| 1 | Visualization of the complex structure and stress field inside rock by means of 3D printing technology显示文摘Accurate characterization and visualization of the complex inner structure and stress distribution of rocks are of vital significance to solve a variety of underground engineering problems. In this paper, we incorporate several advanced technologies, such as CT scan, three-dimensional(3D) reconstruction, and 3D printing, to produce a physical model representing the natural coal rock that inherently contains complex fractures or joints. We employ 3D frozen stress and photoelastic technologies to characterize and visualize the stress distribution within the fractured rock under uniaxial compression. The 3D printed model presents the fracture structures identical to those of the natural prototype. The mechanical properties of the printed model,including uniaxial compression strength, elastic modulus,and Poisson's ratio, are testified to be similar to those of the prototype coal rock. The frozen stress and photoelastic tests show that the location of stress concentration and the stress gradient around the discontinuous fractures are in good agreement with the numerical predictions of the real coalsample. The proposed method appears to be capable of visually quantifying the influences of discontinuous,irregular fractures on the strength, deformation, and stress concentration of coal rock. It is proved that for each distributive hypercontinuous lattice L, the space SpecL of nonunit prime elements endowed with the hull-kernel topology is locally strongly compact, and for each locally strongly compact space X, the complete lattice of all open sets O(X) is distributive hypercontinuous. For the case of distributive hyperalgebraic lattices, the similar result is given. | 9 | Lattice-Boltzmann simulation of microscale CH_4 flow in porous rock subject to force-induced deformation显示文摘Accurate knowledge of the influence of rock deformation on the permeability of fluid flow is of great significance to a variety of engineering applications, such as simultaneous extraction of coal and gas, oil/gas exploitation, CO2 geological sequestration, and underground water conservation. Based on the CT representation of pore structures of sandstones, a LBM(Lattice Boltzmann Method) for simulating CH4 flow in pore spaces at microscale levels and a parallel LBM algorithm for largesize porous models are developed in this paper. The properties of CH4 flow in porous sandstones and the effects of pore structure are investigated using LBM. The simulation is validated by comparing the results with the measured data. In addition, we incorporate LBM and FEM to probe the deformation of microstructures due to applied triaxial forces and its influence on the properties of CH4 flow. It is shown that the proposed method is capable of visually and quantitatively describing the characteristics of microstructure, spatial distribution of flow velocity of CH4,permeability, and the influences of deformation of pore spaces on these quantities as well. | 12 | Gas Film Disturbance Characteristics Analysis of High-Speed and High-Pressure Dry Gas Seal显示文摘The dry gas seal(DGS) has been widely used in high parameters centrifugal compressor, but the intense vibrations of shafting,especially in high-speed condition, usually result in DGS's failure. So the DGS's ability of resisting outside interference has become a determining factor of the further development of centrifugal compressor. However, the systematic researches of which about gas film disturbance characteristics of high parameters DGS are very little. In order to study gas film disturbance characteristics of high-speed and high-pressure spiral groove dry gas seal(S-DGS) with a flexibly mounted stator, rotor axial runout and misalignment are taken into consideration, and the finite difference method and analytical method are used to analyze the influence of gas film thickness disturbance on sealing performance parameters, what's more, the effects of many key factors on gas film thickness disturbance are systematically investigated. The results show that, when sealed pressure is 10.1MPa and seal face average linear velocity is 107.3 m/s, gas film thickness disturbance has a significant effect on leakage rate, but has relatively litter effect on open force; Excessively large excitation amplitude or excessively high excitation frequency can lead to severe gas film thickness disturbance; And it is beneficial to assure a smaller gas film thickness disturbance when the stator material density is between 3.1 g/cm^3 to 8.4 g/cm^3; Ensuring sealing performance while minimizing support axial stiffness and support axial damping can help to improve dynamic tracking property of dry gas seal. | 13 | Solution structure of the RNA recognition domain of METTL3-METTL14 N^6-methyladenosine methyltransferase显示文摘N^6-methyladenosine(m6A),a ubiquitous RNA modification,is installed by METTL3-METTL14 complex.The structure of the heterodimeric complex between the methyltransferase domains(MTDs)of METTL3 and METTL14 has been previously determined.However,the MTDs alone possess no enzymatic