| 1 | Genome-wide Expansion and Expression Divergence of the Basic Leucine Zipper Transcription Factors in Higher Plants with an Emphasis on Sorghum显示文摘Plant bZIP transcription factors play crucial roles in multiple biological processes.However,little is known about the sorghum bZIP gene family although the sorghum genome has been completely sequenced.In this study,we have carried out a genome-wide identification and characterization of this gene family in sorghum.Our data show that the genome encodes at least 92 bZIP transcription factors.These bZIP genes have been expanded mainly by segmental duplication.Such an expansion mechanism has also been observed in rice,arabidopsis and many other plant organisms,suggesting a common expansion mode of this gene family in plants.Further investigation shows that most of the bZIP members have been present in the most recent common ancestor of sorghum and rice and the major expansion would occur before the sorghum-rice split era.Although these bZIP genes have been duplicated with a long history,they exhibited limited functional divergence as shown by nonsynonymous substitutions (Ka)/synonymous substitutions (Ks) analyses.Their retention was mainly due to the high percentages of expression divergence.Our data also showed that this gene family might play a role in multiple developmental stages and tissues and might be regarded as important regulators of various abiotic stresses and sugar signaling. | Jizhou Wang Junxia Zhou Baolan Zhang Jeevanandam Vanitha Srinivasan Ramachandran Shu-Ye Jiang | 2011 | Journal of Integrative Plant Biology2011,53,3: | 41 |

| 3 | Characteristics and dynamics analysis of Populus euphratica populations in the middle reaches of Tarim River显示文摘Populus euphratica Oliv. is widely distributed along the Tarim River. Maintaining stability of P. euphratica population is important to local development. This study explored the static life table, survivorship curves and four function curves (survival rate, cumulative mortality rate, mortality density, and hazard rate), and development index of P. euphratica population in the middle reaches of Tarim River. The results indicated that the age structure of P. euphratica population belonged to positive pyramidal type, which meant young age-class individuals occupied most populations. The number ofⅠ-Ⅱage classes accounted for 66.2% of whole population, and this indicated that there were abundant subsequent seedlings resources to support the growth of P. euphratica population in the middle reaches of Tarim River. The survivorship curve of P. euphratica belonged to the Deevey Ⅲ (concave-type) and the development index was 47.72%. Four function curves revealed that the individuals of P. euphratica sharply decreased at the initial stage and then leveled off at the late stage of survival curve. | 4 | Anti-inflammatory and Immunomodulatory Effects of Marine n-3 Polyunsaturated Fatty Acids on Human Health and Diseases显示文摘The pharmaceutical effects of n-3 polyunsaturated fatty acids(n-3 PUFAs) as dietary nutrients on human health and diseases have gained much attention and are investigated for decades. Docosahexaenoic acid(DHA), eicosapentaenoic acid(EPA) and docosapentaenoic acid(DPA) are the three major n-3 PUFAs enriched in marine organisms, such as fish, shrimp, algae, and so on. It has been well known that n-3 PUFAs, especially DHA and EPA, are beneficial in reducing the risk of cardiovascular and cerebrovascular diseases. Accumulating evidence suggests that n-3 PUFAs might cure inflammatory diseases through several mechanisms, such as plasma membrane remodeling of lymphocytes, down-regulating pro-inflammatory cytokines, and alternating adhesion molecule expressions. Several molecular targets of n-3 PUFAs on immune-regulation have also been identified, such as GPR120(FFA4), protein kinase C(PKC), and PPAR-γ. However, it remains inconclusive if dietary n-3 PUFAs function the same both in vitro and in vivo based on cohort studies. This study focused on the difference of kinematics between mole crickets and normal insects in terrestrial walking. In order to study the function of the specialized foreleg, the adjustments made to the foreleg amputation were discussed by analyzing the motion ability, gait, and body posture of the mole cricket with and without forelegs. The results show that mole crickets walk in terrestrial locomotion by using a special tripod gait, with the clumsy foreleg not as functional as normal hexapods. | 