| 1 | Water quality monitoring in a slightly-polluted inland water body through remote sensing-Case study of the Guanting Reservoir in Beijing,China显示文摘This study focused on the water quality of the Guanting Reservoir,a possible auxiliary drinking water source for Beijing.Through a remote sensing(RS)approach and using Landsat 5 Thematic Mapper(TM)data,water quality retrieval models were established and analyzed for eight common water quality variables,including algae content,turbidity,and concentrations of chemical oxygen demand,total nitrogen,ammonia nitrogen,nitrate nitrogen,total phosphorus,and dissolved phosphorus.The results show that there exists a statistically significant correlation between each water quality variable and remote sensing data in a slightly-polluted inland water body with fairly weak spectral radiation.With an appropriate method of sampling pixel digital numbers and multiple regression algorithms,retrieval of the algae content,turbidity,and nitrate nitrogen concentration was achieved within 10% mean relative error,concentrations of total nitrogen and dissolved phosphorus within 20%,and concentrations of ammonia nitrogen and total phosphorus within 30%.On the other hand,no effective retrieval method for chemical oxygen demand was found.These accuracies were acceptable for the practical application of routine monitoring and early warning on water quality safety with the support of precise traditional monitoring.The results show that performing the most traditional routine monitoring of water quality by RS in relatively clean inland water bodies is possible and effective. | Weiqi HE Shan CHEN Xuehua LIU Jining CHEN | 2008 | Frontiers of Environmental Science & Engineering2008,2,2: | 10 |
| 2 | Clinical relationship between MDR1 gene and gallbladder cancer显示文摘BACKGROUND: The most common mechanisms of mul- tidrug resistance (MDR) in cancer cells is the expression of an energy-dependent exfflux pump. P-glycoprotein (P-gp) encoded by MDR1 gene and multidrug associated protein (MRP) are well known proteins associated with MDR. In human cancers, the MDR1 gene expression is common in patients with intrinsic and acquired MDR. It is a major therapeutic problem in cancer chemotherapy. Previously we found that the MDR of HCC is related to MRP gene ex- pression and initiates the intrinsic MDR. The aim of this study is to study the expression of MDR1 gene encoding P-gp and MDR1 mRNA in primary gallbladder carcinoma, and analyze its clinical significance. METHODS: Immunohistochemistry (IHC) S-P method and in situ polymerase chain reaction (ISPCR) were used to detect the expression of P-gp and MDR1 mRNA in 53 cases of untreated primary gallbladder carcinoma and 12 ca- ses of cholecystitis (archival paraffin-embedded tissues). RESULTS: The positive expression rates of P-gp and MDR1 mRNA in the 53 cases and 12 cases were 60.38%, 71.69% and 25.00%, 33.33%, respectively. There was a significant difference between the two groups (P<0.05). The positive expression rate of P-gp and MDRlmRNA were 69.44%, 83.33% and 41.18%, 47.06% respectively in tissues in stage of Nevin against Nevin , (P<0.05). In well, moderately differentiated gallbladder carcinoma tissues, their expressions were 79.49%, 69.23% against 50.00%, 35.71% in low, undifferentiated tissues (P<0.05). CONCLUSIONS: MDR to gallbladder carcinoma is closely related to the intrinsic MDR and it provides an important evidence to reverse the MDR by detection of the MDR1gene. | 3 | Molecular diversity of arbuscular mycorrhizal fungi at a large-scale antimony mining area in southern China显示文摘Arbuscular mycorrhizal fungi(AMF) have great potential for assisting heavy metal hyperaccumulators in the remediation of contaminated soils. However, little information is available about the community composition of AMF under natural conditions in soils contaminated by antimony(Sb). The objective of this study was to investigate the characteristics of AMF molecular diversity, and to explore the effects of Sb content and soil properties on the AMF community structure in an Sb mining area. Four Sb mine spoils and one adjacent reference area were selected from around the Xikuangshan mine in southern China. The association of AMF molecular diversity and community composition with the rhizosphere soils of the dominant plant species was studied by Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis(PCR-DGGE). Results from all five studied sites showed that the diversity of AMF decreased with increasing Sb concentration. Principal component analysis(PCA) indicated that the AMF community structure was markedly different among these groups. Further redundancy analysis(RDA) showed