1MicroRNA-125b Induces Cancer Cell Apoptosis Through Suppression of Bcl-2 Expression显示文摘MicroRNAs(miRNAs) are small,noncoding RNAs which can often act as an oncogene or a tumor suppressor.Several miRNAs are associated with the development of hepatocellular carcinoma(HCC).We demonstrated that miR-125b significantly suppresses HCC cell proliferation and promotes apoptosis by inhibiting the gene expression of the anti-apoptotic protein,Bcl-2.Bioinformatic analysis indicated that the 3'UTR of Bcl-2 has binding sites for miR-125b.Luciferase reporter assay confirmed the ability of miR-125b to dramatically suppress Bcl-2 transcription,suggesting that Bcl-2 is a target gene for miR-125b.We concluded that miR-125b acts as a tumor suppressor in hepatic tumor development by targeting Bcl-2 and inducing cancer cell apoptosis.Aihua Zhao Quan Zeng Xiaoyan Xie Junnian Zhou Wen Yue Yali Li Xuetao Pei 2012Journal of Genetics and Genomics2012,39,1:30

3Achieving highly uniform two-dimensional PbI2 flakes for photodetectors via space confined physical vapor deposition显示文摘Two-dimensional(2D) PbI_2 flakes have been attracting intensive attention as one potential candidate for the modern optoelectronics. However, suffered from the instability of kinetics-driven growth, the fabricated 2D PbI_2 flakes have a wide dimensional distribution even under the same conditions. Herein, a novel facile space confined physical vapor deposition(PVD) process is provided to synthesize uniform triangle PbI_2 flakes with high quality. The confined space provides a relatively stable growth environment that renders more control on the growth kinetics, leading to highly uniform triangle PbI_2 flakes with the average size of 5 mm and thickness of 17 nm. Moreover, as-fabricated PbI_2-based photodetectors show promising stable and flexible optoelectronic performances to 470 nm light, including high responsivity(0.72 AW^(-1)), large on/off ratio up to 900, fast photoresponse speed(rise time of 13.5 ms and decay time of 20 ms) and high detectivity(1.04×10^(10) Jones). 4Simulation investigation of tractive energy conservation for a cornering rear-wheel-independent-drive electric vehicle through torque vectoring显示文摘Although electric vehicle fully exhibits its comparative merits of energy conservation and environmental friendliness, further improvement of its traction energy efficiency lacks comprehensive investigations in the past. In this paper, the effect of the torque vectoring on traction energy conservation during cornering for a rear-wheel-independent-drive electric vehicle is investigated.Firstly, turning resistance coefficient and energy conservation mechanism of torque vectoring are derived from the single track dynamic model. Next, an optimal torque vectoring control strategy based on genetic algorithm is proposed, with the consideration of the influence of the operation-point change of the in-wheel motors, to find out the best torque vectoring ratio offline. Finally,various simulation tests are conducted to validate the energy conservation effect after Simulink modelling. 5Development and experimental validation of an automatic parking brake system with less driveline sensor显示文摘Electrical Parking Brake(EPB) has been popularly used in passenger cars over the past ten years. With the help of the several kinds of sensors mounted in driveline for the total traction force estimation, EPB can obtain well performance on drive-off assistance and automatic parking brake. Furthermore, its AUTOHOLD function can realize automatic parking brake and ease the driver. However, given that the higher cost and complexity of this traction force estimation method based on the driveline sensors and its slower response resulted by applying maximum parking force for safety parking while driving off, a novel automatic parking brake system without these transmission system sensors, such as clutch position sensor for the cars with manual transmission, is proposed in this paper, including its control scheme and application test. Firstly, the indirect judgement method of the appropriate moment to release the parking brake, which is based on the car pitch moment when it drives off, is introduced according to the force analysis when the car is ready to go. Then a pragmatic mass estimation method for proper brake force calculation is proposed for improving the drive-off performance. In addition, for the convenience and drivability of skillful driver, as well as the system reliability, a mechanical redundant design to reserve the conventional handbrake lever is also described. 