1Spinal cord biological safety of image-guided radiation therapy versus conventional radiation therapy显示文摘Tumor models were simulated in purebred Beagles at the T9-10 levels of the spinal cord and treated with spinal image-guided radiation therapy or conventional radiation therapy with 50 or 70 Gy total radiation. Three months after radiation, neuronal injury at the T9-10 levels was observed, including reversible injury induced by spinal image-guided radiation therapy and apoptosis induced by conventional radiation therapy. The number of apoptotic cells and expression of the proapoptotic protein Fas were significantly reduced, but expression of the anti-apoptotic protein heat shock protein 70 was significantly increased after image-guided radiation therapy compared with the conventional method of the same radiation dose. Moreover, the spinal cord cell apoptotic index positively correlated with the ratio of Fas/heat shock protein 70. These findings indicate that 3 months of radiation therapy can induce a late response in the spinal cord to radiation therapy; image-guided radiation therapy is safer and results in less neuronal injury compared with conventional radiation therapy.Wanlong Xu Xilinbaoleri Hao Liu Ruozheng Wang Jingping Bai 2012Neural Regeneration Research2012,7,35:23
2Active Achilles tendon kinesitherapy accelerates Achilles tendon repair by promoting neurite regeneration显示文摘Active Achilles tendon kinesitherapy facilitates the functional recovery of a ruptured Achilles tendon. However, protein expression during the healing process remains a controversial issue. New Zealand rabbits, aged 14 weeks, underwent tenotomy followed immediately by Achilles tendon microsurgery to repair the Achilles tendon rupture. The tendon was then immobilized or subjected to postoperative early motion treatment (kinesitherapy). Mass spectrography results showed that after 14 days of motion treatment, 18 protein spots were differentially expressed, among which, 12 were up-regulated, consisting of gelsolin isoform b and neurite growth-related protein collapsing response mediator protein 2. Western blot analysis showed that gelsolin isoform b was up-regulated at days 7-21 of motion treatment. 3Retrieval of Total Suspended Matters Using Field Spectral Data in Shitoukoumen Reservoir,Jilin Province,Northeast China显示文摘From August to October in 2006,three times of field spectral measurements with a Field Spec FR spectroradiometer(Analytical Spectral Devices,Inc.,USA) were carried out in Shitoukoumen Reservoir,Jilin Province,Northeast China. Owing to the serious soil and water loss in the upstream,reflectance curves of the reservoir were characterized by high concentrations of total suspended matter(TSM) . Extending the spectral analysis to 1200nm in the near-infrared band,this research revealed an obvious reflectance peak around 1070nm which was caused by the strong backscattering of high TSM. The method of partial least squares(PLS) regression was applied to retrieving the TSM. Reflectance in two spectral bands,i.e.,675-948nm and 1029-1105nm,were used as variables to develop PLS models. Traditional linear regression,first derivative model and logarithmic model were also used for the comparison of different models. Results showed that the PLS model based on Rrs(675) -Rrs(948) gave out best results with high precision and stability. 4Rare inherited missense variants of POGZ associate with autism risk and disrupt neuronal development显示文摘Excess de novo likely gene-disruptive and missense variants within dozens of genes have been identified in autism spectrum disorder(ASD)and other neurodevelopmental disorders.However,many rare inherited missense variants of these high-risk genes have not been thoroughly evaluated.In this study,we analyzed the rare missense variant burden of POGZ in a large cohort of ASD patients from the Autism Clinical and Genetic Resources in China(ACGC)and further dissected the functional effect of diseaseassociated missense variants on neuronal development.Our results showed a significant burden of rare missense variants in ASD patients compared to the control population(P=4.6×10-5,OR=3.96),and missense variants in ASD patients showed more severe predicted functional outcomes than those in controls.Furthermore,by leveraging published large-scale sequencing data of neurodevelopmental disorders(NDDs)and sporadic case reports,we identified 8 de novo missense variants of POGZ in NDD patients.Functional analysis revealed that two inherited,but not de novo,missense variants influenced the cellular localization of POGZ and failed to rescue the defects in neurite and dendritic spine development caused by Pogz knockdown in cultured mouse primary cortical neurons.Significantly,L1CAM,an autism candidate risk gene,is differentially expressed in POGZ deficient cell lines.Reduced expression of L1cam was able to partially rescue the neurite length defects caused by Pogz knockdown.Our study showed the important roles of rare inherited missense variants of POGZ in ASD risk and neuronal development and identified the potential downstream targets of POGZ,which are important for further molecular mechanism studies.Wenjing Zhao Jieqiong Tan Tengfei Zhu Jianjun Ou Ying Li Lu Shen Huidan Wu Lin Han Yanling Liu Xiangbin Jia Ting Bai Honghui Li Xiaoyan Ke Jingping Zhao Xiaobing Zou Zhengmao Hu Hui Guo Kun Xia 2019Journal of Genetics and Genomics2019,46,5:0

7Growth potential of human hepatocarcinoma cells in the liver of neonatal immunocompetent mice and its relation to immunological tolerance显示文摘To determine the pathological behavior of human hepatocarcinoma cells in the liver microenvironment of neonatal non-immunode-ficient mice, three human hepatocarcinoma cell lines (Bel7402, HepG2, and SK-Hep-1), traced by DiI, were transplanted into the intrahepatic or subcutaneous tissue of neonatal and adult Kunming mice. Histopathological observations showed that cells in the adult liver induced a severe immune response as early as the second day after the implantation, while the subcutaneous neoplasm underwent extensive necrosis by the end of the study. Only the cells injected into the neonatal liver underwent a delayed immunologic rejection in the organ microenvironment. These cells retained recognizable tumor features over the first seven days, and displayed an intrahepatic invasive pattern. The expression of tumor markers including alpha-fetoprotein and survivin was maintained. The quantitative ELISA for the expression patterns of IL-2 and IL-10 also confirmed that the intrahepatic immunity was non-susceptive during this period. The high serum alpha-fetoprotein level was inversely correlated with the change in immune response.
