2Aberrant expression and function of TCF4 in the proliferation of hepatocellular carcinoma cell line BEL-7402显示文摘Wnt signaling pathway is essential for development and tumorigenesis,however,this signaling pathway in the progress of hepatocellular carcinoma (HCC) remains unclear. In this paper,we studied the function of human T-cell transcription factor-4 (TCF4),a key factor of Wnt signaling pathway,on the proliferation of HCC cell line. We showed that the expression of TCF4 mRNA in HCC cell line BEL-7402 was higher than that in immortalized normal liver cell line L02. Blockage of Wnt pathway by △NTCF4,a dominant negative TCF4,could suppress BEL-7402 cells growth and decrease the expression of cyclin D1 and c-myc,two of target genes of Wnt pathway. On the other hand,stimulating Wnt pathway by introducing a degradation-resistant β-catenin S37A could increase BEL-7402 cells proliferation. But all the treatments had no effect on L02 cells. 3Optimization of groove texture profile to improve hydrodynamic lubrication performance:Theory and experiments显示文摘It is well known that groove texture with a careful design can be used to enhance the load‐carrying capacity of oil film under the conditions of hydrodynamic lubrication.In this study,a general parametric model was developed,and agenetic algorithm‐sequential quadratic programming hybrid method was adopted to obtain the global‐optimum profile of the groove texture.The optimized profiles at different rotating speeds are all chevrons.The numerical analysis results verified the effect of the optimization.In addition to the numerical optimization,experiments were conducted to validate the superiority of the optimized results.The experimental results show that the optimized groove texture can efficiently reduce the coefficient of friction(COF)and the temperature rise of the specimen.In particular,the optimized groove textures can achieve stable ultra‐low COF values(COF<0.01)under certain conditions.Wei WANG Yongyong HE Jun ZHAO Junyuan MAO Yutong HU Jianbin LUO 2020Friction2020,8,1:13

5Thermodynamic conditions of framework grain dissolution of clastic rocks and its application in Kela 2 gas field显示文摘Feldspar and clastic debris are the most important constituent framework grains of sedimentary clastic rocks and their chemical dissolution plays an essential role in the formation and evolution of the secondary pore in the reservoir rocks. On the basis of thermodynamic phase equilibrium, this study investigates the chemical equilibrium relationships between fluid and various plagioclase and K-feldspar in diagenesis of the sediments, particularly, the impact of temperature and fluid compositions (pH, activity of K+, Na+, Ca2+ and so on) on precipitation and dissolution equilibria of feldspars. Feldspar is extremely easily dissolved in the acid pore water with a low salinity when temperature decreases. The dissolution of anorthite end-member of plagioclase is related to the Ca content of the mineral and the fluid, higher Ca either in the mineral or in the fluid, easier dissolution of the feldspar. Moreover, the dissolution of albite end-member of plagioclase is related to Na of both the mineral and fluid, lower Na out of both the mineral and fluid, easier dissolution of the mineral. Similarly, lower-K fluid is more powerful to dissolve K-feldspar than the higher. The anorthite component of plagioclase is most easily dissolved in ground water-rock system, the albite is the secondary, and K-feldspar is the most stable. Selective dissolution of plagioclase occurs in diagenesis because of the plagioclase solid solution that is mainly composed of albite and anorthite end-members, Ca-rich part of which is preferentially dissolved by the pore water, in contrast to the Na-rich parts. Based on investigation of reservoir quality, secondary pore, dissolution structures of feldspar, clay minerals and ground water chemistry of the Kela 2 gas field of Kuqa Depression in the Tarim basin of Western China, we discovered that the secondary pore is very well developed in the highest quality section of the reservoir, and the plagioclase of which was obviously selectively dissolved, in contrast to the overgrowth of K-feldspar. Chemistry of the ground water out of the highest quality section of the reservoir is consistent with the complete dissolution field of anorthite, partial dissolution field of albite component of plagioclase, and the precipitation of K-feldspar on the temperature-logK phase equilibrium diagram drawn from this study. The thermodynamic modeling calculation gave us an insight into formation of the secondary pore within the reservoir, and the dissolution of plagioclase an important cause of the secondary pore. 6Design and application of a small electrode experimental installation for resistivity measurement of mineral and solid insulating material显示文摘There has not been an effective method to measure the resistivity of small-size sample of mineral and solid insulating material until now.According to the Chinese National Standard(GB/T1410-2006) and features of digital high resistance meter,a small electrode experimental installation was developed;it can work with current high resistance meter;the sample decreases to 18 mm from standard size 100 mm in