6Non-viral gene carrier mediated short hairpin RNA interference for inhibition of tumor cells growth显示文摘A tumor-targeting gene vector G250mAb-PEI-PEG has been prepared by modification of polyethylenimine (PEI) with polyethyleneglycol (PEG) and G250, a monoclonal antibody against the G250 antigen on tumor cell surface. The transfection efficiency was as high as 70% in G250 positive HeLa cells, whereas the transfection efficiency was relatively low (30%) in normal NIH3T3 cells. A plasmid encoding the short hairpin RNA (shRNA) specific for nucleostemin gene (NS) was efficiently transfected into the HeLa cells with this nonviral gene vector. RNA interference down-regulated the expression of NS gene in HeLa cells, inhibited cells proliferation and induced apoptosis. However, the growth and activity of the NIH3T3 cells were not affected under the same treatment. 8A p53/CPEB2 negative feedback loop regulates renal cancer cell proliferation and migration显示文摘The tumor suppressor p53 transactivates the expression of multiple genes to exert its multifaceted functions and ultimately maintains genome stability.Thus,cancer cells develop various mechanisms to diminish p53 expression and bypass the cell cycle checkpoint.In this study,we identified the gene encoding RNAbinding protein cytoplasmic polyadenylation element-binding protein 2(CPEB2)as a p53 target.In turn,CPEB2 decreases p53 messenger RNA stability and translation to fine-tune p53 level.Specifically,we showed that CPEB2 binds the cytoplasmic polyadenylation elements in the p5330-untranslated region,and the RNA recognition motif and zinc finger(ZF)domains of CPEB2 are required for this binding.Furthermore,we found that CPEB2 was upregulated in renal cancer tissues and promotes the renal cancer cell proliferation and migration.The oncogenic effect of CPEB2 is partially dependent on negative feedback regulation of p53.Overall,we identify a novel regulatory feedback loop between p53 and CPEB2 and demonstrate that CPEB2 promotes tumor progression by inactivating p53,suggesting that CPEB2 is a potential therapeutic target in human renal cancer.Jiehui Di Guang Zhao Hui Wang Yaoyao Wu Zhongjun Zhao Bao Zhu Yanping Zhang Junnian Zheng Yong Liu Ying Hu 2021Journal of Genetics and Genomics2021,48,7:1
9Structural Topology and Dynamic Response Analysis of an Electric Torque Vectoring Drive-Axle for Electric Vehicles显示文摘In-wheel motor-drive electric vehicles have the advantage of independently controllable wheel torque and the disadvantages of unsprung mass rise and power restriction.To address the disadvantages,a centralized layout electric torque vectoring drive-axle system(E-TVDS)with dual motors is proposed,which can realize arbitrary distribution of driving torque between the left and right wheels.First,the speed and torque distribution principle of E-TVDS based on velocity diagram are analyzed,and a virtual prototype of the whole vehicle with basic gear ratio relation model of the E-TVDS is built for simulation to verify the theoretical results and the basic effect of E-TVDS on the steering performance of the vehicle.Second,the charac-teristics of 36 types of the novel E-TVDS topology structure are compared and analyzed,and the optimal structure scheme is selected.Third,the accurate multiple degrees of freedom dynamic model for the optimal structure is established by using the bond graph method,and its dynamic response characteristics are analyzed.The results show that the vehicle equipped with the proposed E-TVDS can distribute the driving torque with the almost identical amount but opposite sign between the left and right wheels in any direction,and varying amount according to different chassis dynamics control requirements,and the torque response performance is great with little delay and overshoot.The function and dynamic response of the proposed E-TVDS show that it has potential application value for various performance improvements of electric vehicles.Junnian Wang Shoulin Gao Yue Qiang Meng Xu Changyang Guan Zidong Zhou 2022Automotive Innovation2022,5,2:1

