2Laboratory model study of newly deposited dredger fills using improved multiple-vacuum preloading technique显示文摘Problems continue to be encountered concerning the traditional vacuum preloading method in field during the treatment of newly deposited dredger fills.In this paper,an improved multiple-vacuum preloading method was developed to consolidate newly dredger fills that are hydraulically placed in seawater for land reclamation in Lingang Industrial Zone of Tianjin City,China.With this multiplevacuum preloading method,the newly deposited dredger fills could be treated effectively by adopting a novel moisture separator and a rapid improvement technique without sand cushion.A series of model tests was conducted in the laboratory for comparing the results from the multiple-vacuum preloading method and the traditional one.Ten piezometers and settlement plates were installed to measure the variations in excess pore water pressures and moisture content,and vane shear strength was measured at different positions.The testing results indicate that water dischargeetime curves obtained by the traditional vacuum preloading method can be divided into three phases:rapid growth phase,slow growth phase,and steady phase.According to the process of fluid flow concentrated along tiny ripples and building of larger channels inside soils during the whole vacuum loading process,the fluctuations of pore water pressure during each loading step are divided into three phases:steady phase,rapid dissipation phase,and slow dissipation phase.An optimal loading pattern which could have a best treatment effect was proposed for calculating the water discharge and pore water pressure of soil using the improved multiple-vacuum preloading method.For the newly deposited dredger fills at Lingang Industrial Zone of Tianjin City,the best loading step was 20 kPa and the loading of 40-50 k Pa produced the highest drainage consolidation.The measured moisture content and vane shear strength were discussed in terms of the effect of reinforcement,both of which indicate that the multiple-vacuum preloading method has a better treatment effect not only in decreasing the moisture content and increasing the bearing capacity,but also in increasing the process uniformity at different depths of foundation.Jingjin Liu Huayang Lei Gang Zheng Haizuo Zhou Xuelian Zhang 2017Journal of Rock Mechanics and Geotechnical Engineering2017,9,5:7
3Construction of a recombinant attenuated Salmonella typhimurium DNA vaccine carrying Helicobacter pylorihpaA显示文摘AIM: To construct a recombinant attenuated Salmonella typhimurium DNA vaccine carrying Helicobacter pylori hpaA gene and to detect its immunogenicity.METHODS: Genomic DNA of the standard H pylori strain 17 874 was isolated as the template, hpaA gene fragment was amplified by polymerase chain reaction (PCR) and cloned into pUCmT vector. DNA sequence of the amplified hpaA gene was assayed, then cloned into the eukaryotic expression vector pIRES through enzyme digestion and ligation reactions. The recombinant plasmid was used to transform competent Escherichia coliDH5α, and the positive clones were screened by PCR and restriction enzyme digestion. Then, the recombinant pIRES-hpaA was used to transform LB5000 and the recombinant plasmid isolated from LB5000 was finally used to transform SL7207. After that, the recombinant strain was grown in vitrorepeatedly. In order to iclentify the immunogenicity of the vaccinein vitro, the recombinant pIRES-hpaA was transfected to COS-7 cells using LipofectamineTM2000, the immunogenicity of expressed HpaA protein was detected with SDS-PAGE and Western blot.RESULTS: The 750-base pair hpaA gene fragment was amplified from the genomic DNA and was consistent with the sequence of H pylori hpaA by sequence analysis. It was confirmed by PCR and restriction enzyme digestion that H pylori hpaA gene was inserted into the eukaryotic expression vector pIRES and a stable recombinant live attenuated Salmonella typhimurium DNA vaccine carrying H pylori hpaA gene was successfully constructed and the specific strip of HpaA expressed by pIRES-hpaA was detected through Western blot.CONCLUSION: The recombinant attenuated Salmonella typhimurium DNA vaccine strain expressing HpaA protein with immunogenicity can be constructed and it may be helpful for further investigating the immune action of DNA vaccine in vivo.CanXu Zhao-ShenLi Yi-QiDu Zhen-XingTu Yan-FangGong JingJin Hong-YuWu Guo-MingXu 2005World Journal of Gastroenterology2005,11,1:6
4Genetical study on the parthenogenesis in Laminaria japonica显示文摘The parthenogenesis and natural doubling of chromosomes in wild type female gametophytes of Laminaria japonica were studied. The results indicate that not all the female gametophytes from the wild type hybrid parent can propagate through parthenogenesis. Most parthenosporophytes can mature, their spores germinate into gametophytes, the latter then developed into female sporophytes. To form these parthenosporophytes the natural doubling of chromosomes occurred mainly at the first and second cell divisions of the spores. 