that Sb contamination was the dominating factor influencing the AMF community structure in the Sb mine area. However, the multivariate analysis showed that, apart from the soil Sb content, extractable nitrogen content and organic matter content also attributed to AMF sequence distribution type. Some AMF sequences were only found in the highly contaminated area and these might be ideal candidates for improving phytoremediation efficiency in Sb mining regions. | 7 | Mining of a streptothricin gene cluster from Streptomyces sp.TP-A0356 genome via heterologous expression显示文摘Streptothricins (STs) are used commercially to treat bacterial and fungal diseases in agriculture. Mining of the sequenced microbial genomes uncovered two cryptic ST clusters from Streptomyces sp. C and Streptomyces sp. TP-A0356. The ST cluster from S. sp. TP-A0356 was verified by successful heterologous expression in Streptomyces coelicolor M145. Two new ST analogs were produced together with streptothricin F and streptothricin D in the heterologous host. The ST cluster was further confirmed by inactivation of gene stnO, which was proposed encoding an aminomutase supplying -lysines for the poly-β-Lys chain formation. | 12 | Modeling of nitrobenzene in the river with ice process in high-latitude regions显示文摘To deal with the accidental release of pollutants into frozen rivers, those rivers in high-latitude regions should be treated differently. Pollutants could be stored up in the ice and released when the ice melts in spring, perhaps resulting in secondary pollution in the river. A water quality model of nitrobenzene has been developed in this paper to assess the influence on river water quality due to the freezing process. The model is made up of three modules: thermodynamic module, hydrodynamic module and water quality module. The thermodynamic module considers the complex heat exchange processes between the water body and the atmosphere, the water body and the river bed, the water body and the ice cover, and so on. The growth of the ice cover is simulated in a simplified form whilst taking consideration of heat balance. The hydrodynamic model uses the Saint-Venant equations in non-constant flow and the influence of the ice cover is measured. The degradation, diffusion, absorption and desorption, and the influence of the freezing process are incorporated in the water quality module and the concept of Con- tinuously Stirred Tanked Reactors (CSTRs) model is applied in the model construction. The model has been applied to supporting the management of accidental pollution in the Songhua River. The model was calibrated and validated with monitored data. Regional sensitivity analysis based on a task-based Hornberger-Spear-Young (HSY) algorithm was carried out to examine the model structure and the re- sult showed that the model could describe the system very well. Then the conditioned model was ap- plied to predicting the concentration of nitrobenzene in the water when the ice melted in spring. The predictive result showed that the release of pollutants in the ice could lead to an increase in the con- centration of pollutants in the water, but the increase would be very small because there was only a small amount of pollutants stored in the ice. A secondary pollution could therefore be avoided. | 13 | Lipid-lowering effects of oleanolic acid in hyperlipidemic patients显示文摘Oleanolic acid(OA) is a pentacyclic triterpenoid compound extracted from olea europaeal, a traditional Chinese medicine herb. OA has been used in the clinic as a hepatoprotective medicine in China since 1970 s. In our previous study, we observed that OA could ameliorate hyperlipidemia in animal models. In the present study, we conducted a small-scale clinical trial to evaluate the hypolipidemia effect of OA in hyperlipidemic patients. Hyperlipidemic patients were administrated with OA for four weeks(4 tablets once, three times a day). The blood samples of the patients were collected before and after OA treatment. The biological parameters were measured. Furthermore, three patients' blood samples were studied with DNA microarray. After OA administration, the TC, TG, and HDLC levels in serum decreased significantly. DNA microarray analysis results showed that the expressions of 21 m RNAs were significantly changed after OA treatment. Bioinformatics analysis showed 17 m RNAs were up-regulated and 4 m RNAs were down-regulated significantly after OA treatment. Five m RNAs(CACNA1 B, FCN, STEAP3, AMPH, and NR6 A1) were selected to validate the expression levels by q RT-PCR. Therefore, OA administration differentially regulated the expression of genes involved in lipid metabolism.