12The SKI proto-oncogene restrains the resident CD103^(+)CD8^(+) T cell response in viral clearance显示文摘Acute viral infection causes illness and death.In addition,an infection often results in increased susceptibility to a secondary infection,but the mechanisms behind this susceptibility are poorly understood.Since its initial identification as a marker for resident memory CD8^(+)T cells in barrier tissues,the function and regulation of CD103 integrin(encoded by ITGAE gene)have been extensively investigated.Nonetheless,the function and regulation of the resident CD103^(+)CD8^(+)T cell response to acute viral infection remain unclear.Although TGFβsignaling is essential for CD103 expression,the precise molecular mechanism behind this regulation is elusive.Here,we reveal a TGFβ–SKI–Smad4 pathway that critically and specifically directs resident CD103^(+)CD8^(+)T cell generation for protective immunity against primary and secondary viral infection.We found that resident CD103^(+)CD8^(+)T cells are abundant in both lymphoid and nonlymphoid tissues from uninfected mice.CD103 acts as a costimulation signal to produce an optimal antigenic CD8^(+)T cell response to acute viral infection.There is a reduction in resident CD103^(+)CD8^(+)T cells following primary infection that results in increased susceptibility of the host to secondary infection.Intriguingly,CD103 expression inversely and specifically correlates with SKI proto-oncogene(SKI)expression but not R-Smad2/3 activation.Ectopic expression of SKI restricts CD103 expression in CD8^(+)T cells in vitro and in vivo to hamper viral clearance.Mechanistically,SKI is recruited to the Itgae loci to directly suppress CD103 transcription by regulating histone acetylation in a Smad4-dependent manner.Our study therefore reveals that resident CD103^(+)CD8^(+)T cells dictate protective immunity during primary and secondary infection.Interfering with SKI function may amplify the resident CD103^(+)CD8^(+)T cell response to promote protective immunity.Bing Wu Ge Zhang Zengli Guo Gang Wang Xiaojiang Xu Jian-liang Li Jason K.Whitmire Junnian Zheng Yisong Y.Wan 2021Cellular & Molecular Immunology2021,18,10:1

14Direct transformation of dinitrogen:synthesis of N-containing organic compounds via N-C bond formation显示文摘N-containing organic compounds are of vital importance to lives.Practical synthesis of valuable N-containing organic compounds directly from dinitrogen(N2),not through ammonia(NH3),is a holy-grail in chemistry and chemical industry.An essential step for this transformation is the functionalization of the activated N2 units/ligands to generate N-C bonds.Pioneering works of transition metal-mediated direct conversion of N2 into organic compounds via N-C bond formation at metal-dinitrogen[N2-M]complexes have generated diversified coordination modes and laid the foundation of understanding for the N-C bond formation mechanism.This review summarizes those major achievements and is organized by the coordination modes of the[N2-M]complexes(end-on,side-on,end-on-side-on,etc.)that are involved in the N-C bond formation steps,and each part is arranged in terms of reaction types(N-alkylation,N-acylation,cycloaddition,insertion,etc.)between[N2-M]complexes and carbon-based substrates.Additionally,earlier works on one-pot synthesis of organic compounds from N2 via ill-defined intermediates are also briefed.Although almost all of the syntheses of N-containing organic compounds via direct transformation of N2 so far in the literature are realized in homogeneous stoichiometric thermochemical reaction systems and are discussed here in detail,the sporadically reported syntheses involving photochemical,electrochemical,heterogeneous thermo-catalytic reactions,if any,are also mentioned.This review aims to provide readers with an in-depth understanding of the state-of-the-art and perspectives of future research particularly in direct catalytic and efficient conversion of N2 into N-containing organic compounds under mild conditions,and to stimulate more research efforts to tackle this long-standing and grand scientific challenge.Ze-Jie Lv Junnian Wei Wen-Xiong Zhang Ping Chen Dehui Deng Zhang-Jie Shi Zhenfeng Xi 2020National Science Review2020,7,10:1