5Prediction of anther-expressed gene regulation in Arabidopsis显示文摘Anther development in Arabidopsis, a popular model plant for plant biology and genetics, is controlled by a complex gene network. Despite the extensive use of this genus for genetic research, little is known about its regulatory network. In this paper, the direct transcriptional regulatory relationships between genes expressed in Arabidopsis anther development were predicted with an integrated bioinformatic method that combines mining of microarray data with promoter analysis. A total of 7710 transcription factor-gene pairs were obtained. The 80 direct regulatory relationships demonstrating the highest confidence were screened from the initial 7710 pairs; three of the 80 were validated by previous experiments. The results indicate that our predicted results were reliable. The regulatory relationships revealed by this research and described in this paper may facilitate further investigation of the molecular mechanisms of anther development. 6Comparison of in-vial exhaust method versus conventional exhaust method in the injection of COVID-19 vaccine显示文摘Objective:The objective of the study was to compare the application effects of in-vial exhaust method and conventional exhaust method in the process of coronavirus disease 2019 vaccine injection.Materials and Methods:Using convenient sampling method,102 vaccines were selected as experiment group during the process of vaccine injection,and the in-vial exhaust method was used.One hundred and five vaccines were selected as the control group and the conventional exhaust method was adopted.The incidence of vaccine solution spillage and exhausting time in the two groups during exhaust were observed.Results:The incidence of solution spillage in the experiment group was lower than that in the control group(0 vs.6.67%,P<0.05).The exhausting time of the experiment group was shorter than that of the control group([15.12±4.43]s vs.[22.74±6.53]s,P<0.05).Conclusion:Implementing the in-vial exhaust method in the vaccine injection can effectively reduce the incidence of solution spillage,reduce nucleic acid contamination,and ensure that the vaccine is injected at the prescribed dose.Moreover,the operation is simple and easy,which improves the nurse’s vaccination efficiency,and has a higher promotion and application value.Jingjin XU Hui ZHI Ye LI Jinjing LIU Wen ZHENG Ling TANG 2021Journal of Integrative Nursing2021,3,3:2
7Transient tribo-dynamic analysis of crosshead slipper in lowspeed marine diesel engines during engine startup显示文摘A crosshead slipper-guide system,which bears a significant thrust force,is an essential friction pair in low-speed marine diesel engines.Owing to the low moving speed of the crosshead slipper during engine startup,it is difficult to form good hydrodynamic lubrication between the crosshead slipper and guide.Therefore,a detailed analysis of the crosshead slipper during engine startup is needed.In this study,a new transient tribo-dynamic model for a crosshead slipper during the engine startup process is presented.The model consists of a mixed lubrication model of the crosshead slipper-guide and dynamic models of the piston assembly,crosshead assembly,connecting rod,and crankshaft.The tribo-dynamic performances of the crosshead slipper during startup and under the rated conditions were simulated and compared.The results show that the tribo-dynamics of the crosshead slipper during the startup process are significantly different from those under the rated conditions.Some measures beneficial for the low friction of a crosshead slipper-guide under the rated conditions may significantly increase the friction loss of the crosshead slipper-guide system during the startup process.Rui LI Xianghui MENG Jingjin DONG Wenda LI 2021Friction2021,9,6:2

9Application of five-element music therapy in pain coping skills training in patients with knee osteoarthritis显示文摘Objective:The objective of this study is to assess the application effect of five elements music therapy introduced in the pain coping skills training of knee osteoarthritis(KOA).Materials and Methods:Totally,80 patients with KOA were selected and randomly divided into the experimental group(39 cases)and the control group(41 cases).The control group was only given routine nursing measures,and the experimental group was additionally treated with five-element music therapy on the basis of the control group,twice a day,28 days in total.The Western Ontario and McMaster Universities Osteoarthritis Index(WOMAC)was used to evaluate the functional status of the knee joint of the two groups.The clinical efficacy of the two groups was evaluated by Guiding Principles for Clinical Research of New Chinese Medicine in the Treatment of Osteoarthritis.Results:WOMAC score statistically significantly decreased in the experimental group(35.92±9.48 vs.16.17±5.43,P<0.01)and the control group(36.73±6.42 vs.22.53±7.51,P<0.01)after 28 days of intervention when compared with that before intervention;WOMAC score in the experimental group was lower than that of the control group after 28 days of intervention(16.17±5.43 vs.22.53±7.51,P<0.01).The total effective rate of the experimental group was statistically higher than that of the control group(82.0%vs.51.2%,χ2=11.97,P=0.003).Conclusion:The combination of five-element music therapy and routine nursing measures has better effect in relieving pain and bad emotions of patients with KOA when compared with routine nursing measures alone.Suqian LI Jingjin XU Ling TANG Ye LI Huaxin WANG Lixue ZHAO Jianshuang YAO Shuying WANG Nan LI 2021Journal of Integrative Nursing2021,3,4:2