8 | Kinematics Analysis and Optimization of the Fast Shearing-extrusion Joining Mechanism for Solid-state Metal显示文摘Dynamical Joining of the solid-state metal is the key technology to realize endless hot rolling. The heating and laser welding method both require long joining time. Based on super deformation method, a 7-bar and 2-slider mechanism was developed in Japan, and the joining time is less than 0.5 s, however the length of each bar are not reported and this mechanism is complex. A relatively simple 6-bar and 1-slider mechanism is put forward, which can realize the shearing and extrusion motion of the top and bottom blades with a speed approximately equal to the speed of the metal plates. In order to study the kinematics property of the double blades, based on complex vector method, the multi-rigid-body model is built, and the displacement and speed functions of the double blades, the joining time and joining thickness are deduced, the kinematics analysis shows that the initial parameters can't satisfy the joining process. Hence, optimization of this mechanism is employed using genetic algorithm(GA) and the optimization parameters of this mechanism are obtained, the kinematics analysis show that the joining time is less than 0.1 s, the joining thickness is more than 80% of the thickness of the solid-state metal, and the horizontal speeds of the blades are improved. | 9 | Microstructure Evolution and Mechanical Behavior of Laser Melting Deposited TA15 Alloy at 500℃ under In-Situ Tension in SEM显示文摘TA15 alloy fabricated by laser melting deposition was investigated at 500℃ under tensile deformation. The damage behavior of microstructure was analyzed by the real time observation of the microstructure evolution, microcracks initiation and propagation using in-situ tensile equipment fitted in the SEM chamber. Finally, the mechanism of fracture was discussed. The result showed anisotropic mechanical properties in X-and Z-direction. The existence of columnar β grains and its orientation to the tensile direction were the major factors inducing the anisotropic mechanical properties. As compared to Z-direction specimen, high tensile strength was observed in X-direction specimen due to the resistance in slips propagation provided by the prior-β grain boundaries( β GBs). Accumulation of the cracks at prior β GB caused the shear fracture. In case of Z-direction specimen, parallel orientation of prior β GB and GB α with the tensile direction resulted in a homogeneous deformation. | 10 | The novel ER stress inducer Sec C triggers apoptosis by sulfating ER cysteine residues and degrading YAP via ER stress in pancreatic cancer cells显示文摘Pancreatic adenocarcinoma(PAAD) is one of the most lethal malignancies. Although gemcitabine(GEM) is a standard treatment for PAAD, resistance limits its application and therapy. Secoemestrin C(Sec C) is a natural compound from the endophytic fungus Emericella, and its anticancer activity has not been investigated since it was isolated. Our research is the first to indicate that Sec C is a broad-spectrum anticancer agent and could exhibit potently similar anticancer activity both in GEM-resistant and GEM-sensitive PAAD cells. Interestingly, Sec C exerted a rapid growth-inhibiting effect(80% death at 6 h), which might be beneficial for patients who need rapid tumor shrinkage before surgery. Liquid chromatography/mass spectrometry and N-acetyl-L-cysteine(NAC) reverse assays show that Sec C sulfates cysteines to disrupt disulfide-bonds formation in endoplasmic reticulum(ER) proteins to cause protein misfolding, leading to ER stress and disorder of lipid biosynthesis. Microarray data and subsequent assays show that ER stress-mediated ER-associated degradation(ERAD) ubiquitinates anddownregulates YAP to enhance ER stress via destruction complex(YAP-Axin-GSK-βTr CP), which also elucidates a unique degrading style for YAP. | 11 | Temporal and spatial changes in Bt toxin expression in Bt-transgenic poplar and insect resistance in field tests显示文摘Extensive planting of Bacillus thuringiensis(Bt)-transgenic plants economically benefits society; however, the potential risk they pose is receiving increasing attention. This study used enzyme-linked immunosorbent assay and fluorescence quantitative PCR(RT-PCR) to monitor the temporal and spatial dynamics of the expression of Bt toxic protein in a forest of 6- to 8-year-old trees of transgenic insect-resistant poplar 741 for three consecutive years. The