diameter and reduces by 30.86 times in area.A three-electrode system is supported and precisely positioned by two insulating bases whose diameter is 60 mm and height is 20 mm,which ensures accuracy of device structure and reliability of measuring results.The key technological parameters are as follows:diameter of high voltage electrode is 18mm;diameter of measuring electrode is 14.6 mm;internal diameter and external diameter of guard electrode are 16 and 18 mm,respectively;the gap between guard electrode and measuring electrode is set at 0.6 mm.These parameters are adequate for the measurement of flat specimen of mineral and solid insulating material whose diameter is 18 mm.According to the confirmatory experiment on the volume resistivity and surface resistivity,the measuring results are almost the same,using a small electrode experimental installation and a standard electrode.WANG Ling LUO Ke LI ZiQiang GUAN SongYun GE Wei ZHANG JunYuan 2011Science China(Technological Sciences)2011,54,4:8
7Optimization of Vehicle Side Curtain Airbag Module Based on Computer Aided Engineering显示文摘Compared with other kinds of airbags,curtain airbag(CAB) has more complex structures and larger coverage area. The product development process depends on many module tests,sled tests and full size vehicle tests. Computer aided engineering(CAE) technology can replace tests to a great extent,also save test costs and product development time. This paper introduces the way of setting up simulation models and application of static deployment tests and free motion headform(FMH) tests to verify simulation models. In the CAB simulation,uniform pressure airbag models and computational fluid dynamics(CFD) models are all used. The uniform pressure airbag models are not able to simulate the pressure difference among different parts inside the cushion during inflating process. CFD-based CAB models are used to help the curtain airbag optimization design. Based on effective CAE simulation,the optimization analyses related to diffuser tube parameters,inflator mass flow rate and cushion folding patterns are discussed and performed in different cases. 8Precambrian phosphatized embryos and larvae from the Doushantuo Formation and their affinities, Guizhou (SW China)显示文摘Weng,an phosphates of the Precambrian Dou-shantuo Formation, Guizhou (southwestern China) preserve a large number of exquisite biological structures, which are mostly micro-spherical and represent seaweeds, acritarchs and developing eggs related to various groups of metazoans. Here is a report of a variety of developing eggs and larvae, which are most probably of Cnidarian affinity. The eggs ex-amined in the study are composed of early cleavage embryos and two-layered gastrulae. The early cleavage embryos are radial and total cleavage with equal-size blastomeres. The gastrulae mostly bear a large archenteron, which is filled with yolk-degrading organic matter. Ovoid to fusiform planula-like larvae identified in thin sections under light microscope are mostly mouthless and their gastrovascularcavity is filled with possible yolk-degrading organic matter. They are likely representatives of non-feeding larva. The uncommon planula-like structures are hollow, with each having a mouth-like structure on its narrow end. We inter-pret them as feeding larva. 9Design and Optimization for the Occupant Restraint System of Vehicle Based on a Single Freedom Model显示文摘Throughout the vehicle crash event, the interactions between vehicle, occupant, restraint system (VOR) are complicated and highly non-linear. CAE and physical tests are the most widely used in vehicle passive safety development, but they can only be done with the detailed 3D model or physical samples. Often some design errors and imperfections are difficult to correct at that time, and a large amount of time will be needed. A restraint system concept design approach which based on single-degree-of-freedom occupant-vehicle model (SDOF) is proposed in this paper. The interactions between the restraint system parameters and the occupant responses in a crash are studied from the view of mechanics and energy. The discrete input and the iterative algorithm method are applied to the SDOF model to get the occupant responses quickly for arbitrary excitations (impact pulse) by MATLAB. By studying the relationships between the ridedown efficiency, the restraint stiffness, and the occupant response, the design principle of the restraint stiffness aiming to reduce occupant injury level during conceptual design is represented. Higher ridedown efficiency means more occupant energy absorbed by the vehicle, but the research result shows that higher ridedown efficiency does not mean lower occupant injury level. A proper restraint system design principle depends on two aspects. On one hand,the restraint system should lead to as high ridedown efficiency as possible, and at the same time, the restraint system should maximize use of the survival space to reduce the occupant deceleration level. As an example, an optimization of a passenger vehicle restraint system is designed by the concept design method above, and the final results are validated by MADYMO, which is the most widely used software in restraint system design, and the sled