| 2 | Activin A prevents neuron-like PC12 cell apoptosis after oxygen-glucose deprivation显示文摘In this study, PC12 cells were induced to differentiate into neuron-like cells using nerve growth facto and were subjected to oxygen-glucose deprivation. Cells were treated with 0, 10, 20, 30, 50, 100 ng/mL exogenous Activin A. The 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl tetrazolium bromide assay and Hoechst 33324 staining showed that the survival percentage of PC12 cells significantly decreased and the rate of apoptosis significantly increased after oxygen-glucose deprivation. Exogenous Activin A significantly increased the survival percentage of PC12 cells in a dose-dependent manner. Reverse transcription-PCR results revealed a significant increase in Activin receptor IIA, Smad3 and Smad4 mRNA levels, which are key sites in the Activin A/Smads signaling pathway, in neuron-like cells subjected to oxygen-glucose deprivation, while mRNA expression of the apoptosis-regulation gene caspase-3 decreased. | 4 | Equilibrium Joining Strategies in the M/M/1 Queues with Setup Times under N-Policy显示文摘This paper carries out a game-theoretic analysis of a single-server queueing system with setup times under N-policy by considering both the partially observable and the partially unobservable information scenarios. The server switches off whenever the system becomes empty, and is resumed when the number of customers reaches a certain threshold value. Customers decide whether to join or to balk the system upon arrival based on their available information. The equilibrium joining strategy of customers as well as the systemzs performance measures are derived under different information levels. We find that both Follow-the-Crowd (FTC) and Avoid-the-Crowd (ATC) behaviors exist in our system. | 5 | CDK7-YAP-LDHD axis promotes D-lactate elimination and ferroptosis defense to support cancer stem cell-like properties显示文摘Reprogrammed cellular metabolism is essential for maintaining cancer stem cells(CSCs)state.Here,we report that mitochondrial D-lactate catabolism is a necessary initiating oncogenic event during tumorigenesis of esophageal squamous cell carcinoma(ESCC).We discover that cyclin-dependent kinase 7(CDK7)phosphorylates nuclear Yes-associated protein 1(YAP)at S127 and S397 sites and enhances its transcription function,which promotes D-lactate dehydrogenase(LDHD)protein expression.Moreover,LDHD is enriched significantly in ESCC-CSCs rather than differentiated tumor cells and high LDHD status is connected with poor prognosis in ESCC patients.Mechanistically,the CDK7-YAP-LDHD axis helps ESCC-CSCs escape from ferroptosis induced by D-lactate and generates pyruvate to satisfy energetic demands for their elevated self-renewal potential.Hence,we conclude that esophageal CSCs adopt a D-lactate elimination and pyruvate accumulation mode dependent on CDK7-YAP-LDHD axis,which drives stemness-associated hallmarks of ESCC-CSCs.Reasonably,targeting metabolic checkpoints may serve as an effective strategy for ESCC therapy. | Mengzhu Lv Ying Gong Xuesong Liu Yan Wang Qingnan Wu Jie Chen Qingjie Min Dongyu Zhao Xianfeng Li Dongshao Chen Di Yang Danna Yeerken Rui Liu Jinting Li Weimin Zhang Qimin Zhan | 2023 | Signal Transduction and Targeted Therapy2023,8,9: | 1 |
| 6 | Defect of SLC38A3 promotes epithelial-mesenchymal transition and predicts poor prognosis in esophageal squamous cell carcinoma显示文摘Objective: Solute carrier family 38(SLC38 s) transporters play important roles in amino acid transportation and signaling transduction. However, their genetic alterations and biological roles in tumors are still largely unclear.This study aimed to elucidate the genetic signatures of SLC38 s transporters and their implications in esophageal squamous cell carcinoma(ESCC).Methods: Analyses on somatic mutation and copy number alterations(CNAs) of SLC38 A3 were performed as described. Immunohistochemistry(IHC) assay and Western blot assay were used to detect the protein expression level. MTS assay, colony formation assay, transwell assay and wound healing assay were used to explore the malignant phenotypes of ESCC cells. Immunofluorescence assay was used to verify the colocalization of two indicated proteins and immunopreciptation assay was performed to confirm the interaction of proteins.Results: Our findings