10Structural basis of the ultrasensitive calcium indicator GCaMP6显示文摘GCaMP is one of the most widely used calcium indicators in neuronal imaging and calcium cell biology.The newly developed GCaMP6 shows superior brightness and ultrasensitivity to calcium concentration change.In this study,we determined crystal structures of Ca2+-bound GCaMP6 monomer and dimer and presented detailed structural analyses in comparison with its parent version GCaMP5G.Our analyses reveal the structural basis for the outperformance of this newly developed Ca2+indicator.Three substitution mutations and the resulting changes of local structure and interaction explain the ultrasensitivity and increased fluorescence intensity common to all three versions of GCaMP6.Each particular substitution in the three GCaMP6 is also structurally consistent with their differential sensitivity and intensity,maximizing the potential of using GCaMP6 in solving diverse problems in neuronal research and calcium signaling.Our studies shall also be beneficial to further structure-guided optimization of GCaMP and facilitate the design of novel calcium indicators.DING JingJin LUO Andrew F. 15Association of an Increased Risk of Pre-eclampsia and Fetal Growth Restriction in Singleton and Twin Pregnancies with Female Fetuses显示文摘Objective: To investigate whether the fetal gender affects the incidence of pre-eclampsia (PE) and fetal growth restriction (FGR) in singleton and twin pregnancies.Methods: This was a 10-year single-center, retrospective, cohort study from January 2009 to January 2019. A total of 57,129 singleton and 3699 twin pregnancies aged between 18-55 years old were recruited at the Third Affiliated Hospital of Guangzhou Medical University, China. We used multivariable logistic regression to analyze the effect of fetal gender on the incidence of PE and FGR.Results: In singleton pregnancies, the incidence rates of PE and FGR with a female fetus were higher than those with a male fetus (6.4% (1713/26,793)vs. 5.9% (1803/30,336),P < 0.05 and 3.5% (932/26,793)vs. 2.4% (745/30,336),P < 0.05, respectively). A female fetus was an independent risk factor for either PE or FGR (adjusted odds ratio: 1.169 or 1.563;95% confidence interval: 1.036-1.319 or 1.349-1.810, respectively). In twin pregnancies, the incidence of early-onset PE was greater in pregnancies with two females compared with two males or one male plus one female (4.6% (46/1003)vs. 4.1% (54/1305)vs. 2.4% (33/1391),P < 0.05). 18Long non-coding RNA HCAR promotes endochondral bone repair by upregulating VEGF and MMP13 in hypertrophic chondrocyte through sponging miR-15b-5p显示文摘Endochondral bone formation is an important route for bone repair.Although emerging evidence has revealed the functions of long non-coding RNAs(lncRNAs)in bone and cartilage development,the effect of lncRNAs in endochondral bone repair is still largely unknown.Here,we identified a lncRNA,named Hypertrophic Chondrocyte Angiogenesis-related lncRNA(HCAR),and proved it to promote the endochondral bone repair by upregulating the expression of matrix metallopeptidase 13(Mmp13)and vascular endothelial growth factorα(Vegfa)in hypertrophic chondrocytes.Lnc-HCAR knockdown in hypertrophic chondrocytes restrained the cartilage matrix remodeling and decrease the CD31hiEmcnhi vessels number in a bone repair model.Mechanistically,we proved that lnc-HCAR was mainly enriched in the cytoplasm using fluorescence in situ hybridization(FISH)assay,and it acted as a molecular sponge for miR-15b-5p.Further,in hypertrophic chondrocytes,lnc-HCAR competitively bound to miR-15b-5p to increase Vegfa and Mmp13 expression.Our results proved that lncRNA is deeply involved in endochondral bone repair,which will provide a new theoretical basis for future strategies for promoting fracture healing.Yun Bai Xiaoshan Gong Rui Dong Zhen Cao Ce Dou Chuan Liu Jianmei Li Fei Kang Jingjin Dai Chunrong Zhao Zhansone Tian Jiulin Tan Qijie Dai Shiwu Dong 2022Genes & Diseases2022,9,2:1

20Effects of Melatonin on Growth,Physiology and Gene Expression in Rice Seedlings Under Cadmium Stress显示文摘Melatonin(MLT)is a hormonal substance found in many organisms and can improve plant stress resistance.In this study,the japonica rice variety Y32 and indica rice variety NJ6 were cultivated in hydroponics under different concentrations of CdCl2 at the two-leaf stage.The growth,physiological and biochemical responses of the seedlings and the expression of cadmium(Cd)-related genes under exogenous melatonin(MLT)treatment were assessed.The results indicated that Cd stress destroyed the dynamic balance between reactive oxygen species(ROS)production and removal,resulting in ROS accumulation,membrane lipid peroxidation,and impaired growth and development.Following the application of exogenous MLT to rice seedlings,increases in plant biomass including both underground and above-ground areas were observed.MLT also scavenged the inhibition of superoxide dismutase(SOD)and peroxidase(POD)in a concentration dependent manner in response to Cd stress.Catalase(CAT)activity and malondialdehyde(MDA)expression also decreased following MLT treatment.Amongst the six Cd-related genes assessed,five genes were downregulated and one was up-regulated in response to MLT treatment.Taken together,these data demonstrate that MLT improves the resilience of rice seedlings at the biochemical,physiological,and molecular levels,and diminishes the damage caused by Cd stress.Xiachen Lv Yunxia Fang Lantian Zhang Weiyi Zhang Ling Xu Jingjin Han Bailing Jin Xian Zhang Xiaoqin Zhang Dawei Xue 2019Phyton-International Journal of Experimental Botany2019,88,2:1