test. 11Extended tacrolimus release via the combination of lipid-based solid dispersion and HPMC hydrogel matrix tablets显示文摘The objective of this study is to evaluate the feasibility of obtaining extended release of tacrolimus by a novel combination of lipid-based solid dispersion and matrix-type extended release tablet techniques. Tacrolimus solid dispersion was prepared using glycerylbehenate(Compritol~?ATO888) and Pluronic F127 as the carrier materials with hot-melt method, which was then blended with hydrogel matrix materials, such as HPMC and lactose, the powders were directly compressed into tablets. In vitro drug release tests were carried out to evaluate the performance of the solid dispersions and the tablets. The dissolution rate of tacrolimus was significantly improved by the lipid-based solid dispersion, and the incorporation of HPC into the solid dispersion obviously improved its stability after storage. Extended release tablets loaded with tacrolimus solid dispersion showed prolonged drug release patterns over 24 h, the release patterns of the tablets can be tailored by the compositions of the matrix materials, including the types and content of HPMCs. A modified processing method that directly mixed the melted solid dispersion with HPMC powders improved the uniformity of the solid dispersion inside the tablet matrix and release profile. The release data of the extended release tablet fitted well to the Korsmeyer–Peppas model with n value of 0.85, which suggested diffusion-and erosion-controlled release mechanism. 13Structure-activity relationship optimization for lassa virus fusion inhibitors targeting the transmembrane domain of GP2显示文摘Dear Editor,Lassa virus(LASV)belongs to the Mammarenavirus genus,Arenaviridae family.Arenaviruses are classified into two main groups-Old World(OW)and New World(NW)-based on virus genetics,serology,antigenic properties and geographical relationships.The OW LASV and Lujo virus(LUJV),as well as NW Jurn'n virus(JUNV),Machupo virus(MACV),Guanarito virus(GTOV),Sabia virus(SABV)and Chapare virus(CHAPV),are known to cause severe hemorrhagic fever and are listed as biosafety level 4(BSL-4)agents.The arenavirus glycoprotein complex(GPC)contains three subunits-the retained stable-signal peptide(SSP),the receptor-binding subunit GP1,and the membrane fusion subunit GP2(Lenz et al.,2001).Notably,the proximate external membrane region and TM of GP2,together with the ectodomain loop and TMs of SSP,form an SSP-GP2 interface,playing essential roles in regulating membrane fusion,and providing targets for distinct fusion inhibitors(Larson et al.,2008;Lee et al.,2008;York et al.,2008;York and Nunberg,2009;Thomas et al.,2011;Burgeson et al.,2013a;Shankar et al.,2016;Wang et al.,2016;Wang et al.,2018).Among these inhibitors,ST-161 is LASV specific(Burgeson et al.,2013a).In this study,we conducted structure-activity relationship(SAR)optimization of ST-161.As a result,21 derivatives with IC50 values<1 pmol/L are presented in Table S1.Hit compounds 21,29 and 57 exhibiting robust inhibition of the LASV pseudotype virus(LASVpv,VSV backbone enveloped by LASV GPC with single cycle infection)entry with IC50 values lower than 0.2 nmol/L(Figs.1A and S1),as well as hit compound 72 with an ester bond instead of acylhydrazone,were further investigated.To test whether the four hit compounds inhibit LASV entry by blocking the GPC-mediated membrane fusion,the inhibition effects of these compounds against LASV GPC mediated fusion were quantitatively determined by dual-luciferase assay(Thomas et al.,2011;Wang et al.,2018).Guangshun Zhang Junyuan Cao Yan Cai Yang Liu Yanli Li Peilin Wang Xiaoying Jia Mengmeng Zhang Gengfu Xiao Yu Guo Wei Wang 2019Protein & Cell2019,10,2:3

17Establishment and Validation for the Theoretical Model of the Vehicle Airbag显示文摘The current design and optimization of the occupant restraint system(ORS) are based on numerous actual tests and mathematic simulations. These two methods are overly time-consuming and complex for the concept design phase of the ORS, though they're quite effective and accurate. Therefore, a fast and directive method of the design and optimization is needed in the concept design phase of the ORS. Since the airbag system is a crucial part of the ORS, in this paper, a theoretical model for the vehicle airbag is established in order to clarify the interaction between occupants and airbags, and further a fast design and optimization method of airbags in the concept design phase is made based on the proposed theoretical model. First, the theoretical expression of the simplified mechanical relationship between the airbag's design parameters and the occupant response is developed based on classical mechanics, then the momentum theorem and the ideal gas state equation are adopted to illustrate the relationship between airbag's design parameters and occupant response. By using MATLAB software, the iterative algorithm method and discrete variables are applied to the solution of the proposed theoretical model with a random input in a certain scope. And validations by MADYMO software prove the validity and accuracy of this theoretical model in two principal design parameters, the inflated gas mass and vent diameter, within a regular range.