enrichment, distribution, and degradation of Bt toxic protein and the influence of transgenic poplars on the targeted insect population, Hyphantria cunea, were investigated. The content of Cry1Ac toxic protein dynamically changed in transgenic poplar. During the annual growth cycle, the content initially increased, then decreased in the long and the short branches of the crown and in the root system, peaking in August. During the study, the protein did not accumulate overtime. The m RNA transcription of gene Cry1Ac was almost consistent with the level of the protein, but transcription peaked in July. In the transgenic and control forestland, microscale levels of the Cry1Ac toxic protein were detected from the soil, but increased accumulation was not observed with the planting year of transgenic poplar. Meanwhile, Bt was isolated and detected molecularly from the soil in the experimental forestland. A systematic investigation of the density of H.cunea in the experimental transgenic poplar forest indicated that transgenic Pb29 poplar could resist insects to a certain degree. | 12 | Quantitative Analysis of Faults in Huizhou Sub-basin, Pearl River Mouth Basin显示文摘This study used 2 D seismic profiles to investigate the Cenozoic evolution of faults in the Huizhou Sub-basin. It aims to define the basin structure style through describing the geometric features of the faults and quantitatively analyzing their activities. The results indicate that the boundary faults in the Huizhou Sub-basin display en echelon arrangement in plan view, which indicates that, it was caused by a kind of oblique extension. Calculating the fault slips shows long-term activities of faults occurred in the basin, and some boundary faults kept active after 5.5 Ma. The evolution history of the fault system is reconstructed. Initially, during the Eocene–Early Oligocene, mainly NNE-NE and NW trending faults and NE striking grabens and half-grabens formed in the basin and a series of faults system controlled the deposition. During the Late Oligocene–Early Miocene, the structural activities were relatively weak, the fault activity and the fault growth rate decreased sharply. Finally, in the late stage from Late Miocene to the present, the structure movement was re-activated, and some faults were also reactivated. | 16 | Transfer function based equivalent modeling method for wind farm显示文摘To effectively study the dynamics of power systems with large-scale wind farms(WFs), an equivalent model needs to be developed. It is well known that back-toback converters and their controllers are important for the dynamic responses of the wind turbine(WT) under disturbances. However, the detailed structure and parameters of the back-to-back converters and their controllers are usually unknown to power grid operators. Hence, it is difficult to build an accurate equivalent model for the WF using the component model-based equivalent modeling method. In this paper, a transfer function based equivalent modeling method for the WF is proposed. During modeling, the detailed structure and parameters of the WF are not required. The objective of the method is reproducing the output dynamics of the WF under the variation of the wind speed and power grid faults. A decoupled parameter-estimation strategy is also developed to estimate the parameters of the equivalent model. A WF that consists of 16 WTs is used to test the proposed equivalent model. Additionally,the proposed equivalent modeling method is applied to build the equivalent model for a real WF in Northwest China. | 19 | Dynamic Microwave-Assisted Extraction of Total Ginsenosides from Ginseng Fibrous Roots显示文摘A dynamic microwave-assisted extraction(DMAE) method is established for the extraction of total ginsenosides from ginseng fibrous roots. The extraction process has been simulated and its main affection factors(liquid/solid ratio K of solvent to ginseng powders(V/m), irradiation time, irradiation temperature, extracting solution concentration, flow rate of solvent and microwave power) have been optimized by response surface methodology(RSM). The optimum conditions of extraction are the liquid/solid ratio of 270 m L/g, the extraction temperature of 75 ℃, the extraction time of 35 min, ethanol concentration of 70%(V/V), the solvent flow rate of 1.3 m L/min, and the microwave power of 500 W. The yield of ginsenosides obtained by the proposed DAME method is(15.0±0.7) %, which is well agreement with the yield predicted by the model.
