| 1 | Allogeneic Vγ9Vδ2 T-cell immunotherapy exhibits promising clinical safety and prolongs the survival of patients with late-stage lung or liver cancer显示文摘Vγ9Vδ2 T cells are promising candidates for cellular tumor immunotherapy.Due to their HLA-independent mode of action,allogeneic Vγ9Vδ2 T cells can be considered for clinical application.To apply allogeneic Vγ9Vδ2 T cells in adoptive immunotherapy,the methodology used to obtain adequate cell numbers with optimal effector function in vitro needs to be optimized,and clinical safety and efficacy also need to be proven.Therefore,we developed a novel formula to improve the expansion of peripheralγδT cells from healthy donors.Then,we used a humanized mouse model to validate the therapeutic efficacy of expandedγδT cells in vivo;furthermore,the expandedγδT cells were adoptively transferred into late-stage liver and lung cancer patients.We found that the expanded cells possessed significantly improved immune effector functions,including proliferation,differentiation,and cancer cell killing,both in vitro and in the humanized mouse model.Furthermore,a phase I clinical trial in 132 late-stage cancer patients with a total of 414 cell infusions unequivocally validated the clinical safety of allogeneic Vγ9Vδ2 T cells.Among these 132 patients,8 liver cancer patients and 10 lung cancer patients who received≥5 cell infusions showed greatly prolonged survival,which preliminarily verified the efficacy of allogeneic Vγ9Vδ2 T-cell therapy.Our clinical studies underscore the safety and efficacy of allogeneic Vγ9Vδ2 T-cell immunotherapy,which will inspire further clinical investigations and eventually benefit cancer patients. | Yan Xu Zheng Xiang Mohammed Alnaggar Léonce Kouakanou Jiawei Li Junyi He Jiashuang Yang Yi Hu Yan Chen Li Lin Jianlei Hao Jingxia Li Jibing Chen Man Li Qingling Wu Christian Peters Qinghua Zhou Jianshuang Li Yingqing Liang Xiaohua Wang Baohui Han Meili Ma Dieter Kabelitz Kecheng Xu Wenwei Tu Yangzhe Wu Zhinan Yin | 2021 | Cellular & Molecular Immunology2021,18,2: | 8 |

| 7 | New Metabolic Alterations and A Predictive Marker Pipecolic Acid in Sera for Esophageal Squamous Cell Carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a major histological subtype of esophageal cancer with a poor prognosis.Although several serum metabolomic investigations have been reported,ESCC tumor-associated metabolic alterations and predictive biomarkers in sera have not been defined.Here,we enrolled 34 treatment-naive patients with ESCC and collected their pre-and post-esophagectomy sera together with the sera from 34 healthy volunteers for a metabolomic survey.Our comprehensive analysis identified ESCC tumor-associated metabolic alterations as represented by a panel of 12 serum metabolites.Notably,postoperative abrosia and parenteral nutrition substantially perturbed the serum metabolome.Furthermore,we performed an examination using sera from carcinogen-induced mice at the dysplasia and ESCC stages and identified three ESCC tumor-associated metabolites conserved between mice and humans.Notably,among these metabolites,the level of pipecolic acid was observed to be progressively increased in mouse sera from dysplasia to cancerization,and it could be used to accurately discriminate between mice at the dysplasia stage and healthy control mice.Furthermore,this metabolite is essential for ESCC cells to restrain oxidative stress-induced DNA damage and cell proliferation arrest.Together,this study revealed a panel of 12 ESCC tumor-associated serum metabolites with potential for monitoring therapeutic efficacy and disease relapse,presented evidence for refining parenteral nutrition composition,and highlighted serum pipecolic acid as an attractive biomarker for predicting ESCC tumorigenesis. | Lei Liu Jia Wu Minxin Shi Fengying Wang Haimin Lu Jibing Liu Weiqin Chen Guanzhen Yu Dan Liu Jing Yang Qin Luo Yan Ni Xing Jin Xiaoxia Jin Wen-Lian Chen | 2022 | Genomics, Proteomics & Bioinformatics2022,20,4: | 0 |

| 9 | Effect of Al_(82.8)Cu_(17)Fe_(0.2) alloy doping on structure and magnetic properties of SmCo5-based ribbons显示文摘This work tries to improve the magnetic properties by multi-element doping in the form of a ternary alloy.SmCo_(5+)χwt%Al-Cu-Fe(x=0-7)ribbons melt-spun at 40 m/s were produced by adding Al_(82.8)Cu_(17)Fe_(0.2)alloy into SmCo_(5) matrix,and their phases,microstructure,and magnetic properties were investigated.The results show that both x=0 and 3 ribbons form a cellular microstructure.Al-Cu-Fe addition reduces the content of the Sm_(2)(Co,M)_(7) cell wall,narrows its width,and forms the local disordered micro-regions and solute-segregation nanoclusters in the Sm(Co,M)_(5) grains.With x increasing to5,Al-Cu-Fe addition promotes the phase separation between and within grains of the SmCo_(5)-based alloy.The Al-Cu-Fe addition can simultaneously improve the coercivity and magnetization of the SmCo_(5)-based ribbons,in particular,the magnetization of the x=3 ribbons increases by 35%,while the coercivity of the x=5 ribbons increases by 3.9 times.Finally,the microstructure evolution models are built up,and the relationship between the microstructure and the magnetic properties is discussed. | Zhixin Dong Shu Wang Siyi Chen Yunfei Cao Zihui Ge Jibing Sun | 2022 | Journal of Rare Earths2022,40,1: | 0 |
