| 1 | Evaluation of quantitative contrast harmonic imaging to assess malignancy of liver tumors:A prospective controlled two-center study显示文摘AIM: To establish the extent to which contrast enhancement with SonoVue in combination with quantitative evaluation of contrast-medium dynamics facilitates the detection of hepatic tumors. METHODS: One hundred patients with histologically confirmed malignant or benign hepatic tumor (maximum size 5 cm) were analyzed. Contrast-enhanced ultrasound (bolus injection 2.5 mL SonoVue) was carried out with intermittent breath-holding technique using a multifrequency transducer (2.5-4 MHz). Native vascularization was analyzed with power Doppler. The contrast-enhanced dynamic ultrasound investigation was carried out with contrast harmonic imaging in true detection mode during the arterial,portal venous and late phases. Mechanical index was set at 0.15. Perfusion analysis was performed by post-processing of the raw data time intensity curve (TIC) analysis. The cut-off of the gray value differences between tumor and normal liver tissue was established using Receiver Operating Characteristic (ROC) analysis 64-line multi-slice computed tomography served as reference method in all cases. Magnetic resonance tomography was used additionally in 19 cases. RESULTS: One hundred patients with 59 malignant (43 colon,5 breast,2 endocrine metastases,7 hepatocellular carcinomas and 2 kidney cancers) and 41 benign (15 hemangiomas,7 focal nodular hyperplasias,5 complicated cysts,2 abscesses and 12 circumscribed fatty changes) tumors were included. The late venous phase proved to be the most sensitive for classification of the tumor type. Fifty-eight of the 59 malignant tumors were classified as true positive,and one as false negative. This resulted in a sensitivity of 98.3%. Of the 41 benign tumors,37 were classified as true negative and 4 as false negative,which corresponds to a specificity of 90.2%. Altogether,95.0% of the diagnoses were classified as correct on the basis of the histological classification. No investigator-dependency (P = 0.23) was noted. CONCLUSION: The results show the possibility of accurate prediction of malignancy of hepatic tumors with a positive prognostic value of 93.5% using advanced contrast-enhanced ultrasound. | 2 | Effects of Flower Buds Extract of Tussilago farfara on Focal Cerebral Ischemia in Rats and Inflammatory Response in BV2 Microglia显示文摘Objective: To investigate the effects of the flower buds extract of Tussilago farfara Linné(Farfarae Flos; FF) on focal cerebral ischemia through regulation of inflammatory responses in activated microglia. Methods: Brain ischemia was induced in Sprague-Dawley rats by a transient middle cerebral artery occlusion(t MCAO) for 90 min and reperfusion for 24 h. Twenty rats were randomly divided into 4 groups(n=5 per group): normal, t MCAO-induced ischemic control, t MCAO plus FF extract 300 mg/kg-treated, and t MCAO plus MK-801 1 mg/kg-treated as reference drug. FF extract(300 mg/kg, p.o.) or MK-801(1 mg/kg, i.p.) was administered after reperfusion. Brain infarction was measured by 2,3,5,-triphenyltetrazolium chloride staining. Neuronal damage was observed by haematoxylin eosin, Nissl staining and immunohistochemistry using anti-neuronal nuclei(Neu N), anti-glial fibril ary acidic protein(GFAP), and antiCD11 b/c(OX42) antibodies in ischemic brain. The expressions of inducible nitric oxide synthase(i NOS), tumor necrosis factor(TNF-α), and hypoxia-inducible factor-1 a(HIF-1α) were determined by Western blot. BV2 microglial cel s were treated with FF extract or its main bioactive compound, tussilagone with or without lipopolysaccharide(LPS). Nitric oxide(NO) production was measured in culture medium by Griess assay. The expressions of i NOS, COX-2 and pro-inflammatory cytokines mRNA were analyzed by reverse transcription-polymerase chain reaction. The expression of i NOS, and COX-2 proteins, the phosphorylation of ERK1/2, JNK, and