activity.Here we present the solution structure for the zinc finger domain(ZFD)of METTL3,the inclusion of which fulfills the methyltransferase activity of METTL3-METTL14.We show that the ZFD specifically binds to an RNA containing 5'-GGACU-3'consensus sequence,but does not to one without.The ZFD thus serves as the target recognition domain,a structural feature previously shown for DNA methyltransferases,and cooperates with the MTDs of METTL3-METTL14 for catalysis.However,the interaction between the ZFD and the specific RNA is extremely weak,with the binding affinity at several hundred micromolar under physiological conditions.The ZFD contains two CCCH-type zinc fingers connected by an anti-parallel P-sheet.Mutational analysis and NMR titrations have mapped the functional interface to a contiguous surface.As a division of labor,the RNA-binding interface comprises basic residues from zinc finger 1 and hydrophobic residues fromβ-sheet and zinc finger 2.Further we show that the linker between the ZFD and MTD of METTL3 is flexible but partially folded,which may permit the cooperation between the two domains during catalysis.Together,the structural characterization of METTL3 ZFD paves the way to elucidate the atomic details of the entire process of RNA m6A modification. | Jinbo Huang Xu Dong Zhou Gong Ling-Yun Qin Shuai Yang Yue-Ling Zhu Xiang Wang Delin Zhang Tingting Zou Ping Yin Chun Tang | 2019 | Protein & Cell2019,10,4: | 11 |

Here we show that the M2 isoform of pyruvate kinase (PKM2), one of the rate-limiting enzymes in glycolysis, interacts with mitofusin 2 (MFN2), a key regulator of mitochondrial fusion, to promote mitochondrial fusion and OXPHOS, and attenuate glycolysis. mTOR increases the PKM2:MFN2 interaction by phosphorylating MFN2 and thereby modulates the effect of PKM2: MFN2 on glycolysis, mitochondrial fusion and OXPHOS. | 18 | Diurnal and Seasonal Variations of CO2Fluxes and Their Climate Controlling Factors for a Subtropical Forest in Ningxiang显示文摘In this study, the diurnal and seasonal variations of CO2 fluxes in a subtropical mixed evergreen forest in Ningxiang of Hunan Province, part of the East Asian monsoon region, were quantified for the first time. The fluxes were based on eddy covariance measurements from a newly initiated flux tower. The relationship between the CO2 fluxes and climate factors was also analyzed. The results showed that the target ecosystem appeared to be a clear carbon sink in 2013, with integrated net ecosystem CO2exchange(NEE), ecosystem respiration(RE), and gross ecosystem productivity(GEP) of-428.8, 1534.8 and1963.6 g C m-2yr-1, respectively. The net carbon uptake(i.e. the-NEE), RE and GEP showed obvious seasonal variability,and were lower in winter and under drought conditions and higher in the growing season. The minimum NEE occurred on12 June(-7.4 g C m-2d-1), due mainly to strong radiation, adequate moisture, and moderate temperature; while a very low net CO2 uptake occurred in August(9 g C m-2month-1), attributable to extreme summer drought. In addition, the NEE and GEP showed obvious diurnal variability that changed with the seasons. In winter, solar radiation and temperature were the main controlling factors for GEP, while the soil water content and vapor pressure deficit were the controlling factors in summer. | 19 | Structural Insights into N^6-methyladenosine (m^6A) Modification in the Transcriptome显示文摘More than 100 types of chemical modifications in RNA have been well documented.Recently, several modifications, such as N^6-methyladenosine(m^6 A), have been detected in m RNA,opening the window into the realm of epitranscriptomics. The m^6 A modification is the most abundant modification in m RNA and non-coding RNA(nc RNA). At the molecular level, m^6 A affects almost all aspects of m RNA metabolism, including splicing, translation, and stability, as well as micro RNA(mi RNA) maturation, playing essential roles in a range of cellular processes. The m^6 A modification is regulated by three classes of proteins generally referred to as the 'writer'(adenosine methyltransferase), 'eraser'(m^6 A demethylating enzyme), and 'reader'(m^6 A-binding protein). The m^6 A modification is reversibly installed and removed by writers and erasers, respectively.Readers, which are members of the YT521-B homology(YTH) family proteins, selectively bind to RNA and affect its fate in an m^6 A-dependent manner. | 20 | Investigation of the characteristics and deactivation of catalytic active center of Cr-Al_2O_3 catalysts for isobutane dehydrogenation显示文摘Deactivation mechanism of Cr-Al2O3 catalyst and the interaction of Cr-Al in the dehydrogenation of isobutane, as well as the nature of the catalytic active center, were studied using XRD, SEM, XPS, H2-TPR, isobutane-TPR and TPO techniques. The results revealed that the deactivation of Cr-Al2O3 catalyst was mainly caused by carbon deposition on its surface. The Cr3+ion could not be reduced by hydrogen but could be reduced to Cr2+by hydrocarbons and monoxide carbon. The active center for isobutane dehydrogenation could be Cr2+/Cr3+produced from Cr6+by the on line reduction of hydrocarbon and carbon monoxide.
