1Association of H-FABP gene polymorphisms with intramuscular fat content in Threeyellow chickens and Hetian-black chickens显示文摘Background: To explore the relationship between the heart-type fatty acid binding protein(H-FABP) gene and intramuscular fat(IMF), a polymorphism of the second exon of the H-FABP gene was investigated in 60 Three-yellow chickens(TYCs) and 60 Hetian-black chickens(HTBCs).Results: The IMF contents of the cardiac, chest and leg muscles in HTBC were increased compared with TYC. Both TYC and HTBC populations exhibited Hardy-Weinberg Equilibrium(HWE) according to the χ2test. Three variations of the two birds were detected, namely, G939 A, G982 A and C1014 T. HTBCs with the TT genotypes exhibit increased IMF content in the chest muscles compared with the TC genotype. Thus, the G982 A site could be considered a genetic marker for selecting increased IMF content in the chest muscles of HTBC. The correlation coefficients revealed that H-FABP m RNA expression was negatively correlated with the IMF content in the cardiac, chest and leg muscles of HTBC and in the cardiac and chest muscles of TYC. 2Effects of feeding untreated, pasteurized and acidified waste milk and bunk tank milk on the performance, serum metabolic profiles, immunity, and intestinal development in Holstein calves显示文摘Background: The present experiment was performed to assess the effects of different sources of milk on the growth performance, serum metabolism, immunity, and intestinal development of calves. Eighty-four Holstein male neonatal calves were assigned to one of the following four treatment groups: those that received bunk tank milk(BTM), untreated waste milk(UWM), pasteurized waste milk(PWM), and acidified waste milk(AWM) for 21 d.Results: Calves in the BTM and AWM groups consumed more starter(P < 0.05) than those in the UWM group.Average daily gain in the UWM group was the highest(P < 0.05). Calves exhibited the highest(P < 0.05) serum total protein, albumin, total cholesterol, high density lipoprotein, triglycerides, growth hormone, immunoglobulin(Ig) A and Ig M concentrations in the UWM group, highest malondialdehyde and tumor necrosis factor-α in the PWM group(P < 0.05), and highest glutathione peroxidase and Ig G in the BTM group(P < 0.05). The jejunum and ileum of the calves in all treatments presented a slight inflammatory response. The jejunal inflammation scores were higher(P < 0.05) in the UWM and AWM groups than the BTM group; the ileal inflammation scores increased more(P < 0.05) in the AWM group than the BTM group. Jejunal immunohistochemical scores(IHS) were higher(P < 0.05) in the PWM and AWM groups than the BTM group. Compared to the other three groups, calves feeding on BTM had lower(P < 0.05) ileal IHS. Jejunal interleukin(IL)-1β, IL-8, and IL-10 mRNA expression in the UWM group was the highest(P < 0.05). Calves fed AWM increased(P < 0.05) mRNA expression of IL-8 and toll like receptor 4(TLR-4) in the jejunum and IL-8, IL-1β, IL-6, IL-8, and IL-10 in the mesenteric lymph nodes.Conclusions: Overall, bunk tank milk is the best choice for calf raising compared to waste milk. 3Simultaneous determination of bisphenols, benzophenones and parabens in human urine by using UHPLC-TQMS显示文摘Bisphenols, benzophenones and parabens may alter one or more functions of endocrine system and consequently cause adverse health effects on human. A rapid and sensitive analytical method for the simultaneous determination of six bisphenols, five benzophenones and seven parabens in human urine by using ultra-high performance liquid chromatography coupled with triple quadrupole mass spectrometry was developed and applied for human urine sample analysis. The eighteen targeted compounds were separated within 11 min. Heated-electrospray ionization was operated with multiple reaction monitoring in negative-ion mode. The limit of detection ranged from 0.01 ng/mL to 0.5 ng/mL.Seven isotope-labeled internal standards were used in the quantitative analysis. After the method was validated for sensitivity, linearity, precision, accuracy and matrix effect, it was applied for the analysis of human urine samples to evaluate the profiles of human exposure. 