p38 MAPK and the nuclear expression of NF-κB p65 in BV2 cells were determined by Western blot. Results: FF extract significantly decreased brain infarctions in ischemic rats(P<0.01). The neuronal death and the microglia/astrocytes activation in ischemic brains were inhibited by FF extract. FF extract also suppressed i NOS, TNF-α, and HIF-1α expression in ischemic brains. FF extract(0.2 and 0.5 mg/m L, P<0.01) and tussilagone 20 and 50 μmol/L, P<0.01) significantly decreased LPS-induced NO production in BV2 microglia through downregulation of i NOS m RNA and protein expression. FF extract and tussilagone significantly inhibited LPS-induced expression of TNF-α, IL-1β, and IL-6 m RNA, and also suppressed the phosphorylation of ERK1/2, JNK and p38 MAPK and the nuclear expression of NF-κB in a dosedependent manner. | 7 | Preslaughter diet management in sheep and goats:effects on physiological responses and microbial loads on skin and carcass显示文摘Sixteen crossbred buck goats(Kiko x Spanish;BW = 32.8 kg) and wether sheep(Dorset x Suffolk;BW = 39.9 kg) were used to determine the effect of preslaughter diet and feed deprivation time(FDT) on physiological responses and microbial loads on skin and carcasses.Experimental animals were fed either a concentrate(CD) or a hay diet(HD)for 4 d and then deprived of feed for either 12-h or 24-h before slaughter.Blood samples were collected for plasma Cortisol and blood metabolite analyses.Longisimus muscle(LM) pH was measured.Skin and carcass swabs were obtained to assess microbial loads.Plasma creatine kinase activity(863.9 and 571.7 ± 95.21 IU) and non-esterified fatty acid concentrations(1,056.1 and 589.8 + 105.01 mEq/L) were different(P< 0.05) between sheep and goats.Species and diet treatments had significant effects on the ultimate pH of LM.Pre-holding total coliform(TCC) and aerobic plate counts(APC) of skin were significantly different between species.Goats had lower(P < 0.05) TCC(2.1 vs.3.0 log_(10) CFU/cm^2) and APC(8.2 vs.8.5 log_(10) CFU/cm^2) counts in the skin compared to sheep.Preslaughter skin E.coli counts and TCC were different(P < 0.05) between species.Goats had lower(P < 0.05) counts of E.coli(2.2 vs.2.9 log_(10) CFU/cm^2) and TCC(2.3 vs.3.0 log_(10) CFU/cm^2) in the skin compared with those in sheep.Diet,species,and FDT had no effect(P> 0.05) on E.coli and TCC in carcass swab samples.The APC of carcass swab samples were only affected(P < 0.05) by the FDT.The results indicated that preslaughter dietary management had no significant changes on hormone and blood metabolite concentrations and sheep might be more prone for fecal contamination than goats in the holding pens at abattoir. | Govind Kannan Venkat R Gutta Jung Hoon Lee Brou Kouakou Will R Getz George W McCommon | 2015 | Journal of Animal Science and Biotechnology2015,6,1: | 2 |

| 9 | Gastric hyperplastic polyps causing upper gastrointestinal hemorrhage in a young adult显示文摘Here, we report a case of a young man who presented with a significant upper gastrointestinal bleed treated by endoscopic removal of multiple hyperplastic polyps. Gastric hyperplastic polyps are a relatively uncommon cause of overt gastrointestinal bleeding. While most hyperplastic gastric polyps are asymptomatic, they may present with abdominal pain, iron def iciency anemia or gastric outlet obstruction. These polyps are associated with conditions such as Helicobacter pylori gastritis and atrophic autoimmune gastritis, which predispose the epithelium to chronic inf lammation and epithelial repair.The patient presented to Northwestern Memorial Hospital in July 2011. The polyps were resected by clip-assisted snare polypectomy. Histopathologic assessment of the resected polyps demonstrated multiple, nonulcerative hyperplastic polyps measuring 1.3-1.8 cm in size, without evidence of dysplasia or malignancy. This case describes a young adult patient with multiple, large gastric polyps causing overt gastrointestinal bleeding.