revealed that SLC38 s family was significantly disrupted in ESCC, with high frequent CNAs and few somatic mutations. SLC38 A3 was the most frequent loss gene among them and was linked to poor survival and lymph node metastasis. The expression of SLC38 A3 was lower in tumor tissues compared to that in normal tissues, which was also significantly associated with worse clinical outcome. Further experiments revealed that depletion of SLC38 A3 could promote EMT in ESCC cell lines, and the interaction of SLC38 A3 and SETDB1 might lead to the reduced transcription of Snail. | 11 | CoSe_(2)supported single Pt site catalysts for hydrogen peroxide generation via two-electron oxygen reduction显示文摘Electrocatalytic oxygen reduction reaction(ORR)to prepare H_(2)O_(2)in acidic medium has the advantages of green,safety,and portability,which shows broad development prospects.However,it still suffers from low catalyst activity,insufficient selectivity,and high cost.Herein,Pt_(1)/CoSe_(2)with ultralow 0.01 wt.%Pt atomic distribution was synthesized by a simple hydrothermal method.The Pt_(1)/CoSe_(2)with ultralow Pt content exhibits high activity,high selectivity,and long-term stability for ORR to H_(2)O_(2)in O_(2)-saturated 0.1 M HClO_(4).The onset potential is as low as 0.75 V versus reversible hydrogen electrode(RHE),H_(2)O_(2)selectivity is as high as 84%(0.4 V vs.RHE),and the electron transfer number is 2.3(0.4 V vs.RHE).Moreover,the hydrogen peroxide yield using the flow cell testing is 110.02 mmol g_(cat).^(−1)h^(−1) with high Faradaic efficiency of 78%(0 V vs.RHE)at 0.1 M HClO_(4),and the catalyst did not deactivate significantly after 60 h stability testing.Mechanistic studies and in situ X-ray photoelectron spectroscopy characterization confirm that the ultralow Pt content on CoSe_(2)can effectively regulate the electronic structure of Co as the real active site around the Pt site,which gives a suitable Δ_(dp) value(the difference between the d-band center of the active metal site and the p-band center of the terminal oxygen in*OOH),provides an ideal*OOH binding energy,and inhibits the O-O bond breakage.This work successfully improves the intrinsic activity of the Co active sites around Pt in Pt_(1)/CoSe_(2)for acidic ORR to H_(2)O_(2)by constructing ultralow-content Pt single atom. | Xiao-Dong Zhu Qian Zhang Xiaoxuan Yang Yingnan Wang Jinting Wu Jian Gao Ji-Jun Zou Gang Wu Yong-Chao Zhang | 2023 | SusMat2023,3,3: | 0 |

| 14 | Operating conditions combination analysis method of optimal water management state for PEM fuel cell显示文摘The water content of proton exchange membrane fuel cells(PEMFCs)affects the transport of reactants and the conductivity of the membrane.Effective water management measures can improve the performance and extend the lifespan of the fuel cell.The water management state of the stack is influenced by various external operating conditions,and optimizing the combination of these conditions can improve the water management state within the stack.Considering that the stack's internal resistance can reflect its water management state,this study first establishes an internal resistance-operating condition model that considers the coupling effect of temperature and humidity to determine the variation trend of total resistance and stack humidity with single-factor operating conditions.Subsequently,the water management state optimization method based on the ANN-HGPSO algorithm is proposed,which not only quantitatively evaluates the influence weights of different operating conditions on the stack's internal resistance but also efficiently and accurately obtains the optimal combination of five operating conditions:working temperature,anode gas pressure,cathode gas pressure,anode gas humidity,and cathode gas humidity to achieve the optimal water management state in the stack,within the entire range of current densities.Finally,the response surface experimental results of the stack also validate the effectiveness and accuracy of the ANN-HGPSO algorithm.The method mentioned in this article can provide effective strategies for efficient water management and output performance optimization control of PEMFC stacks. | Wenxin Wan Yang Yang Yang Li Changjun Xie Jie Song Zhanfeng Deng Jinting Tan Ruiming Zhang | 2023 | Green Energy and Intelligent Transportation2023,2,4: | 0 |