4Lactobacillus rhamnosus GR-1 attenuates foodborne Bacillus cereus-induced NLRP3 inflammasome activity in bovine mammary epithelial cells by protecting intercellular tight junctions显示文摘Background:Bacillus cereus is an important pathogen that causes human food poisoning,specifically diarrhea and vomiting.B.cereus can also induce mastitis in dairy cows and has a strong survival ability in milk,as it cannot be inactivated by high-temperature short-time pasteurization.Therefore,B.cereus can enter the market through pasteurized milk and other dairy products,imposing enormous hidden dangers on food safety and human health.Results:In this study,B.cereus 2101(BC)was isolated from milk samples of cows with mastitis.BC grew rapidly with strong hemolysis,making it difficult to prevent mastitis and ensure food security.MAC-T cells were treated with BC and/or Lactobacillus rhamnosus GR-1(LGR-1).Pretreatment with LGR-1 protected the integrity of tight junctions and the expression of zonula occludens-1(ZO-1)and occludin destroyed by BC.Furthermore,LGR-1 pretreatment reduced the expression of NOD-like receptor family member pyrin domain-containing protein 3(NLRP3),caspase recruitment and activation domain(ASC),Caspase-1 p20,gasdermin D(GSDMD)p30,inflammatory factors(interleukin(IL)-1βand IL-18),and cell death induced by BC.Moreover,LGR-1 pretreatment reduced NLRP3 inflammasome activity and increased expressions of ZO-1 and occludin induced by lipopolysaccharides(LPS)+ATP stimulation.MAC-T cells were transfected with NLRP3 si RNA or MCC950 and/or treated with BC and/or LGR-1.NLRP3-si RNA transfection and MCC950 attenuated BC-induced NLRP3 inflammasome activity.Expression of inflammatory cytokines and cell death suggested that the inflammatory pathway might play an important role in the induction of the NLRP3 inflammasome by BC and the protection of LGR-1.Conclusions:These results suggest that LGR-1 might be a probiotic alternative to antibiotics and could be administered to prevent mastitis in dairy cows,thus ensuring food security.Qiang Shan Ning Liu Xue Wang Yaohong Zhu Jinhua Yin Jiufeng Wang 2023Journal of Animal Science and Biotechnology2023,14,1:2
5Prepartum body conditions affect insulin signaling pathways in postpartum adipose tissues in transition dairy cows显示文摘Background: Overconditioned dairy cows are susceptible to excessive lipolysis and increased insulin resistance during the transition period.The associations among body fat reserve,insulin resistance,and lipolysis in adipose tissues(AT) remain to be elucidated.Therefore,this study aimed to investigate whether excessive fat reserves influence the insulin signaling pathway in AT postpartum.Results: Twenty multiparous dairy cows were selected and assigned to one of two groups,according to prepartum body condition score(BCS): Control group(BCS = 3.0–3.5;n = 10) and Overconditioned group(BCS ≥ 4.0;n = 10).Blood samples were collected on days-14,-7,-4,-2,-1,0,1,2,4,7,and 14 relative to parturition.Subcutaneous AT were collected on day 2 following parturition for quantitative real-time polymerase chain reaction and western blot analyses.No differences were observed between the two groups in serum glucose,non-esterified fatty acids,β-hydroxybutyric acid,tumor necrosis factor(TNF)α,insulin,or leptin concentrations during the experimental period.Compared with the control cows,the overconditioned cows had lower serum triglyceride levels and higher adiponectin concentrations.In the AT postpartum,insulin receptor mRNA and protein levels were lower in the overconditioned cows than in the control cows,and no differences were found in glucose transporter 4 mRNA.Compared with the control cows,the overconditioned cows had lower mRNA levels of TNFα and higher mRNA levels of peroxisome proliferator-activated receptor gamma(PPARγ) in AT postpartum.The phosphorylated protein kinase B(AKT) content and phosphorylation rate of AKT were increased in the overconditioned cows compared with the control cows,which suggested that the downstream insulin signaling in AT was affected.Conclusions: In the present study,transition dairy cows with higher BCS did not show more fat mobilization.The changes of insulin signaling pathway in AT postpartum of overconditioned cows may be partly related to the expression of PPARγ and TNFα,and the secretion of adiponectin.Fanjian Zhang Dan Li Qiong Wu Jian Sun Wenyi Guan Yinxu Hou Yaohong Zhu Jiufeng Wang 2019Journal of Animal Science and Biotechnology2019,10,3:2
6A novel index to evaluate discretization methods: A case study of flood susceptibility assessment based on random forest显示文摘The selection of a suitable discretization method(DM) to discretize spatially continuous variables(SCVs)is critical in ML-based natural hazard susceptibility assessment. However, few studies start to consider the influence due to the selected DMs and how to efficiently select a suitable DM for each SCV. These issues were well addressed in this study. The information loss rate(ILR), an index based on the information entropy, seems can be used to select optimal DM for each SCV. However, the ILR fails to show the actual influence of discretization because such index only considers the total amount of information of the discretized variables departing from the original SCV. Facing this issue, we propose an index, information change rate(ICR), that focuses on the changed amount of information due to the discretization based on each cell, enabling the identification of the optimal DM. We develop a case study with Random Forest(training/testing ratio