1Novel discovery of schisandrin A regulating the interplay of autophagy and apoptosis in oligoasthenospermia by targeting SCF/c-kit and TRPV1 via biosensors显示文摘Oligoasthenospermia is the primary cause of infertility.However,there are still enormous challenges in the screening of critical candidates and targets of oligoasthenospermia owing to its complex mechanism.In this study,stem cell factor(SCF),c-kit,and transient receptor potential vanilloid 1(TRPV1)biosensors were successfully established and applied to studying apoptosis and autophagy mechanisms.Interestingly,the detection limit reached 2.787×10^(-15)g/L,and the quantitative limit reached 1.0×10^(-13)g/L.Furthermore,biosensors were used to investigate the interplay between autophagy and apoptosis.Schisandrin A is an excellent candidate to form a system with c-kit similar to SCF/c-kit with a detection constant(K_(D))of 5.701×10^(-11)mol/L,whereas it had no affinity for SCF.In addition,it also inhibited autophagy in oligoasthenospermia through antagonizing TRPV1 with a K_(D) of up to 4.181×10^(-10)mol/L.In addition,in vivo and in vitro experiments were highly consistent with the biosensor.In summary,high-potency schisandrin A and two potential targets were identified,through which schisandrin A could reverse the apoptosis caused by excessive autophagy during oligoasthenospermia.Our study provides promising insights into the discovery of effective compounds and potential targets via a well-established in vitro-in vivo strategy.Lijuan Ma Boyi Li Jinchen Ma Chunyuan Wu Nan Li Kailin Zhou Yun Yan Mingshuang Li Xiaoyan Hu Hao Yan Qi Wang Yanfei Zheng Zhisheng Wu 2023Acta Pharmaceutica Sinica B2023,13,6:1
2Mapping of de novo mutations in primary biliary cholangitis to a disease-specific co-expression network underlying homeostasis and metabolism显示文摘Primary biliary cholangitis(PBC) is an autoimmune disease involving dysregulation of a broad array of homeostatic and metabolic processes. Although considerable single-nucleotide polymorphisms have been unveiled, a large fraction of risk factors remains enigmatic. Candidate genes with rare mutations that tend to confer more deleterious effects need to be identified. To help pinpoint cellular and developmental mechanisms beyond common noncoding variants, we integrate whole exome sequencing with integrative network analysis to investigate genes harboring de novo mutations. Prominent convergence has been revealed on a network of disease-specific co-expression comprised of 55 genes associated with homeostasis and metabolism. The transcription factor gene MEF2 D and the DNA repair gene PARP2 are highlighted as hub genes and identified to be up-and down-regulated, respectively, in peripheral blood data set. Enrichment analysis demonstrates that altered expression of MEF2 D and PARP2 may trigger a series of molecular and cellular processes with pivotal roles in PBC pathophysiology. 4Comparative study on cultivation of microalgae for nutrient removal and lipid production in different artificial wastewaters显示文摘Wastewater contains high concentration of nutrients,like nitrogen and phosphorus,which have been identified as the main reasons for water eutrophication and serious ecological issues.Therefore,cultivating a tolerant and adaptive microalgae strain in wastewater is considered as a promising approach for sustainable biomass/lipid production.The potential usages of Desmodesmus sp.for biomass and lipid production within different artificial wastewater(AW)were investigated and the removal efficiencies of nutrient were compared.The maximum removal rate of chemical oxygen demand,ammonia nitrogen,nitrate nitrogen and phosphate were 272 mg/(L·d),14.021 mg/(L·d),7.774 mg/(L·d)and 3.347 mg/(L·d),respectively in AW2,AW3,AW5 and AW2.Maximum biomass(1.159 g/L)and lipid(280 mg/L)productions were observed in AW5,while the highest lipid content achieved was 37.42%in AW1.Fatty acid analysis showed that lipids extracted from AW-cultivated Desmodesmus sp.contained 59.57%-77.79%polyunsaturated fatty acids(30.6%-44.47%was linoleic acid).Hao Rui Yu Zhen Li Jinchen Gao Min Ma Weiling Zhu Yi 2017International Journal of Agricultural and Biological Engineering2017,10,1:1
