1SARS-CoV-2 promotes RIPK1 activation to facilitate viral propagation显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is the ongoing global pandemic that poses substantial challenges to public health worldwide.A subset of COVID-19 patients experience systemic inflammatory response,known as cytokine storm,which may lead to death.Receptor-interacting serine/threonine-protein kinase 1(RIPK1)is an important mediator of inflammation and cell death.Here,we examined the interaction of RIPK1-mediated innate immunity with SARS-CoV-2 infection.We found evidence of RIPK1 activation in human COVID-19 lung pathological samples,and cultured human lung organoids and ACE2 transgenic mice infected by SARS-CoV-2.Inhibition of RIPK1 using multiple smallmolecule inhibitors reduced the viral load of SARS-CoV-2 in human lung organoids.Furthermore,therapeutic dosing of the RIPK1 inhibitor Nec-1s reduced mortality and lung viral load,and blocked the CNS manifestation of SARS-CoV-2 in ACE2 transgenic mice.Mechanistically,we found that the RNA-dependent RNA polymerase of SARS-CoV-2,NSP12,a highly conserved central component of coronaviral replication and transcription machinery,promoted the activation of RIPK1.Furthermore,NSP12323L variant,encoded by the SARS-CoV-2 C14408T variant first detected in Lombardy,Italy,that carries a Pro323Leu amino acid substitution in NSP12,showed increased ability to activate RIPK1.Inhibition of RIPK1 downregulated the transcriptional induction of proinflammatory cytokines and host factors including ACE2 and EGFR that promote viral entry into cells.Our results suggest that SARS-CoV-2 may have an unexpected and unusual ability to hijack the RIPK1-mediated host defense response to promote its own propagation and that inhibition of RIPK1 may provide a therapeutic option for the treatment of COVID-19.Gang Xu Ying Li Shengyuan Zhang Haoran Peng Yunyun Wang Dekang Li Taijie Jin Zhuohao He Yilun Tong Chunting Qi Guowei Wu Kangyun Dong Jizhou Gou Yang Liu Tongyang Xiao Jing Qu Liang Li Liang Liu Ping Zhao Zheng Zhang Junying Yuan 2021Cell Research2021,31,12:6

4Investigation on Yield Behavior of 7075‑T6 Aluminum Alloy at Elevated Temperatures显示文摘Aluminum alloys have drawn considerable attention in the area of automotive lightweight.High strength aluminum alloys are usually deformed at elevated temperatures due to their poor formability at room temperature.In this work,the yield behavior of 7075 aluminum alloy in T6 temper(AA7075-T6)within the temperature ranging from 25°C to 230°C was investigated.Uniaxial and biaxial tensile tests with the aid of induction heating system were performed to determine the stress vs.strain curves and the yield loci of AA7075-T6 at elevated temperatures,respectively.Von Mises,Hill48 and Yld2000-2d yield criteria were applied to predicting yield loci which were compared with experimentally measured yield loci of the AA7075-T6.Results show that yield stress corresponding to the same equivalent plastic strain decreases with increasing temperature within the investigated temperature range and the shape of yield loci evolves nearly negligibly.The experimental yield locus expands with an increase of equivalent plastic strain at the same temperature and the work hardening rate of AA7075-T6 exhibits obvious stress-state-dependency.The nonquadratic Yld2000-2d yield criterion describes the yield surfaces of AA7075-T6 more accurately than the quadratic von Mises and Hill48 yield criteria,and an exponent of 14 in the Yld2000-2d yield function gives the optimal predictions for the AA7075-T6 at all investigated temperatures.Jianping Lin Xingyu Bao Yong Hou Junying Min Xinlei Qu Zhimin Tao Jiajie Chen 2020Chinese Journal of Mechanical Engineering2020,33,5:1

Background and Methods Total brainstem infarction 14 patients,9 male and 5 female,43 to 80 years old.all patients had been checked by CT or MRI,brainstem foliun scanning 6 cases,checked by MRI 8 canes micbrain infarction 2 cases,pon infarction 5 cases,medulla infasction 7 cases the midbrain infarction were rise rapid,inmediately coma,the mydrasis in defect side,opposite body mucsle tension heighten.then both lower limbs straight,both pathology sign masculine.the two cases are all alive .tocked-in syndrom has appeared in 1 case of pon infarction ,and died of combined illness 1 year later. 1 case defect affect centrum of breath and hearlbeat, coma,breath rhythm malajustment,breath stop.threr were no consciousness clog in the other 3 cases ,vertigo,force head position to trouble side, nystagmus, trouble side face hypalgesia,and all cureed .2 cases of medulla infarction appeared quactriplegia ,swallow hardness,anarthriad and so on, 5 others were hemi Watlenberg syndrom,all wcre cured. Results and Discussion coma or no in brainstem infaciton was related with position.it is reparted that midbrain infarction coma was 7.6 persent of brainstem infarction. consciousness clog is distinctness which defect position in midbrain lateral-back, pon ventro defect, not involved ARAS ,lwas locked-in synxdom. brainstem infarction combined combined with freedom breath clog,in medulla was 16.1percent,midorain was 1 1.6 percent, pons ws 83.96 percent the prognosis was.all right in lightly brainsterm infarction, lf involved in both medulla, ventro pon,the prognosis was bad, and lose quadriplegia. 6Biocompatible carbon dots with low-saturation-intensity and highphotobleaching-resistance for STED nanoscopy imaging of the nucleolus and tunneling nanotubes in living cells显示文摘Many kinds of nano particles and organic dyes as fluorescent probes have been used in the stimulated emission depletion(STED)nanoscopy.Due to high toxicity,photobleaching and non-water solubility,these fluorescent probes are hard to apply in living cell imaging.Here,we reporta new fluorescence carbon dots(FNCDs)with high photoluminescence quantum yield(56%),low toxicity,anti-photobleaching and goodwater-solubility that suitable for live-cell imaging can be obtained by doping fluorine element.Moreover,the FNCDs can stain the nucleolusand tunneling nanotubes(TNTs)in the living cell.More importantly,for STED nanoscopy imaging,the FNCDs effectively depleted backgroundsignals and improved imaging resolution.Furthermore,the lateral resolution of single FNCDs size under the STED nanoscopy is up to 22.1 nm for FNCDs deposited on a glass slide was obtained.And because of their good water dispersibility,the higher resolution of single FNCDs sizein the nucleolus of a living cell can be up to 19.7 nm.After the image optimizati on steps,the fine fluoresce nee images of TNTs diameter with ca.75 nm resolution is obtained living cell,yielding a threefold enhancement compared with that in confocal imaging.Additionally,the FNCDs show excellent photobleaching resistance after 1,000 scan cycles in the STED model.All results show that FNCDs have significant potentialfor application in STED nanoscopy.Hao Li Shuai Ye Jiaqing Guo Huibo Wang Wei Yan Jun Song Junie Qu 2019Nano Research2019,12,12:0