of 7 : 3) to assess flood susceptibility in Wanan County, China.The area under the curve-based and susceptibility maps-based approaches were presented to compare the ILR and ICR. The results show the ICR-based optimal DMs are more rational than the ILR-based ones in both cases. Moreover, we observed the ILR values are unnaturally small(<1%), whereas the ICR values are obviously more in line with general recognition(usually 10%–30%). The above results all demonstrate the superiority of the ICR. 9Environmental siblings of black agents of human chromoblastomycosis显示文摘Agents of human chromoblastomycosis,a skin disease almost exclusively caused by members of the order Chaetothyriales,are assumed to be traumatically inoculated into the skin with sharp environmental materials such as plant thorns or wooden splinters carrying the respective opportunist.In the supposition that such fungi should have their main habitat in the environment,we investigated the occurrence of black fungi in living areas of patients with chromoblastomycosis.In South America Fonsecaea agents are prevalent as agents of the disease,while also related Cladophialophora species,known from other types of skin infections,are known from the continent.Ninety environmental isolates were preliminarily selected as possible agents of chromoblastomycosis,based on morphology.Judging from ITS sequence data isolates were attributed to the genera Cladophialophora,Cyphellophora,Exophiala,Fonsecaea,Phialophora,and Veronaea.A total of 45 fungi morphologically identified as Fonsecaea or Cladophialophora isolated from debris and thorns of living prickly plants in Brazil were processed for taxonomic studies.Phylogenetic analysis revealed that the isolates indeed belonged to the Chaetothyriales,but only rarely an agent of chromoblastomycosis was concerned;only two strains of F.pedrosoi and one F.monophora were isolated from debris plants.The remaining isolates belonged to hitherto unknown molecular siblings of Fonsecaea.Two novel taxa are introduced.V.A.Vicente M.J.Najafzadeh Jiufeng Sun R.R.Gomes D.Robl S.G.Marques C.M.P.S.Azevedo G.S.de Hoog 2014Fungal Diversity2014,,2:1

17High energy density and superior charge/discharge efficiency polymer dielectrics enabled by rationally designed dipolar polarization显示文摘Although many dielectric polymers exhibit high energy storage density(Ue)with enhanced dipolar polarization at room temperature,the substantially increased electric conduction loss at high applied electric fields and high temperatures remains a great challenge.Here,we report a strategy that high contents of medium-polar ester group and end-group(St)modification are introduced into a biode-gradable polymer polylactic acid(PLA)to synergistically reduce the loss and enhance Ue and charge-discharge efficiency(h).The resultant St-modified PLA polymer(PLA-St)exhibits an Ue of 6.5 J/cm^(3)with an ultra-high h(95.4%),far outperforming the best reported dielectric polymers.It is worth noting that the modified molecular structures can generate deep trap centers and restrict the local dipole motions in the polymer,which are responsible for the reduction of conduction loss and improvements in high-temperature capacitive performance.In addition,the PLA-St polymer shows intrinsically excellent self-healing ability and cyclic stability surviving over 500000 charge-discharge cycles.This work offers an efficient route to next-generation eco-friendly dielectric polymers with high energy density,low loss,and long-term stability.Zizhao Pan Yupeng Pan Li Li Xinwei Xu Jiufeng Dong Fei Jin Liang Sun Yujuan Niu Chen Xu Hong Wang 2023Journal of Materiomics2023,9,3:0
18Cyphellophora and its relatives in Phialophora: biodiversity and possible role in human infection显示文摘Cyphellophora is a genus of black yeast-like fungi characterised by having simple phialides with multiseptate,curved conidia.Judging from SSU and LSU data,Cyphellophora was found to be located in a well-supported clade within the Chaetothyriales comprising a number of species occurring on human skin and nail.Cyphellophora is phylogenetically close to Phialophora europaea,P.reptans and P.oxyspora,though morphologically these species produce singlecelled phialoconidia rather than multiseptate ones.Pseudomicrodochium suttonii and P.fusarioides have dark colonies and phylogenetically fit in with Cyphellophora;the type species of Pseudomicrodochium,P.aciculare,has similar,septate conidia but has a hyaline thallus.In the present study,multilocus phylogenetic analyses were combined with morphology and physiology.Sequences of the internal transcribed spacer region,the DNA dependent RNA polymerase II largest subunit and the partial beta tubulin gene were analysed for a set of 30 strains.Two novel species,Cyphellophora pauciseptata and Phialophora ambigua were discovered.Cyphellophora eucalypti was reduced to synonymy of C.guyanensis.The role of the studied fungi between colonization and infection of human skin was discussed.Putative virulence factors for these black yeast-like fungi were hypothesized to be the ability to assimilate monoaromatic hydrocarbons,to produce melanin pigments,and to tolerate the temperature of epidermal human skin.Peiying Feng Qiaoyun Lu M.J.Najafzadeh A.H.G.Gerrits van den Ende Jiufeng Sun Ruoyu Li Liyan Xi V.A.Vicente Wei Lai Chun Lu G.S.de Hoog 2014Fungal Diversity2014,,2:0
