| 9 | Effects of Shengmai Yin(生脉饮)on pulmonary and cardiac function in coronavirus disease 2019 convalescent patients with cardiopulmonary symptoms:a randomized,double blind,multicenter control trial显示文摘OBJECTIVE:To evaluate the efficacy and safety of Shengmai Yin(生脉饮,SMY)on visual analogue scale(VAS)for cardiopulmonary symptoms in coronavirus disease 2019(COVID-19)convalescent patients.METHODS:In this randomized,double blind and multicenter controlled trial,a total of 200 COVID-19 convalescent patients who with cardiopulmonary symptoms were enrolled from three medical centers in Hubei,China.These patients were randomized divided into trial group and the control group,100 patients in each one,SMY and its placebo were applied to respectively,for two weeks.VAS of clinical symptoms included shortness of breath,hidrosis,chest distress,palpitation,and dry cough was performed at 0,1,2 weeks.Decrease in VAS of 30%or more was defined as effective,and a reduction in VAS of 0 was defined as curative.RESULTS:A total of 192 completed the study.The VAS of TCM symptoms showed there was no difference in baseline between the two groups.The VAS in both groups was down-regulated,and there was no significant difference in VAS and cure rates at the first and second week between the two groups.There was no significant difference in breath,hidrosis,palpitation,and dry cough between the two groups but SMY treatment for two weeks has remarkable therapeutic effects in chest distress than placebo.CONCLUSIONS:SMY could effectively ameliorate the symptoms of chest distress,and improve the quality of life of the COVID-19 convalescent patients. | AN Xuedong MAO Lina XIA Ping SU Wen WANG Beibei KOU Leiya ZHANG Zequan QI Meng HU Song CHEN Jing LI Xiujuan LIU Jinwei ZHOU Juan QIAO Jie LUO Dan LUO Guangwei YAN Youqin YANG Guiping DONG Dandan ZHOU Wei TAO Junxiu JIN De TONG Xiaolin WEI Li | 2023 | Journal of Traditional Chinese Medicine2023,43,1: | 1 |
| 10 | A brain structural connectivity biomarker for autism spectrum disorder diagnosis in early childhood显示文摘Background:Autism spectrum disorder(ASD)is associated with altered brain development,but it is unclear which specific structural changes may serve as potential diagnostic markers,particularly in young children at the age when symptoms become fully estab-lished.Furthermore,such brain markers need to meet the requirements of precision medicine and be accurate in aiding diagnosis at an individual rather than only a group level.Objective:This study aimed to identify and model brain-wide differences in structural connectivity using diffusion tensor imaging(DTI)in young ASD and typically developing(TD)children.Methods:A discovery cohort including 93 ASD and 26 TD children and two independent validation cohorts including 12 ASD and 9 TD children from three different cities in China were included.Brain-wide(294 regions)structural connectivity was measured using DTI(fractional anisotropy,FA)together with symptom severity and cognitive development.A connection matrix was constructed for each child for comparisons between ASD and TD groups.Pattern classification was performed on the discovery dataset and the resulting model was tested on the two independent validation datasets.Results:Thirty-three structural connections showed increased FA in ASD compared to TD children and associated with both autistic symptom severity and impaired general cognitive development.The majority(29/33)involved the frontal lobe and comprised five different networks with functional relevance to default mode,motor control,social recognition,language and reward.Overall,clas-sification achieved very high accuracy of 96.77%in the discovery dataset,and 91.67%and 88.89%in the two independent validation datasets.Conclusions:Identified structural connectivity differences primarily involving the frontal cortex can very accurately distinguish novel individual ASD from TD children and may therefore represent a robust early brain biomarker which can address the requirements of precision medicine. | Xi Jiang Xiao-Jing Shou Zhongbo Zhao Yuzhong Chen Fan-Chao Meng Jiao Le Tian-Jia Song Xin-Jie Xu Weitong Guo Xiaoyan Ke Xiao-E Cai Weihua Zhao Juan Kou Ran Huo Ying Liu Hui-Shu Yuan Yan Xing Ji-Sheng Han Song-Ping Han Yun Li Hua Lai Lan Zhang Mei-Xiang Jia Jing Liu Xuan Liu Keith M.Kendrick Rong Zhang | 2023 | Psychoradiology2023,3,1: | 0 |

| 12 | Development of plant cytosine base editors with the Cas12a system显示文摘Base editors of the Cas9 system have been widely used for precise nucleotide substitution in crops. In this study, Cas12a was applied to construct plant cytosine base editors(CBEs). The main elements of Cas12aCBEs were engineered and their efficiency was evaluated in stably transformed rice cells. An optimized ttCas12a-hyA3Bctd editor, consisting of a LbCas12a variant carrying catalytic inactive D832A and temperature-tolerance D156R double mutations, a truncated human APOBEC3B deaminase, a human RAD51 single-stranded DNA-binding domain, and double copies of UGI, outperformed other Cas12aCBEs in base editing efficiency. In T0transgenic rice plants, ttCas12a-hyA3Bctd edited an average of42.01% and a maximum of 68.75% of lines at six genomic targets. A-to-G conversions were generated in rice by an adenine base editor with a similar architecture to the optimized CBE.