17Clinical role of ^(18)F-FDG PET/CT-based simultaneous modulated accelerated radiotherapy treatment planning for locally advanced nasopharyngeal carcinoma显示文摘Objective The aim of this study was to compare the long-term local control, overall survival, and late toxicities of positron emission tomography/computed tomography(PET/CT)-guided dose escalation radiotherapy versus conventional radiotherapy in the concurrent chemoradiotherapy treatment of locally advanced nasopharyngeal carcinoma(NPC).Methods A total of 48 patients with stage III–IVa NPC were recruited and randomly administered PET/CTguided dose escalation chemoradiotherapy(group A) or conventional chemoradiotherapy(group B). The dose-escalation radiotherapy was performed using the simultaneous modulated accelerated radiotherapy technique at prescribed doses of 77 gray(Gy) in 32 fractions(f) to the gross target volume(GTV): planning target volume(PTV) 1 received 64 Gy/32 f, while PTV2 received 54.4 Gy/32 f. Patients in group B received uniform-dose intensity-modulated radiotherapy, PTV1 received 70 Gy/35 f and PTV2 received 58 Gy/29 f. Concurrent chemotherapy consisted of cisplatin [20 mg/m2 intravenous(IV) on days 1–4] and docetaxel(75 mg/m2 IV on days 1 and 8) administered during treatment weeks 1 and 4. All patients received 2–4 cycles of adjuvant chemotherapy of the same dose and drug regimen.Results The use of fluorine-18-fluorodeoxyglucose(^(18)F-FDG) PET/CT significantly reduced the treatment volume delineation of the GTV in 83.3%(20/24) of patients. The 5-year local recurrence-free survival rates of the two groups were 100% and 79.2%, respectively(P = 0.019). The 5-year disease free survival(DFS) rates were 95.8% and 75.0%, respectively(P = 0.018). The 5-year local progression-free survival and DFS rates were significantly different. The 5-year overall survival(OS) rates were 95.8% and 79.2%, respectively. Differences in OS improvement were insignificant(P = 0.079). Late toxicities were similar in the two groups. The most common late toxicities of the two arms were grade 1–2 skin dystrophy, xerostomia, subcutaneous fibrosis, and hearing loss. 19Rare-Earth Metal Boroxide with Formal Triple Metal-Oxygen Orbital Interaction:Synthesis from B(C_(6)F_(5))_(3)·H_(2)O and Radical-Anion Ligated Rare-Earth Metal Amides显示文摘Rare-earth metal boroxides are not only rare and of great synthetic challenge but also important theoretically due to their unrevealed bonding characteristics.In this paper,we report the synthesis,characterization,and bonding characteristics of the 4f-block metal boroxide complexes ^(Mes)DAD_(2)LuOB(C_(6)F_(5))X(4-Lu,X=C_(6)F_(5);5-Lu,X=NC_(13)H_(10))with two chelating radical-anionic 1,4-diazabutadiene(DAD)ligands.The synthesis of 4-Lu or 5-Lu involved three steps,during which the radical-anionic DAD ligands could be kept stable:(1)the reaction of LuCl_(3),^(Mes)DAD,and potassium graphite(KC8)afforded the diradical rare-earth metal chloride ^(Mes)DAD_(2)LuCl·tetrahydrofuran(THF)(1-Lu);(2)the salt metathesis of 1-Lu with two alkali metal amides gave lutetium bis(diazabutadiene)amides 2-Lu or 3-Lu,respectively;and(3)2-Lu or 3-Lu reacted with B(C_(6)F_(5))_(3)·H_(2)O,resulting in the formation of rare-earth metal boroxides 4-Lu or 5-Lu.The radical-anionic nature of DAD ligands in these complexes were proven by their bond lengths and coordination modes in single-crystal structures.DFT calculations clearly show the bonding of 4-Lu can be divided into three major types of Lu-O orbital interactions,which can be categorized asσ-donation,π-donation,and d-π-conjugation.The triple Lu-O orbital interactions cooperatively make a Lu-O single bond and contribute to the linear Lu-O-B structure.Haihan Yan Botao Wu Xiao-Kun Zhao Chao Yu Junnian Wei Han-Shi Hu Wen-Xiong Zhang Zhenfeng Xi 2021CCS Chemistry2021,3,11:1
