The recombinant plasmid and pAAV-RC and pHelper were co-transfected into 293 cell. At the same time, a viral production positive control (rAAV-LacZ) and negative control were performed. The recombinant viruses were used to transfect the cultured rat cardiomyocytes. Site β-Galactosidase staining were performed to observe the transfer efficiency. Reverse transcription-PCR and Western blot were used to determine the mRNA and protein expression of PLB and SERCA. The activity of SERCA and the [Ca^2+]i were measured. Results: The rAAV vectors were constructed successfully and were transfected into rat cardiomyocytes effectively.The PLB mRNA and protein expression were reduced in rat cardiomyocytes transfected by rAAV-asPLB compared with controls. The activity of SERCA was increased. In rest state, the level of [Ca^2+]i in the rAAV-asPLB transfected groupdecreased. | 5 | Computation on Sentence Semantic Distance for Novelty Detection显示文摘Novelty detection is to retrieve new information and filter redundancy from given sentences that are relevant to a specific topic. In TREC2003, the authors tried an approach to novelty detection with semantic distance computation.The motivation is to expand a sentence by introducing semantic information. Computation on semantic distance between sentences incorporates WordNet with statistical information. The novelty detection is treated as a binary classification problem: new sentence or not. The feature vector, used in the vector space model for classification, consists of various factors, including the semantic distance from the sentence to the topic and the distance from the sentence to the previous relevant context occurring before it. New sentences are then detected with Winnow and support vector machine classifiers,respectively. Several experiments are conducted to survey the relationship between different factors and performance. It is proved that semantic computation is promising in novelty detection. The ratio of new sentence size to relevant size is further studied given different relevant document sizes. It is found that the ratio reduced with a certain speed (about 0.86).Then another group of experiments is performed supervised with the ratio. | 6 | Enhancing crystal structure prediction by decomposition and evolution schemes based on graph theory显示文摘Crystal structure prediction algorithms have become powerful tools for materials discovery in recent years, however, they are usually limited to relatively small systems. The main challenge is that the number of local minima grows exponentially with the system size. In this work, we proposed two crossover-mutation schemes based on graph theory to accelerate the evolutionary structure searching by automatic decomposition methods. These schemes can detect molecules or clusters inside periodic networks using quotient graphs for crystals, and the decomposition can dramatically reduce the searching space.