19Indoor metabolites and chemicals outperform microbiome in classifying childhood asthma and allergic rhinitis显示文摘Indoor microorganisms impact asthma and allergic rhinitis(AR),but the associated microbial taxa often vary extensively due to climate and geographical variations.To provide more consistent environmental assessments,new perspectives on microbial exposure for asthma and AR are needed.Home dust from 97 cases(32 asthma alone,37 AR alone,28 comorbidity)and 52 age-and gender-matched controls in Shanghai,China,were analyzed using high-throughput shotgun metagenomic sequencing and liquid chromatography-mass spectrometry.Homes of healthy children were enriched with environmental microbes,including Paracoccus,Pseudomonas,and Psychrobacter,and metabolites like keto acids,indoles,pyridines,and flavonoids(astragalin,hesperidin)(False Discovery Rate<0.05).A neural network co-occurrence probability analysis revealed that environmental microorganisms were involved in producing these keto acids,indoles,and pyridines.Conversely,homes of diseased children were enriched with mycotoxins and synthetic chemicals,including herbicides,insecticides,and food/cosmetic additives.Using a random forest model,characteristic metabolites and microorganisms in Shanghai homes were used to classify high and low prevalence of asthma/AR in an independent dataset in Malaysian schools(N=1290).Indoor metabolites achieved an average accuracy of 74.9%and 77.1%in differentiating schools with high and low prevalence of asthma and AR,respectively,whereas indoor microorganisms only achieved 51.0%and 59.5%,respectively.These results suggest that indoor metabolites and chemicals rather than indoor microbiome are potentially superior environmental indicators for childhood asthma and AR.This study extends the traditional risk assessment focusing on allergens or air pollutants in childhood asthma and AR,thereby revealing potential novel intervention strategies for these diseases.Yu Sun Hao Tang Shuang Du Yang Chen Zheyuan Ou Mei Zhang Zhuoru Chen Zhiwei Tang Dongjun Zhang Tianyi Chen Yanyi Xu Jiufeng Li Dan Norback Jamal Hisham Hashim Zailina Hashim Jie Shao Xi Fu Zhuohui Zhao 2023Eco-Environment & Health2023,2,4:0
20Durable and enhanced immunity against SARS-CoV-2 elicited by manganese nanoadjuvant formulated subunit vaccine显示文摘Dear editor,Vaccines are the most efficient and effective means to prevent infectious diseases,but improving the long-term protective efficacy is still a major challenge in contemporary vaccine development.1 The waning immunity varies depending on the diversification of the pathogen and the number of booster doses.1 Strategies to overcome this warrant is using adjuvants that amplify the immune response,and drive the production of memory B and T cells or long-lived plasma cells that recognize the pathogen for durable protection.2–4 Although existing adjuvants have achieved promising results,research on generating durable protective immunity is lacking in promoting vaccine development and staying ahead of global pandemics such as coronavirus disease 2019(COVID-19).The precisely designed nanoadjuvants can enhance lymph node targeting and increase antigenpresenting cell(APCs)uptake,achieving the co-delivery of adjuvants and antigens and activating innate and adaptive immune responses.5 Previously,we reported a manganese nanoadjuvant(MnARK)and receptor-binding domain(RBD)monomer antigen formulated nanovaccine.6 MnARK transported antigens to lymph nodes,activated the STING pathway,elicited strong neutralizing abilities and increased immune memory T cell percentage against the infection of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).6 Regarding the long-term protection potential of MnARK for subunit vaccine development,we further explored the durable immune regulation abilities of MnARK to a SARS-CoV-2 RBD dimer antigen,which has been used in an approved COVID-19 subunit vaccine ZF2001 with aluminum adjuvant(alum).7,8 TEM result revealed that RBD dimer could interact with BSA on MnARK surface and epitope can be well preserved(Supplementary Fig.1a).The size and zeta potential of MnARK-RBD dimer nanovaccine was~58 nm and-14 mV,respectively(Supplementary Fig.1b,c).Mengyu Guo Mingjing Cao Jiufeng Sun Ziwei Chen Xin Wang Lianpan Dai George F.Gao Yuliang Zhao Yaling Wang Chunying Chen 2024Signal Transduction and Targeted Therapy2024,9,1:0
