| 1 | Generation of new glutinous rice by CRISPR/Cas9-targeted mutagenesis of the Waxy gene in elite rice varieties显示文摘In rice, amylose content(AC) is controlled by a single dominant Waxy gene. We used Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)/CRISPR-associated 9(Cas_9) to introduce a loss-of-function mutation into the Waxy gene in two widely cultivated elite japonica varieties. Our results show that mutations in the Waxy gene reduce AC and convert the rice into glutinous ones without affecting other desirable agronomic traits, offering an effective and easy strategy to improve glutinosity in elite varieties. Importantly, we successfully removed the transgenes from the progeny. | 5 | Corrosion Behavior of Mg-Zn-Y Alloy with Long-period Stacking Ordered Structures显示文摘Mg-Zn-Y alloys with long-period stacking ordered structures were prepared by an ingot casting method. The corrosion performance of Mg-Zn-Y alloys was studied by combining gas-collecting test, immersion test and electrochemical measurements in order to determine the corrosion rate and mechanism of the alloys. The results showed that the volume fraction of Mg(12)YZn phase increased and the shape of the Mg(12)YZn phase changed from discontinuous to continuous net-like with increasing Zn and Y content. The corrosion rate of the alloys greatly depended on the distribution and volume fraction of the Mg(12)YZn phase. Corrosion products appeared at the junction of Mg phase and Mg(12)YZn phase, indicating that the Mg(12)YZn phase accelerated galvanic corrosion of Mg matrix. | 6 | Effects of applying lactic acid bacteria to the fermentation on a mixture of corn steep liquor and air-dried rice straw显示文摘This study was to determine the fermentation quality of a mixture of corn steep liquor(CSL)(178 g/kg wet basis) and air-dried rice straw(356 g/kg wet basis) after being treated with inoculants of different types of lactic acid bacteria(LAB). The treatments included the addition of no LAB additive(control),which was deionized water; homo-fermentative LAB alone(^(ho)LAB), which was Lactobacillus plantarum alone), and a mixture of homo-fermentative and hetero-fermentative LAB(^(he+ho)LAB), which were L. plantarum, Lactobacillus casei, and Lactobacillus buchneri. The results showed that the inoculation of the mixture of CSL and air-dried rice straw with ^(he+ho)LAB significantly increased the concentration of acetic acid and lactic acid compared with the control(P < 0.05). The addition of ^(he+ho)LAB effectively inhibited the growth of yeast in the silage. The concentration of total lactic acid bacteria in the ^(he+ho)LAB-treated silage was significant higher than those obtained in other groups(P < 0.05). The duration of the aerobic stability of the silages increased from 56 h to >372 h. The control group was the first to spoil, whereas the silage treated with ^(he+ho)LAB remained stable throughout the 372 h period of monitoring. | 7 | Position and complications of pedicle screw insertion with or without image-navigation techniques in the thoracolumbar spine:a meta-analysis of comparative studies显示文摘Computer-navigated pedicle screw insertion is applied to the thoracic and lumbar spine to attain high insertion accuracy and a low rate of screw-related complications.However,some in vivo and in vitro studies have shown that no advantages are gained with the use of navigation techniques compared to conventional techniques.Additionally,inconsistent conclusions have been drawn in various studies due to different population characteristics and methods used to assess the accuracy of screw placement.Moreover,it is not clear whether pedicle screw insertion with navigation techniques decreases the incidence of screw-related complications.Therefore,this study was sought to perform a meta-analysis of all available prospective evidence regarding pedicle screw insertion with or without navigation techniques in human thoracic and lumbar spine.We considered in vivo comparative studies that assessed the results of pedicle screw placement with or without navigation techniques.PubMed,Ovid MEDLINE and EMBASE databases were searched.Three published randomized controlled trials(RCTs) and nine retrospective comparative studies met the inclusion criteria.These studies included a total of 732 patients in whom 4,953 screws were inserted.In conclusion,accuracy of the position of grade I,II,III and IV screws and complication rate related to pedicle screw placement were significantly increased when navigation techniques were used in comparison to conventional techniques.Future research in this area should include RCTs with well-planned methodology to limit bias and report on validated,patient-based outcome measures. | Jinshan Tang Ziqiang Zhu Tao Sui Dechao Kong Xiaojian Cao | 2014 | The Journal of Biomedical Research2014,28,3: | 15 |

| 9 | Phosphorylation of LTF1, an MYB Transcription Factor in Populus, Acts as a Sensory Switch Regulating Lignin Biosynthesis in Wood Cells显示文摘Lignin is specifically deposited in plant secondary cell walls,and initiation of lignin biosynthesis is regulated by a variety of developmental and environmental signals.However,the mechanisms governing the regulation of lignin biosynthesis remain to be elucidated.In this study,we identified a lignin biosynthesis-associated transcription factor(LTF)from Populus,LTF1,which binds the promoter of a key lignin biosynthetic gene encoding 4-coumarate-CoA ligase(4CL).We showed that LTF1 in its unphosphorylated state functions as a regulator restraining lignin biosynthesis.When LTF1 becomes phosphorylated by PdMPK6 in response to external stimuli such as wounding,it undergoes degradation through a proteasome pathway,resulting in activation of lignification.Expression of a phosphorylation-null mutant version of LTF1 led to stable protein accumulation and persistent attenuation of lignification in wood cells.Taken together,our study reveals a mechanism whereby LTF1 phosphorylation acts as a sensory switch to regulate lignin biosynthesis in response to environmental stimuli.The discovery of novel modulators and mechanisms modifying lignin biosynthesis has important implications for improving the utilization of cell-wall biomass. | Jinshan Gui Laifu Luo Yu Zhong Jiayan Sun Toshiaki Umezawa Laigeng Li | 2019 | Molecular Plant2019,12,10: | 10 |

| 12 | Control of Surface Thermal Scratch of Strip in Tandem Cold Rolling显示文摘The thermal scratch seriously affects the surface quality of the cold rolled stainless steel strip. Some researchers have carried out qualitative and theoretical studies in this field. However, there is currently a lack of research on effective forecast and control of thermal scratch defects in practical production, especially in tandem cold rolling. In order to establish precise mathematical model of oil film thickness in deformation zone, the lubrication in cold rolling process of SUS410L stainless steel strip is studied, and major factors affecting oil film thickness are also analyzed. According to the principle of statistics, mathematical model of critical oil film thickness in deformation zone for thermal scratch is built, with fitting and regression analytical method, and then based on temperature comparison method, the criterion for deciding thermal scratch defects is put forward. Storing and calling data through SQL Server 2010, a software on thermal scratch defects control is developed through Microsoft Visual Studio 2008 by MFC technique for stainless steel in tandem cold rolling, and then it is put into practical production. Statistics indicate that the hit rate of thermal scratch is as high as 92.38%, and the occurrence rate of thermal scratch is decreased by 89.13%. Owing to the application of the software, the rolling speed is increased by approximately 9.3%. Based on the presented model, the bonding mechanism is simulated and analyzed, different detonation modes are also comparatively studied to indicate the driving force spread in few microseconds. The results show that the three layer plates bond together after many times of impact between the flyers and the base driven by detonation wave, which is damping rapidly at each impact with wavelength decreasing. The pressure at the detonation point is minimal, which induces non-bonding of the plates here. | 19 | Rhizosphere and bulk soil enzyme activities in a Nothotsuga longibracteata forest in the Tianbaoyan National Nature Reserve,Fujian Province,China显示文摘The rhizosphere, distinct from bulk soil, is defined as the volume of soil around living roots and influenced by root activities. We investigated protease,invertase, cellulase, urease, and acid phosphatase activities in rhizosphere and bulk soils of six Nothotsuga longibracteata forest communities within Tianbaoyan National Nature Reserve, including N. longibracteata + either Phyllostachys pubescens, Schima superba, Rhododendron simiarum, Cunninghamia lanceolata, or Cyclobalanopsis glauca, and N. longibracteata pure forest. Rhizosphere soils possessed higher protease, invertase, cellulase, urease,and acid phosphatase activities than bulk soils. The highest invertase, urease, and acid phosphatase activities were observed in rhizosphere samples of N. longibracteata+ S.superba. Protease was highest in the N. longibracteata + R. simiarum rhizosphere, while cellulase was highest in the pure N. longibracteata forest rhizosphere.All samples exhibited obvious rhizosphere effects on enzyme activities with a significant linear correlation between acid phosphatase and cellulase activities(p \ 0.05) in rhizosphere soils and between protease and acid phosphatase activities(p \ 0.05) in bulk soils. | 20 | Effect of different levels of corn steep liquor addition on fermentation characteristics and aerobic stability of fresh rice straw silage显示文摘The object of this study was to determine the proper mixing ratio of fresh rice straw to corn steep liquor(CSL) to obtain a high protein content silage feed. The following experimental silages were generated: the control(C1), composed of fresh rice straw without CSL additive, mixed with CSL in the ratios of 4:1(C4),3:1(C3) and 2:1(C2). Lactic acid bacteria(LAB) inoculant was applied at the rate of 50 mL/kg(fresh basis)of forage to achieve a final application rate of 1 x 10~6 cfu/g of fresh matter(FM). Duplicate silos for each treatment were opened after 0,3, 7,10,20,30,45 and 60 d for microbiological and chemical analysis. The results showed that the addition of CSL significantly increased crude protein(CP) contents, and decreased neutral detergent fiber(NDF) and acid detergent fiber(ADF) contents of treatments after 60 d of ensiling(P < 0.05). The lactic acid contents in C4 and C3 were significantly higher than that in C1(P <0.05). In summary, mixing fresh rice straw with CSL at addition levels of 4:1(C4) and 3:1(C3) can improve the fermentation quality and nutrient composition of fresh rice straw silage.
